Abstract

This study aimed to estimate the aboveground biomass, carbon stock, and carbon dioxide (CO₂) sequestration of coastal she-oak (Casuarina equisetifolia) stands in Muara Cemara, Pantai Panjang, Bengkulu City, and to describe their ecological functions based on field observations. The study was conducted in September 2025 using a non-destructive method based on the allometric equation ln Y = –2.177 + 2.523(ln DBH). Data were collected from 17 systematically established plots, each measuring 20 m × 20 m. The results showed that the stands had an aboveground biomass of 2.37 ± 0.63 t ha⁻¹, a carbon stock of 1.09 ± 0.20 t C ha⁻¹, and CO₂ sequestration of 4.01 ± 1.06 t CO₂ ha⁻¹. The 21–30 cm diameter class contributed the largest proportion of total biomass and carbon stock, indicating the important role of medium-sized trees in carbon accumulation. Ecologically, C. equisetifolia may contribute to reducing coastal erosion, stabilizing sandy substrates, improving microhabitat conditions, and supporting coastal habitat functions. These findings indicate that C. equisetifolia stands have potential as a complementary component of coastal carbon storage, climate change mitigation, and ecosystem-based coastal management in sandy coastal environments.

Abstract

This study aimed to enumerate, detect, and identify potentially pathogenic bacteria contaminating fresh marine fish sold at Kebon Roek Market, Mataram City. This preliminary laboratory-based descriptive survey focused on the fish species most frequently purchased by consumers: eastern little tuna (Euthynnus sp.), sardinella (Sardinella sp.), and Indian mackerel (Rastrelliger sp.). Fish samples were collected from nine purposively selected vendors located in different sections of the market, comprising three vendors each from the front, middle, and rear areas. Laboratory analyses included determination of the Total Plate Count (TPC), bacterial isolation using selective media, and bacterial identification based on cellular morphology, Gram-staining reactions, and biochemical characteristics. The bacterial loads were 3.36 × 10⁶ CFU/g in eastern little tuna, 1.76 × 10⁶ CFU/g in Indian mackerel, and 4.85 × 10⁵ CFU/g in sardinella. The bacterial counts detected in eastern little tuna and Indian mackerel exceeded the maximum limits specified in the Indonesian National Standard SNI 7388:2009. Bacterial identification revealed the presence of potentially pathogenic bacteria, including Enterobacter spp., Citrobacter spp., Escherichia coli, Edwardsiella spp., Vibrio spp., and Aeromonas spp. These findings indicate that fresh marine fish sold at Kebon Roek Market may serve as a vehicle for transmitting pathogenic bacteria to consumers. Therefore, improved hygiene and sanitation practices are required throughout the fish distribution and retail chain to ensure food safety.

Abstract

This study evaluated the effects of coconut water type and storage duration on the vegetative growth of Dendrobium bigibbum. A completely randomized factorial design was employed, consisting of two coconut water types—mature coconut water and young coconut water—and three storage durations: 0, 1, and 2 days. The experiment included six treatment combinations and an untreated control, with three replicates per treatment. The observed growth parameters were plant height, number of leaves, leaf length, and number of shoots. Data were analyzed using two-way analysis of variance at a 5% significance level. No statistically significant differences were detected among treatments for plant height, number of leaves, leaf length, or number of shoots (p > 0.05). Nevertheless, descriptive observations showed that mature coconut water stored for 0 days (AKTL0) produced the highest mean plant height and leaf length, whereas mature coconut water stored for 1 day (AKTL1) produced the highest mean number of leaves and shoots. These findings indicate that, under the conditions of this study, coconut water type and short-term storage for up to 2 days did not produce statistically detectable effects on the vegetative growth of D. bigibbum. However, the numerically higher values observed in the AKTL0 and AKTL1 treatments suggest that fresh or briefly stored mature coconut water warrants further investigation using larger sample sizes, longer observation periods, standardized storage conditions, and laboratory analyses of coconut water quality. This result is consistent with previous evidence that the response of Dendrobium to coconut water depends on its nutrient composition, endogenous hormones, and physicochemical properties, which may change during storage.

