Abstract

Inflamasi merupakan rangkaian perubahan kompleks dalam jaringan akibat cedera yang disebabkan oleh bakteri, trauma, zat kimia, panas dan nyeri.Tumbuhan bambu-bambu (Polygonum pulchrum Blume) adalah salah satu jenis tumbuhan perennial yang mengandung senyawa bioaktif dengan beberapa potensi aktivitas farmakologi, diantaranya sebagai antiinflamasi. Penelitian ini bertujuan untuk mengetahui kandungan metabolit sekunder dan uji  aktivitas antiinflamasi dari daun bambu-bambu (Polygonum pulchrum Blume) dengan menggunakan metode stabilisasi membran sel darah merah secara in vitro. Kandungan  metabolit sekunder diidentifikasi menggunakan metode skrining fitokimia secara kualitatif berdasarkan reaksi pengujian warna. Pengujian potensi aktivitas antiinflamasi dilakukan dengan metode stabilisasi membran sel darah merah secara In Vitro yaitu melalui kemampuan daya hambat ekstrak etanol daun bambu-bambu (Polygonum pulchrum Blume) terhadap lisis sel darah merah akibat induksi larutan hipotonis yang kemudian dibandingkan dengan kontrol positif yaitu natrium diklofenak. Hasil penelitian menunjukkan bahwa ekstrak etanol Polygonum pulchrum Blume positif mengandung flavonoid, tanin, dan saponin. Nilai persentase stabilitas sel darah merah yang dimiliki oleh ekstrak etanol daun Polygonum pulchrum  dengan konsentrasi 50, 100, 200, 400, 800, dan 1600 ppm berturut-turut adalah 58,13%; 67,3%; 75,72%; 83,28%; 87,05%; dan 92,99%. Adapun stabilitas persentase yang dimiliki oleh kontrol positif dengan konsentrasi 50, 100, 200, 400, 800, dan 1600 ppm adalah 58,99%; 66,38%; 73,24%; 80,58%; 82,95% dan 86,73%. Ekstrak etanol daun bambu-bambu (Polygonum pulchrum Blume) pada semua variasi konsentrasi memiliki potensi aktivitas dalam menstabilkan membran sel darah merah.

Abstract

Buah tanaman wualae (bahasa Tolaki) atau Etlingera elatior digunakan oleh masyarakat Tolaki (Sulawesi Tenggara) sebagai bumbu masak dan obat tradisional. Untuk mengungkap potensi tersebut, khususnya buah wualae, dilakukan kajian fitokimia dan farmakologi buah wualae. Kajian fitokimia meliputi isolasi dan penentuan struktur dikerjakan dengan metode kromatografi (kromatografi vakum cair dan kromatografi radial) dan spektroskopi (1H dan 13C NMR). Kajian farmakologi meliputi uji aktivitas biologi terhadap enam jenis bakteri (Bacillus subtilis FNCC 0060, Escherichia coli ATCC 35218, Pseudomonas aeruginosa ATCC 27853, Salmonella enterica ATCC 14028, Staphylococcus aureus ATCC 25923, dan Streptococcus mutans ATCC 25175). Hasilnya menunjukkan tiga senyawa fenolik telah berhasil diisolasi dan diidentifikasi dari buah wualae yaitu asam vanilat (1), asam p-hidroksibenzoat (2), dan asam kumarat (3). Ketiga senyawa tersebut pertama kali dilaporkan berhasil diisolasi dari buah E. elatior. Aktivitas antibakteri ekstrak metanol buah wualae dan tiga senyawa hasil isolasi menunjukkan bahwa sampel tersebut paling aktif terhadap bakteri S. mutans. Dengan demikian aktivitas senyawa-senyawa hasil isolasi dari buah E. elatior  berkorelasi dengan aktivitas ekstrak metanol buah E. elatior  terhadap S. mutans, dimana senyawa paling aktif adalah asam vanilat. Ekstrak yang dominan mengandung asam vanilat dari tanaman ini dapat dikembangkan sebagai herbal antibakteri S. mutans seperti herbal karies gigi dan obat kumur. Kata kunci: Etlingera elatior, wualae, buah, senyawa aromatik, antibakteri. The fruits of wualae (Tolakinese) or Etlingera elatior is utilized by Tolakinese (Sulawesi Tenggara) as spices and traditional medicine. To reveal the potencies, a study on phytochemical and pharmacological aspects have been carried out. The phytochemical study includes isolation and structure determination used chromatography methods (vacuum liquid chromatography and radial chromatography) and spectroscopy methods (1H NMR and 13C NMR). Biological activity was evaluated against some bacteria that are Bacillus subtilis FNCC 0060, Escherichia coli ATCC 35218, Pseudomonas aeruginosa ATCC 27853, Salmonella enterica ATCC 14028, Staphylococcus aureus ATCC 25923, and Streptococcus mutans ATCC 25175). The results showed that three aromatic compounds had been isolated and identified that are vanillic acid (1), p-hydroxybenzoic acid (2) and coumaric acid (3). All compounds are firstly reported that successfully isolated from fruits of Wualae. Antibacterial potency indicated that the methanol extracts and the isolated compounds are the most active towards S. mutans and have good activity correlation between the extracts and the compounds. Vanillic acid is the most active compound from E. elatior fruits. Thus, the extracts contain dominantly vanillic acid from          E. elatior fruits can be developed to be antibacterial herbals caused by S. mutans. Keywords: Etlingera elatior, wualae, fruits, aromatic compounds, antibacterial activity.

