Abstract

Our research focused on the antiproliferative effect of low intensity (18 Vpp) and intermediate frequency (100 KHz) electrostatic wave between two capacitive electrodes on breast tumor cells in vitro and in vivo. In vitro study has been conducted by using MCF-7 cell lines treated with external electrostatic for 24, 48, and 72 hours of treatment and the cells number were calculated during treatment by using hemocytometer and presented as Growth Inhibition (GI)% efficacy. For in vivo, we used female mice (Mus musculus) strain C3H as animal model. The mice were injected with either MCF-7 cells, mammary tumor cells from C3H donor, or NaCl 0.9% (placebo) subcutaneously into the axilla area and exposed by external electrostatic in each cage for 12 hours in 2 weeks before necropsied. The adjacent and breast tissue were collected and stained with Hematoxylin – Eosin then analyzed for histopathological profile. In vitro study revealed the number of exposed cells decreased with lower proliferation rate than the non-exposed cells. Moreover, the external electrostatic caused 28-39% growth inhibition efficacy of MCF-7 cells. After 2 weeks of exposure, placebo mice were physically normal, whereas the tumor undergone significant shrinkage of more than 67% in size. Histopathological analysis of the mammary glands indicated infiltration of macrophages into the tumor area through the blood vessel. No abnormality was found in the skin layer and mammary glands of the breast tissue of placebo mice. Abstract

Background: Animal models are essential for the development and improvement of novel and effective methods for diagnostic and treatment of human diseases. Cynomolgus monkeys have been used as animal model in asthma studies wherein they exhibited different responses to allergen exposure in the airway; some were low responders while others were high responder. CCL2 is a potent chemotactic factor for monocytes and the gene expression was high in animal model of asthma. The aim of this study was to evaluate CCL2 and CCR2 expression between the low and high responders. Methods: Realtime PCR technique was used to evaluate CCL2 and CCR2 gene expression in bronchoalveolar lavage samples. The subject of this study was asthmatic cynomolgus monkeys, consisted of 4 low responders and 4 high responders. This study was held in Biotechnology Laboratory of Primate Research Center LPPM IPB on October 2016- January 2017. Results: CCL2 and CCR2 expression in low responders were higher than those in high responders at 24hours post airway challenge. Conclusion: CCL2 may potentially be developed as target for therapy or a genetic marker for asthma responsiveness in individuals. (J Respir Indo 2018; 38(2): 115-22)

Abstract

Torbangun (Plectranthus amboinicus) is an herbaceous plant that grows in the tropical region. Torbangun can increase breast milk and has antioxidant activity. The objective of this research was to evaluate the antioxidant and cytotoxic activities of methanol extract of torbangun leaves and to identify diff erentially expressed lactation genes upon incubation of MCF-12A human cell epithelial mammary gland with methanolic extract of torbangun leaves. The results showed that the methanolic extract of torbangun leaves had a scavenging activity of DPPH radical with EC50 value of 14.14 μg/ mL. At lower concentration of methanol extracts of torbangun leaves showed no potent cytotoxic eff ect on MCF-12A with IC50 value of 155.24 μg/mL. The methanolic extract of torbangun leaves increased genes expression of prolactin receptor and glucocorticoid receptor in human mammary gland epithelial cells MCF-12A.

Abstract

The use of nonhuman primates as animal models in biomedical research has an important role in the development of human health. A close phylogenetic relationship with humans as well as similarities in genetics, behavior and biochemical activities are some of the things that underlie the selection of nonhuman primates as animal models. Cynomolgus macaques (Macaca fascicularis) is one of Indonesia's endemic primates which is widely used in research. Its current status as Appendix II in Convention International Trade in Endangered Species of Wild Fauna and Flora (CITES) defines that the population remains quite large in nature, which allows this species to be used in research, as long as they are sourced from the captive-bred population. Although the use of nonhuman primates in research is unavoidable, it is warranted that potential harm such as distress, discomfort, stress, pain and even death have to be avoided or minimized. Implementation of ethics is therefore required for the care and use of cynomolgus macaques in research, such as by assuring that the 3R principles (replacement, reduction, refinement) are met. Refinement can be achieved by applying the principles of animal welfare known as the Five Freedoms of Animal Welfare (5F). The active implementation of these ethical principles continues to develop along with the development of science and technology, and receives more spotlight in the world of biomedical research globally. Ensuring adequate implementation of ethical principles in research with nonhuman primates in each research institution must be under the oversight of an IACUC (Institutional Animal Care and Use Committee), which will weigh on the harm to animals compared to the benefits to humans. Active implementation of the principles of ethics and animal welfare will result in increasing the quality of research, serves as an assurance to the society, and in compliance to the requirements for good scientific publication.

