Abstract

Tanaman padi yang menghasilkan beras merupakan komoditas utama di Kecamatan Ledokombo. Kendala yang dihadapi petani dalam budidaya termasuk cekaman abiotik berupa angin kencang pada musim penghujan. Hal ini mengakibatkan tanaman padi lokal dengan postur tinggi rentan mengalami rebah. Untuk mengatasi masalah ini, diperlukan pelatihan terkait penggunaan zat pengatur tumbuh guna mengendalikan pertumbuhan tanaman yang tinggi. Paclobutrazol (PBZ), sebagai salah satu jenis zat pengatur tumbuh, berperan penting dalam mengurangi tinggi tanaman. Pelatihan dilakukan langsung di lahan petani dengan menggunakan lima varietas padi yang berbeda. Hasil aplikasi menunjukkan bahwa padi yang ditanam oleh petani menjadi lebih pendek dibandingkan yang tidak diberi perlakuan serupa.

Abstract

Black rice (Oryza sativa L. indica) is a pigmented rice plant that has high antioxidant content. Drought is an abiotic stress that can inhibit the growth and productivity of rice plants. Planting of local black rice with several drought treatments using PEG 6000 was observed in this study. The aim of this research is to determine the morphological and physiological response of local black rice plants in Purbalingga and Purwokerto at various levels of drought stress. This research used a Completely Randomized Design (CRD) with 2 factors. The first factor is 2 local black rice plants, namely Purbalingga and Purwokerto. The second factor was drought stress treatment via PEG 6000 with 4 treatments, namely 0% PEG (control), 5%, 10%, and 15%. The results showed that when applying 15% PEG, local black rice in Purbalingga and Purwokerto experienced a decrease in plant height and number of leaves but increased root length. Based on the physiological response, when addition 15% PEG there was a decrease in the amount of chlorophyll and an increase in the content of H2O2 and anthocyanins in both Purbalingga and Purwokerto local black rice.   Keywords: Black Rice, Drought Stress, Morphological Response, Physiological Response.

Abstract

Tomato (Solanum lycopersicum L.) is a horticultural commodity which the quality and the yield can be enhanced through the application of the Dutch bucket hydroponic system, along with KH₂PO₄ fertilization and gibberellin hormone treatments. This study aimed to evaluate the response of tomato plants in terms of productivity and quality under these treatments. The experiment was conducted in a greenhouse using factorial randomized complete block design with three replications. The first factor was KH₂PO₄ concentration at three levels: 0 ppm, 150 ppm, and 250 ppm; the second factor was gibberellin concentration at three levels: 0 ppm, 50 ppm, and 100 ppm. Data were analyzed using ANOVA and DMRT on parameters including number of flowers, number of fruits per plant, fruit diameter, average fruit weight, total fruit weight per plant, and total soluble solids value. The results indicated that the interaction of 150 ppm KH₂PO₄ and 50 ppm gibberellin produced optimal values for fruit weight per fruit of 41.32 g, total fruit weight per plant of 398.67 g, and total soluble solids of 6.28 °Bx. Furthermore, either individual effects of KH₂PO₄ or gibberellin concentrations significantly influenced all observed variables except on fruit diameter. Combination KH2PO4 150 ppm and gibbereilin 50 ppm is recommended for enhancing tomato cultivation performance under the Dutch bucket hydroponic system.   ABSTRAK Tomat (Solanum lycopersicum L.) merupakan komoditas hortikultura yang kualitas dan produksinya dapat ditingkatkan melalui penerapan sistem hidroponik Dutch bucket disertai penggunaan pupuk KH₂PO₄ dan hormon giberelin. Penelitian ini bertujuan untuk mengetahui respons tanaman tomat khususnya pada produktivitas dan kualitas terkait perlakuan tersebut. Penelitian dilakukan di dalam greenhouse menggunakan rancangan acak kelompok (RAK) faktorial dengan 3 ulangan. Faktor pertama adalah konsentrasi KH₂PO₄ dengan tiga taraf yaitu 0 ppm, 150 ppm, dan 250 ppm, faktor kedua adalah konsentrasi giberelin dengan tiga taraf yaitu 0 ppm, 50 ppm, dan 100 ppm. Data dianalisis menggunakan ANOVA dan DMRT dengan variabel pengamatan meliputi jumlah bunga, jumlah buah per tanaman, diameter buah, bobot per buah, bobot buah per tanaman, dan total padatan terlarut. Hasil penelitian menunjukkan interaksi antara KH₂PO₄ 150 ppm dan giberelin 50 ppm memberikan hasil terbaik pada variabel bobot per buah sebesar 41,32 g, bobot buah per tanaman sebesar 398,67 g, dan total padatan terlarut sebesar 6,28 °Bx. Selanjutnya, pengaruh faktor tunggal konsentrasi KH₂PO₄ maupun konsentrasi giberelin memberikan pengaruh pada seluruh variabel yang diamati kecuali diameter buah. Penggunaan KH2PO4 150 ppm dan giberelin 50 ppm dapat menjadi rekomendasi terbaik untuk diaplikasikan pada budidaya tomat menggunakan sistem Dutch bucket.   Kata kunci: Dutch bucket, giberelin, hidroponik, KH2PO4, tomat

