Abstract

Melasma adalah kelainan pigmentasi yang diakibatkan sintesis melanin berlebih berupa bercak cokelat disekitar wajah atau leher. Melasma dapat mengenai semua kelompok ras, terutama daerah dengan radiasi ultraviolet tinggi seperti suku Hispanik/Latin dan Asia. Zat yang sering digunakan untuk mencegah hiperpigmentasi adalah asam kojat, asam azelat dan merkuri, akan tetapi zat tersebut bersifat karsinogen jika digunakan terus menerus. Sirih merah (Piper crocatum) memiliki senyawa aktif yang berperan sebagai antioksidan seperti flavonoid dan fenol. Penelitian ini bertujuan untuk mengetahui potensi senyawa aktif daun sirih merah terhadap penghambatan tirosinase. Penelitian ini dilakukan dengan metode in silico dan in vitro. Reseptor yang digunakan adalah tirosinase dengan kode PDB 5M8O, tropolon sebagai native ligand, dan asam kojat sebagai ligan pembanding. Sebanyak 34 senyawa aktif daun sirih merah hasil analisis LC-MS dan GC-MS.digunakan sebagai ligan uji. Penambatan molekuler dilakukan dengan menggunakan AutoDock Vina dan AutoDock Tools, serta divisualisasikan menggunakan Ligplot+ dan PyMOL. Penghambatan tirosinase secara in vitro menggunakan kit tyrosinase activity assay ab252899 dan asam kojat sebagai kontrol positif. Senyawa katekin memiliki energi bebas pengikatan (ΔG) terkecil, yaitu −6.7 kkal/mol, dan kinetika inhibisi sebesar 12.12 µM. Hasil pengujian in vitro, fraksi air memiliki aktivitas inhibisi tirosinase terbesar yaitu 84.84% dan asam kojat memiliki aktivitas sebesar 82.47%.

Abstract

The quantitative analysis of vitamin C content from variations beverage  packaging  containing red guava (Psidium Guajava Linn) fruit juice had been done. In this study, 4 samples were obtained from the shopping center in Garut and Bandung City. Samples were tested quantitatively by 2,6-dichlorophenol indophenol titration method. The results showed different concentration of 4 samples consist of  tetra pack packaging, tin, glass, and plastic bottles, such as; 17.99 mg/100 gr, 31.46 mg/100 gr, 13.00 mg/100 gr, and 12.01 mg/100 gr, respectively. These result indicated that the packaging variations affected to the level of vitamin C content which was characterized by decreased levels of vitamin C. It means the levels of vitamin C from this research were not in accordance to nutritional value information on the packaging. Tetra pack packaging was the most stable compared to other packaging even though it had a shorter expired date than with other.

Abstract

Kebersihan kulit harus dijaga baik dari kotoran ataupun bakteri seperti Staphylococcus aureus. Salah satu cara menjaga kebersihan kulit yaitu dengan sabun batang khususnya sabun batang antibakteri yang mengandung triclosan (non-herbal) ataupun herbal yang mengandung bahan alami dari tumbuhan seperti daun sirih, serai, bunga melati dan buah pepaya. Penelitian ini bertujuan untuk membandingkan aktivitas antibakteri sabun batang non-herbal dan herbal dengan menghitung konsentrasi hambat minimal (KHM) terhadap bakteri Staphylococcus aureus. Metode yang digunakan adalah metode difusi agar dengan cakram kertas. Sampel yang digunakan yaitu 4 sabun batang non-herbal yang mengandung triclosan serta 4 sabun batang herbal yang mengandung ekstrak daun sirih, sereh, bunga melati dan buah papaya. Konsentrasi sampel yang digunakan 1,25%, 0,625%, 0,312%, 0,156% dan 0,078% (b/v) dengan antibiotik klindamisin sebagai pembanding. Hasil penelitian menunjukkan bahwa semua sampel memiliki aktivitas antibakteri. Nilai KHM sabun batang herbal dan non-herbal yaitu pada konsentrasi 0,078% dengan diameter zona hambat 0,163 + 0,012 sampai 0,363 + 0,045 dan 0,323 + 0,012 sampai 0,510 + 0,057 secara berturut-turut. Hasil penelitian menunjukkan bahwa sabun batang non-herbal dengan kandungan triclosan memiliki aktivitas antibakteri yang lebih baik dibandingkan dengan sabun batang herbal karena memiliki diameter zona hambat yang lebih besar dibandingkan dengan sabun batang herbal.

