Abstract

Emulgel merupakan sediaan topikal yang terdiri dari emulsi dan gelling agent. Minyak biji pala mengandung berbagai senyawa yang memiliki aktivitas antioksidan dari golongan fenolik yakni flavonoid. Tujuan penelitian ini untuk memformulasikan emulgel minyak biji pala yang stabil secara fisik. Dengan variasi konsentrasi dari Karbopol pada formula emulgel yang berfungsi sebagai Gelling Agent. Sediaan emulgel dibuat 3 formula yaitu FI (karbopol 0.5%), FII (karbopol 0.75%), FIII (karbopol 1%). Evaluasi sediaan emulgel yang dilakukan yaitu uji organoleptis, uji homogenitas, uji daya sebar, dan uji daya lekat. Dari hasil evaluasi didapatkan semua formula memenuhi rentang stabilitas fisiknya.

Abstract

Uncovering the therapeutic potential of secondary metabolites produced by plants, animals, and microbes constitutes the foundation for the development of novel medications. The objective of this investigation is to discern the classes of secondary metabolites and assess the antibacterial properties of endophytic fungal extracts obtained from Chrysanthemum indicum L. flowers. Through the isolation process, five isolates designated as JEC1, JEC2, JEC3, JEC4, and JEC5 were identified. The cultivation of endophytic fungal isolates spanned a three-week period before undergoing extraction with ethyl acetate. The phytochemical tests revealed the presence of alkaloids, flavonoids, steroids, terpenoids, saponins, and tannins in the ethyl acetate extract. Antibacterial activity was determined using the agar well diffusion method, with ciprofloxacin serving as a positive control. Notably, all ethyl acetate extracts from endophytic fungi exhibited antibacterial activity. The most substantial inhibitory diameter against Staphylococcus aureus was recorded as 19.1±0.8 mm for the JEC3 endophytic fungi, while Escherichia coli exhibited an inhibitory diameter of 16±1.1 mm for the JEC2 endophytic fungi.

Abstract

Chrysanthemum indicum L., commonly known as Chrysanthemum flower, is an herbaceous plant that has a long-established history of medicinal usage. There has been extensive research about C. indicium L, especially about its antioxidant activities, but not much has been done on its fraction. This study aimed to explore the efficacy of the ethanol extract and its fraction derived from Chrysanthemum flowers in scavenging free radicals. The antioxidant potential of the ethanol extract, as well as its aqueous and n-hexane fractions, was evaluated using the 2,2-diphenyl-1-picrilhidrazine (DPPH) method in vitro. The degree of antioxidant activity was quantified by determining the IC50 value, which corresponds to the concentration of the extract or fractions required to inhibit 50% of DPPH free radicals. The results obtained from this investigation provide strong evidence that the ethanolic extract, as well as its aqueous and n-hexane fractions, exhibited significant antioxidant activity. The measured IC50 values for the ethanolic extract, aqueous fraction, and n-hexane fraction were 1.350 µg/mL, 1.109 µg/mL, and 7.588 µg/mL, respectively.

Abstract

Gel preparations usually contain gelling agents which are critical factors that can affect the physical properties of the gel. The purpose to determine the appropriate variation of glucomannan concentration in the formulation of ascorbic acid gel on the physical characteristics of the gel. The research method began with the formulation of gel preparations using glucomannan as a gelling agent with variations in glucomannan concentrations at F1 (1%), F2 (1.5%) and F3 (2%). The gels were evaluated by their physical properties, including in the organoleptic, pH, homogeneity, adhesion, and spreadability tests. The research showed that in the pH and adhesiveness tests, increasing the concentration of glucomannan could increased the pH and adhesiveness of the gel. However, the spreadability of the gel decreased with increasing glucomannan concentration. The gel formula with 2% glucomannan concentration (F3) showed higher adhesion, as well as higher pH, although the gel spreadability decreased. From the evaluation results, all meet requirement for physically stable.

