Abstract

The production of polyploidy with a chemical mutagen such as colchicine is one strategy that can be applied to improve the genetic traits and wood properties of acacia trees. The aim of this study was to determine the effects of colchicine at various concentrations and immersion times on the percentage of surviving explants, percentage of rooted explants, and morphological traits of shoot and root A. crassicarpa in-vitro. A completely randomized design (CRD) with two factors was used in this study. The first factor was the concentration of colchicine solution with five levels (0,00%, 0,02%, 0,04%, 0,06%, and 0,08%), and the second factor was the immersion time with three levels (24, 48, and 72 hours). There were fifteen treatment combinations with each treatment repeated three times. Data were analyzed by ANOVA and Duncan’s multiple range test at 5% significance level. The results showed that the treatment combination of colchicine concentration with immersion time showed a significant effect on the percentage of surviving explants, number of leaves, number of shoots and height of A. crassicarpa explants, but did not significantly affect on the number of roots, length of the root and percentage of rooted A. crassicarpa explants.

Abstract

The floodplain river in Riau is inhabited by various types of vertebrate animals including fish from the genus Ompok. The rapid development of species in the Ompok genus can cause ambiguity in species identity due to inaccurate identification processes. This research aims to analyze the molecular characteristics of two types of selais fish, namely danau selais fish (O. hypophthalmus) and kaporeh selais fish (O. eugeneiatus) from Riau based on 12S rRNA sequences. Methods included sampling of fishes from the Kampar, Tapung and Indragiri Rivers, the total DNA isolation, PCR, electrophoresis, sequencing and data analysis. In this study, the 12S rRNA DNA sequence of the selais fish studied was 939 bp in size. The sequences consisted of 803 bp of conserved sites (86.15%), 127 bp of variable sites (13.52%), 53 bp of parsimony sites (5.64%) and 73 bp of singleton sites (18.57%), with the composition of Adenine 31.7 %, Cytosine 26.9%, Thymine 20.9% and Guanine 20.4%. The average genetic distance between the two species was ±0.034, with the lowest genetic distance identified in the species O. hypophthalmus and O. eugeneiatus from the Indragiri River with a value of ±0.032, while the farthest genetic distance was identified in the species O. eugeneiatus from the Kampar River and the species C. gariepinus (Out group) with a value of ±0.092. Transition substitution (19.50%) occurs more frequently than transversion (10.94%). Phylogenetic reconstruction showed that the two species can be separated into distinct groups based on 12S rRNA sequences.

Abstract

Species within the Pandanaceae family, particularly those in the Pandanus genus, exhibit substantial morphological variation, often leading to challenges in accurate species identification. This study conducted a multilocus DNA barcode analysis using sequences of matK, atpB-rbcL Intergenic Spacer (IGS), and trnL-trnF Intergenic Spacer (IGS) obtained from GenBank. Sequence alignment was performed using MAFFT v.7, and the aligned sequences were concatenated into multilocus datasets. Further analyses included frequency data evaluation and phylogenetic tree construction using the Neighbor Joining method in MEGA11. Results revealed that matK had the highest number of conserved sites and the greatest genetic stability, making it particularly effective for identifying broader taxonomic groups. In contrast, atpB-rbcL IGS and trnL-trnF IGS showed greater genetic variation, useful for distinguishing species with similar morphological features. Phylogenetic trees based on single- or two-locus data often failed to group species accurately, while multilocus combinations yielded well-resolved and more reliable tree structures. Therefore, the combined use of atpB-rbcL IGS + matK + trnL-trnF IGS significantly improves species resolution and reduces taxonomic misidentification within Pandanaceae. This study underscores the importance of multilocus barcoding in addressing limitations of morphological classification and enhancing molecular taxonomy in morphologically complex plant groups.

