Abstract

This study was conducted to evaluate fertility grade of Friesian Holstein cows based on milk urea levels. A number of eight cows which fulfill the criteria of not pregnant and in second to sixth lactation, produced milk of 2.5 liters/day, and body weight ranging from 400-450 kg were used in this research. The cows were given fodder that consisted of 30 kg of forage and 10 kg of a mixture of concentrate (pollard, pulp out, and molasses), in the morning and afternoon. Milk and blood samples were taken once a week for five weeks in the morning before fed. Milk urea levels were measured using the urea FS (DiaSys) and blood samples were tested using enzyme linked immuno-sorbent assay (ELISA) with the progesterone hormone kit (DRG, Germany). The variables measured were pregnancy rate, service per conception (S/C), days open, and progesterone levels. Reproduction and milk urea levels data were analyzed by linear regression to determine the relationship between two variables. Progesterone data were analyzed descriptively. The results showed that the average calving interval, days open, and S/C were 607.5±170.27 days; 341.25±187.65 days; and 2.41±2.32 days, respectively. The average of milk urea levels was 14.16±2.55 mg/dL. Regression analysis showed a significant effect (P0.05) and negative correlation between milk urea level and S/C. The average of non-pregnant progesterone hormone level was 12.04±7.13 ng/mL. In conclusion, the fertility grade of Friesian Holstein cows decreased when the milk urea level at 14.16 mg/dL and non-pregnant progesterone at 12.04±7.13 ng/mL.

Abstract

This research was aimed to study the effect of administration of glycerol and dimetilacetamida (DMA) in BTS and MIIIextender supplemented with sodium dodecyl sulphate (SDS) on boar frozen semen. A number of four boars were used in this study for semen collection (n=20). The collected semen was evaluated both macroscopically and microscopically. In this study, only the semen that demonstrated70% sperm motility, 200.106/mL sperm concentration, and20% sperm abnormalities were used and divided into eight tubes. A number of 4 tubes were diluted with 5 mL of BTS, and the rest with 5 mL MIII. The sampel was stored at 20-22 C for 2 hours, followed by centrifugation for 15 minutes (at 2000 rpm), and taken of pellet with 1 ml supernatant. The pellet that was resulted from centrifugation using BTS, then re-diluted with BTS-glycerol 5% (BTSG), BTS DMA 5% (BTSD), BTS-glycerol 5% and SDS (BTSG-S), BTS-DMA 5% and SDS (BTSD-S). Four other pellets that were centrifuged with MIII also re-diluted with MIII-glycerol 5% (MIIIG), MIII-DMA 5% (MIIID), MIII-glycerol 5% and SDS (MIIIG-S), MIII-DMA 5% and SDS (MIIID-S). Next, all of diluted semen were inserted into 0.5 mL straw and equilibrated for 2 hours (4 C), then frozen and stored in liquid nitrogen. The evaluation of frozen semen quality was conducted at 24 hours after frozen. The result of this study showed that post-thawing motility of spermatozoa in BTSD-S (40.170.2%) was found higher (P0.05) compared to seven other dilution processes. Therefore, it is concluded that the concentration of 5% DMA that supplemented with SDS in BTS dilution much better for maintaining boars frozen semen quality.

Abstract

The objective of this research was to determine the allergy hyperplasma pathologic antibody and to evaluate the immunogenic characteristics. Blood and serum were collected from human suffering rhinitis, skin, eye and asthma allergies. To obtain a cloned allergen plasma protein (protein allergen that had been induced with the serum from human with allergic rhinitis, skin, eye and asthma), the goat was used as an intermediate animal. Hematological analysis showed that the leukocyte cell such as neutrophil, lymphocyte, monocyte, and eosinophil increase in allergic-suffered human. The blood smear test exhibited that the mastocyte cell was dominant which contributes to allergy activities in human body. The sodium dodecyl sulphate-polyacrylimide gel electrophoresis (SDS-PAGE) assay detected protein allergens with molecular weight of 188 kDa (IgE) and 60-62 kDa (mastocyte cell). The reactivity assay using enzyme linked immunosorbent-assay (ELISA) revealed that cloned-allergens (whole hyperplasma allergen from goat isolates) express the best reactivity at various concentrations of IgE than the leukocyte cells. This research concluded that the clones of protein allergen have better immunogenic characteristic and those proteins can be recommended as the candidate of allergen to induce the humoral immunity on host and deliver specific product of anti-allergy such as milk.

