Abstract

Kembang bulan (Tithonia diversifolia [Hemsley] A. Gray) has been used in traditional medicine to treat malaria. Previous research results showed that the ethanol extract of kembang bulan leaves had antimalarial activity in vivo with an ED50 of 113.39 mg/kgBW. Until now, the compounds with antimalarial activity in vivo are unknown. The initial stage that can be done to separate the compounds in the extract is fractionation. This study aims to separate the extract into fractions and test its antimalarial activity. Fractionation was carried out using a vacuum liquid chromatography method using a stationaryphase of silica gel GF254 and a mobile phase of n-hexane, chloroform and methanol in a gradient. In vivo antimalarial activity was tested using the Peter test method. Fractionation produces 5 fractions, each yield is 1.9; 2.1; 17.6; 60.8, and 17.6% respectively for fractions 1-5. The results of the antimalarial activity test for fractions 1-5 with a dose of 2 mg/kgBW showed inhibition of the growth of Plasmodium berghei respectively at 52.3 ± 1.4; 83.5±1.4; 74.6±1.2; 69.5±0.9; 44.8 ± 0.3%. Fraction 2 with the greatest resistance was tested further and showed an ED50 value of 0.52 mg/kgBW.

Abstract

Antiplatelets are crucial medications in the treatment of ischemic stroke patients. The management of stroke cases in Indonesia incurs substantial costs, reaching Rp. 3.2 billion annually. This study conducted a simple pharmacoeconomic research using Cost Minimization Analysis (CMA) method, utilizing medical records and patient financing data of ischemic stroke inpatients at RSUD Gambiran, Kediri City from January to December 2022. The study adopts a hospital perspective with the same outcome of patient recovery. Out of 659 stroke patients, 100 eligible participants, predominantly male aged 46-65 years with Diabetes Mellitus as the most common comorbidity, were included. Based on antiplatelet therapy profiles, the majority received a combination of clopidogrel and aspirin (65%), while 35% received clopidogrel monotherapy. The largest cost component was service cost amounting to Rp 2,898,606 (59.37%). According to CMA analysis, combination therapy showed cost-saving benefits compared to monotherapy in drug and medical equipment, laboratory, service, and total overall costs. Thus, for ischemic stroke patients with the same outcome of recovery, combination therapy of clopidogrel and aspirin isrecommended over clopidogrel monotherapy.

Abstract

Mulberry (Morus alba L.) is a plant from the Moraceae family which has a high flavonoid content. This research aims to determine the ability of NIR spectroscopy and chemometrics to classify mulberry leaf powder based on different planting heights and determine the total flavonoid content using the Uv-Vis spectrophotometric method. The classification of mulberry leaf powder originating from the lowlands, midlands, and highlands provides valid results using NIR spectra and chemometric data from the LDA and SIMCA models with an accuracy value of 100%. The average results of determining total flavonoid content using Uv-Vis spectrophotometry showed that the highest levels were in the highlands, followed by the medium and lowlands with total flavonoid levels respectively, namely 6.453 ± 1.194 mg QE/g powder; 5.889 ± 0.944 mg QE/g powder; and 5.177 ± 0.998 mg QE/g powder. The results of the One Way ANOVA test showed that the total flavonoid content of mulberry leaf powder from different altitude areas did not have a significant difference.

Abstract

Ketoprofen is a potent non-steroidal anti-inflammatory drug compound but is difficult to dissolve in water. This research aims to increase the solubility of ketoprofen through cocrystallization using a urea coformer with the addition of the inorganic salt NaCl. Cocrystallization was carried out using the solvent evaporation method using methanol. The characterization of the cocrystal results included PXRD, DSC, and FTIR. At the same time, the solubility evaluation was carried out using a solubility test in distilled water and a dissolution test in phosphate buffer media pH 7.4. The results showed that ketoprofen forms a cocrystal with the urea coformer, whereas adding NaCl produces a ketoprofen-urea-NaCl cocrystal with a lower crystallinity and melting point. The solubility of ketoprofen-urea-NaCl cocrystals in distilled water was 183.82 ± 2.84 mg/L, while ketoprofen-urea and ketoprofen cocrystals were 174.23 ± 6.83 and 171.12 ± 6.29 mg/L, respectively. The dissolution test in phosphate buffer medium pH 7.4 showed the dissolution rate of ketoprofen-urea-NaCl cocrystals > ketoprofen-urea cocrystals > ketoprofen.

