Abstract

Etlingera is one of the plant genera from the Zingiberaceae family and is widely distributed in Asia to the Pacific Islands, with various species ranging from 150 to 200 species. The Etlingera are commonly used as spices, vegetables, and traditional medicines. Many pharmacological activities have been reported from this genus, including antioxidants, antibacterials, cholesterol-lowering, anticancer, and others. The phytochemical content of Etlingera reported are phenolics, diarylheptanoids, flavonoids, steroids, alkaloids, and terpenoids. Eight species from Etlingera, namely Etlingera elatior, E. pavieana, E. brevilabrum, E. pyramidosphaera, E. megalocheilos, E. coccinea, E. fimbriobracteata, and E. corneri are reported provided anticancer activity. This review article aims to review the phytochemical screening from Etlingera genus, and its anticancer activity.

Abstract

Etlingera alba (EA) is one of the endemic plants grown in Sulawesi. We determined the cytotoxic and anti-metastatic activity of isolates obtained from EA rhizome extract. IC50 value for 1,7-diphenyl-6-heptene-3-one, sitostenone, sinapyl alcohol diacetate, and sinapyl alcohol acetate were 134.59; 170 ; 128.11; and 161.96 μg/mL, respectively. The compounds also had anti-migration activity. 200-μg/mL Sitostenon exhibited the lowest migration rate (3.32%). The free bond energies of sitostenon, 1,7-diphenyl-6-heptene-3-one, sinapyl alcohol acetate, and sinapyl alcohol diacetate were -11.81; -8.25; -6.64; and -6.28 kcal/mol, respectively. All compounds were more effective in stabilizing the Matrix metalloproteinase 1 complex. Isolation from EA rhizomes have the potential to be developed as anti-metastatic for Triple-Negative Breast Cancer.

Abstract

The aim of this study was to determine the best combination of cryoprotectant (Ethylene glycol, EG), dimethyl sulfoxide (DMSO) and propanediol (PrOH) and carrier system (hemistraw and cryotop) in improving ewe oocytes viability during cryopreservation. Oocytes with multi layers of compact cumulus cells were colleted from abbatoir and matured in TCM 199 medium supplemented with 10% fetal calf serum for 24-26 h at 38.5° C under 5% CO2 in the air. Matured oocyte was divided into six parts and vitrified in three different vitrification solutions; (i) 17% EG+17% DMSO with hemistraw as carrier system, (ii) 34% EG with hemistraw as carrier system, (iii) 17% EG+17% PrOH in hemistraw (iv), 17% EG+17% DMSO with cryotop as carrier system (v), 34% EG with cryotop as carrier system (vi), and 17% EG+17% PrOH in cryotop. Oocytes were cryopreserved for one week before revived and evaluated for viability. The result showed that oocytes vitrified in media containing EG and DMSO in cryotop had the highest viability (88.16%) compared to media containing EG only or EG and PrOH (70.95% and 68.76%, respectively) (P0.05). Moreover, oocytes viability that vitrified using cryotop and hemistraw as carrier system were not significantly different. The present results indicated that vitrification using combination of EG and DMSO as permeable cryoprotectant and cryotop as carrier system was the best system to maintain oocyte viability after vitrification-warming.

Abstract

Pendekatan kemotaksonomi genus Etlingera memungkinkan Etlingera rubroloba A.D. Poulsen memiliki kandungan metabolit sekunder sebagai agen imunomodulator. Penelitian ini bertujuan mengetahui efek imunostimulator ekstrak etanol buah E. rubroloba A.D. Poulsen dengan parameter kadar CD8 (Cluster of differentiation) pada tikus jantan galur wistar. Hewan uji tikus sebanyak dua puluh empat ekor dibagi dalam enam kelompok perlakuan yaitu kontrol normal, kontrol pelarut (Na.CMC 0,5%), kontrol positif (ekstrak meniran komersil®), perlakuan ekstrak dosis 200, 300, 400 (mg/kgBB). Perlakuan diberikan selama tujuh hari berturut-turut secara per oral  dan pada hari kedelapan masing-masing kelompok diinfeksi bakteri Staphylococcus aureus 0,5 mL secara intraperitonial. Kadar CD8 diukur dengan metode ELISA dan data dianalisis dengan one way ANOVA. Hasil penelitian menunjukan perlakuan dosis ekstrak etanol buah E. rubroloba A.D. Poulsen memiliki efek imunostimulator berdasarkan kadar CD8 yang berbeda signifikan dengan kontrol pelarut (p<0,05), sehingga berpotensi dikembangkan sebagai imunomodulator alamiah.
