Abstract

The objective of this study was to analyse the effect of standadized pomegranate fruit extracton liver fibrosis due to biliary obstruction.  Liver fibrosis was induced with bile duct ligation (BDL)technique.  Examinations were performed on the expressions of collagen type 1 and the degree ofliver fibrosis. Thirty two male albino rats (Rattus norvegicus), 2.5 month old, weighing 160-190grams were divided into four experimental groups.  The first group (P0) consisted of rats  whichunderwent laparotomy and treated with 2 ml of  carboxymethyl cellulose (CMC) 0.3%.  Three othergroups consisted of rats which underwent laparotomy and bile duct ligation (BDL) but receiveddifferent treatments.  Group P1 was given carboxymethyl cellulose (CMC) 0.3%, P2 was treatedwith standardized pomegranate fruit extract 150 mg/kgBW/po/day within CMC 0.3% , and P3 wasgiven ellagic acid (EA) 60 mg/kgBW/po/day of equal volume.  Treatments were administered on thesecond day after BDL for 21 days.  The livers were excised one day after the last administration.Observations were made on the expressions of collagen type 1 and the degree of liver fibrosis. Theresults showed that standardized pomegranate extract can inhibit the expression of collagen type Isignificantly in the P2 and P3 compared with P1 group (p <0.05). Treatment with standardizedpomegranate fruit extract also significantly suppressed the progression of liver fibrosis comparedto P1, eventhough there was more liver fibrosis compared to P0 (p<0,05).  The degree of liverfibrosis was not significantly different between P2 and P3, and between P3 and P1 (p>0,05).The administration of standardized pomegranate fruit extract 150 mg/kgBW/po/day  exertedantifibrotic effect by inhibiting the increase the expression of collagen type I and the degree of liverfibrosis.

Abstract

A study was carried out to identify the blood groups of dogs and to determine their distribution among dog population as well as their possible relationship with hair color, sexes and location. Fifty dogs were used in this study and they were originated from Jakarta, Bekasi and Bogor, Indonesia. The blood groups of dogs were determined by monoclonal antibodies and a positive reaction was characterized by the presence of agglutination as a result of antigen-antibody reaction. Three blood groups, 1.1B (44.0%), 1.2B (42.0%) and 1(-)B (14.0%) were detected among the 50 dogs examined. No significant relationship between certain the blood groups and the hair color, sexes and their origin were detected in this study.

Abstract

Sambung darah (Excoecaria agallocha L.) leaves are often used as traditional medicine. This experimentwas done to analyze the compounds of E. agallocha extract against  antibacterial activity either on Grampositive bacteria (Staphylococcus aureus, S.  epidermidis, and Streptococcus agalactiae), and Gram negativebacteria  (Escherichia coli and Salmonella sp.). Leaves extract of E. agallocha L. Was diluted with or madewith methanol percolation. Antibacterial activity test was performed by using paper diffusion, whileminimum concentration test was done by using dilution method. The result show that compounds ofsambung darah leaves extract were alkaloid, tanin, flavonoid, tritefenoid, glicosida and saponin. Sambungdarah extract has antibacterial activity on Gram positif bacteria (S. aureus, S. epidermidis, S agalactiae),where as it has not antibacterial activity against Gram-negative bacteria (E. coli and Salmonella sp.).

Abstract

The present study aimed to study the effect of ?maturation media? on maturation rate of goat oocytesafter in vitro maturation. Goat ovaries were collected from a slaughter house in Godean, Sleman. Immediatelyafter slaughter the ovaries were collected, rinsed with physiological NaCl three times then placed in aflask containing the NaCl solutions and hept at 36-370C before transportaion to the laboratory. Oocyteswere observed under stereo microscope and its quality was classified into A, B, and C. Oocytes in vitromaturaion (IVM) was performed in TCM-199 media suplemented with : a) 0.4 mg/ml bovine serum albumin(BSA); and b) 10% newborn calf serum (NCS) then incubated at 38.50C with 5% CO2 for 24-27 h. Followingthis, oocytes were observed under inverted microscope for first polar body extrusion, then stained withaceto-orcein in order to evaluate nuck or maturation. The nuclear maturation stages including : germinalvesicles (GV), germinal vesicles break down (GVBD), metaphase I, apaphase I, telophase I and metaphaseII, respectivelly. The overall results showed that 74-74%, 52-66.6% and 21.5-23.8% of oocytes quality A, B,and C reached maturation at metaphase II, respectivelly. There were no significant differences in oocytesmaturation using media supplemented with either BSA or NCS.

Abstract

Coxiella burnetii belongs to rikettsia group living obligate intracellularly and as the agent of zoonosisQ fever. Enrofloxacine is an antibiotic in quinolon group used to treat infection of C. burnetii in chicken, goat, calve, pig, dog, cat,  and horse. From ruminant practical experience, enrofloxacine if combined withBioATP  can enhance the enrofloxacine activity. Research for the effecivity of enrofloxacine and BioATP totreat C. burnetii has never been carried out. The research was conducted to explore effect of enrofloxacinewith supplementation BioATP against C. burnetii. Enrofloxacine pharmacokinetic study was carried outby using simental beef as an experimental animals. The effectivity of BioATP supplementation onenrofloxacine activity to treat C. burnetii was tested by using Vero cell tissue culture. The results showedthat combination of enrofloxacine and BioATP increased kinetic profile of enrofloxacine in term of onset, duration, pharmacology intensity, and bioavailaibility. Enrofloxacine had activity to treat C. burnetii withvalue of minimal inhibitory concentration (MIC) at 1-2 ppm and value of minimal bactericidal concentrationat 4 ppm. Supplementation of BioATP improved the effectivity of enrofloxacine in treating C. burnetii.