Abstract

Padi merupakan komoditas pertanian utama di Kecamatan Ledokombo. Namun, dalam praktik budidayanya, petani sering menghadapi tantangan cekaman abiotik, terutama angin kencang saat musim hujan. Kondisi ini menyebabkan varietas padi lokal yang memiliki postur tinggi menjadi rentan mengalami kerebahan, yang berdampak pada penurunan hasil. Untuk mengatasi permasalahan tersebut, diperlukan pelatihan penggunaan zat pengatur tumbuh (ZPT) sebagai upaya pengendalian pertumbuhan tanaman. Salah satu ZPT yang efektif adalah Paclobutrazol (PBZ), yang berfungsi menekan pertumbuhan tinggi tanaman. Pelatihan dilaksanakan langsung di lahan pertanian dengan menggunakan lima varietas padi berbeda. Hasil aplikasi menunjukkan bahwa tanaman yang diberi PBZ tumbuh lebih pendek dibandingkan tanaman yang tidak mendapatkan perlakuan, menunjukkan potensi PBZ dalam mengurangi risiko kerebahan.

Abstract

AbstrakTanaman krisan varietas Suciono merupakan tanaman hias yang potensial dikembangkan dan diminati oleh penduduk dunia, yang ditunjukkan dengan permintaan pasar terhadap tanaman krisan meningkat sepanjang tahun. Hal ini menyebabkan perbanyakan krisan secara in vitro sangat penting untuk dilakukan. Penelitian ini bertujuan untuk mengetahui pengaruh Indole Acetic Acid (IAA) dan Benzly Amino Purine (BAP) terhadap multiplikasi tunas krisan varietas Suciono. Desain penelitian ini menggunakan Rancangan Acak Lengkap dengan satu faktor yakni kombinasi hormon IAA dan BAP yang 9 kombinasi, yakni 0 mg/L +  1,5 mg/L; 0 mg/L + 2 mg/L; 0 mg/L + 2,5 mg/L; 0,5 mg/L + 1,5 mg/L; 0,5 mg/L + 2 mg/L; 0,5 mg/L + 2,5 mg/L; 1 mg/L + 1,5 mg/L; 1 mg/L + 2 mg/L; dan 1 mg/L + 2,5 mg/L dimana setiap kombinasi diulang sebanyak 3 kali. Variabel yang diamati meliputi analisis histologis kalus, waktu muncul tunas, jumlah tunas, tinggi tunas, dan jumlah daun. Hasil analisis ragam atau ANOVA menunjukkan bahwa kombinasi IAA 0 mg/L dan BAP 2 mg/L merupakan kombinasi konsentrasi terbaik untuk multiplikasi tunas. Penambahan BAP 2 mg/L mampu menghasilkan persentase muncul tunas. 100%, jumlah tunas 3 per-eksplan, tinggi tunas 7 cm, dan jumlah daun sebanyak 16 helai.AbstractThe variety Suciono of chrysanthemum is a potential ornamental plant that is in demand by the world's population, as indicated by the increasing market demand for chrysanthemum plants throughout the year. This  has led to the importance of in vitro propagation of chrysanthemum. The aim of the this study is to determine the effect of Indole Acetic Acid (IAA) and Benzly Amino Purine (BAP) on the multiplication of chrysanthemum shoots of the Suciono variety. The design of this study used nodus explants, treated with a combination of IAA hormones and BAP. It is make 9 combinations, and then the combinations of hormones is 0 mg/L + 1,5 mg/L; 0 mg/L + 2 mg/L; 0 mg/L + 2,5 mg/L; 0,5 mg/L + 1,5 mg/L; 0,5 mg/L + 2 mg/L; 0,5 mg/L + 2,5 mg/L; 1 mg/L + 1,5 mg/L; 1 mg/L + 2 mg/L; and 1 mg/L + 2,5 mg/L.  The observed variables included histological analysis of callus, time of shoot emergence,  number of shoots, shoots height, and number of leaves. The results of the analysis showed that the treatment with 0 mg/L IAA and 2 mg/L BAP was the best concentration for shoots multiplication. The addition of 2 mg/L BAP was able to produce a percentage of shoots emergence of 100%, 3 shoots per explant, shoots height of 7 cm and 16 leaves.

Abstract

Abstract: Piasa Wetan Village, located in Susukan District, Banjarnegara Regency, Central Java, has great potential for cultivating high-anthocyanin rice, supported by a favorable environment. However, limited knowledge and skills among farmers in organic farming practices and the certification process remain challenges. This community service initiative aims to enhance farmers' skills through comprehensive training that includes organic methods, harvest management, and the importance of certification. The training results showed a significant increase in knowledge, from 7% to 100% after the program's completion. More effective cultivation practices are now being implemented, including the selection of high-quality seeds, optimal land preparation, and more efficient harvest and marketing strategies. Certification is expected to help increase market prices, expand access, and ensure sustainability by enforcing strict organic farming standards. With continued education and improved agricultural techniques, Piasa Wetan Village has the potential to become a leading producer of high-anthocyanin rice in Banjarnegara, ultimately increasing farmers' incomes, supporting food security, and providing health benefits to consumers.

