Abstract

The purpose of this study was to evaluate effect of Coleus amboinicus (CA) stem extracts on uric acid-induced nephropathy by comparingthe levels of Macrophage CD-68 expression and concentration of serum Cystatine C (CYS C ) in Wistar rats. Twenty-four male Wistar rats(Rattus norvegicus) with a body weight (bw) of 200-250 g, were allocated into three groups, with eight animals per group. The rats in controlgroup (PO) received 0.1% carboxymethyl cellulose (CMC) solution orally The rats in group 2 (P1) were orally induced with uric acid (UA) (500mg/kg) and oxonic acid (OA) (750 mg/kg.) and the rats in group 3 (P2) received uric acid (500 mg/kg), oxonic acid (750 mg/kg), and 500 mg/kgof the CA stem extracts for 35 days. Bloods were collected for analysis of serum CYS C expression and concentration of serum creatinine andblood nitrogen urea (BUN). The rats in all groups were sacrificed for kidney tissue extractions for macrophage CD-68 identification andhistopathology analysis. The levels of CYS C concentrations were analyzed by Avidin-Horseradish Peroxidase (HRP) Sandwich-ELISA. Theresults showed that Coleus amboinicus stem extract at dose of 500 mg/kg bw can significantly reduce BUN and creatinine levels (P0.05), whileCys C levels were not different. In the treatment group (P2) compared with group (P1). CD-68 (ED-1) macrophage activity decreasedsignificantly (P0.05) in the treatment group (P2) compared to the control group and (P1). Nephrophaty induction using UA and OA causessevere kidney lesions characterized by degeneration, necrosis and inflammation of the renal tubules and glomerulus in the treatment group.

Abstract

The aim of this research was to analyze the anti-oxidant activities of ethanol extract of mahogany seeds on the liver tissues of diabetic experimental rats to overcome oxidative stress condition. This study used male Rattus norvegicus strain Sprague Dawley which were divided into 5 groups: (i) negative control group (K-), (ii) positive control group/diabetes mellitus (DM) (K+), (iii) DM group treated with 500 mg/kg BW ethanol extract of mahogany seeds (EM), (iv) DM group treated with acarbose (KO), and (v) non-DM group treated with 500 mg/kg BW ethanol extract of mahogany seed (KE). Alloxan with dose of 110 mg/kg BW was used to induce diabetes in rats. The treatments were conducted for 28 days. At the end of treatment, the liver tissues were collected and analyzed for malondialdehyde (MDA) content using thiobarbituric acid reactive substance (TBARS) method and anti-oxidant cooper zinc superoxide dismutase (Cu,Zn-SOD) content using immunohistochemical technique. The result showed that level of MDA were significantly different (P0.05) in K-; K+; EM; KO; and KE with values were 0.810.17; 1.190.26; 1.100.04; 0.950.13; and 0.920.0 mol/gram, respectively. The anti-oxidant content (Cu, Zn-SOD) of rat liver tissue in K+ lower than in K-, while anti-oxidant content in EM and KE were higher compared to K+. The study concluded that ethanol mahogany seeds extract decreased blood glucose levels and increased the anti-oxidant status in the liver tissues of diabetic rats.