Abstract

Telah dilakukan penelitian tentang efek imunomodulator ekstrak etanol daun galing (Cayratia trifolia L. Domin.) terhadap daya fagositosis makrofag mencit jantan. Penelitian ini bertujuan untuk menguji efek imunomodulator ekstrak etanol daun galing terhadap peningkatan aktivitas fagositosis sel makrofag mencit jantan. Ekstrak uji dibuat dengan dua variasi dosis 400 mg/kgBB dan 500 mg/kgBB. Hasil yang didapatkan menunjukkan bahwa semua pemberian perlakuan dosis ekstrak memberikan hasil yang baik terhadap peningkatan akivitas fagositosis sel makrofag, persen aktivitas sel makrofag tertinggi ditunjukkan pada pemberian dosis ekstak 500 mg/kgBB yaitu sebesar 65, 4%, dibandingkan dengan peningkatan aktivitas sel pada dosis 400mg/kgBB dan kontrol positif dengan masing-masing memiliki persen aktivitas sel sebesar 55, 4% dan 60, 5%. Sedangkan nilai aktivitas sel terendah ditunjukkan pada kelompok kontrol negatif (NaCMC) dengan persen aktivitas sel sebesar 37, 8%. Hasil analisis data statistik (ANOVA) menunjukkan bahwa ekstrak etanol daun galing (C. trifolia L. Domin.) dapat meningkatkan imunitas non spesifik (p0,05).

Abstract

Wualae (Etlingera elatior (Jack) R.M. Smith) belonging to Zingeberaceae that spread in South East Sulawesi, empirically used as staple and treatment. The aim of this research to determine the antimicrobial potential of ethanol extract fruit of E. elatior against gram positive (Staphylococcus aureus ATCC 25923 and Bacillus subtilis FNCC 0060), and gram negative (Pseudomonas aeruginosa ATCC 27853 and Salmonella enterica ATCC 14028) using cup-plate method. The antimicrobial activity based on the diameter of inhibition zone. The result shows that extract more potential to inhibit gram positive than gram negative while the highest inhibition illustrate by concentration of 20 mg/mL (inhibition zone= 5 mm). In conclusion, the extract of E. elatior no potent as antimicrobial candidate (category= weak).

Abstract

Vegetable fern leaf (Diplazium esculentumSwartz) is kind of plant that can inhibit Salmonella typhi growth. The study aimed to determine the effect of vegetable fern leaf ethanolic extract on male mice that had been infected by Salmonella typhi bacteria. Vegetable fern leaf powder was macerated with  95% ethanol and was evaporated with vacum rotary evaporator. Dosage variation of vegetable fern leaf ethanolic extract on male mice were (100; 200; 300; 400 and 500 mg/kgbb). Blood of male mice veins was cultured on agar media and  counted bacterial colonies that were formed. Study result showed increasing of vegetable fern leaf ethanolic extract could decrease the colony of Salmonella typhi that infected in male mice. Vegetable fern leaf ethanolic extract with 500 mg/kgbb could decrease 6,21% of Salmonella typhi in male mice. vegetable fern leaf ethanolic extract with dosage variation not significantly  decrease the amount of bacteria colonies in mice. Compound identification by thin layer chromatography (TLC) showed vegetable fern leaf ethanolic extract contained of  flavonoids, saponins, alkaloids, tanins and terpenoids.

Abstract

Reactive oxidative stress (ROS) can lead to cell damage, and one of them is the kidney's cell. Etlingera elatior (Jack) R.M. Smith can be utilized as an agent that can protect the cell from ROS. This study aimed to investigate the protective effect of E. elatior fruit on the kidney's cell. We used experimental animals which were treated with Na-CMC (Group I), Na-CMC (Group II), the extracts of E. elatior fruit 200, 300, 400 mg/kg BW for Group III, IV, and V, respectively, for seven days. The blood was collected after treatment. At day 8, group I, III, IV, and V were induced by CCl4. At day 9, blood was collected and the kidneys were harvested for histopathology analysis. Blood collected were measured for albumin, total protein, urea and creatinine levels. After treatment, albumin and total protein showed no increased levels; urea decreased at doses of 200, 300, and 400 mg/kg BW, respectively, and creatinine levels only decreased at the dose of 400 mg/kg BW (p<0.05). The dose of 200 and 300 mg/kg BW showed protecting effects in the tubular cells of renal. Therefore, the ethanol extract of E. elatior showed a nephroprotective effect by normalizing the urea and creatinine levels of rats and protecting tubular cells of renal.

