Abstract

This research was conducted to investigate the effect of various concentrations of lactose supplementation in Tris extender for maintaining the quality of Etawa crossbreed goat epididymal spermatozoa stored at 3-5° C. Semen in the control group was diluted with a tris extender containing 20% egg yolk without lactose. Semen in the test groups was diluted with a tris extender containing 20% egg yolk and added with 0.3% (0.3 g per 100 mL extender) and 0.6% lactose for group TL1 and TL2, respectively. Parameters evaluated of the fresh epididymal spermatozoa were motility, concentration, percentage of live, and abnormality of spermatozoa, while for diluted-spermatozoa were motility and percentage of live spermatozoa. Spermatozoa observation was conducted until it reaches 40% motility. The results showed that the mean percentage of motility, live sperm, concentration, and abnormality of epididymal spermatozoa were 70%; 81%; 3,220x106 cells/mL; and 4.30%, respectively in all group. After dilution, the percentage of motility and live spermatozoa were also 70% and 81.00±1.58%, respectively in all groups. The decreasing of spermatozoa motility was observed on day 4 of storage, in which percentage of spermatozoa motility in control group (40.00±0.00%) was significantly lower (P0.05) than those in TL1 (44.00±2.24%) and TL2(45.00±0.00%) groups. Percentage of live spermatozoa in control (63.20±2.68%) was not significantly different (P0.05) than TL1(65.40±1.95%) and TL2 (65.60±1.95%). In conclusion, the supplementation of lactose into Tris extender could maintain the epididymal spermatozoa of Etawa crossbreed for 3 days of storage at 3-5° C.

Abstract

Penelitian ini dilakukan untuk mengetahui pengaruh lama maturasi dan lama inkubasi fertilisasi terhadap angka fertilitas oosit sapi peranakan Ongole (PO) secara in vitro. Oosit diaspirasi dari folikel ovarium yang berdiameter 2-6 mm. Oosit dirandom untuk penelitian menggunakan rancangan blok acak sempurna (RCBD), dengan lama maturasi 20, 22 dan 24 jam sebagai blok dan lama inkubasi fertilisasi 4, 5 dan 6 jam sebagai perlakuan. Hasil penelitian menunjukkan angka fertilitas oosit sapi PO dengan lama maturasi 20 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masing-masing adalah 48,86%+3,88, 48,08%+2,22 dan 51,96%+ 2,27. Angka fertilitas oosit sapi PO dengan lama maturasi 22 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masing-masing adalah 50,14%+4,18, 51,14%+4,35 dan 52,27%+2,26. Angka fertilitas oosit sapi PO dengan lama maturasi 24 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masingmasing adalah 51,21%+4,49, 53,49%+2,43 dan 50,96%+4,18. Tidak ada perbedaan yang nyata (P0,05) angka fertilitas antara lama maturasi 20, 22 dan 24 jam dan tidak ada perbedaan yang nyata (P0,05)  angka fertilitas antara lama inkubasi fertilisasi 4, 5 dan 6 jam.

Abstract

The purpose of this study was to obtain Escherichia coli isolates from 11 se’i meat sellers in Kupang City and to observe their resistance to13 types of antibiotics. Escherichia coli were isolated and identified based on SNI 2897: 2008. Antibiotic resistance of the Escherichia colii isolates was determined using the Kirby-Bauer method. The results showed that 13 isolates (39.39%) of Escherichia coli in se’i meat samples had a high level of resistance to erythromycin (100%), tetracycline (76.92%), and doxycycline (61.54%). Isolates demonstrated increased resistance to streptomycin (46.15%), cephalothin (38.46%), trimethoprim-sulfamethoxazole (38.46%), amoxicillin (30.77%), chloramphenicol (30.77%), and choline sulfate (30.77%). The antibiotics nalidixic acid and ciprofloxacin demonstrated low Escherichia coli resistance (7.69%). Escherichia coli are sensitive to cefotaxime and gentamicin. A total of 12 isolates (92.31%) experienced MDR. The presence of non-MDR and MDR resistant Escherichia coli in se’i meat can seriously threaten community health.