Abstract

This study aimed to determine the ability of highly pathogenic avian influenza virus (HPAI) virus subtype H5N1 originated from Indonesia to induce apoptosis in Madin-Darby Canine Kidney (MDCK) cells. Three HPAI virus subtype H5N1 isolates with different genetic characteristic namely A/Bird/Bali1/2011, A/Chicken/East Java/BwiI2/2010 and A/Chicken/West Java/1074/2003, were cultured in MDCK cells. Apoptosis was identified by deoxyribonucleic acid (DNA) fragmentation of infected MDCK cells using Apoptotic DNA Ladder Kit. The results showed that all three HPAI virus isolates used in this study did not able to induce apoptosis in the MDCK cells within 5 to 72 hours post infection.

Abstract

This research was conducted to investigate the effect of various concentrations of lactose supplementation in Tris extender for maintaining the quality of Etawa crossbreed goat epididymal spermatozoa stored at 3-5 C. Semen in the control group was diluted with a tris extender containing 20% egg yolk without lactose. Semen in the test groups was diluted with a tris extender containing 20% egg yolk and added with 0.3% (0.3 g per 100 mL extender) and 0.6% lactose for group TL1 and TL2, respectively. Parameters evaluated of the fresh epididymal spermatozoa were motility, concentration, percentage of live, and abnormality of spermatozoa, while for diluted-spermatozoa were motility and percentage of live spermatozoa. Spermatozoa observation was conducted until it reaches 40% motility. The results showed that the mean percentage of motility, live sperm, concentration, and abnormality of epididymal spermatozoa were 70%; 81%; 3,220x106 cells/mL; and 4.30%, respectively in all group. After dilution, the percentage of motility and live spermatozoa were also 70% and 81.001.58%, respectively in all groups. The decreasing of spermatozoa motility was observed on day 4 of storage, in which percentage of spermatozoa motility in control group (40.000.00%) was significantly lower (P0.05) than those in TL1 (44.002.24%) and TL2(45.000.00%) groups. Percentage of live spermatozoa in control (63.202.68%) was not significantly different (P0.05) than TL1(65.401.95%) and TL2 (65.601.95%). In conclusion, the supplementation of lactose into Tris extender could maintain the epididymal spermatozoa of Etawa crossbreed for 3 days of storage at 3-5 C.

Abstract

The aim of this research was to investigate the potential effect of methanolic extract of Scurrula atropurpurea (MESA) in reducing oxidative stress through observation of superoxide dismutase (SOD) activity and malondialdehyde (MDA) concentration in mice. In this research, 25 male mice were divided into four groups with five replications. Group I (K1, without MESA) was considered as a control group, while Group II (K2), III (K3), and IV (K4) were given 1000, 2000, and 4000 mg MESA/kg BW, respectively. After 14 days, SOD activity and MDA concentration were measured spectrophotometrically. The SOD activity did not significantly increased (P0.05) in group K1, K2, K3, and K4 with values were 19.4692.072, 17.41.373, 20.533.411, and 23.872.528 U/ml, respectively. In contrast, MDA concentration was decreased (P0.05) with values of 492.75019.418, 492.75019.418, 457.12530.095, 454.62519.080, and 475.87537.659 ng/mL in groups K1, K2, K3, and K4, respectively. It can be concluded that the administration of MESA tend to increase SOD activity and decrease MDA concentration in this study, thus might be able to reduce oxidative stress in mice.
