Abstract

Trichodina sp. merupakan ektoparasit yang umum ditemukan menginfeksi ikan budidaya, termasuk ikan nila (Oreochromis niloticus). Ekstrak daun sirih mempunyai potensi untuk digunakan dalam pengendalian Trichodina sp. pada benih ikan nila. Penelitian ini bertujuan untuk mengetahui dosis ekstrak daun sirih (EDS) yang tidak menyebabkan mortalitas benih ikan nila, mengetahui pengaruh EDS terhadap prevalensi, intensitas, dan kelimpahan Trichodina sp., dan menentukan dosis optimal EDS untuk mengendalikan Trichodina sp. Hasil pengamatan menunjukkan dosis aman EDS untuk perendaman selama 2 jam adalah maksimal 200 mg/L. Berdasarkan hasil tersebut maka EDS dengan dosis 0 mg/L (kontrol), 50 mg/L, 100 mg/L, dan 200 mg/L digunakan untuk penelitian utama. Metode experimental berdasarkan Rancangan Acak Lengkap digunakan dengan 4 perlakuan dan 10 ulangan. Hasil penelitian menunjukkan bahwa perlakuan dosis 100 mg/L dan 200 mg/L menurunkan prevalensi parasit hanya pada insang dan tidak pada permukaan tubuh. Selain itu, kedua dosis tersebut secara signifikan menurunkan intensitas dan kelimpahan Trichodina sp. di permukaan tubuh tetapi tidak dapat secara total membebaskan permukaan tubuh dari parasit tersebut. Berdasarkan hasil tersebut, dosis optimal EDS untuk mengendalikan Trichodina sp. pada benih ikan nila belum dapat ditentukan dalam penelitian ini.

Abstract

Keberadaan bakteri proteolitik pada komoditas akuakultur penting untuk dipelajari, salah satunya terkait dengan praktek budidaya ikan skala kecil di daerah pedesaan. Penelitian ini bertujuan untuk mengetahui keberadaan dan melakukan identifikasi secara molekuler bakteri proteolitik yang diisolasi dari saluran pencernaan ikan nila (Oreochromis niloticus). Sampel ikan nila diambil dari tiga unit kegiatan akuakultur yang menggunakan pakan berbeda di Kabupaten Banyumas yaitu dari Desa Pandak (dengan probiotik, pakan pellet), Desa Beji (tanpa probiotik, pakan tumbuhan) dan Desa Tambaksogra (dengan probiotik, kombinasi pakan pellet dan tumbuhan). Jumlah bakteri, proporsi bakteri proteolitik, dan indeks aktifitas proteolitik diamati dari usus bagian anterior, middle, dan posterior. Sampel isolat bakteri  proteolitik dikelompokkan berdasarkan hasil analisis restriksi 16S rDNA menggunakan software PhyElp. Bakteri dari setiap kelompok diidentifikasi berdasarkan sekuen gen 16S rDNA dengan menggunakan analisis BLAST dan analisis filogenetik. Jumlah bakteri di saluran pencernaan ikan nila dari tiga tempat relatif sama dan cenderung meningkat ke arah posterior. Hasil penelitian menunjukkan ikan nila dari Desa Pandak memiliki proporsi bakteri proteolitik yang lebih tinggi dibandingkan sampel ikan dari Desa Beji dan Tambaksogra. Nilai aktivitas bakteri proteolitik saluran pencernaan ikan nila dari Desa Pandak relatif lebih tinggi dibandingkan dari dua desa lainnya. Bakteri proteolitik dari saluran pencernaan ikan nila dapat dikelompokkan menjadi 15 kelompok berdasarkan polimorfisme hasil digesti fragment gen 16S rDNA. Sampel dari 15 kelompok tersebut memiliki sekuen 16S rDNA yang mirip dengan Pseudomonas aeruginosa (4 isolat), Plesiomonas shigelloides, Escherichia coli, Aeromonas veronii, Klebsiella variicola, Enterobacter ludwigii, Enterobacter hormaechei (2 isolat), Enterobacter cloacae, Bacillus subtilis, Bacillus amyloliquefaciens dan Bacillus sp.

