Abstract

High mortality in the larvae stage was a major problem in fish hatchery efforts. Efforts that can be done through the hormonal stimulation with the utilization of thyroxine hormone. The purposes of this research was to determine the effect of immersing eggs in a solvent of thyroxine hormone at different doses on hatchability, growth and survival nilem fish larvae (Osteochilus hasseltii). The research method use a complete randomized design (RAL). Treatments in this research were A (control), B (0.05 mg / L), C (0.10 mg / L), D (0.15 mg / L). The research variables observed were hatchery, an absolute long-term growth, relative long-term growth and the survival. The results of this research showed that the soaking of eggs in thyroxine hormone at different doses had no significant effected on hatchability (P> 0.05) but had a significant effected on growth, and survival of nilem fish larvae (P

Abstract

The administration of chromium picolinate (Cr-Pic) is expected to positively influence the growth of eel (Anguilla bicolor) and the expression of the growth related gene. The purpose of this research was to study the effect of chromium picolinate (Cr-Pic) on the growth performance and expression of growth hormone encoding gene of eel (Anguilla bicolor), a species of fish that is in the process of domestication. The experiment consisted of four treatments: without Cr-Pic (P0), with Cr-Pic of 1 mg / kg of feed (P1), 2 mg / kg of feed (P2), and 3 mg / kg of feed (P3). Eel growth performance was analyzed based on absolute and daily growth rates. Observation of the level of growth hormone (GH) gene expression was performed using real-time RT-PCR method. The results showed that administration of Cr-Pic did not significantly increase eel growth rate. The expression level of growth hormone genes in eel fed 2 and 3 mg Cr-Pic / kg was higher than the expression level of this gene in eel fed without Cr-Pic. The results showed that the level of growth hormone expression did not appear to be related to growth rate of the eel.

Abstract

Aeromonas hydrophyla is a gram negative bacteria, which often infects tilapia and causes internal damage to the liver, spleen and kidneys. Tilapia Sultana, Nirwana and Larasati are superior strains that have fast growth and are resistant to disease. This study was conducted to observe and compare the physiological response of the three strains to Aeromonas hydrophyla infection through blood profiles. The blood profile can describe the health status of the fish. The parameters of the blood profile observed were the number of erythrocytes, hemoglobin, hematocrit, blood glucose, lymphocytes and monocytes. The results showed that the erythrocytes, monocytes and polymorphonuclear counts of the three strains were significantly different, while for hemoglobin, hematocrit, glucose, and lymphocytes were not significantly different.

Abstract

Ikan guppy HB Gold (Poecilia reticulata) merupakan salah satu strain guppy yang mudah untuk dibudidayakan serta memiliki permintaan yang tinggi. Tujuan penelitian ini adalah untuk menambah wawasan tentang cara budidaya ikan guppy HB Gold (Poecilia reticulata) skala rumah tangga dengan substrat dasar yang berbeda, dari penelitian ini diharapkan dapat diaplikasikan oleh masyarakat umum untuk memulai usaha perikanan dengan modal kecil. Metode yang digunakan menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan 2 ulangan. Hasil penelitian dianalisis secara deskriptif serta di bandingkan dengan literatur terdahulu. Hasil penelitian menunjukkan bahwa semua perlakuan mampu menghasilkan benih ikan dan tumbuh dengan baik. Dimana P1A menetaskan 27 larva; P2A 0 larva; P3A 26 larva, sedangkan pada P1B dan P2B masing-masing 4 larva dan P3B sebanyak 21 larva. Parameter kualitas air diperoleh suhu akuarium pemijahan dan pembesaran antara 25-26°C, dan pH antarar 6,5-8. Maka dari itu dapat diketahui bahwa budidaya ikan guppy skala rumah tangga dapat diterapkan.

Abstract

The presence of vibrio bacteria in the breeding pond is one of the important factors that affect the activities of vaname shrimp breeding. Vibrio sp. bacteria in waters can cause losses if the number is above the threshold that should be. The high abundance of Vibrio sp. bacteria in the cultivation environment is an indicator that shows the potential for disease that can reduce the growth rate and survival of shrimp. The purpose of the study was to determine the abundance and dominance of vibrio bacteria in vaname shrimp breeding ponds using the TPC (Total Plate Count) calculation technique. The methods used include bacterial sampling, preparation of tools and materials, making bacterial culture media (NA and TCBS), dilution, bacterial inoculation, and calculating the number of bacterial colonies. The results of the calculation of bacterial abundance showed that there were 9 breeding ponds with different abundances. The highest abundance was found in pond BV1 with a total bacterial value of 9.6 x 105 CFU/ml. Meanwhile, the highest vibrio dominance was found in pond BV4 with a percentage of 43.69%.

Abstract

Tilapia is a commodity cultivated in the rice-fish farming area of Panembangan, Banyumas. Information related to the presence of potential bacteria in the intestines of fish cultivated in rice-fish areas is still low, especially proteolytic bacteria. The purpose of this study was to determine the proportion and activity index of proteolytic bacteria in the intestines of fish. This study used an observation method with a purposive sampling technique. Proteolytic bacteria were isolated from the intestines of tilapia in blocks A and E. The bacteria that were successfully isolated were grown on 2% skim milk agar media with an incubation time of 48 hours and observed at 24-hour intervals. Based on the results of the study, the proportion of proteolytic bacteria in the intestines of tilapia from both blocks was 28% in block A and 44% in block E, respectively. The proteolytic activity index of both blocks had an average of 0.48 for block A and 0.75 for block E. The highest proteolytic activity index was from block A, namely 0.83, and in block E 3.0. Meanwhile, the smallest proteolytic index is 0.17 block E and 0.22 block A.

