Abstract

Comercial toothpaste contain fluoride to prevent dental caries. The usage of fluoride in large quantities may cause irreversible enamel fluorosis, brittle bones, brittle teeth, premature aging, spontaneous abortion, and carcinogenic. Objective this experiment is knowing the extract concentration of finger roots and lemongrass which is equivalent to positive control and then performed optimization of toothpaste preparation formula with different concentrations of CMC-Na and carbomer with pH and viscosity parameters using the Simple Lattice Design method. The antibacterial test of lemongrass extract, finger roots and its combination with a concentration of 5% against Streptococcus mutans bacteria, showed the average diameter of the largest clear zone: finger roots extract was 11.167±0.288mm; combination of lemongrass extract:finger roots extract (1:2) was 10.83±0.288mm; combination of lemongrass extract: finger roots (2:1) was 10,067±1,006mm; lemongrass extract was 9.33±0.751mm; The combination of lemongrass extract:finger roots extract (1:1) was 9,133±0,321mm.. Post Hoc test results shows comparable to positive controls were 5% finger roots extracts, combination of lemongrass:finger roots 1:2 combination, and 2:1 combination. The optimum formula was obtained using the SLD method with a combination of 0.900% CMC-Na and 1,100% carbomer as a binding agent. The optimum pH formula of toothpaste was 7,613±0,076 and Viscosity 2,43,104cps±2571,640. The paired t-test pH obtained p-value 0.222 and the viscosity obtained p-value 0.934. The experimental results were not significantly different from the results predicted by the software simply latex design. The 5% finger roots extract can be developed into an herbal toothpaste with binding agent CMC-Na and a carbomer ratio of 0,9:1,1.

Abstract

Diabetes Mellitus (DM) is one of the non communicable diseases (NCDs) with the largest number of deaths in the world. The Macaranga hispida (Blume) Mull. Arg is a source of phenolic compounds. Phenolic is grouped as polyphenols group that widely functioned as antidiabetic. The α-glucosidase enzyme plays a role in the forming of glycoproteins and glycolipids. The α-glucosidase enzyme can be used for antidiabetic activity assay because of its capability on breaking down carbohydrate into glucose in the human small intestine. This study aimed to determine phenolic content, total flavonoid content, and antidiabetic activity of M. hispida leaves extract and its fraction. The extract and fraction were tested for the phenolic levels using Folin-Ciocalteau method, the total flavonoid using alumunium chloride method, and the antidiabetic activity using α-glukosidase method. The yield of methanolic extract of the Macaranga hispida was 10.23%. Phenolic content of the ethyl acetate fraction from the methanolic extract was 8.411 mg, equivalent to gallic acid/100 mg extract. Total flavonoid content was 6.14 ± 0.31b/b μg/mL. Antidiabetic activity assay of the extract and the fractions of Macaranga hispida resulted the highest antidiabetic activity (IC50) was in the ethyl acetate fraction with IC50 value 21.91 μg / mL.

Abstract

Research on the safety of Jamu (herbal medicine) formula is important to guarantee its use in community. The Jamu Saintification development program has been resulted a cholesterol-lowering herb containing of 7 plants. This study aimed to determine the safety of the cholesterol-lowering herb. This study is a phase one of clinical trial with a pre-post group design, a single arm, involving 50 subjects with mild hypercholesterolemia without any complications and comorbidities. The intervention was conducted for 28 days at the Hortus Medicus Herbal Medicine Research House (RRJ) in 2014. Outcome parameters included clinical signs and symptoms, liver function laboratory tests (SGOT, SGPT), renal function (ureum, creatinine) and routine blood counts (hemoglobin, leukocyte, platelets, hematocrit). Subjects were requested to boil the herb everyday and drank it twice a day. The data were analyzed descriptively and statistically using SPSS. The results showed increasing in bowel frequencies of 92% subjects and abdominal pain in bowel movements of 34% subjects. These effects were still in a normal range and can be tolerated by the subjects. The paired t test analysis showed no differences in average laboratory parameters on day (D)-14 and D-28 compared to the baseline (p > 0.05). The cholesterol-lowering herbal medicine does not change the function of the liver, kidneys and blood, with mild side effects of increasing the frequencies of bowel movements and stomachaches.

Abstract

Laboratory work of Pharmaceutical Immunology, Faculty of Pharmacy UGM aims to provide skills of applying antigens, antibodies, and immunological techniques in pharmaceutical; and ideally each student should be able to perform it completely. However, the limitation of time and facilities making the learning process ineffective. An innovative learning method such as hybrid e-learning is expected to increase the learning effectiveness. Learning materials and online quizzes/discussions were uploaded and carried out using elisa.ugm.ac.id (eLisa), respectively, combined with on-site works in the laboratory. The method was evaluated by examination scores and final grades. Student participations in online quizzes increased from first to fourth quiz (average participation rate 98.86±0.87%). Compared to the conventional method applied in 2016 (n=207), the examination average score did not increase; however, the percentage of students whose score is 90-100 were higher in the hybrid e-learning. The average final grade of the hybrid e-learning was 81.43±0.70; whereas the conventional method of 2016 was 80.22±5.22. More than 80% students (n=48) expressed easiness in following courses and were satisfied with the online learning materials’ quality. Hence, the hybrid e-learning results positive effects in increasing the effectiveness of the learning process. Development of more interactive online platforms is needed to optimize the learning process.

