Abstract

Research concerning of propagation of a wild-extinct fern Lygodium circinnatum had been done at Faculty of Agriculture, Udayana University, Denpasar Bali Indonesia. At some places in Indonesia, as well as Bali, this species is used as materials for making handicraft. In Bali, the species grows wildly in the forest and it is almost extinct due to over gathering. This study aimed to find out method for domestication of L. circinnatum, therefore this wild species can be cultivated, provided materials for making handicraft and might solve the problem of extinction. Various media for growing spores of L. circinnatum in vitro and ex vitro were trialed. In conclusion, full strength of MS media without sugar was the most appropriate media for growth and development of spores of L. circinnatum in vitro. While for ex vitro, the appropriate media were paddy silt-soil and decomposed leaf either with or without addition of foliar fertilizer. However, we suggested ex-vitro cultivation was more appropriate, the technique was much easier and the spores grew faster compared to those of in-vitro.

Abstract

Study of Sterilization and Induction of Red Grape Shoots (Vitis vinifera L. var. Prabu Bestari) with Several Types of Cytokinins In Vitro This study aims to determine the most suitable sterilization method for the sterilization of red grape explants and to determine the effect of giving growth regulators BAP, 2iP, and TDZ in the induction of red grape shoots. This research was conducted from January to June 2020 at the Tissue Culture Laboratory, Faculty of Agriculture, Udayana University. The design used in this study used a completely randomized design (CRD). The parameters observed in this study were the percentage of contamination, when the shoots appeared, when the roots appeared, the percentage of shoot growth explants, the percentage of root growth explants, and the percentage of leaf growth explants. This research is discussed descriptively. The results of the analysis of the research on the sterilization method showed that the sterilization method I through the maintenance of mother plants with 80% mankozeb and the use of clorox immersion in gradual concentrations was able to reduce the percentage of contamination to 61.2% and the use of TDZ 1 ppm growth regulators in combination with 0.01 ppm NAA on WPM medium was able to induce shoots and roots in red wine explants.

Abstract

Shoot Induction of Sandalwood (Santalum album L.) With GA3 Treatment In Vitro Sandalwood (Santalum album L.) is a woody plant with high economic value whose population is decreasing due to overexploitation. Propagation of sandalwood generatively and conventionally has been carried out but the success rate is still low due to the difficulty of obtaining quality adult individuals. This study aims to find the best concentration of GA3 in inducing sandalwood shoots in vitro. This research used Completely Randomized Design with 5 levels of treatment concentration of GA3 hormone, E0: WPM without GA3 (control), E1: WPM + 1 ppm GA3, E2: WPM + 2 ppm GA3, E3: WPM + 3 ppm GA3, E4: WPM + 4 ppm GA3. The results showed that the addition of GA3 concentration succeeded in inducing sandalwood shoots. The best 1 ppm GA3 for the variable when the fastest shoots were formed, which was 2.6 (DAT), and the variable number of shoots was (10). The best GA3 concentration of 2 ppm was on the number of explants that grew shoots (2.4), the percentage of explants sprouted (0.2%) and shoot length (1cm). GA3 1 ppm and 2 ppm succeeded in growing leaves.

Abstract

Mikropropagation Plant Strawberries (Fragaria sp.) Through Induction of Organogenesis Problems for farmers strawberry in Bali are the difficulty of get healthy strawberries  seeds a with prices are relatively cheap. Micropropagation method is ways to multiplay seeds for solving the problem. Research aims to understand the influence of concentration ZPT NAA + BAP to organogenesis on explant leaves strawberries in vitro. Experiment arranged using a Randomized Block Design (RBD). Medium the base used is MS medium with adding of 2 gL- 1 of activated charcoal, and 0.1 ppm NAA. As the treatment is BAP with concentration of 5 ppm, 10 ppm and 15 ppm with 5 replications. The results showed that the use of 0,1 ppm NAA + 15 ppm BAP most effective in deflection (3,0 DAP) with the percentage of 90,00%. The use of the NAA 0,1 ppm + 15 ppm BAP also most effective in a time of swelling (7,6 DAP) with the percentage of 82,95 % , while only the use of the NAA 0,1 ppm + 5 ppm BAP that will be capable of eliciting shoots at 23 DAP time.

