Abstract

This study directly tested the hypothesis that the induction of oocyte maturation on catfish Clariasgrapienusby insulin aspartresulted in a transient decrease of oocyte cyclic AMP (cAMP) level due to the increase ofphosphodiesterase (PDE) activity. Under the influence of insulin the germinal vesicle (GV) of the oocyte migrated towards the animal pole (GVM), reached the micropyle, and then dissolved (GVBD). By using different concentrations of insulin aspart i.e., 0 U/ml, 0.01 U/ml, 0.1 U/ml, 1 U/ml and 10 U/ml, optimum amount required was found to be 1U/ml. The number of oocytes incubated for 24 hours with insulin aspartic treatment that undergo meiotic re-initiation can reach the percentage of meiotic resumption oocytes was 82%. The results suggest that catfish oocytes have responsive to sensitive adenylatecyclase and also an active phosphodiesterase system. Addition of the maturation-inducing steroid, a progesterone, stimulated (P0.01) GVBD of catfish oocytes and its combination with insulin aspart showed an additive effect. This information will be important in hormonal manipulation during induced breeding of Indonesian commercial catfish.

Abstract

This study was aimed to determine the influence of the washing method under running water on nitrite levels of edible birds nest (EBN). Total of 40 samples of EBN were divided into four groups with different washing frequency, control group without washing treatmet (P0), once, twice, and three times washing treatment (P1, P2, and P3) respectively. Each washing was performed for 30 seconds under running water. Nitrite levels assessment was carried out by spectrophotometry at 540 nm of wavelength. The results showed that the average nitrite levels of EBN in P0, P1, P2, and P3 were 93.124.40 ppm, 65.243.38 ppm, 63.603.81 ppm, and 30.872.11 ppm, respectively. The nitrit level in edible birds nest decreased significantly (P0.05) by using three times washing.

Abstract

Penelitian ini bertujuan mengetahui efek injeksi inhibin B terhadap penurunan konsentrasi follicle stimulating hormone (FSH) di dalamserum pada tikus putih (Rattus norvegicus). Dalam penelitian ini digunakan 24 ekor tikus putih berjenis kelamin jantan dengan strain Wistar berumur 4 bulan dengan bobot badan 150-200 g. Tikus-tikus dikelompokkan secara acak ke dalam 4 kelompok, yaitu KK0, KP1, KP2, dan KP3, masing-masing kelompok terdiri atas 6 ekor. Kelompok KK0 merupakan kelompok kontrol hanya diinjeksi dengan phosphate buffer saline (PBS), sedangkan kelompok KP1, KP2, dan KP3 diinjeksi dengan inhibin B dengan dosis berturut-turut 25, 50, dan 100 pg/ekor. Injeksi inhibin B dilakukan secara intraperitoneum sebanyak 5 kali selama 48 hari dengan interval waktu 12 hari. Injeksi pertama inhibin B dilarutkan dengan0,05 ml PBS dan 0,05 ml Freuds complete adjuvant (FCA). Injeksi kedua sampai kelima, inhibin B dilarutkan dengan 0,05 ml PBS dan 0,05 ml Freuds incomplete adjuvant (FICA). Pada hari ke-6 setelah injeksi inhibin B terakhir, tikus dikorbankan secara dislocatio cervicalis,lalu darah dikoleksi langsung dari jantung dan didiamkan hingga didapatkan serum untuk pemeriksaan konsentrasi FSH dengan menggunakan metode enzyme-linked immunosorbent assay (ELISA). Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan konsentrasi FSH secara nyata (P0,05) bila dibandingkan dengan kelompok kontrol. Berdasarkan hal tersebut, inhibin B berpeluang untuk dikembangkan sebagai kandidat kontrasepsi pria hormon berbasis peptida.

