Abstract

The knowledge on the species adaptation factor of avian influenza virus of H5N1 subtype (AIV H5N1)is very important as a signal for the emergence of a new strain with pandemic potential. This research wasconducted to find out the sequence variation of the Non-Coding Region (NCR) and Coding Region (CR) of 5’-terminal cRNA of the polymerase acidic (PA). Total RNA from twenty six (26) avian influenza virussubtype H5N1 isolates were amplified using reverse-transcriptase-polymerase chain reaction (RT-PCR)with a universal forward primer for influenza virus and specifically designed backward primers. Nineteen(19) gene fragments of PA could be amplified. RT-PCR products were sequenced and analyzed using Mega4 software. The length of NCR of PA gene was found to be 24 bases and conserved. A/T composition of PANCR was 58.3%. Species and geographical specificity could not be found in the genetic distance, the aminoacid polymorphism, as well as the phylogenetic analysis of the CR. RNA sequencing is discussed andrecomended to be studied further.

Abstract

The effectiveness of homeopathy combination in eliciting nonspecific cellular defense mechanism indairy cows is demonstrated in this study. Two groups of 2-7 years old, in 3-5 months term of pregnancydairy cows which suffering clinical mastitis were used id this study. At 3 weeks and 4 weeks prae partusanimals received their first homeopathy treatment, i.e. Group I: Coenzyme comp®, and Group II: a combinationof Mucosa comp® and Traumeel®, respectively. At one week and two weeks post partus all animals in thetwo groups received their second treatment, i.e. a combination of Lachesis comp® and Traumeel®.Subsequently, at 3 and 4 weeks post partus all animals in the two groups received their third treatment,i.e. a combination of Carduus comp® and Coenzyme comp®, respectively. All homeopathy solutions weregiven at 5 ml subcutaneously. Ten ml of blood samples for haptoglobin analysis were drawn from eachanimal and blood smears for White-Blood-Cell counting and differentiation and phagocytosis activity andcapacity observation were prepared from each animal at 3 and 4 weeks prae partus; and at one week, 3, 5,and 7 weeks post partus, respectively. Our results indicated that although there were non significantdifference in the phagocytosis activity and capacity between animals in Group I and Group II; animals inGroup II showed a marked increase in phagocytosis activity and capacity. Based on haptoglobin analysis,animals in Group II showed a steadily low haptoglobin level in the blood (0.08 mg/ml) up to 3 weeks postpartus and a tendency to further decline until the end of the observation. Based on the WBC counts anddifferentiation, animals in Group II also showed a better response compared to animals in Group I.In conclusion, the use of homeopathy combination is capable in eliciting nonspecific cellular defensemechanism; this phenomena were clearly shown in the combination of Mucosa comp® and Traumeel®. Theuse of this homeopathy combination should be considered at prae partus allowing the recognition of thehost defense mechanism.

Abstract

Fracture healing is a complex process that requires the ordered synthesis of matrix proteins and deposition of mineral. Complications such as delayed union or non-union may arise from excessive flexibility bone because low mineral content of bone or apposition of both fracture fragments is not complete yet. The aim of this study is to determine the effect of excessive calcium carbonate (CaCO3) on the healing femoral fracture in rat by examining histopathological change of femoral fracture. Twelve male Sprague Dawley rats, 12 weeks old were subjected to this investigation. The entire rats model at 13 weeks old were treated reposition of femoral dexter fractures with intramedullary pin fixation. The rats model were evenly randomly divided into three groups of four at 15 weeks old ; group I treated with normal diet, group II with normal diet + CaCO3 supplement 75 mg/day, group III with normal diet + CaCO3 supplement 225 mg/day per oral. At the end of the experiment (19 weeks old), the rats were killed, subsequently dexter femur were taken out for determining the histopathological examination. The results showed that excessive suplementation CaCO3 to description of histopathological fracture healing on group III had even better, callus was able to bridge the fracture fragment with completely. The proportion woven bone and trabeculae were higher, while group II were cartilage dominate in callus, and group I were higher fibrous tissue. From the results can be concluded that given CaCO3 225 mg/ day accelerate the healing process of femoral fractures.

