Abstract

Penelitian ini bertujuan untuk menganalisis profil nutrisi dari non edible portion ikan betik (Anabas testudineus) dan mengkaji potensinya sebagai suplemen penambah nafsu makan. Pembuatan ekstrak tulang dan sisik ikan betik dilakukan dengan beberapa tahapan yaitu tahap persiapan, ekstraksi, pengujian proksimat, pemberian perlakuan, penimbangan sisa pakan serta penimbangan berat badan hewan coba. Data berat badan hewan coba dianalisis menggunakan uji Kruskal Wallis. Hasil analisis proksimat menunjukkan bahwa komponen tulang dan sisik ikan betik mengandung Kadar Air sebesar 5,18%, Kadar Abu 47,58%, Kadar Lemak 7,41%, Kadar Protein sebesar 33,19%, Kolesterol (-), Fe 21,29mg/100g, Se 0,50mg/g, Zn 1,10mg/100g, Ca 22,7g/100g, Fosfor 9,38g/100g, K 0,28g/100g, Na 1193 mg/kg, Karbohidrat 6,64%. Hasil uji Efektivitas Ekstrak tulang dan sisik ikan betik sebagai suplemen penambah nafsu makan didapatkan kelompok dosis 12mg memiliki nafsu makan tertinggi. Namun berdasarkan hasil uji Kruskal-Wallis menunjukkan bahwa tidak ada perbedaan yang signifikan di antara perlakuan dengan nilai signifikansi sebesar 0.121>0.05.

Abstract

The process of composting inked waste paper can be done because inked waste paper contains organic material in the form of cellulose, hemicellulose, lignin and ash compounds. But unlike other organic materials inked used paper has undergone various chemical processes in its manufacture so it needs to be done soaking treatment in advance to reduce the content of various chemicals. In this case inked scrap paper is soaked with distilled water added with activated charcoal, corn cobs charcoal, PGPR and rumen liquid. The PGPR was then added to a phosphate buffer of pH 7 and incubated for 7 days at room temperature without exposure to sunlight. Rumen fluid is taken at the Jenu Slaughterhouse in Tuban Regency, this liquid is taken directly after the cow is slaughtered and put into a flask then phosphate buffer pH 7 is added and incubated for 7 days at room temperature and not exposed to sunlight. Provision of activated charcoal, corn cobs charcoal and PGPR can be done in the process of precomposting inked waste paper with a cow rumen liquid starter without causing pH and temperature fluctuations. Abstract

The community service activity was carried out in Margosuko Village, Bancar District, Tuban Regency aims to improve the soft skills and hard skills of farmers group to reduce environmental pollution caused by poorly managed livestock waste. Livestock waste will be processed into solid and liquid organic fertilizers, so it will reduce the dependence on chemical fertilizers through the initiation of independent organic fertilizer production from livestock waste. The implementation method begins with participatory preparation (coordination of farmer groups, preparation of Local Microorganisms (MOL) from mimosa roots, and provision of a compost reactor). The activity then continued with blended learning training for 20 farmers with theoretical material on the impact of waste and simultaneous fermentation practices for 2 weeks. The next stage is 2 months of intensive mentoring to solve technical problems in the field. The successful of the program has seen from the increase in the level of empowerment in managing livestock waste. The indicators used are the level of knowledge and the amount of livestock waste that can be managed. The evaluation results show that there has been an increase level of knowledge regarding cattle farm waste management as seen from the average pretest score which was initially 60.1 to 87.55 in the posttest. Meanwhile, the amount of livestock waste that has been successfully processed on to organic fertilizer is around 30%.

Abstract

Tujuan penelitian untuk mengukur efektivitas ekstrak kulit buah gayam sebagai agen antibakteri pada bakteri pembusuk ikan tongkol. Pembuatan ekstrak kulit buah gayam dilakukan dengan beberapa tahapan yaitu (1) sterilisasi alat bahan, (2) peremajaan isolat bakteri Staphylococcus sp. dan Bacillus sp. pembusuk ikan tongkol, (3) ekstraksi kulit buah gayam, dan (4) uji aktivitas antibakteri. Data uji efektivitas daya hambat ekstrak kulit buah gayam pada bakteri pembusuk ikan tongkol dengan konsentrasi 40%, 50%, 60%, dan 70% menunjukkan terbentuknya zona bening di sekitar paper disc biakan bakteri genus Staphylococcus dan genus Bacillus dengan hasil kategori lemah yaitu ≤5mm. Penelitian ini juga menunjukkan hasil bahwa kulit buah gayam mengandung senyawa metabolit sekunder berupa flavonoid. Tidak terdapat perbedaan bermakna pada aktivitas antibakteri ekstrak kulit buah gayam dengan berbagai konsentrasi untuk tumbuhnya bakteri Staphylococcus sp. dan Bacillus sp. secara in vitro.

