Abstract

In Planta Transformation of SoSPS1 Gene Through Agrobacterium tumefaciens in Balinese Grapevine (Vitis Vinifera L. var. Alphonso Lavalle). Vitis vinifera L. var. Alphonso Lavalle is one of the cultivar of grapevine cultivated in Bali. The sour taste of fruit is one of the problem of these cultivar that make it is less prefer by consumer. Research regarding improvement of fruit quality through genetic engineering has been investigated. The aims of the research were to find out the most proper methods of in planta transformation mediated with Agrobacterium tumefaciens applied on V. vinifera. The T-DNA harbored the SoSPS1 gene under the control of promoter of the CaMV from Cauliflower Mosaic Virus and contained the NPTII gene, a kanamycin resistant gene as a selectable marker for transformant selection. V. vinifera seedlings from stem cutting aged 40-50 days were ripped and then dipped with the bacterial suspension for 1 hour, 2 hours, 1 day, and 2 days. Visualization of V. vinifera samples after PCR showed that ripping and dipping for 2 hours method was positively integrated with NPTII gene. The dipping of plants wound for 2 hours treatment was effective for inserting gene of interest in ripping and dipping methods.

Abstract

Applications of Extracts of Fermented Immature Corn Seed and BambooSprout For Improvement of Table grape Quality of Vitis vinifera L. var.Alphonso Lavallee. The objective of the research was to determine the effect ofapplication of extracts of fermented immature corn seed and bamboo sprout for improvementof table grape quality. The research was designed as Completely Randomized Block Design,with five treatments and ten replications. The treatments were K0 (control), J50 (extract offermented immature seed of corn with concentration of 50%)), J100 (extract of fermentedimmature seed of corn with concentration of 100%), R50 (extract of fermented bamboo sproutwith concentration of 50%) and R100 (extract of fermented bamboo sprout with concentrationof 100%). The results showed that all of the treatments improved sugar content of the fruit.However, the treatment of J100 resulted in the greatest increase of berry number per-bunchand the lowest number of seed per-berry. In conclusion, for improvement of the table grapequality, the treatment of J100 was the most appropriate among other treatments used in thecurrent research.

Abstract

Identification And Characterization Of Local Fruits Genetic Resources In BangliRegency. Local fruit is all kinds of fruits that are cultivated in Bali. This study aims toidentify the types of local fruits grown in Bangli Regency; to create profile about those localfruits based on morphological and agronomic character, usability, production, andphotographs of genetic resources; to create a map regarding geographical destination of thefruit. This study was conducted in Bangli Regency consisting four districts i.e District ofBangli, Susut, Kintamani, and Tembuku. The experiment was conducted for six months, fromJanuary to June 2015. The research consisted of three stages of activities: (1) collection ofsecondary data, (2) surveys the types of genetic resources and distribution, (3) identificationof morphological and agronomic characters of genetic resources, growing environment andbenefit of local fruits. The results showed that the observations in the field found 36 speciesand 68 sub-species of local fruits spread over four districts. Based on the value of LocationQuotient (LQ) Bangli District has two main commodities including citrus, in which Awanvillage is the central production region, and banana, in which Subaya village in the centralproduction region.

Abstract

Due to the high cost of the most often used basic media in tissue culture, it is vital to identify more affordable alternatives. This research aimed to determine the best alternative culture media for the protocorm and plantlet growth of Coelogyne pandurata Lindl. It employed a completely randomized design, eight treatments and four replications. The treatments referred to the different types media and consisted of M1 = New Phalaenopsis (NP) medium, M2 = a medium made from foliar fertilizer (FFM), M3 = NP + 2 cc L-1 AB mix solution (a media for hydroponics), M4 = FFM + 2 cc L-1 AB mix solution, M5 = NP + 50 cc L-1 of tomato extract, M6 = NP + 50 cc L-1 of bean sprout extract, M7 = FFM + 50 cc L-1 of tomato extract and M8 = FFM + 50 cc L-1 of bean sprout extract. The M4 medium exhibited the best results in terms of average leaves count (4.80), average shoot length (2.68 cm), average root length (4.35 cm), the average fresh weight per plantlet (214.5 mg) and dry weight of plantlets (73.1 mg). The average number of roots per plantlet was 4.25, acquired using the less expensive M8 treatment, which also produces a negligible number of leaves (4.50). In conclusion, the M4 medium is the most appropriate medium for growing protocorm and plantlet of C. pandurata. The experiment also found that the FFM basic medium combined with 50 cc L-1 of bean sprout extract can be used as another cheaper alternative for growing protocorms of C. pandurata.