Abstract

Penelitian ini bertujuan mengetahui respons superovulasi dan pengaruh kehadiran folikel dominan pada saat inisiasi superovulasi sapi aceh yang diinduksi superovulasi dengan follicle stimulating hormone (FSH). Dalam penelitian ini digunakan 7 ekor sapi aceh betina yang telah didiagnosis sehat reproduksinya, umur 5-8 tahun, mempunyai berat 150-250 kg, dan mempunyai minimal dua siklus reguler. Seluruh sapi disuperovulasi dengan FSH dosis menurun pada hari ke-9 siklus estrus (3-3, 2-2, 1-1, dan 0,5-0,5) ml. Kehadiran atau ketiadaan folikel dominan pada saat inisiasi superovulasi diobservasi melalui penggunaan ultrasonografi (USG). Sapi yang mempunyai folikel kecil (3-8 mm) berjumlah 10 folikel dikategorikan mempunyai folikel dominan, sedangkan sapi yang mempunyai folikel kecil (3-8 mm) 10 folikel dikategorikan tidak mempunyai folikel dominan. Koleksi embrio dilakukan pada hari ke-7 setelah inseminasi secara non surgical menggunakan kateter Foley. Sapi yang disuperovulasi tanpa kehadiran folikel dominan menghasilkan korpus luteum (6,7+0,58 vs 4,5+1,73),folikel anovulasi (9,7+8,0 vs 19,5+6,8), total embrio (11,0 vs 3,0), dan embrio kualitas baik (6,0 vs 2,0) dibandingkan dengan sapi yang disuperovulasi dengan kehadiran folikel dominan. Dapat disimpulkan bahwa induksi superovulasi dengan FSH dengan kehadiran folikel dominan pada saat inisiasi superovulasi menurunkan respons superovulasi sapi aceh.

Abstract

The propagation of E. mivati local strain was used single cell inoculation. The objective of this study was to determine the pathogenicity of Eimeria mivati inoculation in broiler chicken by measuring the growth rate, oocysts production and levels of changes in small intestine. The chickens were devided into two age groups of infection, three and teen days old. Each group was received 1x104, 1x105 and 1x106 oocysts/chick. The chichen were raised in wire cages and sampled accordingly. Data collected was analyzed using analysis of variance. A large number of oocysts production, intestinal villi lesions and depression of growth rate were seen after infection. Most of the peak production of oocysts were accured after the 9th day of infection. However, after the peak production of oocysts, oocysts production decreased extremely. The choice of parasitic development were in the surface epithelial cells of the villi and the region dominantly effected was the third anterior of small intestine. The result revealed no significant differences regarding photogenicity findings (P0.05) between chicken aged three and ten days after infected with E. mivati.

Abstract

Penelitian ini bertujuan mengetahui gambaran histopatologis hepatopankreas udang windu yang terinfeksi hepatopancreatica parvovirus (HPV). Sebanyak 10 ekor udang windu yang berasal dari tambak rakyat digunakan sebagai sampel penelitian terdiri atas 5 sampel positif terinfeksi HPV dan 5 sampel normal. Semua udang dinekropsi untuk diambil hepatopankreas dan difiksasi dengan larutan Davidson. Selanjutnya hepatopankreas dibuat preparat histopatologis sesuai dengan prosedur teknik yang biasa dilakukan di Laboratorium Patologi Fakultas Kedokteran Hewan Universitas Syiah Kuala Darussalam Banda Aceh dan diwarnai dengan hematoksilin eosin (HE) serta diamati di bawah mikroskop. Hasil penelitian menunjukkan bahwa pada hepatopankreas udang windu yang terinfeksi HPV ditemukan adanya hipertrofi basofilik badan inklusi intranuklear, degenerasi, dan lisis sel.

Abstract

This study aimed to evaluate the physiological response of female beef cattle to peatland microclimate stress in Central Kalimantan. This study used direct observation on small holder beef cattle farm. Microclimate data on 41 units cattle barns and physiological parameters of female cattle were collected in the morning, at noon and in the afternoon. The physiological parameter measurements involved 215 female beef cattle, consisting of 119 Bali and 96 crossbred cattle with different physiological stages including pregnant cows, lactating cows, dry cows, heifers and calves. The microclimate condition within cattle barns on peatland of Central Kalimantan is not the comfort zone for beef cattle. It is characterized by high air temperature and relative humidity, and low wind speed, which result in high temperature humidity index (THI). The barn with gable roof type and asbestos materials gave the lowest THI. This Microclimate caused heat stress to beef cattle reared on the peatlands, indicated by the high rectal temperature, heart rate, respiratory rate, and heat tolerance coefficient, although it was still categorized as mild to moderate stress. Bali cattle showed better physiological responses to microclimate stress than crossbred cattle. Pregnant crossbred cows were the most susceptible to peatland microclimate stress.