Abstract

Binahong (Anredera cordifolia (Ten) Steenis) is a natural ingredient that can prevent tooth decay due to plaque. The purpose of this study was to formulate and evaluate the physical properties of binahong extract paste and chicken egg shells with various concentrations of CMC Na as a binder. The paste was formulated with various concentrations of CMC Na, namely 1.5% (F1), 2% (F2) and 2.5% (F3). The physical properties of the paste studied included organoleptic, homogeneity, pH, viscosity, spreadability and foam height. The results showed that variations in the concentration of CMC Na affected the organoleptic physical properties of color, viscosity and foam height of the paste. However, variations in the concentration of CMC Na had no effect on the pH and spreadability of the paste.

Abstract

Tuberculosis (TB) is an infectious disease caused by the bacteria Mycobacterium tuberculosis (Mtb) and can attack other body organs. Transmission occurs through splashes of phlegm when coughing, sneezing or talking. TB treatment using antituberculosis drugs (OAT) has side effects ranging from mild to severe side effects, namely hepatotoxicity. Therefore, alternative therapy from natural ingredients is needed because natural ingredients have few side effects when used appropriately. This research aims to determine the class of secondary metabolite compounds in the extract and fraction of the Pacing plant (Costus speciousus, J.Sm) and determine its effectiveness against Mycobacterium tuberculosis H37Rv. The extraction method used is remaceration with methanol solvent. The concentrations of the samples tested varied with the positive control of isoniazid. The research results showed that Pacing plant extracts and fractions contained alkaloids, flavonoids, saponins and tannins. Terpenoid compounds were not detected in the samples tested. The antituberculosis test showed that the n-hexane fraction had the best anti-TB activity, although the IC50 value of the extract and fraction was still lower than that of isoniazid as a positive control. However, Pacing extract and fraction still have potential as a very strong anti-TB agent because the IC50 value is below 50 ppm.

Abstract

Facial skin problems are often caused by skin frequently exposed to free radicals, causing damage to skin cells. Usually, skin damage is characterized by a non-smooth skin structure, the appearance of dark spots, wrinkled skin, and dull skin. Antioxidants are one of the compounds that can protect cells from the influence of free radicals and slow down or prevent the oxidation process. To help reduce, stop, and slow down the adverse effects of free radical exposure, antioxidant essence preparations are used. In this study, glycerin and propylene glycol have been optimized in the preparation of sappan wood extract essence as an antioxidant. The purpose of this study is to determine the optimum concentration of glycerin and propylene glycol as a humectant in the essence preparation of sappan wood extract. In this study, the evaluation carried out includes pH tests, viscosity, and IC 50 antioxidant activity. In this study, essence characterization tests were also carried out including organoleptics, homogeneity, and dispersibility. From the results of the pH test, it was found that all essence formulas had a pH in the range of 5.5-6.0; viscosity in the range of 1.0-2.0 dPa.s; and IC50 test of antioxidant activity in the range of 100-125 μg/mL. The optimum essence formula has a propylene glycol concentration of 10% with a desirability of 0.871, with a predicted pH value of 5.658, a viscosity value of 1.256 dPa.s, and an IC50 value of 106.473 (μg/mL).

Abstract

Smoking has become a lifestyle that affects human health and causes many diseases, including respiratory system disorders, cancer, infertility, and cardiovascular diseases. Cigarette smoke contains harmful ingredients that trigger the formation of free radicals that can damage cells. Dates become a source of antioxidants. Some studies showed that dates have high antioxidant activity due to their compounds, such as carotene, flavonoids, and phenolic acids. Date palm fruit juice is widely sold on the market, but date research on fruit juice is still limited. The present study was designed to evaluate the preventive effect of date palm fruit juice in lung histopathology of mice induced by cigarette smoke. Twenty-four mice were exposed to cigarette smoke and then treated with date palm juice in different doses (5, 10, and 20 ml/kg BW) daily for 14 days. On the 15th day, mice were sacrificed, and the lung organ was separated for microscopic observation. The results showed that starting a low dose of date fruit juice treatment can minimize lung damage comparable to vitamin C, based on the histopathological profile and scoring of the lung damage. These findings suggest that date juice has been proven to have a protective effect in preventing and reducing lung damage due to exposure to cigarette smoke through its antioxidant activity.
