Abstract

Pisang Raja is an important banana cultivar for the Javanese people since ancient times, with many morphological variations and local names. This study aims to determine the genetic variation and phylogeography of Pisang Raja based on Internal Transcribed Spacer (ITS) sequences. The results of the research can be used as a reference for developing superior Pisang Raja cultivars that are resistant to disease and make farmers easier to choose Pisang Raja cultivars that suit with geographical conditions on Java Island. This study used 9 samples of Pisang Raja (in-group) and 2 samples of wild bananas (out-group). The research method started with DNA isolation, qualitative and quantitative tests, ITSL and ITS4 primer amplification, and sequencing. Data analysis performed included polymorphism, phylogenetics (MP, ML, NJ and BI) and haplotype. The results showed that the ITS sequences in Pisang Raja was amplified with a length of 600-700 bp and GC content of 61.70%. Polymorphism analysis showed that the genetic variation of Pisang Raja in Java was very high. Phylogenetic analysis resulted in 4 phylogenetic trees with relatively same topologies, where the in-groups were divided into 2 clades, namely clade I (AAA and AAB) and clade II (AAA and ABB). The highest genetic similarity was showed by Raja Brentel and Raja Bali (89.43%) and the least was Raja Gareng and Raja Seribu (56.82%). Haplotype analysis formed 11 haplotypes which grouped into 4 groups. Pisang Raja evolution comes from two ancestors, i.e. M. acuminata and M. balbisiana. The phylogeography of ITS gene flow in Pisang Raja originates from Central Java then spread to East Java and Jakarta. This is presumably due to the high utilization followed by the planting of Pisang Raja by the people of Central Java for ritual and ceremonial events in the past which influenced the domestication process. The results of this study can served as the reference for genetic conservation strategy of Pisang Raja.

Abstract

The plantation sector is one of the area utilization activities that can significantly contribute to the income of rural communities, both in terms of providing employment and earning potential foreign exchange. These conditions are not supported by digitally integrated regional development information. This study aims to map the potential development of coffee plants in the Sambas sub-district based on the physical properties of the land presented in digital maps. The research was conducted in six villages within the Sambas sub-district administrative area using purposeful sampling. Within every village, three locations were chosen for soil sampling plots, each plot covering an area of 1 ha, and the sampling was done diagonally. The distance between each sampling plot is 5 km. Soil samples from each plot were then analyzed in the soil laboratory. The analysis results were processed using quantum GIS software to generate a digital map of land development potential. The results show a variety of land capability classes (classes II, III, and IV) with their respective limiting factors, namely drainage, flood threat, and soil texture. Land capability class II-d2 consists of locations A1, A2, A5, and A6. Land capability class III-d3O2 is at location A3, and capability class IV-t3.d4.O3 is at location A4.

Abstract

Sourdough yeast is a natural yeast for making bread that aims to slow down the decay process. The fermentation process can be assisted by using tomato extract. This study aims to obtain the best quality yeast from various types of flour with the addition of tomato extract (Solanum lycopersicum). This experimental study was designed with independent variables in the form of flour types (wheat, wheat, glutinous rice), control variables in the form of tomato extract and the ratio of material composition (1:1), and response variables in the form of yeast quality (color, odor, shape, presence of foreign objects, and yeast activity). Data were analyzed descriptively and compared with yeast quality standards based on SNI (Indonesian National Standard). The results showed that wheat flour produced the best sourdough yeast. The results showed that wheat flour met the yeast requirements based on SNI 01-2982-1992, had a yellowish white color with a normal odor, good acidity, in the form of coarse powder with no foreign objects and good yeast activity. Unlike wheat flour, which has a brownish white color, a very sour normal odor, a coarse powdery texture, and no foreign objects, but its activity is not good so it does not meet the SNI 01-2982-1992 standard. Glutinous rice flour is white, has a slightly sour normal odor, a fine powdery form, and no foreign objects, its activity is very good, but does not meet the SNI 01-2982-1992 standard because of its color and texture.

Abstract

Dragon fruit is a fruit that contains many benefits inside it. One part of the dragon fruit that has high nutritional value is the skin. The skin can be processed into products made into flour which can be used as an ingredient in cakes. The utilization of this dragon fruit skin to cakes can also add nutritional benefits to cakes itself. Dragon fruit skin flour itself has a rich content of antioxidants. These antioxidants have the benefit of protecting the body from various kinds of disease and can protect the body against the effects of free radicals. Apart from that, the utilization of this dragon fruit skin can be used to reduce dragon fruit waste which often pollutes the surrounding environment. This dragon fruit waste is still minimally utilized in the wider community so that accumulation often occurs. Therefore, holding this research can add efficacy and nutritional value to cakes and reduce the amount of dragon fruit peel waste in the surrounding environment to create a clean and eco-friendly environment. The purpose of this study is to analyze the characteristics and also the antioxidant activity contained in dragon fruit peel flour which would be made in the form of pukis brownies. These pukis brownies were divided into 3 concentrations, contain with 100% wheat flour, 50% dragon fruit peel flour, and 100% dragon fruit peel flour by analyzing the starch content and the influence of the quality of the pukis brownies in terms of shape, color, taste, smell and texture.