Abstract

This study aimed to analyze the effect of moringa leaf flour addition at concentrations of 0%, 5%, 10%, and 15% on the sensory quality of crackers and to identify the most acceptable formulation based on consumer preference. The study employed a Completely Randomized Design (CRD) with three replications. Organoleptic evaluation was conducted with 30 untrained panelists using a 5-point hedonic scale to assess color, aroma, taste, and texture. The data were analyzed using one-way analysis of variance (ANOVA), followed by Tukey’s HSD test at a significance level of α = 0.05. The results showed that moringa leaf flour addition had a significant effect on all organoleptic attributes (p < 0.05). The formulation containing 10% moringa leaf flour showed the most balanced sensory profile, with mean hedonic scores of 3.60 for aroma, 3.47 for texture, 3.10 for taste, and 3.03 for color. These scores ranged from neutral to approaching like, indicating that the 10% formulation was relatively the most acceptable formulation compared with the other treatments.

Abstract

This study aimed to evaluate the effects of growth medium formulation, storage temperature, and preservation period on the spore viability of the white oyster mushroom (Pleurotus ostreatus). The study employed an experimental method using a split-plot design involving three types of preservation media: Potato Dextrose Broth (PDB), PDB supplemented with 15% paraffin, and corn decoction medium. The treatments were evaluated under two storage temperature conditions, namely 4°C and room temperature at 26°C, across four preservation periods: 2, 4, 6, and 8 weeks. The observed parameters included the time of colony emergence, colony number, and microscopic hyphal morphology. The results showed that all media stored at 4°C were able to maintain spore viability for up to 8 weeks. PDB produced the highest colony numbers, ranging from 224 to 593 colonies, and showed the fastest colony emergence, occurring on days 1 to 2. PDB supplemented with 15% paraffin produced 128 to 248 colonies, with colony emergence on day 2, whereas the corn decoction medium produced 89 to 142 colonies, also with colony emergence on day 2. All treatments stored at room temperature at 26°C became contaminated and therefore failed to maintain spore viability. Microscopic observations indicated that hyphal structures remained normal in all treatments stored at 4°C. Based on colony number and growth rate, PDB stored at 4°C demonstrated better preservation performance than the other media in maintaining the spore viability of Pleurotus ostreatus for up to eight weeks of storage.

Abstract

This study aimed to analyze the effect of implementing the Problem-Based Learning (PBL) model on eighth-grade students’ science learning outcomes. This study employed a quantitative quasi-experimental approach with a nonequivalent control group design. The sample consisted of 53 eighth-grade students who were assigned to an experimental class and a control class. The experimental class was taught using the PBL model, whereas the control class received conventional instruction. Data were collected using a multiple-choice test administered before and after the treatment as a pretest and posttest. The test was designed to measure students’ cognitive learning outcomes in science. The data were analyzed using an independent samples t-test after the initial abilities of the two groups were examined. The results indicated that the two groups had relatively equivalent initial abilities before the intervention. After the treatment, the experimental class achieved a higher posttest mean score than the control class, and the difference between the groups was statistically significant. These findings suggest that PBL is more effective than conventional instruction in improving students’ science learning outcomes.

Abstract

This study investigated the effect of an iNaturalist-assisted Project-Based Learning (PjBL) model on students’ biodiversity literacy in the context of biodiversity education. A quasi-experimental method with a nonequivalent control group design was employed. The participants were Grade 10 students at SMA Negeri 8 Tasikmalaya during the 2025/2026 academic year, selected using purposive sampling. The experimental group received iNaturalist-assisted PjBL, the positive control group received PjBL without iNaturalist, and the negative control group received Discovery Learning. Biodiversity literacy was assessed using a validated and reliable 36-item multiple-choice test. The data were analyzed using analysis of covariance (ANCOVA) at a 5% significance level, followed by the Least Significant Difference (LSD) post hoc test. The ANCOVA results demonstrated that the iNaturalist-assisted PjBL model had a significant effect on students’ biodiversity literacy (F = 39.992, p < 0.001). The LSD test further confirmed that the experimental group achieved the highest mean score (32.66), significantly exceeding the PjBL group without iNaturalist assistance (23.85) and the Discovery Learning group (19.73). These findings indicate that integrating citizen science technology into the PjBL framework significantly enhances students’ biodiversity literacy. The statistically significant differences among the three groups provide empirical evidence that combining project-based instruction with digital species-identification technology is more effective in facilitating students’ mastery of biological concepts than the other instructional approaches examined.