Abstract

Vanda tricolor  Lindl. var. suavis  is an Indonesian wild orchid which is now extremely rare in nature due to its habitat  destruction.  Development  of  an  appropriate  method  for  improving   Vanda  orchid  through  genetic modification could be valuable for horticulture and, indirectly, also for conservation. In this research, a method of  Agrobacterium-mediated transformation of two  Vanda  tricolor  obtained  from  Salak Mount, West Java and merapi Mount, Yogyakarta in Indonesia protocorms was improved using acetosyringone (AS). Concentrations of 0 and 25 ppm AS were used in transformation of pG35S binary vector containing kanamycin resistance gene into V.  tricolor  protocorms.  The  result  showed  that  25  ppm  AS  was  required  on  innoculation  with Agrobacterium  solution, without AS on cocultivation. Five week s after  treatment  on the 300  ppm kanamicyncontaining medium, green protocorms were obtained, that  was  11.01% for  V.  tricolor  from Salak Mount with pre-culture treatment prior to innoculation, 9.39% for  V.  tricolor  from Merapi  Mount with pre-culture treatment prior to  innoculation,  and  1.37%  for  V.  tricolor  from  Merapi  Mount  without  pre-culture  treatment  prior  to innoculation. The  best  condition  to  set  high  efficiency  of  transformation  is  pre -culture  protocorms  prior inoculation, soaking protocorm on 25  ppm  AS for 30 minutes, then cocultivate its on AS-free  callus induction medium Key words: Vanda tricolor, Agrobacterium, orchid protocorms, acetosyringone

Abstract

Mangga memiliki potensi sebagai sumber aktioksidan dengan mutu yang baik karena mengandung senyawa asam askorbat, karotenoid dan fenolik yang cukup tinggi. Bagian tanaman mangga yang diketahui menghasilkan antioksidan antara lain daun, batang dan buahnya. Berbagai antioksidan baik dari kelompok fenolik maupun flavonoid merupakan senyawa metabolik sekunder. Senyawa metablik sekunder secara umum dikenal sebagai senyawa kimia yang mempunyai kemampuan bioaktif sehingga banyak digunakan sebagai obat tradisional. Artikel ini menelaah ragam dan potensi antioksidan pada mangga dan biosintesisnya pada mangga. Jenis dan potensi kandungan senyawa antioksidan pada mangga beragam menurut vaietas dan bagian tanaman. Biosintesis senyawa antioksidan pada mangga diperkirakan sama seperti pada tanaman lainnya, dimulai dari jalur sikhimate.

Abstract

Tanaman mangga meupakan salah jenis tanaman yang banyak dikembangkan di Kabupaten Buleleng. Umumnya, pengembangan mangga tidak didasarkan pada analisa kesesuaian lahan, tetapi berdasarkan coba- coba atau meniru petani lain yang telah berhasil. Artikel ini merupakan hasil studi pustaka guna menelaah kondisi eksisting pengembangan tanaman mangga di Buleleng, dan kecocokan kondisi faktual pengembangan tanaman mangga di Buleleng dengan Petunjuk Teknis Evaluasi Lahan untuk tanaman mangga. Pada tingkat ordo, lahan-lahan di wilayah Kabupaten Buleleng tergolong cocok (S) untuk tanaman mangga. Tetapi, pada tingkat klas, kesesuaian lahan untuk tanaman mangga di Buleleng cukup bervariasi. Wilayah yang paling sesuai untuk tanaman mangga adalah Kecamatan Buleleng, Sawan dan Kubutambahan.