Abstract

 Mesenchymal stem cells (MSC) derived from white adipose tissue are considered to be an appropriate therapeutic candidate for various human diseases due to their natural ability to maintain homeostasis. They have also been considered for use as a therapeutic alternative due to their sufficient availability and ease of harvesting. MSC play an important role in the process of adipogenesis and energy metabolism that is directly related to the pathogenesis of various degenerative diseases and metabolic syndromes. The purpose of this study was to obtain MSC cultures from white adipose tissue biopsied from three adult males Macaca fascicularis and to evaluate these cultures qualitatively and quantitatively regarding their characteristics and ability to differentiate. Polymerase chain reaction (PCR) was used to confirm the expression of mesenchymal stem cell-specific gene markers, and a specific growth medium was used to differentiate these cells into adipocytes, chondrocytes, and osteocytes. The results obtained showed that the MSC culture from M. fascicularis WAT had a fibroblast-like morphology with a spindle shape. The MSC markers of CD73, CD90 and CD105, were also expressed. The MSC culture was also able to differentiate into adipocytes, chondrocytes, and osteocytes. Based on these results, white adipose tissue from Macaca fascicularis was found to have potential as a source of MSC.

Abstract

Pencitraan Ultrasonografi (USG) ginjal dapat memberikan informasi struktur internal organ yang bermanfaat, terlebih apabila dikombinasikan dengan parameter kimia darah seperti profil Symmetric Dimethylarginine (SDMA). Penelitian ini bertujuan untuk deteksi dini perubahan morfometri dan fisiologis pada ginjal serta mengevaluasi apakah terdapat kaitan mengenai kesehatan ginjal dengan latihan fisik. Sampel yang digunakan adalah 6 ekor anjing ras Belgian malinois dengan umur 3 - 4 tahun. Tiga ekor anjing merupakan anjing yang aktif dilatih dan 3 ekor lainnya adalah anjing yang tidak aktif dilatih. Pemeriksaan USG dilakukan pada posisi hewan right and left lateral dengan microconvex probe frekuensi 5 – 8 MHz. Sampel darah diambil sebanyak 0.5 ml melalui v. cephalica dan dilakukan pemeriksaan SDMA. Hasil pengukuran menunjukan rasio panjang ginjal:aorta pada anjing aktif adalah 6.02, 6.85, dan 6.87, sedangkan pada anjing tidak aktif adalah 5.76, 6.36, dan 5.5. Nilai SDMA pada anjing aktif adalah 10, 11, dan 8 µg/dL, sedangkan pada anjing tidak aktif adalah 12, 11, dan 15 µg/dL. Hasil ini menunjukan bahwa anjing yang aktif berlatih memiliki ukuran ginjal yang lebih besar dibandingkan dengan anjing yang tidak aktif dilatih. Hasil SDMA pada ketiga anjing yang aktif berada pada rentang normal. Terdapat 1 ekor anjing yang memiliki nilai SDMA diatas rentang normal pada anjing yang tidak aktif dilatih, yang mengindikasikan mengalami chronic kidney disease (CKD) stage 1 menurut International Renal Interest Society (IRIS). Berdasarkan penelitian ini, aktifitas fisik mempengaruhi morfometri ginjal tanpa mengganggu fungsi ginjal anjing Belgian Malinois di Badan Narkotika Nasional (BNN).

