Abstract

This study explored the analgesic effectiveness of ethanol extract of red betel leaf (Piper crocatum) in white rats (Mus musculus L.) induced with acetic acid. A total of 15 rats were divided into five groups with different treatments, and the number of twitching movements as a pain response was measured for one hour. The extract was prepared using 96% ethanol solvent, and the analgesic effect was analyzed using the writhing test method. The results showed that the extract contained flavonoids, alkaloids, tannins, and saponins, which might contribute to its analgesic properties. It was found that the higher the dose of extract administered, the greater the inhibition of pain that occurred, with a dose of 800 mg/kgBB showing optimal analgesic effectiveness, almost equivalent to paracetamol as a positive control. Statistical analysis showed significant differences between treatment groups, with doses of 200 mg/kgBB and 400 mg/kgBB showing lower effectiveness. This study recommends further exploration of the therapeutic benefits of red betel leaf extract and formulation development for wider medical applications.

Abstract

Pain is the body's response to tissue damage that is often addressed using analgesic compounds. Purslane plants (Portulaca oleracea L) are known to contain flavonoids, saponins, tannins, and alkaloids that have potential as analgesics. This study aims to test the analgesic effectiveness of purslane ethanol extract by writhing test method using white mice (Mus musculus) induced by 1% acetic acid. A total of 15 mice were divided into 5 treatment groups, namely negative control, positive control (paracetamol), and three groups with doses of purslane extract 200 mg/kgBB, 400 mg/kgBB, and 800 mg/kgBB. The number of writhes was observed every 15 minutes for 1 hour, and the percentage of protective power was calculated. The results showed that the dose of 800 mg/kgBB gave the highest analgesic effect with a percent protection power of 65.28%, followed by doses of 400 mg/kgBB (52.4%) and 200 mg/kgBB (30.6%). The highest analgesic effect was achieved at a dose of 800 mg/kgBB, almost equivalent to the positive control. This shows the significant potential of purslane ethanol extract as an analgesic agent.

Abstract

The pharmaceutical industry operates under a strict regulatory framework to ensure that the products produced are safe, of high quality, and effective. These regulations are continuously updated to keep up with advancements in science, technological developments, and global demands for higher quality standards. The pharmaceutical industry also faces the challenge of global regulatory harmonization, such as standards set by WHO and PIC/S. These regulations are designed to ensure drug quality from development to distribution. Changes in Good Manufacturing Practices (GMP) regulations are crucial for improving pharmaceutical production quality in Indonesia. This study aims to conduct a gap analysis between the 2018 GMP and the draft 2024 GMP Annex 1, particularly in terms of equipment, supporting facilities, and personnel. The method used is a comparative analysis by comparing the provisions of the 2018 GMP with the draft 2024 GMP. The gap analysis results between the 2018 GMP and the draft 2024 GMP Annex 1 in terms of equipment, supporting facilities, personnel, and sterile product completion indicate significant changes in the adopted standards. The proposed 2024 GMP revision introduces substantial improvements in sterile product manufacturing standards. As part of efforts to enhance global competitiveness, the Indonesian pharmaceutical industry must continue to adapt to dynamic technological and regulatory changes. By implementing various strategies—including substantial investments in modern equipment, adjustments to supporting facilities, and capacity building for personnel—the Indonesian pharmaceutical industry can meet global standards while ensuring the availability of safe, high-quality, and effective sterile products for the public.

