Abstract

Nitrogen fixation bacteria are bacteriathat are able to convert free nitrogen into nitrogen that can be used by theplants. This study aims to determine the presence of bacteria fixationnitrogen fixation bacteria in hydroponic and conventional rice roots and can identify the bacteriaidentify the bacteria. Isolation was done from the roots of rice (Oryza sativa) with the scatter blot method on NA (Nutriet Agar), and then tested for thethen tested for the ability to fix nitrogen using mineral-free nitrogen media added with Brontomimineral-free nitrogen medium added with Brontomiul Blue and glucose, which was incubated for 24 hours, thenincubated for 24 hours and then biochemical tests. From the results of the isolation there are 2 isolates that are able to bind nitrogen and have the ability ofHOS 833 ppm and KOS 1,128 ppm. The results of the characteristics and biochemistrycharacteristics and biochemistry showed that the isolate HOS is the genusBacillus genus and KOS is Azotobacter genus.

Abstract

This study aimed to determine the diversity, biomass potential, and carbon stocks of understory vegetation and trees in the Serdang Village Forest Area of Tahura, Karo Regency, North Sumatra. The methods used were destructive and non-destructive methods with a purposive sampling technique. Sampling was conducted by establishing measurement plots at three observation locations with a transect length of 150 m. Each location consisted of 10 plots, including 5 plots measuring 20 m × 20 m for tree observations and 5 plots measuring 5 m × 5 m for understory vegetation observations. The results showed that 25 understory plant species belonging to 20 families were identified with a diversity index of 2.66, while 22 tree species belonging to 16 families were identified with a diversity index of 2.38. The biomass of understory vegetation was 0.48 tons/ha with an average of 0.136 tons/ha, whereas tree biomass reached 2366.11 tons/ha with an average of 107.55 tons/ha. The carbon stock of understory vegetation and trees was 1183.194 tons/ha with an average of 591.597 tons/ha. Environmental factors influencing the diversity and biomass of understory vegetation and trees included temperature, humidity, light intensity, and altitude. The study concluded that the Serdang Village Forest Area of Tahura, Karo Regency, has a moderate level of biodiversity with high biomass and carbon stock potential, indicating its important role in carbon storage and ecosystem balance. Abstract

Usaha laundry adalah suatu jasa layanan yang terdapat sarana pencucian pakaian. Angka pengguna jasa laundry meningkat setiap tahunnya di kota Medan. Limbah dari laundry umumnya tidak diolah terlebih dahulu yang kemudian dapat menimbulkan pencemaran baik untuk tanah maupun air. Tujuan dari penelitian ini untuk memperoleh dan mengetahui informasi konsentrasi berapakah yang paling baik dalam menurunkan kadar Fosfat dengan menggunakan populasi mikroorganisme pseudomonas putida dan pengaruh penambahan waktu inkubasi. Penelitian ini merupakan penelitian eksperimental dengan menggunakan rancangan kuantitatif dan analisis statistik SPSS ANOVA (Analysis of Variance) One Way, untuk melihat pengaruh penambahan variasi konsentrasi Bakteri P.putida selama proses bioremediasi. Penelitian dilakukan selama 4 Minggu dengan memvariasikan penambahan bakteri P.putida sebanyak 0% (kontrol), 5%, 10%, dan 15% dengan Rancangan Acak Lengkap (RAL) 3 ulangan. Berdasarkan hasil penelitian penambahan variasi konsentrasi bakteri Pseudomonas putida dilakukan pungujian ANOVA One-Way memiliki hasil sig. 0,002. Konsentrasi 15% bakteri Pseudomonas putida dinyatakan konsentrasi terbaik yang dapat menurunkan kadar fosfat hingga melampaui batas baku mutu.Pemberian waktu inkubasi pengukuran penurunan kadar fosfat tidak homogen karena Hasil uji ANOVA One-Way memiliki hasil yang tidak sig, 0,302. Penurunan kadar fosfat teringgi pada minggu ke -4 sebesar 1,7 mg/L.

