Abstract

Many pathogenic microorganisme were found to be resistant to penicillin, to prevent the occurrence of penicillin resistance is then produced semisynthetic penicillins, basic materials used for the manufacture of semisynthetic penicillins a 6-aminopenisilanat acid (6-APA). 6-APA is the result of hydrolysis of penicillin by the enzyme activity of penicillin G acylase, Penicillin G acylase produced by various microorganisme, such as fungi and bacteria. This study aimed to determine the effect of pH on the activity of penicillin G acylase resulting bacterial isolates from stem water Kurao Padang. This study was conducted from October 2012 to January 2013 in the Laboratory Kopertis X Padang region of West Sumatra. This type of research is that the experimental treatment pH variation between (6,5 - 8,0) of the PGA activity resulting bacterial isolates and calculate the resulting enzyme activity using the method of Kornfeld is a method that has a high sensitivity and fast in detecting compounds 6-aminopenisilanat acid (6-APA). Results showed pH affects the activity of PGA by bacterial isolates from water with pH variation stems Kurao PGA 0.0101 6.5 activity units / ml, at pH 7.0 activity PGA 0.0269 units / ml, where as at pH 7.5 PGA activity of 0.0200 units / ml, and the pH 8.0 activity PGA 0.0064 units / ml. Based on the research that has been done can be concluded that the activity of Penicillin G Acylase resulting bacterial isolates from water Kurao Batang Padang has an optimum pH of 7.0 with enzyme activity (AE) PGA 0.0269 units / ml.

Abstract

Natural Penicillins (penicillin G) is known as an antibiotic which is chemically unstable, such as the onset of allergic in the body and its narrow spectrum. We perform the repairproperties of penicillin that makes penicillin G derivatives, in order to obtain a semisyntheticpenicillin that are better. Development of new penicillin can be reached by modifying thechemical spectrum penicillin G into 6-APA which is at the core of penicillin. To hydrolyzepenicillin G into 6-APA required the PGA enzyme produced by bacteria. PGA is anequilibrium reaction activity is influenced by pH. Under these conditions, has done research onthe effect of pH on PGA activity by bacterial isolates from Air batang Aru Lubuk BegalungPadang. The purpose of this study was to determine the effect of pH on PGA activity bybacterial isolates from Air Batang Aru. The research was conducted in the laboratory KopertisX from October 2012 to January 2013. This study is an experimental study with the method ofKornfeld on variation between pH (6.5, 7.0, 7.5, and 8.0). These results indicate that the pHeffect on the activity of PGA at pH 6.5 variation (0.0076 U / ml), pH 7.0, (0.0114 U / ml), pH7.5 (0.0609 U / ml ), and pH 8.0 (0.0200 U / ml). PGA optimum activity produced by bacteriaisolates were at pH 7.5

Abstract

Abstract Chilli is one of commodity that never left by people. As the development of technology , instant production greatly favored by the people. One of that is minced chili that made from red chilli. From the market survey to minced red chilli tradisional traders at traditional market of Padang is that red chilli processing into minced chilli for public consumption still does not fulfill the requirements in terms of hygiene.This study aimed to test the bacteriological to minced red chlli ( Capsicum annum L.) from some traditional market in Padang City, which was conducted in June2013  at  the  Microbiology  Laboratory of  the  Department  of  Biology,  Andalas  University.This research was conducted with descriptive methods. Sample taking was picked up by purposive sampling which 2 sample were taken from each 5 traditional market in Padang, consist of Bandar Buat market, Gaung market, Lubuk Buaya market, Simpang Haru market, and Siteba market.Bacteriological testing is then performed with the steps :1.estimation test ( presumtive test ),2 . confirmation test ( confirmed test ) and 3 . completion test (completed test ) using the MPN method (Most Probable Number) by using a combination of 5:1:1. From the research, results obtained that minced red chili sample contain bacteria with the range number 43 x        cell/ml - 157 x         cell/ml. Minced red chili sample from five traditional market in Padang city have coliform bacteria MPN values range from 12.0 to 240.0 ml sample, and E. Coli bacteria from 7.5 to 240.0 cells / 100 ml sample. Concluded that minced red chili that sold in some traditional market in Padang city is generally not suitable for consumption because it contains E. coli > 3 colonies / g and coliform bacteria > 100 colonies / g.

Abstract

ABSTRACTHazelnut is the spice material include dried spices sold in seed fotm. Many hazelnut used commoncy in cook eat like that, curry goat, rendang, asam padeh and etc by people. Hazelnut had solded in the Market in the open area and dont use of packaging. The condition like that hazelnut is dray will be easy damp, because it deals directly with the muggy air. It is will make the microorganism easily grow. This study for know the types of moulds contaminating Hazelnut (Aleurites moluccana Willd.) sold in Market. The study used survey method in the field with the simple random sampling technic. Descriptive observation of the isolated moulds that grow on the media, make oblique and identify to species lievel. Result of isolation and identification of a sample Hazelnut (Aleurites moluccana Willd.) found five types of kapang. It is find commoncy contaminants in foods like that, Aspergillus niger, Aspergillus sp.1 suspected Aspergillus flavus, Aspergillus sp.2 suspected Aspergillus oryzae, Penicillium sp. and Rhizopus oryzae. In conclusion hazelnut orkeon in that sold in the Market contaminated by moulds contaminations.

