Abstract

ABSTRACTExploration of Soil Actinomycetes Diversity from Ternate as Indigenous Antibiotic Sources.Actinomycetes of soil samples from Ternate, North Moluccas were isolated using SDS-YEmethod in humic acid vitamin agar. Ternate has high abundance of Actinomycetes, approximately6.00 ? 487 x 104 CFU/ g soil, depends on habitat types. We have selected 60 isolates andconducted antibiotic screening against pathogenic bacteria and fungi using agar diffusionmethod and found both narrow and broad antibiotic spectrum types . Based on 16S rDNAanalysis, all Actinomycetes with antibiotic activities are belong to the genus Streptomyces. .Minimum Inhibitor Concentration (MIC) value was determined by broth microdilution method.It was found that MIC values varied, depended on microbial tested. We found two isolateswith higher antibiotic activity compared to the commercial antibiotics (chloramphenicol,erythromycin for antibacterial and nystatin, kabicidin for antifungal). Cell destruction analysiscaused antibiotic activities was conducted through leak of protein and nuclatic acid.Key words : Actinomycetes, soil, Ternate, antibiotic, cell distruction

Abstract

The objective of study was to investigate the microbial transformation reaction of 2E-6E-farnesol by the endophyticfungi isolated from temu hitam (Curcuma aeruginosa ROXB. The transformation was carried out in PDB with (2E-6E-farnesol), incubated at room temperature (25-32o C) under shaking condition at 120 rpm for two days produceda major biotransformed product. Structure elucidation based on 1D- and 13C-NMR analysis showed that the biotransformedproduct was 10,11-dihydroxi-2E-6E-farnesol. It It was verified that biotransformation reaction of 2E-6E-farnesol into 10,11-dihydroxi-2E-6E-farnesol through an intermediate 10,11-epoxi-2E-6E-farnesol.Keywords: Temu hitam; Curcuma aeruginosa; endophytic fungi; biotransformation; 2E,6E-farnesol; 10,11-dihydroxi-2E,6E-farnesol; 10,11-epoxi-2E-6E-farnesol.

Abstract

ABSTRACTThe objectives of study were to identify antimicrobial producing Actinomycetes using 16S rDNA analyses and morphology and physiology characteristics. Eight Actinomycetes strain with the higest antibacterial and antifungal activity were selected and identified using six primers (20F, 520F, 920F, 1500R, 920R, and 520R). Morphological observation and physiology analyses were performed to the selected strain to accurately identify the strains. Morphological characters observed were aerial mycelium, spore chain, colony form, and pigment production. Physiological characterizations were antimicrobial properties, growth temperature, pH tolerance, salinity concentration for growth, sugars assimilation, and some enzymes production (arginine dihydrolase, urease, ß-glucosidase, protease, ß-galactosidase). Based on homology search by BLAST program and phylogenetic tree analyses, all of isolates were identified as the genus Streptomyces. They belong to eight different spesies. Isolates RC-SS-37-4, RC-SS-37-16 and BL-22-3 have been identified as Streptomyces costaricanus (100 %), Streptomyces costaricanus (99.8 %) and Streptomyces parvulus (98.6 %), respectively. Five isolates were identified as Streptomyces spp. (BL-36-1, BL-20-2, BL-14-2, BL-22-1 and BL-06-5) and can be presumed as new species because of the low homology value to their closest related spesies. Keywords : actinomycetes, antimicrobial, morphology, phylogenetic, physiology, 16S rRNA gene.

Abstract

Endophytic fungi have been reported possess an interesting ability to mimic their host plant metabolites. Several fungi also show their specific capability to biotransform the chemical constituents of the host plant. The endophytic fungus Diaporthe sp. E isolate obtained from young stem of a tea plant (Camellia sinensis) show their unique capability to biotransform (-)-epigallocatechin-3-O-gallate [(-)-EGCG] into a major product in glucose-peptone-yeast extract medium that incubated under dark condition at 27 oC for 48 h. The major biotransformation product were isolated and purified through column chromatography techniques using Sephadex LH-20 and silica gel. The chemical structure of the major product were elucidated as (-)-2R,3S-dihydromyricetin based on their IR, FAB-MS, 1D- and 2D-NMR spectra. Abstract

Endophytic fungi have been recognized as source of broad range biological active metabolites with high chemical structure diversity. The purpose of this research is to isolate and characterize major metabolite produce by the endophytic fungi Penicillium sp isolated from a tea plant. Cultivation of endophytic fungus Penicillium sp (AB2245443) in liquid medium PDB on a rotary shaker at 100 rpm, temperature 25 – 30oC for 7 days, produced a yellow metabolite. Separation of metabolite through chromatography technique and followed by chemical structure elucidation based on MS, IR, NMR spectra and published data showed that the yellow metabolite is (-)-citrinin.

Abstract

The whole nine isolates of endophytic fungi have been obtained from leaves and roots of pandan wangi (Pandanus amarylifolius) collected from Bogor Botanical Garden. Identification of the fungi isolates were conducted based on their morphological characteristics, and revealed that one fungus belongs to the genus of Fusarium, one of Dreschlera, and 7 fungi isolates belong to Coelomycetes. The ethyl acetate extract of the fungi cultures in PDB and GYP media showed a diverse secondary metabolite profiles. Ethyl acetate extract derived from the culture of PWA-2 fungus in GYP medium showed an antifungal activity against Saccharomyces cerevisiae, but not to Fusarium oxysporum and Candida tropicalis. The GC-MS analysis of the active extract showed eugenol as an active component.