Abstract

This study aimed to assess water quality based on physicochemical and microbiological parameters, identify the dominant social factors influencing pollution, and evaluate the performance of a Constructed Wetlands (CW) model as a domestic wastewater treatment solution. Water sampling was conducted at three stations along the Aur River in accordance with Indonesian National Standards (SNI). Dominant factor analysis was performed using linear and logistic regression with SPSS version 24, while CW performance was evaluated using independent t-tests in horizontal and vertical flow systems planted with three types of vegetation: water spinach, water hyacinth, and water lily. The results showed that the highest total coliform concentration was recorded at the downstream station, reaching 9 × 10⁶ MPN/100 mL. Septic tank ownership was identified as the most dominant variable associated with total coliform concentration (OR = 9.470). All water quality parameters decreased significantly after CW intervention (p = 0.000), with removal efficiencies of 91.43% for BOD, 86.41% for COD, 94.24% for ammonia, and up to 96.05% for total coliform. No significant difference in treatment performance was observed between the horizontal and vertical configurations, indicating that CW design selection may be guided by cost considerations and site-specific field conditions.

Abstract

This study aimed to evaluate the potential of Streptomyces sp. isolated from the rhizosphere of the mangrove Avicennia marina as a biological control agent against Sclerotium rolfsii, the causal agent of basal stem rot in bird’s eye chili (Capsicum frutescens L.). The study was conducted using a non-factorial completely randomized design consisting of a control treatment and root-soaking treatments in a Streptomyces sp. suspension for different durations: 15, 30, and 45 minutes. Observational data were analyzed using ANOVA at the 5% significance level with IBM SPSS Statistics version 27. The results showed that Streptomyces sp. inhibited the growth of S. rolfsii on artificial medium by up to 52.33%, indicating strong antagonistic activity. The in vivo assay further demonstrated that the 45-minute root-soaking treatment (SW3) delayed disease incubation until the final day of observation, prevented the development of disease symptoms throughout the observation period, and enhanced vegetative growth in bird’s eye chili plants. These findings suggest that Streptomyces sp. from the rhizosphere of A. marina has promising potential as a biological control agent for managing basal stem rot caused by S. rolfsii in bird’s eye chili.

Abstract

This study aimed to characterize microplastics based on variations in their physical form and size and to assess their abundance in the brackish waters of Kendari Bay. Purposive sampling was conducted at three stations representing port and shipping activities, residential areas, and mangrove habitats. Three biological replicates and three technical replicates were collected, yielding 18 samples comprising nine surface-water samples and nine sediment samples. The data were analyzed using descriptive frequency analysis, and the coordinates of all sampling locations were recorded using a Global Positioning System (GPS). The highest microplastic abundance was observed in the port and shipping zone, with 11,460 particles per 150 mL of water and 83 particles g⁻¹ of sediment, indicating that this zone was the primary accumulation area. This was followed by the mangrove area, with 10,620 particles per 150 mL of water and 77 particles g⁻¹ of sediment, and the residential area, with 10,200 particles per 150 mL of water and 68.4 particles g⁻¹ of sediment. Four microplastic forms were identified: fibers, films, fragments, and foams. Foam was the dominant form in both environmental matrices, indicating a substantial contribution from expanded polystyrene (EPS) waste. Most particles were smaller than 0.27 mm, suggesting advanced fragmentation processes in the environment. Based on their color characteristics, the microplastics were predominantly black (37%) and transparent (31%), indicating extensive environmental weathering, followed by yellow (13%), green (13%), and blue (6%). These findings provide an evidence-based foundation for developing policies to control EPS packaging waste across upstream and coastal areas. They also highlight the urgent need to restore mangrove forests as natural filters to reduce the accumulation of microplastics in Kendari Bay.