Abstract

Stunting is a growth and development disorder in children caused by chronic malnutrition and recurrent infections characterized by substandard height. School children generally neglect breakfast because they are busy in the morning, easily bored with food consumed daily which is traditionally processed by boiling, stir-frying, frying, steaming. The majority of children and teachers in this school do not know clearly about stunting, its causes, impacts, and how to overcome it. This service aims to provide counseling on education about stunting and introduce local foods that can function as stunting prevention by making functional food innovations. The method was carried out by measuring the child's weight, height, and arm circumference as early indicators of stunting. Providing counseling to children and teachers about stunting, its causes, impacts, and how to overcome it and providing functional food innovation products processed from surrounding plants. The results of this service activity showed that the results of monitoring the measurement of weight, height, and arm circumference were 4 students whose body condition was below the ideal standard of children at MIN 1 Minahasa school. No body condition was found below normal standards at Ra Nurut Takwa Kindergarten school. Children's knowledge about stunting and the benefits of local plants in Sea Village that can be used as functional food has increased. Nutritious gifts containing high protein were also distributed to children in the form of milk, vitamin C, cheese, milk candy, jelly candy as a result of the Team's functional food product innovation whose material was sourced from comedy shows. This service can be concluded that education about stunting in children in schools has had a positive impact on improving health, increasing knowledge, monitoring potential and further prevention of stunting.

Abstract

Breast cancer remains a major cause of mortality among women, particularly the aggressive subtypes HER2-positive and triple-negative breast cancer (TNBC). Fermented foods such as tempeh contain bioactive peptides with potential therapeutic properties, including anticancer activity, yet their molecular mechanisms in cancer remain unclear. This study aimed to investigate the potential of tempeh-derived peptides as anti-breast cancer agents using a network pharmacology approach integrated with molecular docking. Tempeh peptides were collected from previously published literature. Target genes of tempeh-derived peptides were predicted and compared with breast cancer–associated genes to identify overlapping candidates. These were analyzed through protein–protein interaction networks and subjected to functional and pathway enrichment to uncover key molecular mechanisms. The results showed that tempeh-derived peptides are closely linked to key oncogenic pathways, including PI3K-Akt, ErbB, MAPK, JAK-STAT, and general cancer signaling. Protein–protein interaction network analysis highlighted AKT1, SRC, STAT3, and PIK3CA as central hub proteins with well-established roles in regulating proliferation, migration, angiogenesis, and survival. AKT1 is strongly connected to HER2-driven signaling, SRC is involved in both HER2+ and therapy-resistant TNBC, STAT3 is critically implicated in TNBC biology, and PIK3CA functions as a pivotal upstream regulator of AKT1, underscoring their therapeutic significance. Molecular docking confirmed strong binding affinities of peptides such as Trp-Met-Phe-Asp-Trp, Pro-Phe-Tyr-Phe, and Trp-Met-Gly-Pro-Tyr to these hubs, suggesting disruption of phosphorylation-dependent activation and downstream oncogenic cascades. These findings support the potential of tempeh-derived peptides as multi-target modulators in aggressive breast cancer subtypes and highlight the need for experimental validation to advance their therapeutic application.

Abstract

Daging buah pala memiliki aktivitas antioksidan yang kuat dibandingkan dengan bagian biji, akar dan batang. Oleh karena itu, dilakukan penelitian pembuatan minuman instan terenkapsupsulasi dari buah pala yang kaya akan antioksidan. Minuman instan herbal pala (MHP) dibuat dengan proses enkapsulasi menggunakan maltodekstrin : susu skim (2:4). Pengujian antioksidan dengan metode DPPH, dan kandungan total fenolik dan flavonoid menggunakan spektrofotometer UV-Vis. Korelasi aktivitas antioksidan dengan kandungan total fenolik dan total flavonoid didasarkan pada persamaan regresi (total fenolik/ flavonoid vs antioksidan). Berdasarkan hasil pengujian MHP dengan konsentrasi 25% memiliki aktivitas antioksidan yang sangat kuat (IC50 : 21,20361 μg/mL.) Kekuatan antioksidan yang diberikan sampel lebih rendah dibandingkan dengan vitamin C (0,568 μg/mL), namun aktivitas antioksidan sampel masih tergolong sangat kuat. Aktivitas antioksidan yang diberikan dipengaruhi oleh kandungan fenolik dan flavonoid yang mana sekitar 39,07% kandungan total fenolik mempengaruhi aktivitas antioksidan. Sementara kandungan total flavonoid sebesar 39,5% adalah bagian dari senyawa fenolik yang mempengaruhi aktivitas antioksidan. Oleh karena itu, herbal pala dapat dikembangkan sebagai minuman fungsional antioksidan bernilai gizi.