Abstract

Maman plant (Cleome gynandra L.) is an underutilised leafy vegetable that has a high nutritive value and contains phenolic compounds that are essential in reducing or preventing the occurrence of chronic and infectious diseases. In this study, we investigate the effect of drying methods on the total phenolic content of maman plant from Riau. The drying treatment process for maman leafs was tried using three methods: room air-dried without sunlight exposure at 21-24oC for seven days, freeze-dried at -40oC for four days, and oven-drying at 40oC for three days. Freeze-dried maman leafs exhibited significantly (p < 0.05) higher total phenolics (5,90 mgGAE/g) than those dried using other methods, followed by oven-dried (5,14 mgGAE/g) and room-air dried (4,50 mgGAE/g). Drying maman leaves for further research is recommended using the freeze-dried method to obtain optimal phytochemical content.

Abstract

Ikan betok (Anabas testudineus, Bloch 1792) is one of fishes which is a member of family Anabantidae. Betok is an endemic fish in Riau Province. The scientific study about DNA barcoding of betok fish is still rarely. This research aims to analyze the DNA barcode sequence of cytochrome c oxidase I (COI) on betok fish. Methods were used in this study such as sampling, total DNA extraction, PCR, electrophoresis, sequencing and data analysis. The COI sequence studied had size of 694 bp. The BLASTn analysis showed that the betok fish had the highest similarity of 99.7% with A. testudineus and the lowest 93.00% with A. cobojinus. There was one nucleotide that characterize the betok fish based on the COI sequence namely nucleotide number 309. This study may enrich the DNA barcode database of betok fish in GeneBank.

Abstract

The traditional agroforestry industry is part of the Forestry and Other Land Use Net Sink 2030 (FOLU Net Sink 2030) strategy for low carbon development and climate resilience in Indonesia. This study aims to estimate the value of biomass, carbon stock, and carbon sequestration in traditional agroforestry systems in Rimbo Panjang Village, Kampar. Six plots (each measuring 20 x 20 meters) are used; three of the plots are 15 years old and three are 9 years old, respectively, for the conventional agroforestry system.  Biomass, carbon stock and carbon sequestration were estimated at the tree, pole and sapling levels.  Biomass calculations used allometric equations based on diameter at breast height (DBH) data. The results showed that carbon sequestration in Dusun 2 (618.2 tons/ha) was greater than in Dusun 1 (399 tons/ha), with an average carbon sequestration in both dusun of 508.6 tons/ha.  Trees contribute the highest value of biomass, carbon stock, and carbon sequestration, which is 74%, The difference in the value of biomass, carbon stock, and carbon sequestration in traditional agroforestry systems in Rimbo Panjang Village is significantly influenced by the average stem diameter. The various trees that make up traditional agroforestry in Rimbo Panjang Kampar, which is generally in the yard, should ideally be maintained and developed because of their high carbon sequestration potential.

Abstract

The kitolod plant (Hippobroma longiflora) is a traditional medicinal plant originating from the Campanulaceae family. DNA barcoding is a technique for identifying an organism using short nucleotide sequences known as DNA barcoding. One of the DNA barcodes in plants is matK. This research aims to analyze DNA barcode sequence in the matK region of the kitolod plant using the DNA barcode. Samples were taken from the area of Tarai Bangun Village, Kampar Regency, Riau Province, Indonesia as many as two different individuals. The research stages carried out are sampling, followed by DNA isolation using the Geneaid Mini Plant kit, PCR follow thermo scientific instructions, gel agarose electrophoresis, sequencing, and data analysis using the bioinformatics program, namely BioEdit, BLASTn, and MEGA 6.0. The matK sequence of the kitolod plant obtained was 841 bp. The result showed that no identity value was found to reached 100%. The highest identity value (99.88%) was found in H. longiflora NC_035361.1. 34 nucleotide variations were found with one critical nucleotide for H. longiflora from Riau and six critical nucleotides for the H. longiflora. The matK DNA sequence from Kitolod Riau in this study is the first sequence reported in database GenBank.
