Abstract

Abstract. Saifudin, Shafira S, Dwiranti A, Salamah A. 2021. Short Communication: Analysis of the chromosome numbers of Zinnia elegans Jacq. in single, double, and pom-pom flowers. Biodiversitas 22: 2771-2777. Zinnia elegans Jacq. is highly valued as an ornamental plant with a variety of flower colors, sizes, and shapes. Polyploidization has been reported in Z. elegans with pom-pom flowers, nevertheless, the variation in chromosome numbers of various flower shapes has yet to be investigated. This study aimed to analyze the chromosome numbers of Z. elegans Jacq. cultivar “California Giant,” “Lilliput,” and “Cactus Flowered Mix” with single, double, and pom-pom flowers to determine their variations and identify the morphology of the flowers. Chromosomes were prepared using the squashing method, and images were analyzed using the Chromosome Image Analyzing System (CHIAS) IV. The minimum of 5 slides was prepared for each flower type from each cultivar. The results show that the three cultivars are diploid plants (2n = 24) with varying flower morphology. The single and double flowers of Z. elegans “Lilliput” and the single flower of “Cactus Flowered Mix” showed no variation in chromosome numbers. In contrast, chromosome number variation was found in the pom-pom flower of Z. elegans “California Giant” (2n = 22, 24, 48) and the double flower of “Cactus Flowered Mix” (2n = 9, 13, 15, 24). Two cultivars, Z. elegans “California Giant” and Z. elegans “Cactus Flowered Mix,” were successfully analyzed using CHIAS IV. Statistical analysis using a t-test (? = 0.05) showed that the total chromosome length of Z. elegans “California Giant” (2n = 24) was significantly greater than that of Z. elegans “Cactus Flowered Mix” (2n = 24). Chromosome satellites were found in both cultivars.

Abstract

Abstract. Abinawanto A, Lestari S, Bowolaksono A, Dwiranti A, Lestari R, Gustiano R, Kristanto AH. 2023. Title The effect of different honey concentrations on the ultrastructure profile of spermatozoa in Dewa Mahseer (Neolissochilus soro). Biodiversitas 24: 1025-1031. Dewa Mahseer (Neolissochilus soro) is one of Indonesia's endemic freshwater fish at risk of population decline due to water pollution and asynchronous maturation of gonads. One of the efforts to conserve this species is preserving their spermatozoa using honey as a protector at certain temperatures. Therefore, this study aims to evaluate the effect of multiflora honey concentrations on ultrastructure spermatozoa profile of Dewa Mahseer. The temperature was maintained at 4°C and stored for 48 hours, followed by the cement dilution (1:10) in a stock solution containing fish ringer and honey. Transmission electron microscopy (TEM) examined the spermatozoa profiles at a magnification of ×1,500. Furthermore, ANOVA one-way analysis of variance was conducted to test the effect of head length, width, and area of spermatozoa on honey. The results showed a statistically significant difference in head width (F=13.929, p=0.001). Also, there is a difference in area between honey spermatozoa, fresh spermatozoa, and without honey at a cold of 4°C. The fresh spermatozoa stored at cold temperatures without honey observed considerable membrane damage. The novel aspect is an investigation of the effect of honey as a natural protector in the storage of Neolissochilus spermatozoa at 4°C for 48 hours. This study can be applied as a guide for the 48 hours of spermatozoa in a cold temperature in fish farming.

