Abstract

This research was aimed to discover the traditional knowledge of Enggano people when using plants as concoction after giving birth. Data was collected through in depth interview, open-ended, and literature studies. The results showed that Enggano people used specific plants as concoction in childbirth process. There were two types of the concoction. The first was used after giving birth to 40 days, jarak pagar or tiarak (Jatropha curcas L.). The second was used after 40 days to 60 days or after recovery, such as dukung anak (Phyllanthus niruri Linn.), kumis kucing (Orthosiphon stamineus Benth.), temu lawak (Curcuma xanthoriza Roxb.), alang-alang (Imperata cylindrica (L) P. Beauv.), banana (Musa sp.), kencur (Kaempferia galanga L.), turmeric (Curcuma longa L.), and ginger (Zingiber officinale Roscoe). Results from our literature studies confirmed the efficacy of plants used by Enggano people is scientifically acceptable. Further study on phytochemical aspects of these medicial plants is therefore required to be carried out.

Abstract

Totally 15 isolates of endophytic fungi were obtained from leaves and young stems of a medicinal plant Albertisia papuana Becc.(Menispermaceae). The antibacterial screening of the ethyl acetate extract derived from the fungi cultures in potato dextrose broth (PDB)were performed on a non eluted Thin Layer Chromatography Bioautography assay (TLC bioautography assay). From the screening test, itwas found that the ethyl acetate extract of the fungus Xylaria sp. DAP-KRI-5 culture showed strong antibacterial activity against Staphylococcus aureus. The isolation of active metabolite based on the bioautography guided assay from ethyl acetate extract of the Xylaria sp. DAP-KRI-5, led us to identify phloroglucinol as a main antibacterial compound. The chemical structure of phloroglucinol was deduced from it spectral data, including UV-Vis, 1H and 13C-NMR, GC-MS and published data elsewhere.

Abstract

Fungal endophytes have been known as sources for bioactives with high chemical structure variability. This study aimed to screen extracts of some endophytic fungi associated with plants from Enggano Island as antioxidant and antibacterial using thin layer chromatography (TLC) bioautography method. Antibacterial ativity was performed against Escherichia coli and Staphylococcus aureus. Seventeen out of 22 extracts showed growth inhibition activity against both bacteria, while 16 extracts were active as DPPH free radical scavenging agents. Further determination on IC50 value of four prominent extracts revealed that fungal endophytes AK3018-1, FC-1, KC-4, and SO-3 have IC50 value of 85, 84, 704, and 347 ?g/ml, respectively in DPPH radical scavenging method. Extracts of fungal endophytes CR-3, CS-2, and SM-2 showed prominent antibacterial activity among other extracts, indicated by wide clear white zone around the spot. Further evaluation on minimum inhibitory concentration (MIC) of those three extracts by microdilution method showed that CR-3, CS-2, and SM-2 have MICs value of 512, >512, and 64µg/ml, respectively against S. aureus.Fungal endophytes AK3018-1, FC-1, KC-4, SO-3, CR-3, CS-2, and SM-2 were respectively isolated from Dioscorea bulbifera tuber, Fibraurea chloroleuca twig, Knema cinerea twig, Smilax odoratissima stem, Cryptocarya sp. twig, Calophyllum soulattri twig, and Smilaxmacrophylla stem.

Abstract

Three species of Smilax spp. (Smilax macrophylla, S. odoratissima and S. zeylanica) collected from Enggano island were evaluated for their potential as an antibacterial and antioxidant. Stems and leaves of three species of Smilax spp. were extracted successively with nhexane,chloroform, ethyl acetate and methanol. Antibacterial activity was evaluated by TLC-bioautography against Eschericha. coli InaCC B5 and Staphylococcus. aureus Ina-CC B4. The antioxidant activity was analyzed by DPPH free radical activity by bioautography method. The value of minimum inhibitory concentration (MIC) and IC50 of active extracts were done by serial dilution in 96- well microplate. The results showed that 20 extracts have antioxidant activity, 13 extracts inhibited the growth of S. aureus Ina-CC B4, and 14 extracts inhibited the growth of E. coli Ina-CC B5. MIC values of active extracts against S. aureus Ina-CC B4 were in the range of 128 - > 512 µg / ml, while the values of MIC against E. coli B5 Ina-CC were > 512 µg / ml. IC50 values of extracts that has antioxidant activity were in the range of 184.11-4549.34 mg/L.

Abstract

Cultivation of an endophytic fungus Schizophyllum sp.D, isolated from a tea plant Camellia sinensis (L.) O.K. in liquid medium PDB, on a rotary shaker at 100 rpm at room temperature (26 - 30 oC) produced a major metabolite with a production capacity of 56.7 mg/L. Structure elucidation based on MS, NMR spectra and published data showed that metabolite is nerolidol. This chemical compounds is one of important aromatic chemical constituent that contribute to tea aroma and defense system in a tea plant.

Abstract

The endophytic fungus Diaporthe sp.E show a unique ability to biotransform (-)-epicatechin into (-)-(2R,3R,4R)-leucocyanidin in a semisynthetic medium. Extension of the incubation time gave a major product (-)-(2R,35)-dihydroquercetin which was identified by spectroscopic methods.

Abstract

Five kinds of endophytic filamentous fungi i.e. Pestalotiopsis sp. GNC, Fusarium sp. GNC-A, Fusahum sp. GNC-B, Pestalotiopsis sp. GUC and Fusarium sp. GUC were isolated from surface sterilized of steems of gambir nasi and gambir udang (Uncaria gambier Roxb.) collected from Rengat, Riau Province,Indonesia. Identification of the endophytic fungi were done through morphologically observations and chemotaxonomically approach. Abstract

Terpenoid is a fundamental cells constituent in living organisms. In living organisms, terpenoid biosynthesize via classical pathway of mevalonate and via deoxyxylulose diphosphate (DXP). The distribution patterns of both pathways are unique and specific in living organisms. In human and animal, terpenoid biosynthesize via mevalonate and IPP isomerase type I enzyme system. In plant, terpenoid biosinthesize via combination of the mevalonate and DXP pathways with IPP isomerase type I enzyme system. In parasitic protozoa like Plasmodium falciparum and in major human bacterial pathogen such as Mycobaclerium tuberculosis, Salmonella typhii and Helycobacter pylori, terpenoid biosynthesize via DXP and repeatitive IPP isomerase type II enzymesystem. The specific diversity of the terpenoid biosynthetic pathways in living organisms can use as targets for development ofnovel antiinfective drugs.

Six kinds of the endophytic fungi have been isolated from a tea plant. One of them identified as Diaporthe sp. E based on their morphological characters and rDNA sequens. By the cultivation in liquid medium, the endophytic fungus produced (+)-epiepoxydon besides gentisyl alcohol and toluquinol.

Abstract

Aril and seeds of "pala wegio" {Myristica fatua Houtt),as one of endemic plants from Moluccas, contained 0.81% and 0.73%essential oils respectively.Gas Chromatography & Mass Spectrometry (GCMS) analyses indicated two similar major components (>10%) in aril and seeds essential oils of "pala wegio", (-) a-Copaene (aril 36.11% & seeds 29.81%) and caryophyllene (aril 18.05% & seeds 34.62%).However, the seeds essential oil had the third major component unindentified with 17.58% content.