Abstract

Inflammation is a normal protective response to tissue damage that mediated by Cyclooxygenase-2 (COX-2) enzyme. The enhancement incidence and impact of inflammatory diseases have encouraged the search for new pharmacological strategies to overcome the problem. Ketumbar (Coriandrumsativum L.) has been traditionally used as an anti-inflammatory and antioxidant, thus giving chance for anti-inflammatory studies. This study aims to determine the ethanol extract of Ketumbar (Coriandrumsativum L.) leaves anti-inflammatory activity and inhibitory effect on COX-2 enzyme. This study uses male wistar rats with 180–220 g body weights. The rats is divided into six groups by the dose of 125 mg/kg BW, 250 mg/kg BW, and 500 mg/kg BW; positive control (Carrageenan 1%); negative control (Na. CMC 0,5%); and comparative group (Celecoxib 9 mg/kg BW). Determining the anti-inflammatory activity, the oedema formation is measured using a plethysmometer and the inhibition activity is determined by ELISA methods. The results showed that the ethanol extract of Ketumbar leaves by the dose of 125 mg/kg BW, 250 mg/kg BW, and 500 mg/kg BW was significantly inhibitted COX-2 valued 65,61%, 76,83%, and 62,93%. The anti-inflammatory activity was shown by inhibiting oedema valued of 62.26%, 70.59%, and 54.90% respectively. These findings suggested that the ethanol extract of Ketumbar leaves had anti-inflammatory activity, that was possibly mediated through inhibition of COX-2 enzymes

Abstract

Diabetes mellitus (DM) is a chronic disease with a very high complication rate and the third-highest cause of death in Indonesia. The management of DM cannot be separated from the action of drug therapy. It is necessary to have pharmaceutical care in handling drug related problems (DRPs) in order to achieve optimal drug effects on patients. The purpose of this study is to describe the characteristics and incidence of DRPs in type 2 DM patients at the Tuminting Public Health Centre, Manado. Sampling of 42 samples is conducted with a purposive sampling method using medical records and patient medical treatment history data for type 2 DM patients retrospectively from January to June 2019. The data is analyzed descriptively. The results obtained from the incidence of type 2 diabetes mellitus in women were 53.10% and men 46.90%. Most patients with type 2 diabetes were at the age of >60 years, 66.67%. The highest co-morbidity were cardiovascular disorders 24,24%. The most widely used antihyperglycemic drug was metformin 72.50%. The most DRPs incidence occurred in May 2019 at 36.36%, with 59.05% was needing drugs was, 13.64% did not need drugs, 20.45% wrong drugs, 2.27% less dose and 4.55% over dose.

Abstract

The high prevalence of infectious disease in Indonesia causes increased use of antibiotics. It has serious consequences since it can cause germicidal resistance rapidly increased, significant morbidity and mortality, as well as high additional medical costs per year. This study aims to evaluate the appropriateness of antibiotic use inpatients in the internal medicine ward of Dr. H. Abdul Moeloek in Lampung Province. This research was observational (non-experimental) with a descriptive evaluative research design. The data were obtained from the retrospective tracing of medical records that collected by a purposive sampling technique on July-December 2017. A total of 163 inclusion samples are analyzed qualitatively and quantitatively. The qualitative analysis uses standard guidelines for the use of antibiotics and quantitative analysis uses the anatomical therapeutic chemical (ATC)/Defined Daily Dose (DDD) method. The results showed that out of 168 medical records, 19 types of antibiotics were used with a 118.57 DDD value of 100 patient days. The most widely prescribed was ceftriaxone (49.09%). The highest number of infections was diabetic ulcers with an incidence of 42 cases (25%). Out of 168 evaluated cases, 166 cases (98.8%) were appropriate indication, 168 cases (100%) were appropriate patients, 150 cases (89,29%) were appropriate medication, and 89 cases (52,97%) were appropriate dosage. These results indicated that the use of antibiotics inpatients in the internal medicine ward of Dr. H. Abdul Moeloek in Lampung Province were rational but it was necessary to consider selectivity regarding the choice of antibiotic use for infected patients.

Abstract

Type 2 diabetes mellitus is a chronic disease of the digestive system characterized by high blood glucose levels. The main enzyme in carbohydrate metabolism is α-glucosidase. One of the therapeutic approaches to treat T2DM is to make glucose uptake into the blood delayed through inhibition of the α-glucosidase enzyme activity. Neem leaves and mango ginger are reported to reduce blood glucose levels. This study aims to determine the potential inhibitor of neem leaves extract and mango ginger extract and their respective fractions on α-glucosidase activity. Simplicia of neem leaves and mango ginger were macerated using 96% ethanol for 24 hours. The ethanol extract of neem leaves and mango rhizome were fractionated using silica gel 60 GF254 as adsorbent and a combination of ethyl acetate and n-hexane as eluent. The extracts of neem leaves and mango rhizome and fractions of both plants were tested for α-glucosidase inhibition with acarbose as a comparison. The results showed that both plants provided inhibitory activity on α-glucosidase with the lowest IC50 value from the semi-polar fraction of neem leaves about 24.16±4.58 μg/mL. Neem leaves and mango ginger have potential as α-glucosidase inhibitors to treat type 2 diabetes mellitus.