Abstract

The aim of this research was to isolate and characterize the virus based on macroscopic examination of native chickens embryo infected with the AI virus. This research used 10 infected native chickens from Ateuk Pahlawan and Doi villages. All chickens were examined to identify the present of AI using rapid test and in ovo virus propagation. Rapid test showed 7 chickens from Ateuk Pahlawan villageand 5 chickens from Doi village were AI positive. Propagation of the viruses in 11 days old chicken embryo in ovo showed that embryo died 5 days post inoculated whereas macroscopic examination showed dwarfed embryo, hemorrhagic, and loss of feather.

Abstract

Penelitian ini bertujuan membuktikan pengaruh alanin-glutamin dipeptida 7% terhadap perbaikan fungsi hipokampus melalui pengamatanbeberapa parameter kinerja motorik seperti jarak tempuh (JT), waktu stereotipe (WT), waktu ambulatori (WA), dan waktu istirahat (WI). Penelitian ini dilakukan dengan memberikan alanin-glutamin dipeptida 7% selama 12 hari pada tikus umur 12 dan 24 bulan yang mengalami penuaan fisiologis dan penuaan akibat stres oksidatif. Pengamatan terhadap semua parameter kinerja motorik dilakukan dengan alat Opto-Varimex yang dikoneksikan dengan software Autotract. Hasil penelitian menunjukkan alanin-glutamin dipeptida 7% berpengaruh terhadap perbaikan kinerja motorik pada kondisi penuaan fisiologis dan penuaan akibat stres oksidatif.

Abstract

The objectives of this research were to examine the effect of Sechium edule ethanolic extract to repair beta pancreas cells damage and tomeasure nitrogen oxide (NO) concentration in streptozotocin-induced rats. Type I diabetic rats were obtained by intraperioneally injected therats with multiple low dose streptozotocin (MLD-STZ) for 5 days. In this research, rats were assigned into 5 treatment groups: untreated rats ingroup 1 were considered as negative control, while MLD-STZ induced type 1 diabetic rats in group 2 were considered as positive control. Inanother 3 groups, type 1 diabetic rats were orally treated with three doses of Sechium edule ethanolic extract (14, 28, 42 mg/kg bw) respectivelyfor 7 consecutive days. Repairing of beta pancreas cells were analyzed descriptively whereas concentration of NO were analyzed using one wayANOVA and continued with LSD test (= 0.01). The result showed that the administration of Sechium edule ethanolic extract significantlydecreased NO concentration (P0.01) up to 67.9 M and repaired beta pancreas cells damage. Dose of 42 mg/kg bw of Sechium edule ethanolicextract is the effective dose for diabetic rat therapy. Key words: Sechium edule extract, diabetic mellitus, rat, beta pancreas cells

Abstract

Penelitian ini bertujuan mengetahui efektivitas pengobatan endometritis dengan menggunakan kombinasi antibiotik (gentamicine, flumequine) dan analog prostaglandin F2 alfa (PGF2) berdasarkan pengamatan dinamika ovarium dengan metode ultrasonografi (USG). Enam ekor sapi endometritis dibagi dalam 2 kelompok perlakuan. Kelompok I (K1, n= 3) diterapi dengan 250 mg Gentamicine, 250 mg Flumequine, dan 12,5 mg PGF2 secara intra-uterus. Kelompok II (K2, n= 3) diterapi menggunakan antibiotik dengan dosis dan cara pemberian yang sama seperti pada Kelompok I. Hasil pengamatan terhadap sapi-sapi endometritis K1 dan K2 memperlihatkan rataan panjang siklus estrus selama 18 hari. Hasil analisis statistik menunjukkan tidak ada perbedaan yang nyata (P>0,05) terhadap kelas folikel pada kedua kelompok perlakuan. Folikel besar (DF) dan folikel besar kedua (SF) pada ke-3 gelombang folikel yang muncul tidak memperlihatkan perbedaan yang nyata antara K1 dan K2 (P>0,05), tetapi terdapat perbedaan yang nyata (P