Abstract

Colon cancer is one of the degenerative diseases caused of death after lung and breast cancer. One of the food ingredients that potentially inhibites the occurrence of colon cancer is black rice bran. The aim of this study was to found the effect of black rice bran on apoptosis and cell proliferation on BALB/c mice. In this study, BALB/c mice were divided into three groups, they are normal group (K-), AOM/DSS induced tumor group (K+), and feeding black rice bran with induction of AOM/DSS (BRB). At the end of the study, all mice were terminated and the presence of nodule were analysed in colon tissue used imageJ application. The expression of caspase-3, caspase-8, and PCNA were analysed in colon tissue used q-RT PCR. The data obtained by analyzes of variance (ANOVA) and continued by Tukey HD different test at 5% level. Administration of BRB significantly decreased the expression of PCNA in comparison with that of control mice (K+) (0.58 ± 0.09 vs 5.22 ± 0.80). The expression of caspase 3 (0.91 ± 0.20) and caspase 8 (0.36 ± 0.15) of BRB significantly increased comparasion with control mice K+. Our results thus indicated that black rice bran performed to suppresed proliferation and induced apoptosis

Abstract

Macaca nemestrina, also known as the pigtail macaque, plays a crucial role in drug development and serves as a highly valuable animal model for studying various human diseases due to its phenotypic similarities to humans, including those related to the aging process. Hematology is a critical component of clinical assessments for this spesies, serving both as a diagnostic tool to determine their health status and to characterize disease models, including degenerative and geriatric diseases. As anemia is a common phenomenon in the elderly, it is also important to determine whether M. nemestrina exhibit a similar condition with age. The aim of this study was to determine the hematological profiles of clinically healthy female M. nemestrina across young, adult, and elderly age groups, particularly related to anemia parameters. This study was conducted at the Primate Research Center, IPB University, Bogor, Indonesia. The animals were part of a breeding colony and were divided into three groups based on age: young (5–7 years), adult (12–14 years), and elderly (18–21 years). Following anesthesia with ketamine, a physical examination was performed to determine clinical status, and blood samples were collected for a complete blood count (CBC) evaluation using an automated analyzer. The results showed that total erythrocytes, hemoglobin concentration, hematocrit value, and mean corpuscular hemoglobin (MCH) were significantly different (P<0.05) in the elderly group compared to the other groups. Additionally, the red cell distribution width (RDW) percentage and leukocyte count differed significantly between the adult and elderly groups. The results showed that the erythrogram differs with age in clinically healthy M. nemestrina, suggesting the importance of determining appropriate baseline reference values in macaques of different ages.

Abstract

Pengembangan berbagai obat, vaksin dan produk kesehatan memerlukan tahap penelitian dan pengujian pada hewan laboratorium/hewan coba. Kesejahteraan hewan selama pemeliharan dan penggunaannya dalam studi merupakan salah satu faktor penting yang menentukan kesahihan hasil, dan pemenuhannya semakin menjadi sorotan publik dan tuntutan global. Diperlukan aturan hukum, kebijakan dan pedoman nasional untuk menjamin implementasi prinsip kesejahteraan hewan yang terstandar di berbagai institusi di Indonesia, dan mengacu pada standar internasional. Kajian ini bertujuan menelaah aspek legal di Indonesia yang mengatur implementasi prinsip etik dan kesejahteraan hewan di bidang penelitian biomedis, dan mengidentifikasi kebijakan dan pedoman yang diperlukan untuk mencapai standarisasi penjaminan tersebut.

Abstract

Anjing jantan ras dachsund (Teckel) berwarna hitam, berusia 4 tahun dengan bobot badan 7,5 kg dibawa pemiliknya datang ke klinik hewan dengan keluhan muntah berulang. Hasil pemeriksaan hematologi menunjukkan polisitemia, leukositosis dan jumlah granulosit yang tinggi. Hasil uji biokimia darah menunjukkan peningkatan pada aktivitas SGPT/ALT, GAMA-GT, total bilirubin, total protein, albumin, rasio ALB/GLOB dan rasio BUN/CRE. Hasil radiografi menunjukkan adanya bentukan membulat radiolucent didekat organ hati. Diagnosa kasus klinis pada anjing ini yaitu extrahepatic biliary duct obstruction dan terapi simptomatik dilakukan dengan memberikan gastroprotektan, antiemetik, serta obat-obatan untuk membantu memperbaiki fungsi hati. Pengobatan dilakukan selama 2 minggu dan menunjukan perbaikan pada organ hati berdasarkan hasil pemeriksaan biokimia darah, diikuti dengan tidak ada dilatasi kantung empedu pada radiogram.