Abstract

Mangrove merupakan ekosistem yang sangat produktif. Sumber daya mangrove mempunyai beberapa peran baik secara fisik, kimia, dan biologi yang sangat berperan penting dalam menyediakan kebutuhan hidup masyarakat dan memiliki fungsi sebagai penyanggah keseimbangan ekosistem di wilayah pesisir. Ada berbagai macam organisme salah satunya bakteri pelarut fosfat (BPF) yang dapat diisolasi dari tanah rhizosfer mangrove. Penelitian ini bertujuan untuk mendapatkan isolasi bakteri pelarut fosfat dan uji potensinya. Metode yang digunakan adalah metode kuantitatif dimana merupakan informasi yang bisa diberi nilai yang diukur. Keanekaragaman dan populasi bakteri pelarut fosfat terhadap perbedaan dari beberapa jenis mangrove hasil yang diperoleh berbeda sangat nyata, hal ini diuji dengan uji beda nyata terkecil 5%. Pada penelitian ini diperoleh 12 isolat BPF dengan kode BFM 01, BFM 02, BFM 03, BFM 04, BFM 05, BFM 06, BFM 07, BFM 08, BFM 09, BFM 10, BFM 11 dan BFM 12. Kemampuan bakteri dari tanah magrove di pulau kampai memiliki diversitas indeks yang rata-rata berpotensial. Isolat yang memiliki indeks diversitaspaling tinggi dengan kode isolat BFM 04 yaitu sebesar 8,50 mm dan BFM 12 yaitu sebesar 8,25 mm sedangkan yang memiliki indeks diversitas paling rendah BFM 09 yaitu sebesar 2,25.

Abstract

Tempoyak adalah makanan tradisional melayu populer yang memiliki cita rasa yang enak dan aroma yang unik dan gurih. Tujuan dari penelitian ini adalah untuk mengidentifikasi jenis isolat serta ciri dan potensi probiotik yang terdapat pada durian fermentasi. Penelitian dilakukan di Laboratorium Mikrobiologi FMIPA USU. Bakteri diisolasi dari sampel tempoyak dengan metode pour plate dan dimurnikan dengan metode streak plate untuk menghasilkan biakan murni pada substrat agar laut. Ciri-ciri bakteri dan morfologi koloni diamati melalui pengujian morfologi, pewarnaan gram, dan uji biokimia. Sementara itu, uji pH rendah 3,0 dan kontrol 7,0 digunakan untuk menilai potensi probiotik, serta aktivitas bakteri petogenik. Hasil penelitian menghasilkan empat isolat murni dari genus bakteri Lactobacillus. Batang gram positif, koloni berwarna putih susu, putih, dan coklat, serta tahan asam atau pH rendah 3,0 dan kontrol 7,0 merupakan ciri dan morfologinya. Ini memiliki sifat antibakteri terhadap kuman berbahaya.

Abstract

Biology lessons for class Learning carried out by teachers to students is considered important because it is in accordance with the concepts, ideas and processes that must carry out the implementation of a plan that has been prepared carefully and in detail. This research aims to analyze 2 textbooks used in carrying out learning, including: 1) books Biology for SMA/MA Class XI by Djubaedah and Purnami with publisher Grafindo Media Pratama; 2) Biology book for SMA/M A Class XI by Irnaningtyas and Istiadi with publisher Erlangga. This research uses qualitative methods with data analysis in this research using descriptive statistics. The scientific literacy analysis is intended to determine the level of scientific literacy in class XI biology textbooks. Analysis was carried out by calculating the percentage of scientific literacy in each book analyzed. This research has the results that the level of scientific literacy in class XI high school biology textbooks on endocrine material used at MAPN 4 Medan is quite good according to the assessment criteria. The scientific literacy that often appears in the analyzed biology textbooks is science as a body of knowledge, followed by science as a way of investigation, science as a way of thinking and science and its interaction with technology and society

Abstract

One of the learning resources for biology is biodiversity. Region-specific biodiversity differs. Thus, in light of biodiversity, many students still struggle to comprehend the material covered in biology lectures, particularly at SMA Negeri 1 Natal.The research uses the ADDIE model research and development (R&D) method. The instrument of this research is a questionnaire sheet for material experts, media experts, teachers and students to test practicality and pre-test and post-test to test effectiveness. Class X MIPA-1 SMA Negeri 1 Natal students as test subjects. Testing was carried out on 30 students and biology teachers to see the practicality of the module using student and biology teacher response questionnaires. Based on the validation results from material experts, a percentage of 96.25% was obtained with the criteria "Very Valid". The validation results from media experts obtained a result of 81.2% with the criteria "Very Valid". The results of the teacher's response to the module were 87.7% for the "Very Practical" criteria. The results of student responses to the module obtained a percentage of 93.5% with the criteria "Very Practical". And the effectiveness results were 77.4% with effective criteria. Thus, biology learning modules based on problem based learning and local potential modules on biodiversity material are very practical and effective for teachers and students to use in biology learning