Abstract

Sandalwood (Santalum album L.) grows naturally in Indonesia, especially in Nusa Tenggara Timur area and this is one of 22 Santalum genus species in the world. Aim of the study is to determine the effect of 2-iP and the best concentration of 2-iP to stimulate callus explant shoots of Santalum album L. This studies carried out from September 2019 up to February 2020, the explants used were callus from bud culture from previous studies. The explants were planted at the Tissue Culture Laboratory, Faculty of Agriculture Udayana University Pulau Moyo street, Denpasar. This study used a completely randomized design (CRD) with 5 treatments, namely: (2-iP 0.0 ppm, 2-iP 0.5 ppm, 2-iP 1.0 ppm, 2-iP 1.5 ppm, 2-iP 2.0 ppm). The results showed that the addition of ZPT 2iP on WPM + 0.01 ppm NAA had a significant and very significant effect on the number of shoots, number of leaves and shoot length and had no significant effect on the emergence of shoots. The M1 treatment (WPM + 0.01 ppm NAA + 0.5 ppm 2-iP) showed the fastest time (1,55 MST) for the emergence of shoots on sandalwood callus explants. Treatment without the addition of zpt 2-iP (M0) could increase the number of shoots (2.87 pieces), number of leaves (5.59 strands) and shoot length (1.64 cm).

Abstract

Callus Induction and Multiplication of Eucalyptus sp. on Various Media Callus Induction Medium (CIM) In Vitro. Callus induction in vitro can be done by planting specific part of plant such as leaf, root, stem, etc in aseptic condition by using medium that contains 2,4-D, a plat regulator for promoting callus formation. One of the objectives of callus production in vitro is to form secondary metabolites, because eucalyptus is known as a producer of essential oils. This research is an initial study for in vitro production of secondary metabolites for further research. The aim of this study was to determine the most suitable callus induction medium for callus formation and callus multiplication for Eucalyptus grown in vitro. This research was designed as a Completely Randomized Design (CRD), five treatments, repeated five times. Woody Plant Medium (WPM) was used as the basic media. Several Callus induction Medium (CIM) were used as treatments, i.e. CIM1 = 1 ppm 2,4-D; CIM2 = 1,5 ppm 2,4-D; CIM3 = 2 ppm 2,4-D; CIM4 = 1 ppm NAA+1 ppm BAP; and CIM5 = 2 ppm NAA+ 2 ppm BAP. Variables observed were the time of curly leaf formation, time of the emerging callus, colour and texture of the callus, callus’s weight, callus surface area, the weight of callus after multiplication. The result showed that CIM4 and CIM5 had the fastest emerging callus (7 days after planting), CIM1 and CIM2 formed green and crumb texture of callus, CIM1 produced the highest of callus’s weight, and CIM2 resulted in the highest of callus’s surface area. In conclusion, the most suitable CIM for callus induction was CIM1, while for callus multiplication was CM2.

Abstract

Abstracts The community service activity entitled "Training for Acclimatization of Strawberry Seeds from Meristem Culture for Pancasari Strawberry Farmers" has been completed. This activity was motivated by the problem of seeds by strawberry farmers in Pancasari, where farmers needed healthy seeds from tissue culture, because through propagation with runners, crop yields decreased drastically due to virus attacks. The solution is the use of seeds from meristem culture. Acclimatization training by farmers is important because removing seeds from culture bottles and then growing them on media ex vitro requires skills and understanding of acclimatization theory. The purpose of this service was to provide strawberry acclimatization skills to farmers, so that farmers can acclimatize themselves to meristem cultured strawberries produced by the Tissue Culture Laboratory of the Faculty of Agriculture Udayana University. This activity was based on the results of strawberry meristem culture research in 2019 funded by the ministry of research technology and higher education Republic of Indonesia through a national competitive grant. The results of the current community service concluded several things as follows: The training participants, especially farmers, have succeeded in carrying out their own acclimatization practice; Strawberry seedlings grown in the highlands (Pancasari) had better vigor than those grown in the lowlands (Denpasar).