Abstract

This study was conducted to compare Ventral Median Approach (OHM) and Lateral Flank Approach (OHF) for feline ovariohysterectomy (OH). Fifteen healthy local female cats (Felis catus) with body weights ranged 2-4 kg and aged 1-2 years were divided into two groups, OHM (n= 8) and OHF (n= 7). Prior to OH, the cats were anesthethized using ketamine-xylazine and the sugery was performed by the same operator. The wound scoring showed a significantly higher difference in the OHM group on day 3 and 4 post-surgery. Scores of erythematous, swelling and dehiscence showed no significant differences. The 4Avet pain scale was significantly higher in OHF at the 2, 6, and 12-hours post surgery. The pain response was evaluated using baseline tactile Semmes-Weinstein Monofilaments and was significantly higher in OHF than OHM on day 1 to day 5 post surgery. Blood hematology, blood glucose and wound healing duration did not differ significantly between groups. The OHF approach showed greater post-operative pain than the OHM approach. Wound dehiscence was observed in two cats (25%) in OHM group from day 4 post-operative. The OHM approach showed a greater risk of complications than the OHF group and therefore requires better post-operative care. The OHF group showed a smaller risk of complications but requires better post-operative analgesic therapy.

Abstract

The aims of this study were to detect Jembrana antigen with polyclonal antibody and to describe antigen distribution in the Bali cattle organ that positively infected with Jembrana disease. Spleens, lungs, and livers were harvested from 10 naturally infected Bali cattle whose infection was confirmed through positive Polymerase Chain Reaction (PCR) result for Jembrana virus. Immunohistochemistry test was performed using polyclonal antibodies produced in rabbits. The results showed that cells infected by Jembrana viruses displayed positive reaction with a reddish brown color. Immunohistochemistry methods using polyclonal antibodies can detect Jembrana antigens in the spleen, liver, and lung with the highest average detection score (P0.05) was found in spleen, followed by liver and lungs. There was significant difference in the distribution of Jembrana antigens between the spleen, liver, and lungs with spleen having the highest antigen density.

Abstract

This research was aimed to find out a substance that was useful in early pregnancy diagnosis of Friesian Holstein cows. Pregnancy Specific Protein B (PSPB) was isolated, purified and partially characterized from cotyledon Friesian Holstein cows. Characterization of PSPB protein was conducted using SDS-PAGE and Western Blot. Antisera were developed against PSPB and by immunohistochemical techniques the protein was localized to the binucleated cells of the cotyledon. The estimated molecular size of Friesian Holstein cows PSPB was 59.88 kDa with the concentration of PSPB protein in cotyledon was 6480 ng/ml. PSPB protein could induce anti-PSPB antibody with the value of optical density (OD) were 0.1790.0102 (before immunization); 1.4660.3288 (3rd week after immunization); 1.9360.4754 (1st week after booster;) and 2.2560.4842 (2nd week after booster).

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian antiserum inhibin terhadap kinerja reproduksi kambing lokal. Dalam penelitian ini digunakan 12 ekor kambing betina lokal yang dibagi dalam 2 kelompok, kelompok kontrol dan perlakuan (KI dan KII), masing-masing terdiri atas 5 dan 7 ekor kambing. Seluruh kambing diinjeksi dengan 0,5 ml PGF2 (LutalyzeTM) secara intramuskulus, 2 kali injeksi dengan interval 10 hari. Pada kelompok perlakuan (KII), kambing diinjeksi dengan 500 g antiserum inhibin pada hari ke-9 siklus dan diikuti penyuntikan 0,5 ml PGF2 48 jam kemudian. Inseminasi dilakukan 10 jam setelah awal berahi dan diulang 12 jam kemudian. Parameter yang diamati adalah persentase berahi, kebuntingan, kelahiran, kelahiran kembar, jumlah anak total, dan jumlah anak per kelahiran. Hasil penelitian menunjukkan semua kambing (100%) pada kedua kelompok memperlihatkan berahi. Persentase kebuntingan pada KI dan KII masing-masing adalah 60,0 dan 57,1%. Angka kelahiran KI dan KII masing-masing adalah 100% dan persentase kelahiran kembar masing-masing adalah 0,0 dan 50,0%. Total jumlah anak pada KI dan KII masing-masing adalah 3 dan 6, dan rata-rata jumlah anak per kelahiran masing-masing adalah 1,0 dan 1,5 ekor. Perlakuan dengan antiserum inhibin dapat meningkatkan kinerja reproduksi kambing lokal.