Abstract

Cassava Plants (Manihot esculenta Crantz), traditionally cultivated and offering many benefits as food ingredients, animal feed, and industrial raw materials. Cassava is also known to be rich in carbohydrates, soluble dietary fiber, and beneficial micronutrients for the body. Morphological and genetic diversity in cassava plants is crucial for the development of superior and improved varieties. This study aims to identify the morphological diversity of cassava leaf plants (Manihot esculenta Crantz) and analyze dendrogram relationships using RAPD (Random Amplified Polymorphic DNA) markers. The research was conducted in Serdang Bedagai Regency and the laboratory of the State Islamic University of North Sumatra, Medan. The methods used included sample collection, DNA isolation, PCR amplification, electrophoresis, and data analysis. The results showed variations in leaf morphology as observed in the number of leaflets, color, and shape of leaves in cassava varieties Genderuwo, Roti, Keriting, and Jari. Morphologically, the Genderuwo and Roti cassava leaves were the most similar. The dendrogram analysis using RAPD primers OPA 2, OPA 3, OPA 5, and OPA 7 showed a close relationship between Genderuwo and Roti varieties (coefficient 0.85), while the Jari variety had the most distant relationship (coefficient 0.60). This research can serve as a reference in cassava plant breeding to produce superior varieties

Abstract

Fingerprinting is a technique in DNA technology that can be used to see individual diversity and can also differentiate individuals from each other even though they are very closely related. This study aims to see the results of DNA fingerprinting of starfruit genotypes with the genus Averrhoa using Random Amplified Polymorphic DNA (RAPD) primers and to see the polymorphism value of each Random Amplified Polymorphic DNA (RAPD) primer. The research method used is Fingerprint DNA with Random Amplified Polymorphic DNA (RAPD) primers, namely OPA-2, OPA-3, OPA-5, OPA-7, OPD-11 and OPD-13. The results of the research show that the fingerprint results can be seen clearly. using electrophoresis. The best results were the primers OPA 2, OPA3 with 6 band patterns, followed by the second highest number of OPA5, OPD 11 with 5 band patterns, and the third highest number of OPA7, OPD 13 with 4 DNA fingerprint band patterns, the results of the kinship relationship between each Averrhoa have The polymorphic value shows that sweet starfruit (Averrhoa carambola L.) has a relationship of around 0.62% with starfruit. wuluh ( Averrhoa bilimbi L.). The Demak starfruit (Averrhoa carambola Cultivar Demak) has a relationship of around 0.71% with the Sembiring starfruit (Averrhoa carambola).

Abstract

Sweet potato (Ipomea batatas L.) is a national food security supported by one of the potential plant resources. After rice, corn, and cassava, sweet potato (Ipomea batatas L.) contains the fourth highest carbohydrate. Sweet potato is a good source of thiamine (vitamin B1), riboflavin, vitamin A, vitamin B, and vitamin C. The purpose of this study was to determine the genetics of the analysis results presented in sweet potato (Ipomea batatas L.) using dendograms and DNA banding patterns in sweet potatoes based on RAPD markers. The research method used is an experimental method to collect data by conducting direct experiments, isolating DNA, and conducting PCR using the RAPD technique; Six RAPD primers were used: OPA 2, OPA 3, OPA5, OPA7, OPD 11, and OPD 13. The study resulted in genetic diversity seen from the DNA banding pattern that forms polymorphism. RAPD produces bands measuring 100-900 bp. The genetic diversity of 4 types of Ipomea batatas L. plants analyzed using the NTSYS pc-2.02i application on the kinship relationship displayed with a dendogram shows the same diversity, namely orange sweet potatoes with yellow sweet potatoes, which have the highest level of similarity because they join at a coefficient value of around 0.91. While in purple sweet potatoes joining the group (yellow sweet potatoes and orange sweet potatoes) at a coefficient value of 0.86 indicates that purple sweet potatoes are slightly more different but still in the group. Then in white sweet potatoes have a connection coefficient value of around 0.81 which indicates that this sample is the most different compared to the others)
