Abstract

Protein-based therapies have already brought about a significant transformation in the field of medicine, and their use continues to grow. The development of protein therapeutics has revolutionized the treatment of various diseases, such as cancer, autoimmune disorders, viral infections, cardiovascular disease, myocardial infarction, etc. Consequently, it is crucial to ensure the stability of formulations, given their rapid growth. Preserving the structural integrity of protein-based medicines is a significant challenge in developing a stable and high-quality formulation. This challenge arises during various stages, including manufacturing procedures, storage, handling, distribution, and delivery due to proteins' intricate and delicate nature. Hence, it is crucial to thoroughly understand the various degradation mechanisms that impact protein stability to enhance different variables and minimize the formation of degradation products, such as aggregation, oxidation, deamidation, etc., which could potentially have clinical implications. This review provides an important step in understanding the process of protein degradation and offers a beneficial approach to investigate the degradation of proteins, specifically aggregation, through several analytical and biophysical methods. This is important for facilitating the further advancement of protein-based therapies.

Abstract

Superoxide dismutases (SODs) are metalloenzymes that defend the body against reactive oxygen species and contribute to combating inflammation. Human cells have three distinct SODs, i.e., manganese SOD (MnSOD), extracellular SOD(ECSOD), and copper-zinc SOD (Cu-ZnSOD) or SOD1. The crystal structure of human SOD1 in a complex with a naphthalene-catechol-linked compound revealed hydrogen bonds and hydrophobic interaction. Catechins arepolyhydroxylated polyphenols contained in the leaves of Camellia sinensis L. This work aims to study the binding mode and molecular dynamics of two major catechins, epigallocatechin (EGC) and epigallocatechin gallate (EGCG) with human SOD1 (in complex with SBL1, a naphthalene-catechol linked compound). Both catechins demonstrated a binding mode with the enzyme, in terms of hydrogen bonds and hydrophobic interaction, similar to the native ligand (SBL1). Of the two catechins, EGC possesses a better binding affinity (docking score of -4.15 kcal/mol) for human SOD1 compared to EGCG (docking score of -4.02 kcal/mol), thus the EGC-SOD1 complex was continued in MD simulation to investigate the conformational stability and time-dependent ligand binding ability in the binding pocket. The molecular dynamics simulation confirmed that EGC is more stable than the native ligand, SBL1, with the RMSD average value of SBL1 and EGC being 1.1669 Å and 0.5607 Å, respectively. Taken together, this study confirms the antioxidant activity of catechins in C. sinensis L.

Abstract

Sasar penelitian ini ialah untuk mengetahui dosis terbaik untuk histologi pankreas dan metabolits sekunders pada konsentrat etanolik daun pepaya (Carica papaya L). Penelitian ini merupakan percobaan laboratorium dengan 30 ekor tikus sebagai hewan uji. Hewan uji di bagi menjadi enam kelompok perlakuan masing-masing lima ekor tikus. Suspensi Na-CMC diberikan pada kelompok 2 (control negatif), glibenklamid diberikan pada kelompok 3 (control positif), dan 100, 200, dan 300mg/kgBB konsentrat etanol daun pepaya (Carica papaya L) diberikan pada kelompok 4, 5, dan 6. Konsentrat etanol daun pepaya mengandung senyawa metabolits sekunders antara lain alkaloids, flavonoids, saponins, dan tanins, sesuai temuan. Pepaya Etanol daun pepaya secara bermakna mempengaruhi gambaran histopatologi pankreas, Etanol daun pepaya terpisah pada porsi 300mg/kgBB merupakan bagian yang mempengaruhi gambaran histopatologi pankreas dengan skor tipikal 1,2.

Abstract

Luka bakar derajat dua merupakan luka yang sering terjadi akibat kecelakaan dan memiliki nilai mortalitas yang tinggi. Pengembangan pembalut luka bakar primer seperti “film polielektrolit” dapat dilakukan untuk mengontrol pelepasan zat aktif dan menjaga kelembaban luka. Efektivitas film ini dapat ditingkatkan dengan penambahan getah dari tanaman Jarak Pagar (Jatropha curcas) yang secara ilmiah memiliki aktivitas dalam penyembuhan luka seperti Flavonoid, Saponin dan Tanin. Matriks dari Film Polielektrolit memiliki kemampuan yang baik apabila dibentuk dari biopolimer yang biodegredibel dan biokompatibel serta nontoksik seperti Karagenan dan Kitosan. Karagenan diekstrak dari Alga Merah (Eucheuma spinossum) dengan metode maserasi. Karagenan dikombinasikan dengan Kitosan sebagai polimer penyusun film polielektrolit dievaluasi meliputi Uji Organoleptik, Uji Ratio Swelling, dan Uji Fraksi Gel. Formula terbaik di uji iritasi selama 3 x 24 jam dan kemudian diuji efektivitas terhadap 5 kelompok hewan uji mencit dengan perlakuan berbeda yakni PE/JS0 (film polielektrolit); JS5 (getah 5%); JS10 (getah 10%); PE/JS5 (film polielektrolit dengan getah 5%); dan PE/JS10 (film polielektrolit dengan getah 10%). Dimana pengukuran penurunan dievaluasi secara statistik menggunakan metode One Way – Anova (α=0,05). Hasil menunjukan formula PE/JS10 memiliki aktivitas penyembuhan luka sebesar 100% selama 14 hari.

