Abstract

This study examined the tyrosinase inhibitory potential of bengkoang peel standardised ethanolic extract and ethyl acetate fractions. 450 g of bengkoang peel powder (Pachyrhizus erosus L.) was macerated with 96% ethanol for 2 x 24 h, filtered, and concentrated till viscous. Extract standardisation uses specific and non-specific parameters. Microplate colorimetric tyrosinase inhibition assays were performed. The result of plant determination identified the bengkoang peel as Pachyrhizus erosus (L.) Urb. The organoleptic test revealed a brownish-yellow viscous extract, fresh smell, and bitter taste. Ethanol-soluble extract was 47.60% and water-soluble 22.42%. Bengkoang peel ethanolic extract includes alkaloids, flavonoids, saponins, tannins, and steroids. The non-specific parameters showed loss of drying (9.71%), water content (7.28%), total ash (9.91%), acid-insoluble ash (3.68%), no residual solvent content, lead content (-0.2324±0.1729), cadmium content (-0.0841±0.3418), TPC of 1 x 101 CFU/g, and TYMC of 2.26 x 102 CFU/g. The ethanol extract (103.9263) and ethyl acetate fraction (81.8606) IC50 values of bengkoang peel decrease tyrosinase enzyme activity compared to control (25.1235 ppm). These results show that the ethanol extract met both particular and non-specific quality standards. Bengkoang peel ethyl acetate inhibited tyrosinase.

Abstract

Hypertension has been an increasing global epidemic problem over years and considered as one of the deadly disease with a high cardiovascular complication risk. Knowledge and adherence play an important role in patient's clinical impact, which determined the level of medication’s adherence. Thus, adherence level determines the achievement of clinical outcome. The optimum clinical optimum will be achieved if the adherence level is at good level. The method in this study was using observational analytics with a cross sectional approach. The sampling method was consecutive sampling. The subjects were the adult hypertensive patients at Puskesmas Jetis Kota from August until October 2020. Univariate analysis was conducted in patient’s and medication characteristics. Kendal Tau test was used to analyze the relationship between adherence and clinical outcome. 81 hypertensive patients were included in this study. The highest prevalence of patient’s characteristics were shown on the women’s group: 45-59 years old, senior high school education background, and no occupation. Amlodipine was the highest medication used as a single therapy. It was found that there is no relationship between knowledge and adherence level. In addition to that, there was no relationship between adherence level and clinical outcome measured by systole and diastole.

Abstract

Diabetes Mellitus, one of the non-communicable diseases (NCDs), is the leading cause of death in addition to cardiovascular disease, cancer, and chronic respiratory disease. The Basic Health Research (Riskesdas) recorded that Yogyakarta experienced a prevalence increase of 97% in 2013-2018, the second-highest in Indonesia. Uncontrolled sugar levels cause deterioration of the immune system. This study aims to determine the pattern of prescribing antibiotics and the occurrence of comorbid complications, both infectious and non-infectious. This was a non-experimental descriptive study with a cross-sectional design. The data was collected from patient's medical records retrospectively from July to December 2019. Furthermore, 65 medical records were taken using a simple random sampling method. A descriptive statistical analysis was performed, and the results were presented as percentages. The percentage of infectious and non-infectious comorbid diabetes mellitus II in Panembahan Senopati Bantul Hospital was 22.2% and 77.8%, respectively. The prescribing pattern primarily used was ceftriaxone (23.5%). The antibiotic class mainly prescribed was cephalosporin (49%).

Abstract

Aloe vera (Aloe barbandesis Mill.) is a plant with many benefits, such as an antibacterial agent. A gel dosage form has many advantages, such as being easy to use, not sticky, and quickly washed with water. This pure experimental study aims to obtain the optimal composition of CMCNa as a gelling agent and glycerin as a humectant to produce a gel with good physical properties and physical stability using the Simplex Lattice Design. The results of the responses of spreadability, viscosity, viscosity shift, and spreadability shift were used to determine the optimum formula and were analyzed using Software Design Expert. The results of the research found that the optimum formulas were formulas 1, 2, and 3 with a concentration of CMC-Na and glycerin, respectively, which were 1.500 g and 1.750 g; 1.563 g and 1.688 g; and 1.625 g. In addition, formulas 1, 2, and 3 fulfill the gel's physical properties and stability parameters.

Abstract

Propolis is a widely used medicine that may be found in both its pure form and when combined with othernatural ingredients in over-the-counter preparations, cosmetics, and health foods. Some customers develop alcohol sensitivity after using ethanol, which is a common solvent to extract propolis. As propolis solvents, various Natural Deep Eutectic Solvents (NaDES) were examined in this study. Choline chloride, propylene glycol, glycerol, citric acid, and 1,2-propanediol were combined in molar ratios to create some of the NaDES that were employed. Propolis and solvents were used in a kinetic maceration ratio for the extraction process. The Folin-Ciocalteu method was used to measure the total phenolic content (TPC), and the DPPH assay was used to measure antioxidant activity. In ethanol and NaDES solvents, the TPC of propolis extract ranged from 136.52 ± 27.9 to 365.8 ± 20.54 mgGAE/g. The values for antioxidant activity were 45.94 to 183.76 ppm. The propolis extract with the lowest IC50 and the highest TPC content was choline chloride-glycerol (CCG). It was discovered that the optimum NaDES solvent for extracting propolis was CCG-based. NaDES may be a potential solvent for use in Indonesian propolis, both as an extraction medium and as a formulation element.