Abstract

The purpose of this study was to evaluate the effect of cooling and freezing of ewes ovarian tissue ontheir follicles morphology. The study was carried out in two experiments. Experiment I, ovaries weremaintained in Phosphate Buffered Saline (PBS) at -20oC and room temperature (RT) for 24 h, and at 5oCfor 24 h and 72 h, respectively. After storage, follicles were histologically evaluated. Experiment II, theovarian cortex was isolated and tissue slices (±1 mm3) were prepared. Following this tissues were loadedinto hemistraw then transferred to equilibration solution (PBS+20% FBS+7,5% EG+7,5 % DMSO) atroom temperature and held for 10, 20, 30 minutes, respectively. Afterward tissues were tranferred tovitrification solution (PBS+20%FBS +15%EG+15%DMSO ) for 3 minutes, then the hemistraw was placeddirectly into liquid nitrogen. After thawing, the tissues were prepared for histological examination. All ofthe follicles were deteriorated after 24 h storage at RT. The percentage of morphologically normal follicleswas significantly reduced when ovarian tissues were stored at -20oC for 24 h and at 5oC for 24 h and 72 h.However, it seemed to have a minor deterioration effect when the tissues were kept at 5oC for 24 h(P<0.05). Antral follicles were damaged in all of the treatments. Primordial follicles preserved theirmorphology intactness better than growing follicles. Exposing tissues to equilibration medium for 10minutes seemed to produce higher numbers of morphologically normal follicles (P<0.05), compared towhen tissues were exposed for 20 minutes and 30 minutes (P>0.05). It can be concluded that exposingtissues to equilibration solution for 10 minutes prior to freezing would kept the ovarian follicles morphologyin good condition.

Abstract

Keamanan bahan pangan merupakan masalah yang amat penting bagi konsumen dan industri pangan. Cemaran bakteri Escherichia coli dan Coliform dianggap sebagai indikator sanitasi dalam proses pengolahan bahan pangan. Pelacakan bakteri patogen dalam pangan juga telah dilakukan secara rutin, termasuk yang bersifat zoonosis seperti Escherichia coli O157:H7. Bakteri ini menghasilkan toksin yang dikenal dengan Shiga toxin. Toksin ini dapat menimbulkan diare berdarah, colitis haemorrhagi dan hemolytic uremic syndrome (HUS) pada manusia. Dalam penelitian ini dipelajari hubungan antara tingkat cemaran dan insidensi Coliform, E.coli, E.coli O157 dan E.coli O157:H7 pada daging sapi. Bakteri pertama ditumbuhkan pada media EMBA, selanjutnya dipupuk pada media SMAC dan diakhiri dengan uji aglutinasi lateks untuk memastikan keberadaan bakteri E.coli O157 dan uji antiserum H7 untuk memastikan isolat yang diisolasi merupakan isolat E.coli O157:H7. Hasil isolasi dan identifikasi terhadap 89 sampel daging sapi diperoleh hasil rata-rata tingkat cemran coliform dan E.coli sebesar 93,01+ 2,64x103 cfu/g dan

Abstract

A severe and extensive intestine damages can occur in dogs suffering from volvulus , strangulation,intersuception and neoplasia. Extensive enterectomy is the most common medical treatment for dogssuffering from such disorder. A study was therefore conducted to compare the serum electrolyte level ofextensively enterectomyzed dogs after being treated with different level (0%, 0.05% and 0.5 mg/bodyweight) of lactoferin. As many as 9 dogs at 3-4 moth-old and with the body weights ranging from 4-5 kgwere used. After being enterectomized to the extent of 75% out of the total length of the intestine, the dogswere divided randomly into 3 groups (I, II and III) each of which consisted of 3 dogs. The three groups weretreated for 30 days respectively with 0.0 mg/body weight, 0.05mg/kg body weigth and 0.5mg/kg bodyweight of lactoferin. The electrolyte levels (Na, K and Cl ion) of dogs were determined at days 1, 15 and 30of during the treatment. The data collected from this study were analysed by Analysys of Variance (Anova)proceded by Duncan Multiple Range test (DMRT). The result showed that at days 15 and 30 of thetreatment, the electrolyte levels of the dogs with lactoferin (0.05 and 0.5 mg/body weight) were significantlyhigher than in dogs without lactoferin (0mg/bodyweight). The level of Na ion of dogs with 0,5, 0,05, dan 0mg/kg bw lactoferin were 143, 143,4, dan 141,7 mEq/L respectively at day 15 and 147, 150, dan 137,7mEq/L respectively at day 30. The levels of K ion for those dogs were 5,17, 4,97, dan 3,83 mEq/Lrespectively at day 15 and 30: 5,1, 5,13, 3,73 mEq/L respectively at day 30. Meanwhile, their Cl levelswere 113,7, 114,3, 104 mEq/L respectively at day 15 and 115,3, 117,3, dan 91,3 mEq/L respectively atday 30. Hasil penelitian ini menunjukkan bahwa memperian lactoferin memperbaiki kadar elektrolitserum anjing yang dienterektomi secara ekstensif. It was evidence that lactoferin treatment can improvedthe electrolyte profiles of extensively enterotomized dogs.