Abstract

Sejauh ini inovasi raw material pasta gigi herbal yang berasal dari limbah masih jarang dikembangkan, cangkang kepiting menjadi material yang berpotensi untuk menjadi bahan baku dalam pembuatan pasta gigi herbal. Tujuan penelitian ini yaitu mengembangkan limbah cangkang kepiting menjadi material yang memiliki nilai ekonomi. Penelitian ini dilakukan dengan ekstraksi dan sintesis sederhana. Karakterisasi XRD, FTIR, dan SEM-EDX digunakan untuk menganalisis kristalinitas, gugus fungsi, dan morfologi kitosan. Hasil analisis menunjukkan struktur kitosan membentuk kristalin yang ditunjukkan dengan terkonfirmasinya tiga puncak tajam pada 2-teta 19o, 26o, dan 29o. Hasil analisis puncak serapan FTIR menunjukkan terdeteksinya 5 gugus fungsi utama kitosan pada bilangan gelombang 3458 cm-1, 2895,15 cm-1, 1654,92 cm-1, 1587,42 cm-1, dan 1386,82 cm-1 adalah OH stretching, CH(CH3) bending, C=O (-NHCOCH3) stretching amida I, NH (-NHCOCH3) bending amida II, CH (-CH2) bending sym secara berturut-turut. Morfologi kitosan yang disintesis dari cangkang kepiting berpori, bergelombang, dan bentuknya tidak teratur serta unsur yang terkandung yaitu C, O, Ca, dan Si.

Abstract

Katekin adalah senyawa flavonoid yang memiliki aktivitas antioksidan, antiinflamasi, antikanker, antimikroba, dan antidiabetes. Senyawa ini dapat ditemukan pada kulit kacang tanah. Oleh karena itu, perlu dilakukan uji aktivitas antioksidan dan analisis secara kualitatif senyawa katekin kulit kacang tanah Kawangkoan. Proses ekstraksi menggunakan empat pelarut yaitu etanol, methanol, aseton dan aquades dengan metode maserasi. Pengujian aktivitas antioksidan dilakukan berdasarkan metode 2,2-difenil-1-pikrihidrazil (DPPH) dan Asam 2,2-Azinobis (3-etilbenzatiazolin)-6-sulfonat (ABTS). Analisis kualitatif senyawa katekin dilakukan berdasarkan panjang gelombang maksimum (λmax) pada spektrofotometri UV-Vis menggunakan metode spiking. Berdasarkan hasil penelitian diperoleh rendemen ekstrak aquadest, methanol, etanol, dan aseton secara berturut-turut sebesar 5,75%, 10,56%, 3,46%, dan 3,74%. Aktivitas antioksidan terbesar diberikan oleh ekstrak aseton dengan IC50 sebesar 52,832 ± 2,143 µg/mL untuk uji DPPH dan 22,078 ± 5,690 µg/mL untuk uji ABTS. Hal ini berbanding lurus dengan keberadaan senyawa katekin yang hanya terdapat pada ekstrak aseton yang ditandai dengan 1 puncak pada panjang gelombang maksimum 535 nm. Hal ini menunjukkan bahwa ekstrak aseton kulit kacang tanah kawangkoan memiliki aktivitas antioksidan sangat kuat yang dipengaruhi oleh senyawa katekin.

Abstract

This study developed a Self-Nanoemulsifying Drug Delivery System (SNEDDS) of red ginger (Zingiber officinale var. rubrum) to enhance its stability and bioavailability. 8 formulations (F1–F8) were tested with varying Tween 80 : PEG 400 ratios (Smix 4.5 mL). Formula F7 (2.8 : 1.8 mL) produced a water-clear emulsion with pH 5.20 ± 0.02 and excellent stability for 28 days. PSA results showed droplet size 20.33 ± 0.57 nm, PDI 0.430 ± 0.053, and zeta potential −19.58 ± 1.50 mV. These findings indicated that F7 is the optimum, physicochemically stable SNEDDS formulation, potentially improving solubility and oral bioavailability of red ginger’s active compounds as a phytopharmaceutical antidiabetic candidate Keywords: red ginger, SNEDDS, nanoemulsion, physicochemical stability