Abstract

Abstract. Syahrani L, Dwiranti A, Asih PBS, Rozi IE, Permana DH, Bowolaksono A, Syafruddin D. 2025. Susceptibility of the Punctulatus group (Diptera: Culicidae) to insecticide carbamate in Papua, Indonesia. Biodiversitas 26: 1376-1383. Insecticide Treated Nets (ITNs) and Indoor Residual Spraying (IRS) are the primary vector control tools recommended for reducing malaria transmission in endemic areas. However, the emergence and spread of pyrethroid resistance among the Anopheles punctulatus group poses a significant challenge for malaria vector control in Papua, Indonesia. While carbamate-based IRS is a potential strategy to mitigate resistance, its effectiveness within malaria control programs requires continuous monitoring. This study assessed the susceptibility of the Punctulatus group to bendiocarb (0.1%) and analyzed the underlying resistance mechanisms. Bioassays were conducted in three sub-districts of Keerom District, Papua. Additionally, biochemical assays were performed to measure the activity of detoxifying enzymes, including esterases, Glutathione S-Transferases (GSTs), and acetylcholinesterase. Ace-1 fragments were amplified using Polymerase Chain Reaction (PCR), followed by sequencing to detect nucleotide substitution at codon 119. Results indicated full susceptibility (100%) of the Punctulatus group to bendiocarb at all study sites. Compared to unexposed controls, bendiocarb-exposed populations exhibited significantly higher GST (p: 1.403e-06), elevated esterase activity (p: 0.247), and reduced acetylcholinesterase inhibition (p: 0.002). PCR sequencing of 29 samples confirmed that all carried the wild-type Ace-1 allele (GGG, GGC, and GGT). These findings provide crucial evidence for malaria control programs in selecting appropriate vector control products and managing insecticide resistance. Despite full susceptibility to bendiocarb, elevated detoxification enzyme activity suggests potential early-stage metabolic resistance. Further molecular studies are recommended to investigate metabolic resistance mechanisms, along with rigorous monitoring using bioassays, biochemical assays, and molecular tools to track insecticide susceptibility trends over time.

Abstract

Our study aims to scrutinize the hormonal and laboratory profile of women with PCOS visiting a fertility center in Indonesia. A computerized database of 120 PCOS women and 120 controls was retrospectively reviewed and analyzed. According to analysis, the concentration of AMH and basal LH were significantly more elevated in the PCOS group than in the control group (6.77 vs. 2.05, P<0.001; and 6.8 vs. 5.3, P<0.001, respectively). The level of basal FSH was significantly lower in the PCOS group (5.83 vs. 7.86; P<0.001). After an IVF cycle, PCOS women had significantly higher retrieved oocytes and mature oocytes compared to controls (18 vs. 8, P<0.001; 12 vs. 6, P<0.001). The number of fertilizations, cleavages, and top-quality cleavages were also higher in the PCOS group than in the control group (10 vs. 4, P<0.001; 10 vs. 4, P<0.001; 4 vs. 2, P<0.001). At the blastocyst stage, patients with PCOS had a higher number of blastocysts and top-quality blastocysts than control (9 vs. 4, P<0.001); 4 vs. 1, P<0.01). However, the clinical pregnancy was significantly higher in the control group than in the PCOS group (52.6% vs. 16.7%; P = 0.045).

Abstract

Cancer is a disease characterized by uncontrolled cell growth, often leading to death. The high incidence of cancer has led to continued exploration of potential therapeutics, particularly those derived from natural products. One of the potential natural products is sericin, which has been shown to have anticancer effects against several types of cancer through various in vitro studies. Previous studies generally indicate that sericin's anticancer effects are reflected in its ability to reduce cancer cell viability and induce apoptosis by regulating the expression levels of its related proteins. Studies have suggested that the expression levels of apoptosis-related proteins are known to influence the metastatic potential of cancer cells. Although the specific antimetastatic effects of sericin, an industrial waste by-product, remain largely unexplored, this review comprehensively evaluates its role as an anticancer agent and its underlying cellular mechanisms. Sericin modulates the apoptotic pathway by shifting the Bax/Bcl-2 ratio, specifically through upregulating the pro-apoptotic Bax and suppressing the anti-apoptotic Bcl-2, leading to the sequential activation of pro-caspase-9 and downstream effector caspases. Beyond its established pro-apoptotic activity, we highlight the ability of sericin to restore the apoptotic threshold, which may effectively limit the survival of circulating tumour cells and their capacity to metastasize. Consequently, while sericin offers a promising frontier for future therapeutic investigation, realizing this remarkable potential requires addressing critical experimental gaps, specifically the current lack of dedicated in vitro migration/ invasion assays and in vivo metastasis models to evaluate sericin impact on disrupting the metastatic cascade.