Abstract

The aim of this study was to determine the best concentration of skim milk in maintaining the quality of Kampung chicken’s spermatozoa during storage at 5 oC. Semen was collected from five roosters which used massage method. Furthermore, semen was divided into five tubes as treatment (no extenders, extenders containing 0%, 6%, 9% and 12% skim milk concentrations), the liquid semen was stored at 5 oC. Observations of motility, viability and abnormalities of spermatozoa were carried out at 0, 12, 24, 36, dan 48 hours. The results showed that there was a very significant difference (P <0.01) among the treatment of motility, viability and abnormalities of Kampung chicken spermatozoa after dilution during storage. Storage of spermatozoa in 6% skim milk concentrate for 48 hours was resulted in motility of 49,00±1,30%, viability of 52,13±1,53% and abnormalities of 20,47±0,47%. The use of lactated ringer extenders with a concentration of 6% skim milk can generally maintain sperm quality which was better than other treatments during storage.

Abstract

Isoflavone compounds have been known to have biological effects in improving health. The purpose ofthis study was to determine the bioavailability of isoflavone in plasma and its role as lowering bloodglucose levels in hyperglycemia rats. Bioavailability of isoflavones in plasma was performed in 18 ratsstrains Spraque Dawley age 3 months with 200 g of body weight on average and were given 300 ppm oralof isoflavone single dose. The analysis of isoflavon as done at 0, 1, 6, 12, 18 and 24 hours using HPLCmethod. Study effect on blood biochemical values used 25 male rats, which was divided into 5 treatmentgroups. One group as a negative control (K1) was given aquades; positive group hyperglycemia (K2), and 3other groups (K3, K4 and K5) were a positive groups of hyperglycemia and given isoflavone different doses,i.e. 1.0, 1.5 and 2.0 mg/200g bw per oral. Blood glucose were analyzed by using glucose oxidase biosensor,while cholesterol, HDL (Hight-density lipoprotein) and triglycerides were analyzed enzymatically using aspectrophotometer. The results showed the analyzes isoflavone genistein was more efficiently absorbedthan daidzein and glisitein. Isoflavone as found at highest concentration in plasma at 6 hours after oraladministration i.e: genistein of 11,17 ppm, daidzein of 2,86 ppm and glisitein of 2,47 ppm. Theadministration isoflavones at dose 2 mg/200g bw could decrease blood glucose levels in hyperglycemiarats, and also has hipocholesterolemia effect.

Abstract

The objective of this study was to evaluate the development of ewe follicles and oocytes viabilityfollowing an intrauterine transplantation of the ovaries into pseudopregnant rabbit. The experiment wasconducted in day 1 of pseudopregnant rabbit. Following transplantation for 5, 7, and 9 days, the ovarieswere recollected. The development of follicles determined by counting the number of follicles in paraffinembedded ovaries after staining with haematoxylin-eosin (HE). The viability of oocytes was determined byslicing the ovaries. Oocytes were incubated in CO2 incubator with 5% CO2, 38°C for 24 hours. After maturation,the oocytes were stained with 2% aceto-orcein to determine the nuclear oocytes status. The result showedthat follicles were detected in all stages of their development (primordial, primary, preantral, and antralfollicle stages), but their number decreased significantly (P<0.05) 5, 7 or 9 days after transplantation,except for those at primordial stage which at day 5 post-transplantation (634.7±56.88) were not significantlydifferent to the control (683.7±61.55). After maturation, the oocytes that were able to reach the M-II phaseat day 5 and day 7 post-transplantation were 35.05% and 35.24% respectively. They were significantly(P<0.05) lower than the control (56.65%). In conclusion, the development of follicles and oocytes viability inthe ewe ovaries in pseudopregnant rabbits was still preserved during intrauterine transplantation.

Abstract

Antlers are deer’s bony organ that follows a cycle of growing, hardening, casting and regroupingwithin a certain period. The effect of consuming velvet antler from temperate origin has beenknown scientifically to have positive effect for rheumatism and metabolic disorder sickness therapy.However, the role of velvet antler originated from tropical deer has not yet been explored. Thisstudy aimed to assess the potential of the velvet antler of sambar deer (Rusa unicolor) which wasexperimentally fed to laboratory rats. The assessment was made based on the animals growth rate(i.e. femur length, weight of testicle, body eight) and physical endurance (i.e. swimming test).Laboratory rats at 21 days old were allocated into four different groups and each group consisted offive rats were fed with powder of soft and hard parts of velvet antler at dose of 0, 1, 2, and 3 g/kgbody weight, respectively. Animals were examined for eight weeks the body weight was examinedweekly and the dose at velvet antler supplement was adjusted accordingly. At the end of the studythe rat were put on endurance swimming test and then euthanized, for measurement of femurbone length and weight of testis. The results showed that there were no differences in the bodyweight. However at dose of 2 g soft part/kg BW indicating a consistently higher live weight gainsacross the observation time. Testis weight showed no significant differences between the treatments,but the length of femur bone showed a significant effect (p <0.05) with the doses level, with thehighest score being at 3 g hard part / kg BW. Physical endurance showed a significant effect (p<0.05) with the doses level, with the level of 1 g soft part/kg BW gave the best performance.