Abstract

Tuberculosis is an infectious disease that is a leading cause of death globally, as well as one of the top ten causes of death from a single infectious agent. Antibiotics treatment for tuberculosis reduces pro-inflammatory cytokines, which is one of the reasons for dysbiosis. The proportion of Actinobacteria, Firmicutes, and Bacteroidetes in the gut microbiota distinguishes between drug-sensitive and drug-resistant tuberculosis. The gut–lung axis theory explains how tuberculosis alters the gut microbiota while also altering the immune response. Probiotics have immunostimulatory and immunoregulatory properties that affect the immune system. The gut-lung axis is a two-way system that enables microbial products, endotoxins, metabolites, hormones, and cytokines to reach the bloodstream that connects the intestines and lungs, where they exert effects on both. According to the gut–lung axis theory, probiotics may play a role in tuberculosis immune responses. This review includes studies conducted in English and Indonesian from 2010-2022. The Cochrane Library, Scopus, Medline, PubMed, and grey literature databases will be used in the review. Studies that use specimens from pulmonary tuberculosis patients, healthy volunteers induced by Mycobacterium tuberculosis, volunteers with a history of pulmonary tuberculosis disease, and volunteers who had close contact with pulmonary tuberculosis patients were considered eligible. The current review highlights the immune modulation induced by probiotics usage in tuberculosis. Accordingly, probiotics have been shown to enhance the immune response against tuberculosis. More studies are needed to understand probiotic’s role in different types of tuberculosis, and the influence of different probiotic bacteria on immune modulation.

Abstract

Pemanfaatan supernatan Bacillus subtilis kini telah banyak dimanfaatkan dalam bidang kesehatan sebagai probiotik karena mengandung senyawa bakteriosin serta enzim enzim. Senyawa tersebut mampu menghambat mikroorganisme lain dan mudah terdegradasi oleh enzim proteolitik. Pemanfaatan nanopartikel polimer cukup efektif untuk melindungi probiotik dari kondisi gastrointestinal. Penelitian ini bertujuan untuk membuat sediaan nanopartikel supernatan B. subtilis dengan pembawa PLGA dan dilakukan karakterisasi. Nanopartikel dibuat dengan metode solvent evaporation menggunakan variasi konsentrasi PLGA. Hasil karakterisasi nanopartikel supernatan B. subtilis memiliki ukuran nanopartikel 545,45 ± 90,73 nm dengan nilai indeks polidispersitas (PDI) kategori monodisperisi yaitu 0,503 ± 0,065 dan zeta potensial -14,57 ± 3,08 mV. Morfologi yang dihasilkan berbentuk sferis dan cukup seragam. Analisis Fourier transform infrared spectroscopy (FTIR) menunjukkan pita-pita spektrum mirip dengan nanopartikel PLGA dan menunjukkan beberapa puncak spesifik supernatan B. subtilis dengan sedikit pergeseran. Analisis Differential Scanning Calorimetry (DSC) pada termogram tidak menunjukkan adanya puncak endotermik dari supernatan B. subtilis. Formulasi nanopartikel supernatan B. subtilis telah ter-loading-kan ke dalam sistem nanopartikel melalui analisis FTIR dan DSC.

Abstract

Immunomodulators operate through various mechanisms, one of which involves enhancing phagocytic activity. The purple button herb (Borreria laevis Lamk.) is recognized for containing secondary metabolites that may function as immunomodulators; however, its influence on phagocytosis has not been previously assessed. Consequently, a study explored the immunomodulatory effects of distinct fractions of the purple button herb on macrophage phagocytosis in male mice. Over seven days, the mice were orally administered ethanolic extract, ethyl acetate, and n-hexane fractions of the purple button herb. On the eighth day, the subjects were infected with Staphylococcus aureus. After euthanization, intraperitoneal fluid was collected to analyze macrophage phagocytosis activity. The study's results suggested that the ethanolic extract, ethyl acetate, and n-hexane fractions of the purple button herb tended to enhance macrophage activity, with the n-hexane fraction achieving the highest phagocytosis rate of 84.7%. The corresponding p-value > 0.05 in the LSD test indicated no statistically significant difference when compared to the positive control. In conclusion, the n-hexane fraction of the purple button herb demonstrates potential as an immunostimulant by augmenting macrophage activity.

Abstract

Kompleksitas patogenesis kanker membuat kanker sulit untuk menemukan target terapi yang tepat, sehingga tingkat keberhasilan obat baru dalam uji klinis rendah. Oleh karena itu, studi pengembangan teknik intervensi yang baru untuk pasien kanker sangat penting. Large-type Amino Acid Transporter 1 (LAT-1) diekspresikan secara berlebihan dalam sel kanker jika dibandingkan dengan sel normal, hal ini menjadikannya salah satu target molekuler khusus untuk terapi kanker. Fungsi LAT-1 adalah memberikan nutrisi pada sel kanker untuk berkembang biak secara masif sehingga penghambatan LAT-1 dapat dijadikan alternatif terapi kanker. Penelitian ini bertujuan untuk mendapatkan senyawa potensial yang akan digunakan untuk radioteranostik dengan dua senyawa inhibitor LAT-1 sebagai senyawa pembawa, yaitu 5-Benzyloxytryptophan (5-BOTP) dan asam (S)-2-amino-4-(3,5-diklorofenil) butanoat (ADPB) yang dikonjugasikan dengan senyawa linker dan berbagai agen pengkelat bifungsional (BFCA) terhadap situs antiporter LAT-1. Metode penelitian terdiri dari mode pengikatan senyawa 5-BOTP dan ADPB terkonjugasi dengan LAT-1, simulasi penambatan molekuler, dan analis pre-ADMET. Hasil penelitian menunjukkan bahwa 5-BOTP-6AHA-H2CB-DO2A dan ADPB-6AHA-H2CB-DO2A memiliki afinitas terbaik dan kompleks pengikatan yang terbaik dengan LAT-1 sehingga keduanya berpotensi sebagai agen radioteranostik kanker dengan menghambat LAT-1.Kata kunci: Kanker, radioteranostik, LAT-1, 5-Benzyloxytryptophan, asam (S)-2-amino-4-(3,5-diklorofenol) butanoat.