Abstract

ABSTRACT Ground  cayenne  pepper  is  one  of  the  preparations  that  are  sold  in  the  markets. Cayenne is one food that is often contaminated by soil, dirt and microorganisms. Contamination  can  occur  during  harvesting,  transport,  processing  and  marketing.  The purpose of this study to determine the types of molds contaminating ground pepper. This study uses purposive sampling method with mold growing on Potato Dextrose Agar media and create later identify pure culture. Results of this study are 5 types of molds are milled pepper pengkontaminsi Aspergillus ustus, Aspergillus niger, Aspergillus sp, Rhizopus stolonifer and Penicillium corylophilum.

Abstract

ABSTRACT Lemang is a traditional food from West Sumatera. Generally, lemang is made of black sticky rice or white sticky rice. In Talang Babungo, is made of black rice (not sticky rice) and still can be consumed eventhough it has been kept for days and has be taken by mould. The purpose of this research is to discover the varieties of mould which exists in black rice lemang.  This  research  uses descriptive survey method  by isolating mould,  making pure seedling, and then identifying. The result of isolating and identifying are found five varieties of mould in that lemang, those are Aspergillus niger, Aspergillus ustus, Aspergillus sp, Mucor racemosus, and Penicillium expansum.

Abstract

ABSTRACK Lemang beras hitam (Oryza sativa Siarang) of Kenagarian Talang Babungo is one of money distinctive Minangkabau foods. Lemang moldy left to get a different taste from the usual lemang. This fungus occurs on chemical procces aided by the amylase enzyme that  functions  to  decompose starch  into  to  units  of simple sugars. Research  has  been performed to determine the activities of fungal amylase enzyme which is isolated from black rice lemang. Researcher was performed in microbiology laboratory of match and science faculty of Andalas Univercity Padang from March from to April in 2013. The researcher used descriptive metode by growing the fungal isolates on rice flour medium (ATB) that aims to identify fungal amylase enzyme which is characterized by the formation of clears areas surrounding the fungal colony. From this research has been successful to rejuvenert the five isolates of the fungus. Amylolitik index is the highest is Penicillium expansum (1,5) and amylolitik index is lowest is Mucor racemosus (0,5).

Abstract

ABSTRACT Bayam ungu (Alternanthera brasiliana Kuntze.) efficacious for treating various diseases . Parts  used  are the leaves.  Spinach  purple   has a chemicals  such  as  tannins,  saponins,  and flavonoids. Escherichia coli is a bacteria found in that stout opportunist in human colon as normal flora. Escherichia coli suspected as a cause of diarrhea that sometimes arise in humans and animals . Research has been conducted in order to determine the inhibitory infusa purple leaf spinach (Alternanthera brasiliana Kuntze.) on the growth of Escherichia coli. This research was conducted at the Laboratory of Basic Biology Education Program STKIP PGRI West Sumatera in January 2013 , using a completely randomized design with 6 treatments and 4 replications with each infusion concentrations were 10 % , 20 % , 30 % , 40 % , 50 %. Analysis of the data showed that treatment with each test level α f 5 % and followed by BNT test. The results showed that infusion violet leaf spinach (Alternanthera brasiliana Kuntze.) could inhibit the growth of Escherichia coli.

Abstract

ABSTRACT There is no teacher makes character modules on subjects of biology. The use of a limited number of books and supporting students who came from the printing module used student does not meet the actual module components. There are also weaknesses in the present picture of them colorless, the concepts of the material in less clear learning objectives, in addition to supporting books and modules that are used do not seem character values. As a result students difficulty doing exercises or questions that the teacher and students are not stimulated to develop character values both at school and in public life. To the researchers have developed teaching materials in the form of character modules. This research aims to produce valid character module practical matter respiratory system. This type of research is the development of research (research and development) with 4-D models that define (definition) consists of the analysis of the curriculum, desaign (design) module and enter the value of character, develop (development) and dessiminate (spread). This research has been carried out and campus STKIP PGRI and SMAN 6 Padang, from the month of March-April 2013. At this stage of develop test the validity (2 teachers and 2 professors) and the practicalities by two biology teachers and students of class XI IPA4  many as 33 people. Character module on material generated respiratory system is very valid criteria for high school and very practical, with an average of 87.08 % validation (very valid) and the average value of the practicalities of 93.58 % (very practical). Expected products resulting from the research is the development of character modules can be used by teachers in the learning process, especially in SMA N 6 Padang, the teacher should remind students to practice honesty in filling out worksheets and do not see the answer key sheet module before work is      completed.