Abstract

The rhizome of Etlingera elatior or Wualae (Tolakinese) has many advantages on traditional remedies and cooking in Sulawesi Tenggara. To support those advantages, two secondary metabolites derived from steroid and phenylpropanoic acid classes, stigmast-4-en-6β-ol-3-one (1) and p-coumaric acid (2), respectively, have been firstly isolated and identified from the E. elatior rhizome. Isolation of these two compounds was performed using several chromatography techniques, including thin layer chromatography (TLC), vacuum liquid chromatography (VLC) and radial chromatography (RC). Identification of isolates was carried out using 1 H and 13C NMR spectroscopy and comparing the spectroscopy data with the library. The potency of antibacterial of the methanol extract of Wualae rhizome and the isolates were evaluated against Pseudomonas aeruginosa ATCC 27853, Bacillus subtilis FNCC 0060, Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 35218, Salmonella enterica ATCC 14028, and Streptococcus mutans ATCC 25175 using agar diffusion method. Antioxidant activity was evaluated against DPPH radicals (2,2-diphenyl-1-picrylhydrazyl). The results show that the antibacterial potential of Wualae methanol extract is better than compound 1 and 2. Furthermore, the antioxidant properties of Compound 2 (IC50 159.47 μg/mL) was stronger than the antioxidant properties of Compound 1 (IC50 219.95 μg/mL) and the methanol extract (IC50 586.38 μg/mL).

Abstract

Cancer is a devastating disease characterized by abnormal cell growth. Due to several safety issues associated with current cancer treatment, identification of new strategies for cancer therapy is considered necessary. Natural substances in plants m and fruits have the potentials to be developed as anticancer drugs. One of them is Cayratia trifolia (C. trifolia). This study was aimed to determine the cytotoxic activity of stem and leaf ethanol extract of C. trifolia against zygote cells of sea urchin or Tripneustes gratilla (T. gratilla). Extraction of stem and leaf of C. trifolia was conducted using maceration method in ethanol solvent. Thin layer chromatography was performed to screen phytochemicals contained in the extract. Zygoteof T. gratilla was used to conduct antimitotic activity test. We found that stem and leaf ethanol extract had IC50 of 169.82 μg/ml and 208.92 μg/ml, respectively. In comparison with positivecontrol of vincristine which had IC50 value of 3.23x10-3 μg/ml (very strong category), this extract showed moderate antimitotic activity. In conclusion, C. trifolia is potential to be developed as anticancer treatment.Keywords: cancer, Cayratia trifolia, antimitotic

Abstract

The potential toxicity effect presents in the medicinal plants is important to be identified forthe safety assurance. Accute toxicity study is an initial step in the drug safety assurance test.Thus, the aim of this study is to identify the potential acute toxicity of roots, stems, leaves,and flowers ethanol extracts of bamboo bamboo plants (Polygonum pulchrum Blume). Thisstudy used brine shrimp lethality test (BSLT) method. The total number of larvae used ineach concentrations in three times replications was 330 larvae. Each group was given consecutivelyroots, stems, leaves and flowers ethanol extracts of P. pulchrum Blume plants withvariation concentrations 12.5 μg/ml, 25 μg/ml, 50 μg/ml, 100 μg/ml, 125 μg/ml, 250 μg/ml, 500 μg/ml, 1000 μg/ml, 2000 μg/ml, and 4000 μg/ml. The number of dead larvae after24 hours treatment were calculated to obtain the mortality percentages and to determine theLC50 value, which were determined by probit analysis using MiniTab application version17.1.0. Ethanol extract of P. pulchrum roots and stems resulted LC50 values of 933.08 μg/mland 919.58 μg/ml, respectively. While the LC50 values of leaves and flowers extracts were2207.06 μg/ml and 1081.90 μg/ml, respectively. In conclusion, P. pulchrum Blume roots andstems were classified as toxic according to BLST method, while leaves and flowers were not.Keywords: acute toxicity, brine shrimp lethality test, Polygonum pulchrum Blume

Abstract

The emergence of resistant bacteria strain has become a global health concern. It encourages the exploration of potential antibacterial agents, particularly from natural sources. The aim of this study was to investigate the antibacterial activity of ethanol extract of root, stems, leaves, and flowers of Polygonum pulchrum Blume against Staphylococcus aureus and Escherichia coli, through disc diffusion method using cup-plate method. Inhibition zone against S. aureus from roots, stems, leaves, and flowers ethanol extract were 3.5 mm, 2.5 mm, 2.25 mm, and 2.62 mm, respectively, while the inhibition zone against E. coli were 2.25 mm, 2.12 mm, 1.62 mm, and 1.75 mm, respectively. In conclusion, ethanol extract of root, stem, leaves, and flower of P. pulchrum Bl possessed weak antibacterial activity against S. aureus and E. coli.Keywords: P. pulchrum Bl, antibacterial, E. coli, S. aureus, cup-plate technique