Abstract

The immune system is vital in protecting the body from pathogens and harmful foreign substances. This study aims to evaluate the immunomodulatory activity of Arenga pinnata leaf extract on macrophage phagocytosis activity in mice (Mus musculus) and determine its optimal dosage. Extraction was conducted using the maceration method with 96% ethanol as the solvent. The immunomodulatory test was performed by measuring macrophage phagocytosis activity. Male mice were divided into six groups: a negative control (0.5% Na-CMC), a positive control (Stimuno® 0.013 mg/kgBW), and four treatment groups receiving Arenga pinnata leaf extract at doses of 100 mg/kgBW, 200 mg/kgBW, 300 mg/kgBW, and 400 mg/kgBW. After seven days of treatment, the mice were injected intraperitoneally with Staphylococcus aureus bacteria, and peritoneal fluid was collected for macrophage phagocytosis activity analysis under a microscope. The results showed that Arenga pinnata leaf extract significantly increased macrophage phagocytosis activity at 300 mg/kgBW and 400 mg/kgBW compared to the positive control. Phytochemical analysis revealed the presence of flavonoids, saponins, tannins, and triterpenoids, which contribute to its immunomodulatory effects. In conclusion, Arenga pinnata ethanol leaf extract exhibits potential as an immunomodulatory agent, particularly at 300 mg/kgBW and 400 mg/kgBW doses. However, further studies are necessary to explore its pharmaceutical applications and elucidate its mechanisms of action.

Abstract

Bakteri dan jamur merupakan penyebab utama penyakit infeksi, pencegahan dapat diatasi dengan penggunaan hand sanitizer, tetapi dapat menimbulkan efek samping iritasi, maka diperlukan bahan alternatif alam. Asap cair kayu sengon memiliki kemampuan sebagai antiseptik. Penelitian ini bertujuan untuk mengetahui efektivitas asap cair kayu sengon sebagai antiseptik terhadap pertumbuhan mikroba secara in vitro dan in vivo. Hasil uji in vitro didapatkan zona hambat tertinggi pada asap cair kayu sengon grade 1 terhadap E. coli ATCC 25922 konsentrasi 75% 5,8 mm.  S. aureus ATCC 25923 konsentrasi 75% 10 mm. A. flavus ATCC 9643 konsentrasi 75% 22,8 mm. C. albicans ATCC 10231 konsentrasi 75% 3,6 mm. Grade 2 terhadap E. coli ATCC 25922 konsentrasi 75% 7,5 mm. S. aureus ATCC 25923 konsentrasi 75% 12 mm. A. flavus ATCC 9643 konsentrasi 75% 27,8 mm. C. albicans ATCC 10231 konsentrasi 75% 5,1 mm. Hasil uji in vivo menunjukan efektivitas asap cair kayu sengon grade 2 pada bakteri sebesar 54,51%, jamur sebesar 84,37%. Dari data kuesioner responden diperoleh 75% menyukai warna, 41% menyukai aroma, 75% tidak menimbulkan efek kekeringan, 92% tidak mendapatkan efek samping. Dapat disimpulkan asap cair kayu grade 2 konsentrasi 75% memiliki kemampuan dalam menghambat pertumbuhan mikroba.

Abstract

Hibiscus surattensis L. leaves have been known as a folk medicine. Several previous reports have shown several potential therapeutic effects, including antidiabetic, antibacterial, anti-diarrheal, analgesic, hepatoprotective, and antioxidant, but no toxicity study is similar to the current one. Therefore, it is necessary to investigate the cytotoxic effect of these commonly consumed medicinal leaves. This study aimed to investigate the cytotoxicity of Hibiscus surattensis L. leaves extract (HSL). The cytotoxic effect was determined through brine shrimp lethality bioassay (BSLT) and in vitro Presto Blue assay of MCF-7 and Hela cell.  HSL extract was obtained by maceration using 96% ethanol. The BSLT results showed that the HSL has LC50 value of 224.814 μg/mL. MTT assay results showed the IC50 value of the HSL extract on MCF-7 cells was 1419.83 μg/mL, and Hela cells were 2050.50 μg/mL. Based on the LC50 value that has been analyzed against HSL extract shows an LC50 value of less than 1000 μg/mL which can be concluded while having potential cytotoxic activity against experimental animal larvae of A. salina Leach. The results of the cytotoxic test with Presto Blue assay on two cancer cells used showed that the HSL extract was not toxic.