Abstract

The study was conducted to investigate the effects of caffeine treatment during organogenesis period to the fetal birth weight, using rat (Rattus norvegicus) as the animal model. Thirty-six primipararat obtained from Unit Pengembangan Hewan Percobaan, Gadjah Mada University (UPHP-GMU), 3 month old, 165-200 g body weight, were divided into 6 groups, consisted of 6 rats each. Six of the ratshave been selected based on the estrous cycles, and only rat with regular estrous were use for theexperiment. The rat then were mated, and during day 6-14 of the pregnancies were treated orally withcaffeine diluted in aquadest in dosage: placebo (1 cc aquadest) for group I (control), and 5.4, 10.8, 16.2,21.6, and 27 g/200 g body weight/day for treatment groups II-VI respectively. The pregnant rat bodyweights were determined at day 6 of pregnancies for calculating the caffeine treatment dosages. At day 20thof the pregnancies all of the pregnant rats were caesarotomized, and all of the fetuses were removed and weighed. The results showed that all of the treatment groups have significantly lower birth weightcompare to the groups control group. More over, fetal obtained from the treatment groups also showedserious subcutaneous hemorrhagic.Keywords: organogenesis, Rattus norvegicus, birth weight

Abstract

Penelitian ini bertujuan mengetahui pengaruh penyuntikan dan dosis optimal ekstrak hipofisa sapi terhadap peningkatan produktivitas ayam petelur fase akhir produksi. Sebanyak 60 ekor ayam petelur berumur 22-44 bulan yang telah mengalami penurunan produksi sekitar 50-60% yang terdapat pada peternakan ayam petelur Jantho Farm, Aceh Besar digunakan dalam penelitian ini. Seluruh ayam dibagi 6 kelompok perlakuan, masing-masing kelompok terdiri atas 10 ekor ayam petelur. Kelompok I, II, III, IV, V, dan VI masing-masing disuntik dengan 0,1 ml NaCl fisiologis, 15 IU PMSG; 0,1 ml ekstrak hipofisa sapi; 0,2 ml ekstrak hipofisa sapi; 0,3 ml ekstrak hipofisa sapi; dan 0,4 ml ekstrak hipofisa sapi. Injeksi dilakukan secara intramuskular pada otot dada setiap dua minggu sekali selama enam minggu.Total rata-rata produksi telur pada kelompok I; II; III; IV; V; dan VI masing-masing adalah 5,81+1,10; 4,28+1,04; 4,60+2,04; 5,43+1,45; 6,29+1,34; dan 5,74+1,17 butir. Total rata-rata berat telur pada kelompok I; II; III; IV; V; dan VI masing-masing adalah 65,27+1,61; 63,66+1,86; 65,38+3,51; 64,01+3,91; 66,20+1,67; dan 65,21+1,91 g sedangkan total rata-rata ketebalan cangkang telur pada kelompok I; II; III; IV; V; dan VI masing-masing adalah 0,43+0,02; 0,42+0,03; 0,43+0,02; 0,42+0,02; 0,43+0,02; dan 0,42+0,02 mm. Dari hasil penelitian dapat disimpulkan bahwa ekstrak hipofisa sapi dapat meningkatkan produktivitas ayam petelur pada fase akhir produksi dan dosis optimal ekstrak hipofisa adalah 0,3 ml.

Abstract

This study aimed to evaluate anticancer activity and apoptosis induction of ethanolic extract of Arcalengisia flava (AF) roots on HepG2 cancer cell lines. The AF roots were extracted by maceration using ethanol 80%. MTT assay method was used to evaluate the anticancer activity and the proliferation of HepG2 cells. Flow cytometry method was used to assess the induction of HepG2 cells apoptosis. This study showed that the IC50 of AF ethanol extract against HepG2 cells was 109.14 g/ml. With IC50 treatment, the apoptosis assay showed a significant decrease in intact cells (80.101.7%) and a significant increase in early apoptosis (7.90.7%) and late apoptosis cells of HepG2 cancer cells (4.90.35%) compared to control cells. Moreover, the proliferation of HepG2 cells was declined significantly in 48 and 72 hours after treatment with IC50 (77.55.76% and 64.35.37%, respectively) and 2xIC50 of the extract (75.91.79% and 70.54.27%, respectively). This research suggests that the ethanolic extract of AF roots can potentially be used for hepatocarcinoma treatment.