Abstract

Salt and herbal supplementations in fish feed are expected to promote aquaculture productivity, including for slow-growing fish such as Bonylip barb (Osteochilus vittatus). The objective of this study was to evaluate the growth performance and occurrence of Aeromonas spp. in the intestine of Bonylip barb fed a diet supplemented with salt and herbs. This experiment was conducted using a completely randomized design with four treatments: 1) control, 2) salt 3% supplementation, 3) herbal supplementation, and 4) Salt 3% and herbal supplementation. Growth performance evaluation included weight gain, relative growth rate, and specific growth rate after 30 and 60 days of rearing.  Bacterial samples were isolated on trypticase soy agar (TSA) and glutamate starch phenol red agar (GSP) media. Yellow colonies on GSP were counted and confirmed as Aeromonas spp. via molecular identification based on 16S rDNA sequence. Results showed that herbal supplementation, either alone or in combination with 3% salt, insignificantly increased the fish growth performance in the short-term (30 days), but tended to decrease it over the long-term (60 days). Salt supplementation consistently decreased fish growth performance but improved fish survival rates. Aeromonas isolates comprised more than 60 % of the culturable intestinal bacteria in all treatments. Furthermore, herbal supplementation appeared to increase the percentage of Aeromonas spp. in the intestine, with identified species including Aeromonas veronii, Aeromonas caviae, and Aeromonas jandaei.

Abstract

Ikan Kerapu Cantang merupakan hasil pesilangan dari ikan Kerapu Macan betina dan ikan Kerapu Kertang jantan. Ikan Kerapu Cantang merupakan salah satu komoditas perikanan budidaya yang memiliki nilai ekonomis tinggi. Penelitian ini bertujuan untuk mengetahui keberadaan bakteri selulolitik serta aktivitas bakteri selulolitik pada saluran pencernaan ikan Kerapu Cantang. Metode yang digunakan pada penelitian ini adalah metode observasi dengan pengambilan sampel secara purpose random sampling. Variabel utama yang diamati pada penelitian ini yaitu mengamati proporsi bakteri selulolitik dan aktivitas bakteri selulolitik pada saluran pencernaan ikan Kerapu Cantang. Untuk variabel pendukung pada penilitian ini yaitu kelimpahan bakteri, uji gram KOH, proporsi Bacillus Serta pewarnaan gram. Keberadaan bakteri Selulolitik pada saluran pencernaan ikan Kerapu Cantang pada penelitian ini ditunjukkan dengan adanya zona bening di sekeliling koloni bakteri yang ditumbuhkan pada media spesifik ( CMC 1%). Hasil penelitian ini untuk proporsi keberadaan bakteri selulolitik pada bagian anterior 49,33% middle 38,66% dan posterior 28%. Aktivitas bakteri selulolitik dari penelitian ini menunjukan hasil dengan indeks hidrolisis pada bagian anterior sebesar 0,14 – 1,4, midddle 0,1 – 3,3 dan posterior 0,2 – 1,6.

Abstract

Guppy fish (Poecilia reticulata) is a type of ornamental fish that is in great demand, because of its small size and beautiful color. The color of ornamental fish will generally fade, due to a lack of carotenoids in their feed. The purpose of this study was to determine the effect of giving Tubifex sp. in commercial feed to increase the color intensity of guppy fish and the best dose of Tubifex sp. The test fish used were male guppy fish strain HB Red. This study used a completely randomized design (CRD) method with five treatments and three replications, that is commercial feed with the addition of Tubifex sp. with doses of 0% (P1), 2% (P2), 4% (P3), 6% (P4), and 8% (P5). Parameters observed were color intensity (chroma value), survival, and water quality. Addition of Tubifex sp. in commercial feed gave the effect with the highest yield on P5 at a dose of 8%, with an increase in color intensity (chroma value) of 4,21±0,25d. At P1 it gave an increase of 1,19±0,02a, P2 was 1,34±0,04a, P3 was 1,81±0,21b, and P4 was 2,88±0,18c. So, the best treatment is P5 (8%). Survival showed the results were not significantly different, that is 100%. Water quality is included in the tolerance limit of fish with the results of temperature 26,6 – 27,4°C, pH 7,4 – 7,9, and DO 6,2 – 7,7 mg/L.

Abstract

Amylolytic bacteria play an important role in the ecosystem, especially as probiotic and bioremediation agents in cultivation, as examples can be found in Mina Padi culture. The purpose of this study was to determine the amylolytic bacteria in the waters of the Mina Padi pond. Bacterial isolation began with bacterial sampling, inoculation and isolation of bacteria, calculation of the total abundance of bacteria, observation of bacterial morphology and bacterial purification, and isolation of amylolytic bacteria. The results of the isolation of amylolytic bacteria obtained 3 bacterial isolates capable of producing amylolytic enzymes, namely BA5, BA6, and BA7. The highest index of amylolytic activity was obtained by isolates of BA6 with a medium category of 2.3 cm, and the lowest index was obtained by BA5 with a weak category of 0.3 cm. The average bacterial abundance from each dilution was 2.5 x 103 CFU/mL.