Abstract

Prevalensi luka bakar masih menjadi perhatian kaitannya dengan perubahan fisik pasca luka bakar yakni contractures, hypertrophic scar dan keloid yang menurunkan fungsi, kualitas hidup dan estetika kulit. Herba pegagan (Centella asiatica) melalui senyawa triterpen telah dikenal untuk mempercepat luka bakar. Namun, sifat senyawa triterpen yang polar identik dengan keberadaan klorofil dan senyawa lainnya yang sulit dipisahkan. Masih adanya kontroversi terkait protein yang berperan dalam kesembuhan luka bakar membutuhkan kajian untuk menentukan protein potensial. Penelitian ini bertujuan untuk mengeksplorasi metode baru pada ekstraksi herba pegagan menggunakan karbon aktif serja kajian farmakologi jejaring untuk menentukan protein potensial. Herba pegagan diekstraksi menggunakan metode maserasi pada pelarut metanol dan dilanjutkan dengan penambahan karbon aktif. Hasil purifikasi diidentifikasi menggunakan KLT dengan fase gerak kloroform: asam asetat glasial: metanol: akuades (60:32:12:8 v/v) dan pereaksi semprot anisaldehid-asam sulfat. Kajian farmakologi jejaring dilakukan secara in silico untuk mengkaji interaksi protein-protein dan senyawa-protein berkaitan dengan luka bakar. Hasil skrining fitokimia menunjukkan keberadaan asaticoside pada ekstrak terpurifikasi dan maserat dengan nilai Rf yang relatif identik. Senyawa non aromatik berperan dalam mempercepat kesembuhan luka bakar dengan mempengaruhi protein IL1β, IL6, MAPK8, PTGS2, NOX4, IFN-γ, NFKB1, TGFβ1, MMP13, dan TNF.

Abstract

The Indonesian bay leaf (Syzygium polyanthum) are one of the traditional cooking spices used in several countries in Southeast Asia, and this spice gives a distinctive herbaceous aroma. Bay leaves are often used as a medicinal plant for generations by the community to reduce cholesterol, diabetes, hypertension, gastritis, and diarrhea. The purpose of this study was to compare the content of polyphenols in Indonesian bay leaf decoction and bay leaf tea. The total phenolic content was determined by Folin-Ciocalteu’s method. The test results showed that the highest polyphenol content was found in bay leaf tea at 19.19 mg/kg, and then bay leaf decoction at 6.36 mg/kg. Based on the statistical analysis, it was stated that there was a difference in the levels of polyphenols in each sample, meaning that there was a significant difference. The implications of this study can be to develop processing techniques for Indonesian bay leaf which contain the best polyphenols for public consumption.

Abstract

Pasien geriatri memiliki resiko lebih besar menerima terapi polifarmasi akibat penyakit dan komplikasi yang dideritanya yang membuat pasien geriatri lebih rentan terhadap penggunaan obat yang berpotensi tidak tepat sehingga meningkatkan terjadinya risiko efek samping obat.  Penelitian ini dilakukan untuk mengetahui karakteristik pasien dan mengidentifikasi terjadinya Potentially Inapropriate Medication (PIM) pada pasien geriatri rawat inap dengan diagnosis gangguan kardiovaskuler menggunakan kriteria Beers dan STOPP di RSUD Inche Abdoel Moeis tahun 2022. Penelitian ini adalah penelitian observasional dengan menggunakan data retrospektif yang teknik pengambilan datanya dilakukan secara purposive sampling. Setelah dilakukan penelitian, didapatkan hasil berupa data pasien sebanyak 87 pasien yang meliputi karakteristik pasien geriatri dengan jenis kelamin terbanyak adalah perempuan sebanyak 51 pasien (59%) dengan rentang usia 65-74 tahun sebanyak 64 pasien (74%), riwayat pendidikan pasien terbanyak adalah SMA sebanyak 25 pasien (25%) dengan pekerjaan pasien terbanyak adalah Ibu Rumah Tangga sebanyak 45 pasien (52%). Berdasarkan analisis Potentially Inappropriate Medications (PIM) menggunakan kriteria Beers dari 72 kasus ditemukan sebanyak 13 kejadian (18,06%) dan berdasarkan kriteria STOPP dari 48 kasus ditemukan 3 kejadian (6,25%).

