Abstract

Backgrounds: Heavy metal that pollutes the river area affects living organisms which reside in it. Contamination of heavy metal in Ciliwung River leads to the presence of heavy metal elements (Cd, Hg, and Pb) inside the body of the plecos (Pterygoplichtys Pardalis) which inhabits that area. Hence, the utilization of plecos flesh and bones as a raw material in processed food products (e.g. shredded fish, shumai, and fish flour) might be harmful for human. Accumulated load of heavy metal in human's body would likely trigger health problems. Insufficient data on heavy metal concentration in Ciliwung River plecos-based food products underlies this research, with the aim to calculate the concentration of heavy metal Cd, Hg, and Pb on the previously mentioned plecos-based shredded fish, shumai, and flour. Methods: Heavy metal Cd, Hg, and Pb concentration analysis using X-Ray Fluoresence (XRF) methods conducted on PAIR Batan. Results: Value of Cd on shredded fish <0.5 mg/kg, shumai 0,7 mg/kg, head+tailbone and body skeleton flour <0.3 mg/kg. Value of Hg on shredded fish and shumai <0.7 mg/kg, head+tailbone flour 0.3 mg/kg, body skeleton flour 0.4 mg/kg. Value of Pb on shredded fish 1.3 mg/kg, shumai 0.8 mg/kg, head+tailbone flour 2.3 mg/kg, body skeleton flour 1.6 mg/kg. Conclusions: Heavy metal concentration on Ciliwung River plecos-based processed food products (shredded fish, shumai, and flour) has exceed maximum limit of Cd, Hg, and Pb determined by SNI, BPOM, and FAO.

Abstract

Eggshells are part of urban wastes. The nutrition content in eggshells is 94% calcium carbonate, 1% potassium phosphate, and 1% magnesium carbonate. One alternative to overcome this waste is to utilize it as eggshell flour. This study aims to determine the acceptability, analyse the nutritional content, and assess the contribution of nutrients to the Reference Nutrition Label (ALG) of onion chips substituted with eggshell flour as an alternative food source of calcium. This type of research is experimental with a completely randomized design. The eggshell flour used was commercial and non-commercial eggshell flour with a total of eight (8) formulas. Panelists were 30 students of University Al-Azhar Indonesia with two replicates. The results of the organoleptic test (hedonic and hedonic quality) were analyzed using the Kruskal-Wallis test, with the Mann-Whitney further test at a real level of 5%. The results showed that the A1B1 formula with the addition of eggshell flour as much as 10% and the use of commercial eggshell flour as the selected formula based on the Exponential Comparison Method (MPE). The results of the analysis of the nutritional content of the selected formula per 100 grams are energy 505 kcal, protein 9 grams, fat 31,5 grams, carbohydrates 46 grams, water content 4,3 grams, ash content 9,5 grams, and calcium 414,8 mg. Based on the nutrient contribution analysis, the product contributed 15,1% of calcium. Keywords – Eggshell, Onion Crisps, Substitution, Nutrient Content, Calcium

Abstract

The Mx gene is an antiviral gene used to determine the resistance or the susceptibility to different types of viruses, including the Avian Influenza (AI) virus subtype H5N1. The AI virus subtype H5N1 infection in chickens causes Mx gene polymorphism. The Mx+ gene shows resistant to the AI virus subtype H5N1, whereas the Mxgene shows signs of susceptible. The objective of this research was to detect the Mx gene in wild aquatic birds using the Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method with the primer pairs F2 and NE-R2/R and the RsaI restriction enzyme. DNA samples were obtained from eight species of wild waterbirds with positive and negative exposure to the AI virus subtype H5N1. DNA amplification results showed that the Mx gene in wild aquatic birds is found in a 100 bp fragment, which is the same as the Mx gene found in chickens. However, unlike chickens, the Mx gene in wild aquatic birds did not show any polymorphism. This study proves that Mx- based resistance to AI virus subtype H5N1 in different in wild birds than in chickens.