Abstract

The purpose of this research was to determine the quality of spermatozoa of Nilem Fish (Osteochillus hasselti) after storage in extender palm juice, milk and egg yolk with different dilution ratios. The treatment given was in the form of different dilution ratios between sperm and extender palm juice, milk and egg yolk, namely P1: dilution ratio 1:10, P2: dilution ratio 1:15 and P3: dilution ratio 1:20. The data obtained were analyzed by means of variance (ANOVA). Data that showed a significant effect were continued with further Duncan testing. The results showed that the treatment had a significant effect on motility and viability, but did not have a significant effect on fertility and hatchability.

Abstract

The purpose of this study was to determine the effect of adding honey with different concentrations in the extender on the percentage of fertility and hatchability of nilem fish (Osteochillus vittatus) eggs fertilized by stored sperm. This study used a completely randomized design (CRD) with a factorial pattern of 2 x 4 with 4 replications. The type of extender (Factor A) consisted of egg yolk milk and coconut water, while the concentration of honey (Factor B) used was 0%, 3%, 6%, and 9%. Observations on the percentage of fertility and egg hatchability were carried out to determine the sperm performance of nilem fish. Data analysis was carried out statistically using statistical analysis of variance (ANOVA) and further tested using multiple distance tests (Duncan). Data analysis showed that there was an interaction between the two factors that gave a significant effect on the percentage of fertility and egg hatchability. The combination of milk and egg yolk extender with 6% honey concentration gave the highest yield of 61.75% for fertility and 23.08% for temporary hatchability, for the lowest percentage of fertility was 4% for coconut water without honey and the lowest percentage was for hatchability egg yolk milk extender without honey was 7.51%.

Abstract

Hydrolytic bacteria are bacteria that play an important role in helping the digestive process of fish. This study aims to determine the hydrolytic activity of bacteria in the digestive tract of Nilem fish seen from amylolytic, proteolytic, lipolytic and cellulitic indices. The method used is exploratory and data analysis using parametric and non-parametric to explain the research data. Exploration carried out in this study included the total number of bacteria, proportion of hydrolytic bacteria, activity index of hydrolytic bacteria and molecular identification of bacteria with the best activity index. Fish were taken at random from fish farming ponds in Beji and Singasari with three fish each. The number of bacteria found an increasing pattern to the posterior intestine of fish from Beji and a pattern that varied between parts of the intestine of fish from Singasari. Hydrolytic bacteria were isolated from each part of the intestine from both locations which showed a decreasing pattern in each type of hydrolytic and intestinal tracts of fish from Singasari and varied patterns in the intestines of fish from Beji. The hydrolytic activity index showed a stable average value in each part of the intestine and the sampling location. Based on the best activity index, seven isolates were identified from three different genera, namely 1) Bacillus subtilis, 2) Enterobacter mori, Enterobacter cloacae, and 3) Aeromonas hydrophila (3), Aeromonas veronii. The bacteria obtained are classified as non-pathogenic bacteria that have potential in the field of aquaculture and normal pathogenic bacteria in the fish intestine

Abstract

Durian leaves have been used extensively to aid in the anesthetic process in various types of fish. The purpose of this study was to determine the immotilization capacity and stress response and survival of Tilapia under anesthesia. The RAL study design (4x4) was carried out with the durian leaf infusion treatment at concentrations of 2%, 4%, 6% and 8%. The fastest induction and sedation times were at 8% and 6% concentrations, which were 12.33 minutes and 13.35 minutes, respectively. The highest survival rate was at a concentration of 2% which reached 100%. Blood glucose levels increased due to anesthesia, ranging from 67.75 to 134.25 mg/dL

Abstract

Information on the existence of Parathelpusa spp crabs in the Banjaran River, Banyumas Regency is still very limited, so research is needed on this matter. The aim of this study was to identify and describe species richness, sex proportion, distribution, and abundance. Sampling was taken by Cluster Random Sampling, from 5 selected stations, repeated 7 times, an interval of 2 weeks, in the period August-November 2021. Samples were taken using a hand net, fresh and brought to the laboratory for identification. The physicochemical conditions measured were air temperature, water temperature, pH, dissolved oxygen and current velocity. The richness and abundance of Parathelpusa spp species obtained was calculated based on the number of species and individuals obtained, while their presence at each station became the basis for calculating the longitudinal distribution. Correlation between physico-chemical factors of the river with species richness and abundance was analyzed using Principal Component Analysis (PCA). During the study, 158 crabs were obtained consisting of Parathelpusa convexa and P. bogorensis. Both species were found at all sampling sites and P. convexa dominated the catch. Overall and in several locations, the number of male crabs caught was higher. The existence of each species in the Banjaran River is influenced by different physicochemical factors. P. convexa is affected by bottom substrate, air temperature, and current speed, while P. Bogorensi is affected by water depth, bottom substrate, and DO. These results can be used as a reference in selecting habitats to find P. convexa and P. bogorensis in river waters.