Abstract

Background: Preterm labor (PTL) remains a major global health concern, contributing significantly to neonatal morbidity and mortality. Asymptomatic bacteriuria (ASB), has been implicated as a predisposing factor to PTL. This research investigates association between ASB and PTL, emphasizing inflammatory mediators such as high-sensitivity C-reactive protein along with tumor necrosis factor- α. Methods: This observational case-control study was conducted at Tikrit Teaching Hospital, Iraq, involving 100 pregnant women (50 PTL cases and 50 full-term controls). Demographic, clinical, and laboratory data were collected. Urine cultures identified bacterial isolates. In addition, levels of TNF-α and hs-CRP in serum were measured using ELISA. Statistical analyses were performed using SPSS.  Results: ASB was significantly associated with PTL, with 30% from PTL cases having a positive urine culture compared to 8% in controls (p = 0.009). Escherichia coli was the predominant pathogen (53%). TNF-α and hs-CRP levels were significantly elevated in PTL cases (p < 0.001), suggesting an inflammatory pathway in PTL pathophysiology. Conclusions: ASB is a significant risk factor for PTL, likely mediated by systemic inflammation. Routine ASB screening and targeted antimicrobial therapy may reduce PTL risk. Further study is required to explore the mechanistic connections amongst microbial infection and inflammatory responses in PTL.

Abstract

This meta-analysis evaluates the effectiveness of community-based interventions in addressing complex trauma among children in Gaza through a systematic analysis of 42 studies (N=4,873) published between 2010 and 2024. The findings reveal a significant treatment effect (g=0.78, 95% CI [0.65, 0.91], p<.001) with moderate heterogeneity (I²=68%). School-based interventions demonstrate the greatest effect (g=0.89), followed by family interventions (g=0.76) and peer-group interventions (g=0.65). Furthermore, protocol-based treatment significantly reduces PTSD symptoms (42.3%), anxiety (38.7%), and depression (35.2%) among children in Gaza. Moderator analysis identifies intervention duration (β=0.34, p<.01) and family involvement (β=0.29, p<.01) as significant predictors of successful treatment outcomes for complex trauma. Lastly, meta-regression results indicate a linear relationship between intervention intensity and symptom improvement (R²=0.45, p<.001). These findings expand on previous research by Thompson et al. (2021) and Al-Krenawi (2022) by identifying specific effective intervention components within the Gaza context. Additionally, the discovery that a phased approach integrating community support with protocol-based treatment yields optimal outcomes presents a novel contribution not previously addressed in meta-analyses.

Abstract

Diabetes mellitus is one of the chronic diseases whose prevalence is increasing worldwide. This disease is characterized by high blood glucose levels due to impaired insulin production or function, the hormone that regulates blood sugar levels. This study aims to examine the effect of wild betel leaf extract (Piper aduncum) on the inhibition of glucose transport using the ileum of white rats (Rattus norvegicus) with the infusion method. This research is a pre-experimental study with a posttest-only control design using white rats and wild betel leaves. The study demonstrates that wild betel leaves (Piper aduncum L.) and red betel leaves (Piper crocatum) have significant potential in inhibiting glucose transport and reducing blood sugar levels. This is evident from the significant decrease in the rate of glucose transport after the administration of wild betel leaf extract, indicating an inhibitory effect on glucose transporters such as SGLT-1 and GLUT-2.

Abstract

One of the health issues that can lead to significant consequences, including heart disease and organ damage, is hypertension. For hypertensive patients, non-pharmacological treatments such as progressive muscle relaxation (PMR) offer an alternative to reduce discomfort and blood pressure. This study uses a quantitative methodology and a quasi-experimental design. For two weeks, 66 hypertensive patients from Puskesmas Bangetayu Semarang participated in the ROP intervention. The T-Test and Wilcoxon statistical tests were used to measure blood pressure and pain levels before and after the intervention. According to the Numeric Rating Scale (NRS), ROP therapy reduced pain from 2.68 to 1.76 and systolic blood pressure from an average of 150.15 mmHg to 133.64 mmHg. With a p-value of less than 0.05, statistical analysis revealed a significant difference. It has been proven that progressive muscle relaxation helps hypertensive individuals with their pain and blood pressure. This approach can be used both independently and in healthcare settings as a simple and safe non-pharmacological treatment. the effect of progressive muscle relaxation on pain and blood pressure in hypertensive patients