Abstract

Some topical preparations for acne have been developed in the form of gels and creams. However,it is still rarely developed in the form of a patch dosage form. Waste from silkworm cocoons has theactivity of inhibiting the growth of acne-causing bacteria, can be formulated into patch preparations.This study aims to formulate anti-acne patches from silkworm cocoon waste with variations of HPMCand PVP. The formulation of the anti-acne patch from silkworm cocoon waste was made by mixingthe lyophilisate of silkworm cocoon waste with a patch base with variations of HPMC and PVP. Theformulations were divided into 3 with different concentrations of HPMC and PVP additives, namelyformulas 1 (4:1), 2 (2,3:1), and 3 (1:1), respectively, The evaluations carried out included organolepticexamination, patch thickness, weight uniformity, fold resistance, moisture absorption, drying shrinkage.The results obtained showed that the concentration of the combination of HPMC and PVP polymersin the patch preparation gave different results from all evaluations that had been carried out, includingorganoleptic evaluation, weight uniformity, fold resistance, and drying shrinkage were in accordancewith the requirements of a good patch. The optimum concentration of the combination of HPMC andPVP as a polymer in the anti-acne patch preparation from silkworm cocoons is formula 2 with a ratioof (2,3:1) as novelty in this study.

Abstract

Pengelolaan diabetes melitus tipe II yang tepat sebaiknya mengikuti pengobatan yang rasional. Penggunaan obat antidiabetik yang rasional menjadi hal penting dalam mencapai terapi yang efektif dan efisien. Penelitian ini bertujuan untuk mengetahui profil dan rasionalitas penggunaan antidiabetik, serta korelasi rasionalitas pengobatan terhadap kadar glukosa darah pasien. Penelitian ini menggunakan desain penelitian non-eksperimental observasional dengan rancangan penelitian retrospektif, kemudian hasil yang diperoleh di analisis secara deskriptif. Uji korelasi pada penelitian ini menggunakan uji Chi-square. Teknik pengambilan sampel menggunakan purposive sampling dengan jumlah sampel 82 pasien diabetes rawat inap di RSUD Kota Kendari Tahun 2021-2022. Hasil penelitian menunjukkan penggunaan antidiabetik kombinasi (36,58%) dan antidiabetik tunggal (63,41%). Berdasarkan kerasionalan penggunaan antidiabetik yaitu tepat obat (17,07%),  tidak tepat obat (82,92%,) tepat pasien 10,97%, tidak tepat pasien 89,02%, tepat dosis dan tepat interval waktu pemberian (100%). Penelitian ini menunjukan tidak terdapat hubungan antara rasionalitas tepat obat dengan kadar glukosa darah (p>0,05) dan rasinolitas tepat pasien dengan kadar glukosa darah (p>0,05), namun terdapat hubungan antara rasionalitas tepat obat dengan komorbid (p< 0,05) dan rasionalitas tepat pasien dengan komorbid (p< 0,05). Kesimpulan dari penelitian ini didapatkan sebagian besar pasien diabetes rawat inap RSUD Kota Kendari berusia 45-59 tahun dengan jenis kelamin perempuan serta hipertensi sebagai penyakit penyerta terbanyak, rasionalitas penggunaan obat secara umum masih dibawah 100% dan tidak terdapat hubungan antara rasionalitas pengobatan dengan kadar glukosa darah, namun terdapat hubungan dengan komorbid.

Abstract

Sunscreen is applied to the skin to protect it from UV rays. Wungu leaves (Graptophyllum pictum (L.) Griff), which are rich in flavonoids and exhibit antioxidant activity, are one of the substances that can be used to create sunscreen formulations. The purpose of this research is to determine how varying concentrations of wungu leaf extract affect the physical qualities of sunscreen spray formulations. This research method involves extraction, extract assessment (organoleptic, yield, and water content), formulation with different extract concentrations (500 ppm, 1,000 ppm, and 1,500 ppm), and physical evaluation (organoleptic, homogeneity, pH, viscosity, and spraying pattern). Data analysis use linear regression equations in the software Microsoft Excel. All formulations with different extract quantities yielded green, transparent, liquid, homogenous solutions with a pronounced smell of wungu leaf extract. The results of pH and viscosity tests on formulations including diverse extracts satisfied the requirements for topical spray preparations, with pH and viscosity values sequentially dropping (Formulas 1, 2, and 3) with slope values of -0.43 and -4.835, respectively. The results of the spray pattern test indicate an oval to circular spread with increased diameter values in the extract variation and slope values of 0.55, 0.568, 0.6685, and 0.4865 at spraying distances of 3, 5, 10, and 15 cm, respectively. On the basis of the study's findings, it can be inferred that differences in the content of wungu leaf extract have an effect on the physical properties of spray sunscreen formulations, as measured by pH, viscosity, and spray pattern diameter.