Abstract

Induction of Black Raspberry Buds (Rubus occidentalis L.) Through Direct Organogenesis In Vitro. Supervised Black Raspberry plants are sources of various bioactive compounds such as phenolic compounds or phenolic acids, flavonoids, anthocyanins, flavonols, and tannins that are used as medicine. Black Raspberry also have a wide market share, the price reaches Rp 200,000 per kg. Propagation of black raspberry in a conventional way requires a long time. This study aims to obtain black raspberry seedlings in a relatively short time and large quantities through direct organogenesis in vitro. This research was divided into two, namely research on the sterilization method and the effect of gowth regulators with six treatments. The data were analyzed descriptively. The results of this study indicated that sterilization using tween 20 and vitamin C could reduce the occurrence of contamination and browning in explants respectively by 33.3% and 100%. The used of MS media + 2 ppm BAP + 150 ml coconut water + 0.01 ppm NAA (K4) treatment showed a response of gowth of shoots and leaves on the plantlet of 100%.

Abstract

The Effect of GA3 Concentration on The Induction of Grapevine (Vitis vinifera L.) Shoots Grown In Vitro The research concerning ‘The Effect of GA3 Concentration on the Induction of Grapevine (Vitis vinifera L.) Shoots Grown In Vitro’ have been conducted during period of February to August 2016 at The Laboratory of Plant Tissue Culture, Faculty of Agriculture, Udayana University. The objective of the research was to find out the most appropriate concentration of GA3 in stimulating of grapevine shoot grown in vitro. The experiment was laid out in a Completely Randomized Design with 5 treatments of GA3 concentration. The treatments were 0, 10, 20, 30, 40 ppm of GA3, each was replicated five times. The results showed that the treatment of 20 ppm GA3 was the most appropriate concentration in stimulating growth of grapevine shoots. The highest percentage of explants growing shoots i.e. 33.3% was obtained with the treatment of 20 ppm of GA3, compared to 6,6% (0 ppm), 0% (10 ppm), 20% (30 ppm), 6,6% (40 ppm).

Abstract

Influence of 2,4-D to induce callus from strawberry leaves on alternative media of AB Mix hydroponic nutrientThis research was conducted to find out the most optimum 2,4-D concentration to induce callus from strawberry leaves on alternative media of AB Mix hydroponic nutrient. This research used Completely Randomized Design with 5 levels of treatment concentration of 2.4-D hormon, first: AB Mix hydroponic nutrients (as basic media) without 2,4-D, second: basic media + 0.5 ppm 2.4 -D, third: basic media + 1 ppm 2,4-D, fourth: basic media + 1.5 ppm 2,4-D, fifth: basic media + 2 ppm 2,4-D. The variables observed were the day explant curled up, day explant swelled, percentage of eksplan curled up (%), percentage of explants swelled (%), the day callus appears, and percentage of explant browning (%). The best percentage of the treatment in callus induction of the strawberry leaves is on the P3 treatment with the concentration of 2,4-D as much as 1.5 ppm, with the fastest curled up explants time of 3 days after culture, the highest curled up eksplants (93.3%), the fastest swelled explants (17.0 days after culture), and the highest swelled percentage (40.0%). Callus formed 32 days after swelling. Successful callus formed only in treatment basic media with 1,5 ppm 2,4-D concentration.