Abstract

This study aimed to determine the occurences of mice brain ventricles dilatation that congenitally infected with Toxoplasma gondii (T. gondii) as a marker of hydrocephalus and cellular changes in the brain. A total of twenty pregnant mice (11.5 days pregnacy) were divided into 2 groups, which were control (P1) group and treatment (P2) group. The mice in the treatment group were infected with 101 tachyzoites of T. gondii. All mice were maintained until delivery. The newborn mice were sacrificed and their brain were removed and fixed in 10% buffered formalin to prepare histology slides with HE staining for observation of ventricular width, TUNEL assay for apoptosis observation, and immunohistochemistry for the expression of transforming growth factor beta (TGF-β) observations. The data were analyzed using t test and linear regression. The results showed that ventricular width and apoptosis index significantly increased (P0.01) in the treatment group compared to control group, but there was no difference in the expression of TGF-β (P0.05) in both groups. Dilatation of ventricle correlated with the apoptotic index of brain cells but did not correlated with the expression of TGF-β.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak vesikula seminalis terhadap penurunan konsentrasi progesteron serta diameter korpus luteum pada tikus putih. Dalam penelitian ini digunakan 30 ekor tikus putih (Rattus norvegicus) betina dewasa, galur Wistar, berumur 3-4 bulan dengan berat badan antara 200-250 g dan dibagi atas dua kelompok (K1 dan K2) masing-masing diberi 25 μg cloprostenol dan 0,2 cc ekstrak vesikula seminalis secara intravaginal pada hari ke-7 kebuntingan. Tiga ekor tikus masing-masing kelompok dibunuh pada jam ke-0, 3, 6, 12, dan 26. Pemeriksaan progesteron dilakukan menggunakan metode enzymelinkedimmunosorbantassay (ELISA). Konsentrasi progesteron pada kelompok perlakuan PGF2α dan ekstrak vesikula seminalis pada lima periode waktu pengukuran yakni jam ke-0, 3, 6, 12, dan 26 memperlihatkan perbedaan yang signifikan (P0,05). Ekstrak vesikula seminalis mempunyai kemampuan yang sama dengan PGF2α komersial dalam menurunkan diameter korpus luteum yang ditandai secara mikroskopis dengan berkurangnya vaskularisasi darah menuju ovarium (P0,05). Disimpulkan bahwa ekstrak vesikula seminalis mempunyai kemampuan yang lebih baik dibandingkan dengan PGF2α komersial dalam menurunkan konsentrasi progesteron tikus putih dan mempunyai kemampuan yang sama dengan PGF2α komersial dalam menurunkan diameter CL.

Abstract

This study aimed to determine the presence of Salmonella spp. in duck meat and to identify the resistance of Salmonella spp. against several antibiotics in duck meat associated with food safety. Total of 52 meat samples of ducks was collected from 5 subdistricts in Bogor District, i.e., Cariu-Jonggol, Gunung Sindur, Klapanunggal, Parung Panjang, and Ciomas based on the assumption of 95% confidence level, 50% predicted prevalence, and 10% standard error. The results showed that three samples (5.8%) were positive Salmonella spp. The majority of Salmonella spp. isolated from duck meat showed resistance against erythromycin (66.7%), streptomycin (33.3%), and chloramphenicol (33.3%). Nevertheless, Salmonella spp. was still sensitive againts enrofloxacin, trimethoprim sulfamethoxazole, cephalothin, ampicillin, nalidixid acid, tetracycline, and gentamicin.

Abstract

Penelitian ini bertujuan mengetahui efek antikolesterol fraksi n-heksana ekstrak rumput kebar pada hewan model hiperlipidaemia. Delapan kelinci dewasa dibagi dalam dua kelompok. Kelompok kontrol (K1) hanya diberikan diet tinggi lemak selama 1 bulan dan pada kelompok perlakuan (K2) diberikan diet tinggi lemak ditambah fraksi n-heksana ekstrak rumput kebar. Uji keberhasilan penelitian dilakukan dengan pemeriksaan kolesterol total serum darah. Hasil penelitian menunjukkan konsentrasi kolesterol total serum darah pada K1 dan K2 masing-masing adalah 100,00±18,28 dan 47,75±12,53 mg/dl. Dari hasil penelitian disimpulkan bahwa fraksi n-heksanaa mampu menurunkan tingkat kolesterol total dalam serum darah.