Abstract

The aims were to do an ectoparasite and pathogenic bacteria inventory of Bonylip barb reared in Banyumas district. Total of 118 fish were randomly taken from Singasari (30 individual), Pabuaran (30 individual), Beji (30 individual) and Jipang (28 individual) villages. This work observed ectoparasite species, prevalence, and intensity as well as pathogenic bacteria species. Ectoparasites were identified based on morphological characteristics observed microscopically and their prevalence and intensity were calculated. Bacterial pathogen was isolated from fish samples showing symptoms of bacterial disease and identified molecularly based on 16s rDNA sequence using BLAST dan phylogenetic analysis. Results showed Trichodina sp. and Monogeneans were found infected fish samples from all location with prevalence was 70-100%. Ichthyophthirius multifiliis was found from all location except Singasari village.  Some of fish samples showed fin necrosis and five bacterial isolates were obtained. 16s rDNA marked gene was successfully amplified and sequenced. Based on sequence analysis, five isolates could be clustered into two groups, the first was found only in Singasari village and the second was found in Singasari and Beji villages.  Similarity value of 16s rDNA gene sequence these two groups was 99,8%. Result of BLAST and phylogenetic analysis showed that the bacterial pathogens were identified as group of Aeromonas hydrophila complex.

Abstract

The administration of chromium picolinate (Cr-Pic) is expected to positively influence the growth of eel (Anguilla bicolor) and the expression of the growth related gene. The purpose of this research was to study the effect of chromium picolinate (Cr-Pic) on the growth performance and expression of growth hormone encoding gene of eel (Anguilla bicolor), a species of fish that is in the process of domestication. The experiment consisted of four treatments: without Cr-Pic (P0), with Cr-Pic of 1 mg / kg of feed (P1), 2 mg / kg of feed (P2), and 3 mg / kg of feed (P3). Eel growth performance was analyzed based on absolute and daily growth rates. Observation of the level of growth hormone (GH) gene expression was performed using real-time RT-PCR method. The results showed that administration of Cr-Pic did not significantly increase eel growth rate. The expression level of growth hormone genes in eel fed 2 and 3 mg Cr-Pic / kg was higher than the expression level of this gene in eel fed without Cr-Pic. The results showed that the level of growth hormone expression did not appear to be related to growth rate of the eel.

Abstract

Penelitian ini bertujuan untuk mengetahui pertumbuhan dan kelangsungan hidup ikan Gurame(Osphronemus gouramy Lac.) yang diberi pakan komersial dengan penambahan enzim papain.Penelitian dilaksanakan secara eksperimental berdasarkan RAL dengan 4 perlakuan dan 3ulangan. Perlakuan yang diteliti adalah penambahan dosis enzim papain yang berbeda yaitu 0%(kontrol), 1,5%, 2,0%, dan 2,5%. Variabel yang diteliti adalah nilai pertumbuhan mutlak dankelangsungan hidup. Hasil penelitian menunjukkan ada perbedaan yang nyata terhadap keduavariabel yang diteliti. Penambahan enzim papain sebanyak 2,0% secara nyata menghasilkanpertumbuhan mutlak terbaik ikan Gurame yaitu sebesar 3,06±0,62 g. Nilai kelangsungan hidupperlakuan kontrol, 2,0% dan 2,5% tergolong baik dan berkisar 70,00-76,67%. Namun, nilaikelangsungan hidup pada perlakuan penambahan enzim papain 1,5% tergolong sedang yaitusebesar 33,33±15,28%. Enzim papain sebanyak 2,0% dapat ditambahkan ke dalam pakankomersial untuk meningkatkan pertumbuhan ikan Gurame.