Abstract

Prevention of DM by controlling sugar and lipid levels is the main therapy. The traditional medicine diabetes herb from Kaliputih Batur, Banjarnegara, Central Java consists of 11 kinds of medicinal plants that are empirically proven to be efficacious for diabetes mellitus. The potion is prepared by the infundation method. Three control groups (normal, positive, and negative) and three test groups with different doses (18, 36, and 54 mL/kg BW) that had previously been induced by streptozotocin were created using Sprague Dawley (SD) albino rats. Using HE staining, observations were obtained on fasting blood glucose (FBG), superoxide dismutase (SOD), HbA1c, and pancreatic organs. At doses of 36 and 54 mL/kg bw, results analysis of biochemical parameters of FBG levels revealed a substantial decline (P<0.05). At all dosages of the herb, there was a significant change in SOD activity (P<0.05), and at a dose of 18 mL/kg BW compared to negative control, there was a significant difference in HbA1c levels (P<0.05). HE staining results revealed no distinctpathological alterations in pancreatic organ at the test dose compared to negative control with necrosis in the Langerhans insula. Herbal medicine for diabetes traditional medicine from Kaliputih Batur, Banjarnegara, Central Java has decreased blood glucose.

Abstract

Robusta green coffee beans (Coffea canephora) have strong potential to be used as antioxidants which can inhibit free radicals that causes premature aging. The peel-off gel mask is a cosmetic product that increases the penetration of active ingredients, is easy to use, and maintains the natural moisture of the skin. This research aimed to examine the possibility of the ethanolic extract of Robusta green coffee beans into a peel-off gel preparation. A peel-off gel mask made from the ethanolic extract of Robusta green coffee beans was formulated using 3 polyvinyl alcohol (PVA) concentrations to obtain the best preparation, which were 10% (F1), 12% (F2), and 14% (F3). All preparations produced a clear yellowish gel. Only F3 met the qualifications for all evaluation tests. The results of a mechanical property test showed that F3 has the best value for tensile strength and elongation. There were statistically significant differences between the results of the evaluation tests before and after the stability test. The results of irritation tests showed that none of the formulas caused irritation. The best peel off gel mask preparation made with ethanolic extract of Robusta green coffee beans was formula F3 that used 14% PVA.

Abstract

The active metabolites of Colocasia esculenta L. and Zingiber officinale L. have been reported to reduce pain and exert anti-inflammatory effects. This study aimed to examine the anti-inflammatory and analgesic effects of a combination of C. esculenta and Z. officinale extract. Thirty rats and mice were each divided into 6 groups (n=5), namely the normal group, negative control, positive control (Nadiclofenac), and 3 test groups were given a extract combination (dose 1.3 mg/20 gBW, 2.6 mg/20 gBW, and 5.2 mg/20 gBW). Winter's method was used for anti-inflammatory tests by inducing carrageenan in rat paws, and Sigmund's method was used for analgesic tests by the intraperitoneal induction of acetic acid in mice. The percentage of inhibition of leg edema in rats was 11.33%, 18.90%, and 19.10% for the three doses of the combined extracts of C. esculenta and Z. officinale, and 22.72% for Na-diclofenac (p<0.05). The percentage inhibition in the analgesic test in the positive control group and the three test groups was 61.17%, 41.19%, 51.79%, and 52.35%, respectively (p<0.05). The combination of the C. esculenta and Z. officinale extracts exhibited anti-inflammatory and analgesic effects. Dose of 2.6 mg/20 gBW and 5.2 mg/20 gBW as effective as Na-diclofenac.

Abstract

Black Sea cucumbers (Holothuria atra) are a potential source of active metabolites such as flavonoids widely used for nutraceutical, pharmaceutical, and cosmeceutical products. The type of nanoparticle that is often used in cosmetics and skin products is Solid Lipid Nanoparticles (SLN). In this study, the SLN of black sea cucumber extract will be prepared in the form of a peel-off gel mask as an antioxidant. Black sea cucumbers were extracted using the maceration method, while SLN preparation used high-shear homogenization and freeze-drying methods. Crude extract and SLN were formulated into peel-off gels and then evaluated, including organoleptic, homogeneity, viscosity, pH, spreadability, drying time, tensile strength test, and antioxidant activity using the DPPH method. The IC50 values of blank (F1), peel-off gel extract (F2), and nanoparticle peel-off gel extract (F3) were each 600.8727 ± 0.43; 580.7849±0.7; and 592.76±0.55 µg/mL. In conclusion, the SLN method can be applied to the formulation of black sea cucumber extract nanoparticles. However, peel-off gel masks from the crude and nanoparticle extracts have very weak antioxidant activity.

Abstract

Averrhoa bilimbi L. is generally used as a food flavor enhancer and traditional medicine to treat inflammation, cancer sores, cough, fever, gout, rectal bleeding, and hemorrhoids. In vivo and in vitro studies on Averrhoa bilimbi L. have shown anti-inflammatory activity, but the active compounds that play a role in anti-inflammatory activity have not been reported. This study aimed to analyze sixtyfour (64) bioactive compounds in the Averrhoa bilimbi L. plant as cyclooxygenase-2 (COX-2) enzyme inhibitors using in silico approach and predict the pharmacokinetic and toxicological profiles of each compound. The cyclooxygenase-2 enzyme is an enzyme that plays a role in the inflammatory process by converting arachidonic acid into prostaglandin. Increased prostaglandins will cause inflammation. The research method used molecular docking with the application of YASARA, PLANTS, Marvinsketch, Pymol, visualization with PLIP and prediction of ADMET with pkCSM. Control compound used celecoxib. The results showed that there were 13 test compounds that were predicted to have better COX-2 inhibitor activity than celecoxib with good pharmacokinetic properties. Erucic acid has the best pharmacokinetic and toxicity profile. Erucic acid has the potential to be developed as a cyclooxygenase-2 enzyme inhibitor drug.