Abstract

Propagule Induction of Node Culture of Grape (Vitis vinifera) varieties Prabu Bestari at Various Concentration of BA and NAA Propagation of grape seeds still uses conventional methods such as cuttings and grafting. In vitro culture technology can be a solution for plant propagation techniques that using small explants and in small quantities but capable to producing large amounts of seeds. Propagules are new formations that arise from explant tissue, can be in the form of shoots or callus that can occur through direct or indirect organogenesis. The aim of the research was to obtain the best culture medium in the formation of propagules. The research was conducted in two stages, the first was propagule induction and the second was subculture. The research used a Completely Randomized Design. Propagule induction was used a factor with combination of BA and NAA. The levels use of ZPT BA are: 2 ppm, 4 ppm, 6 ppm and the levels use of ZPT NAA are: 1 ppm, 3 ppm, and 5 ppm. The total combination treatment was 9 treatments and the addition of treatment K0 (control). Each treatment was repeated 3 times for a total of 30 experimental units. The subculture medium used was WPM + 1 ppm TDZ. The results showed that MS treatment with a combination of ZPT BA and NAA succeeded in forming propagules in the form of meristemoids and shoots. K3 (MS + 2 ppm BA + 5 ppm NAA) was the treatment that gave the best response to the percentage of meristemoid formation of 100%. K4 (MS + 4 ppm BA + 1 ppm NAA) was the treatment that succeeded in inducing 33.3% shoots.

Abstract

Fertilization outside the season of Siam Orange (Citrus microcarpa L.) with Induction of Flowering and Dormancy Breaking Substances. The Siam orange (Citrus microcarpa L.) produces seasonal fruit with the on-season harvest from July to August while the off-season harvest, i.e. from February to September there is hardly any fruit, or very limited supply. The purpose of this study was to examine the response of flowering induction and dormancy-breaking substances to produce off-season fruits of Siam orange. This research began in April to December 2016. The experiment was arranged factorially with two factors using a randomized block design (RBD), with nine combination treatments and six replications. Flowering induction factors consist of three levels, namely Ik = Control, IP = Paclobutrazol at a dose of 7.5 g/l, and Im = cutting the tip of dormant twigs. Dormancy-breaking substances factor consists of three levels, namely Dk = Control, Db = BAP 200 ppm, and Dn = KNO3 300 ppm. The results of the experiment showed that the highest fruit weight per tree at off-season production was obtained by induction with paklobutrazol (28.86 kg) and the lowest was in control (12.75 kg). Flowering induction treatments was not significantly different effect on fruit quality of Siam orange, represented by weight per fruits, fruit diemater, total soluble solid and vitamine C content was not siginificant different among levels of treatment. Dormancy-breaking substances treatment also not significantly effect on fruit quality of Siam orange. Based on this research it is recommended to induce flower for off-season produce of Siam orange by pruning the tips of dormant twigs or by using paklobutrzol.

Abstract

Induction of Organogenesis on Micropropagation of Vanda tricolor Orchidand Detection of Genetic Variation of the Progenies Using RAPD Marker.Research concerning on micropropagation of Vanda tricolor orchid and detection of geneticvariation among the progenies has been conducted in the year of 2015. The objective of theresearch was to obtain the most appropriate explant type for inducing organogenesis and toobserve genetic variation of the progenies using Random Amplification of Polymorphic DNA(RAPD). Various pieces of organ from 18 month old-seedlings were used as material ofmicropropagation, i.e. shoot tip, shoot base, leaf tip and leaf base. Explants were planted onMS medium without plant growth regulator. The percentage of explant producing shoots andthe average number of shoots per-explant were observed at 12 weeks after planting. Progeniesresulted from the micropropagation were then analysed using RAPD with 5 primers, i.e.OPA8 (5’ GTGACGTAGG 3’), OPA9 (5’ GGGTAACGCC 3’), OPA10 (5’ GTGATCGCAG3’), OPA11 (5’ CAATCGCCGT 3’), OPA13 (5’ CAGCACCCAC 3’). The results showedthat the most appropriate explant for inducing organogenesis was shoot base. Analysis ofRAPD showed that the slight variation was detected between mother plant and the progeniesand also among the progenies. The results obtained from the current research are promisingand suggests that RAPD markers can be utilized as a simple molecular tool to assess thegenetic variation of plants derived in-vitro.