Abstract

Increasing lettuce production will be done by improving cultivation inputs, such as increased nutritional needs. An alternative organic fertilizer that can be used is made from spoiled milk, eggs, Trichoderma sp. molasses, and fruit waste, or STMJ Plus for short. STMJ Plus Fertilizer acts as an organic nutrient to encourage plant growth. This study aimed to determine the effect of the interaction between STMJ Plus fertilizer concentrations and lettuce varieties on the growth and yield of lettuce plants. The experiment was carried out in a factorial manner using a completely randomized design (CRD) consisting of 2 factors with 4 replications. The first factor is the concentration of STMJ Plus fertilizer consisting of 4 levels: P0: without fertilizer; P1: 5 ml L-1; P2: 10 ml L-1; and P3: 15 ml L-1. The second factor was the variety, which consisted of 2 levels: V1: Grand Rapids and V2: Red Rapid. The results of this study that the interaction of STMJ Plus fertilizer concentrations and lettuce varieties showed highly significant differences in plant height and plant fresh weight variables. The combination that gave the best effect was the concentration of 15 ml L-1 on the Red Rapid (P3V2).

Abstract

Identifying pathogenic fungi at the species level based on morphology, especially for leaf rust, is quite difficult. Further investigation, such as molecular techniques, is required to identify leaf rust fungi precisely. One way that can be employed is DNA barcoding. This study seeks to detect leaf rust fungi (Puccinia spp.) on sugarcane. Accurate species identification allows early detection of Puccinia kuehnii, helping farmers and extension agencies to apply targeted control strategies. A fungal sample was collected from sugarcane fields in Bondowoso, East Java, Indonesia. Morphological identification was done with an SEM, whereas molecular identification was done through DNA extraction, amplification, visualization, sequencing, and phylogenetic tree creation. The primers employed are obtained from the ITS region of ribosomal DNA, which contains enough diversity to differentiate fungi at the species level. The amplification findings revealed a DNA band of 500 bp at both loci. Sequencing results indicate that samples The sequencing results indicatethat the samples from Bondowoso are closely related to the Puccinia kuehnii sequence from NCBI. The findings of the phylogenetic tree construction revealed that samples from both locations are still associated with P. kuehnii sequences from other nations. This is the first study documenting P. kuehnii in Indonesia, especially East Java, that integrates morphological and molecular characterization.

Abstract

Lettuce (Lactuca sativa L.) is a leaf vegetable that has a high level of consumer demand and commercial value and can be cultivated using a combination of hydroponic substrate and drip irrigation methods. This research aims to determine the effect of the time interval for providing nutrients on the growth and harvest results of three lettuce plant varieties. Plants were cultivated on cocopeat and husk charcoal media (ratio 1:3) and arranged using a Randomized Block Design (RAK) consisting of two treatments. The first treatment is the nutritional interval which consists of an interval of 2 hours with a discharge of 17 ml (I1), 3 hours with a discharge of 25 ml (I2), and 4 hours using a discharge of 34 ml (I3). The second treatment is the plant variety which consists of the Grand Rapids variety (V1), the Green Coral variety (V2), and the Red Coral variety (V3). The data is analyzed using analysis of variance and if the results obtained are significantly different then a DMRT test will be carried out. The research results showed that the Green Coral lettuce variety was better than the Grand Rapids and Red Coral varieties. This is shown by the results with the highest and best values for the observation variables of number of leaves, plant fresh weight, and chlorophyll content. The nutritional interval which consists of an interval of 2, 4, and 6 hours gave results that were inversely proportional to the variety treatment, that is, they were not significantly different in all observed variables. Keywords: lettuce plants, plant varieties, nutritional interval, substrate hydroponics

Abstract

The purpose of this study was to characterize the active compounds in Indonesian local rice that are resistant to environmental stress in order to identify the nature of resistance to environmental stress, one of the indicators of which is the presence of flavonoid compounds. This step is one of the efforts that will lead to the acquisition of gene donors in the assembly of varieties resistant to biotic stress (pests and diseases) and abiotic stress (drought, high salinity, low temperature), which are very necessary in rice breeding programs. The method used is to plant 8 local rice varieties that are resistant to environmental stress in buckets in greenhouses. Next, analyze the content of active compounds by extracting the leaves and stems of rice plants, then macerate with methanol for 48 hours and test the content of active compounds. Then the total flavonoid content test was carried out using a quercetin standard and measured using a spectrophotometer. Whatever the results of this study, all samples showed active compounds containing flavonoids, alkaloids, tannins and saponins which were indicated by indicators of color change when tested with certain reagents. The results of the Flavonoid content analysis showed that the pendok variety had the highest flavonoid content, namely 3.9253 ± 0.06, and the Sigupai variety had the lowest flavonoid content, with a value of 1.8073 ± 0.07.