Abstract

Schoutenia ovata Korth. atau Walikukun merupakan tumbuhan yang banyak ditemukan di Indonesia. Kayu dari tumbuhan ini digunakan oleh masyarakat sebagai gagang perkakas dan juga sebagai bahan kerajinan tangan. Namun demikian, hingga saat ini, penelitian mengenai potensi farmakologis Walikukun belum dieksplorasi lebih jauh. Oleh karena itu, pada penelitian kali ini dilakukan skrining aktivitas antibakteri dan antioksidan dari daun dan batang Walikukun yang dikoleksi dari Mandalika, Nusa Tenggara Barat. Simplisia kering dari daun dan batang Walikukun diekstraksi secara bertingkat dengan pelarut n-heksana, diklorometan, etil asetat dan metanol. Analisis profil kimia dari daun dan batang Walikukun dilakukan dengan metode kromatografi lapis tipis. Uji aktivitas antibakteri dilakukan terhadap Staphylococcus aureus dan Escherichia coli sedangkan uji aktivitas antioksidan dilakukan terhadap radikal bebas 1,1-diphenyl-2-picrylhydrazyl (DPPH). Penentuan kadar hambat minimum (KHM) antibakteri serta penentuan indeks aktivitas antioksidan (IAA) dilakukan dengan metode mikrodilusi. Hasil pengamatan menunjukkan bahwa ekstrak daun dan batang dari Walikukun menunjukkan aktivitas antibakteri yang lemah hingga moderat dengan nilai KHM terendah sebesar 128 µg/ml. Ekstrak batang Walikukun menunjukkan aktivitas antioksidan yang potensial. Ekstrak metanol batang Walikukun menunjukkan aktivitas antioksidan terbaik dengan nilai IAA 8.275 ± 0.012 (aktivitas antioksidan sangat kuat). Hasil penelitian ini menunjukkan bahwa batang Walikukun merupakan sumber senyawa antioksidan yang potensial.Kata kunci: Schoutenia ovata Korth., Staphylococcus aureus, Escherichia coli, DPPH

Abstract

Totally 15 isolates of endophytic fungi were obtained from leaves and young stems of a medicinal plant Albertisia papuana Becc.(Menispermaceae). The antibacterial screening of the ethyl acetate extract derived from the fungi cultures in potato dextrose broth (PDB)were performed on a non eluted Thin Layer Chromatography Bioautography assay (TLC bioautography assay). From the screening test, itwas found that the ethyl acetate extract of the fungus Xylaria sp. DAP-KRI-5 culture showed strong antibacterial activity against Staphylococcus aureus. The isolation of active metabolite based on the bioautography guided assay from ethyl acetate extract of the Xylaria sp. DAP-KRI-5, led us to identify phloroglucinol as a main antibacterial compound. The chemical structure of phloroglucinol was deduced from it spectral data, including UV-Vis, 1H and 13C-NMR, GC-MS and published data elsewhere.

Abstract

Plants from Ixora genus exhibit a variety of pharmacological activities including antioxidant, antibacterial, and antitumor activities. This plants may contain many interesting bioactive compounds, especially phenolics and terpenoids groups. Ixora cumingiana is one of the Ixora species whose pharmacological effect has not been explored comprehensively. This study aimed to evaluate the antibacterial and antioxidant activity of eight extracts from bark and leaf of I. cumingiana. The plant samples were successively extracted using n-hexane, dichloromethane, ethyl acetate and methanol. The antibacterial evaluation was carried out against Staphylococcus aureus and the antioxidant activity was evaluated using a radical scavenging assay against 1,1-diphenyl-2-picrylhydrazyl (DPPH). Initial activity screening was performed using thin layer chromatography-bioautography followed by the determination of minimum inhibitory concentration (MIC) and the determination of antioxidant activity index (AAI) using microdilution technique. The dichloromethane extract of the bark as well as the n-hexane and dichloromethane extract of the leaf of I. cumingiana exhibited moderate antibacterial effect with MIC value of 128, 128, and 256 µg/ml, respectively. The methanol extract from the bark displayed a strong antioxidant activity (AAI = 1.5±0.13) and possessed the highest total phenolic content (43±0.91 mg AGE/ g extract). These experimental results showed that I. cumingiana is potential to be developed as an antioxidant agent rather than as an antibacterial agent.

Abstract

Aril and seeds of "pala wegio" {Myristica fatua Houtt),as one of endemic plants from Moluccas, contained 0.81% and 0.73%essential oils respectively.Gas Chromatography & Mass Spectrometry (GCMS) analyses indicated two similar major components (>10%) in aril and seeds essential oils of "pala wegio", (-) a-Copaene (aril 36.11% & seeds 29.81%) and caryophyllene (aril 18.05% & seeds 34.62%).However, the seeds essential oil had the third major component unindentified with 17.58% content.