Abstract

The objective of this research was to investigate the capability of the donor cattle to produce embryos by comparing three cattle breeds, namely Bos taurus beef cattle (Limousin), Bos taurus dairy cattle (Friesian Holstein/FH), and Bos indicus beef cattle (Peranakan Ongole/PO). This research was observational method with in vivo embryo production method and a nonsurgical embryo collection method (flushing). The data collected were then processed using the one way ANOVA method. The results showed that the number of CL, response rate, total number of embryos, and recovery rate in PO cattle, FH cattle, and Limousin cattle did not differ significantly (P0.05). The appropriate number of embryos to be transferred and degenerative embryo were significantly different among PO cattle, FH cattle, and Limousin cattle (P0.05), while the unfertil ova, fertilization rate, and transferable embryo did not differ among cattle breed (P0.05). This research concluded that each breed of the donor cattle produced embryos with varies quality and the best quality of embryo production is found in Limousin cattle.

Abstract

This study aimed to determine the antibacterial activity of soursop flower extract (Annona muricata L.) and hematological profile of rats (Rattus norvegicus) due to administration of soursop flower ethanol extract and Salmonella enteritidis infection. The concentrations of soursop flower ethanol extract used for the antibacterial activity test were 25%, 50%, 75%, 100%, with the antibiotic ampicillin 10 μg/disk was used s a positive control (PC) and dimethyl sulfoxide (DMSO) 10% as a negative control (NC). For examination of hematological features, 15 male rat aged two months were used. All rats were divided into 5 treatment groups, each consisting of three rats. The NC group was not given soursop flower ethanol extract and S. enteritidis infection. The PC group was not given soursop flower extract but was given S. enteritidis infection. Groups P1, P2, and P3 were given ethanol extracts of soursop flower at a dose of 0.18, 0.36, and 0.72 g/rat/day peroral for a week using gastric sonde. On the following day (after administration of soursop flower extract) the first blood drawing was performed. All rats, except NC group, were then infected with S. enteritidis intraperitonially at dose of 3x108 CFU/mL dose (0.5 mL McFarland). One week after being infected with S. enteritidis, a second blood drawing was performed. The results of the antibacterial activity test showed that there was no antibacterial activity was observed since no inhibition at various concentrations was formed. The administration of soursop flower extract at various dosage levels was able to maintain the number of leukocytes but reduced the number of erythrocytes, hemoglobin concentration, hematocrit value and the number of platelets in rats; whereas S. enteritidis infection decreased all the hematologic features of lab rats.

Abstract

The objective of this study is to observe the relationship of development of adrenocorticotropic hormone (ACTH) cells of adenohypophysis–adrenal cortex axis during pre and postnatal period of longtailed monkey (Macaca fascicularis). Five fetuses aged 70, 85, 100, 120, and 150 days of gestation and 2new born aged 10 and 105 days old were used as research materials. The sections of the pituitary gland stained immunohistochemically using human ACTH antiserum and the adrenal glands stained by Hematoxylin-Eosin procedures. At the fetus 120 days old, the ACTH cells and blood vessel of fetaladenohypophysis developed well and its similar that found in older age. This condition suggested distributing the ACTH to whole body optimally. At the same time, the adrenal cortex cells of fetus 120 days old were active in proliferation and differentiation processes to form transition zone that lies between definitive and fetal zones, and central vein also developed in adrenal gland. In fetus aged 150 days, the cortex adrenal structure was similar with found in older fetus. The adenohypophysis ACTH cells developed earlier than adrenal cortex and their secretion suggested influencing the development of adrenal cortex cells of long-tailed monkey.