Abstract

Kombucha merupakan salah satu produk minuman yang dihasilkan melalui proses fermentasi. Fermentasi kombucha melibatkan penggunaan SCOBY yang mengandung bakteri dan ragi. Kombucha awalnya dikembangkan dengan fermentasi terhadap daun teh dan diketahui memiliki banyak manfaat Kesehatan salah satunya sebagai antioksidan. Saat ini eksplorasi terhadap bahan dalam pembuatan kombucha terus dikembangkan. Salah satu bahan potensi dalam pengembangan produk kombucha adalah sabut siwalan. Sabut siwalan merupakan salah satu limbah agroindustri yang keberadaannya cukup melimpah dengan pemanfaatan yang masih belum optimal. Sabut siwalan diketahui memiliki beberapa kandungan bioaktif, sehingga potensial untuk dikembangkan sebagai bahan pembuatan kombucha. Penelitian ini bertujuan untuk mengevaluasi karakter sensoris dan aktivitas antioksidan dari kombucha sabut siwalan. Metode dalam penelitian ini adalah eksperimental dengan Rancangan Acak Lengkap. Perlakuan dalam penelitian ini adalah lama waktu fermentasi yang terdiri dari 7, 10, dan 14 hari dengan ulangan sejumlah 6. Evaluasi karakter sensori dilakukan dengan uji organoleptik yang melibatkan 15 responden agak terlatih. Sedangkan aktivitas antioksidan diuji dengan metode DPPH. Hasil penelitian menunjukkan bahwa lama waktu fermentasi berpengaruh terhadap karakter aroma, rasa, dan aktivitas antioksidan, akan tetapi tidak berpengaruh terhadap karakter warna. Waktu fermentasi 10 hari menghasilkan rerata skor tertinggi dalam parameter warna dan rasa, sedangkan skor tertinggi parameter aroma adalah waktu 7 hari fermentasi. Sementara aktivitas antioksidan tertinggi ditunjukkan oleh kombucha sabut siwalan yang difermentasi selama 14 hari.

Abstract

This research aims to determine the characterization of the rhizosphere bacteria of the roots of Mimosa pudica in its contribution as a decomposer agent that will be used in making of biourine. The research held in June-August 2023, was carried out in several stages, like the preparation stage, the bacterial isolation stage, and observing the macroscopic and microscopic characteristics of the bacteria. The results of the macroscopic and microscopic characteristics obtained during the observations showed that the isolates in the MOL of the roots of Mimosa pudica resembled the characteristics of bacteria from the genera Bacillus (MAP6), Rhizobium (MAP4), Pseudomonas (MAP1, MAP2, MAP3, MAP5, and MAP8), and Klebsiella (MAP7). These four genera are types of soil bacteria that are capable of producing plant hormones that can speed up the process of nutrients absorption and breakdown of organic material in the soil optimally.

Abstract

Pioneer plants are plants with high adaptability, so they can grow in disturbed environments, such as ex-limestone mining areas. This adaptability is supported by the ability of these plants to associate with soil bacteria in their rhizosphere area. The presence of these bacteria can support the growth of pioneer plants on disturbed land. The purpose of this study was to characterize the soil bacteria around the rhizosphere of pioneer plants in the former limestone mining areas. Soil samples in the rhizosphere area of 5 pioneer plants from ex-mining lime were then isolated using the Pour plate method to obtain bacterial isolates. The bacterial isolates obtained were then identified their colony morphology. Based on the results of colony morphological identification, 21 isolates were obtained from the rhizosphere of 5 pioneer plants from the former limestone mining area. The highest number of isolates was obtained from the rhizosphere of the Waltheria indica with 9 isolates, 8 isolates of each Lamium barbatum and Digitaria sanguinalis, 7 isolates of Chromoleana odoratum, and 5 isolates of Tridax procumbens.

Abstract

Environmental damage included the limestone mining process would remove the topsoil layer, reduce fertility, reduce microbial diversity and eliminate the vegetation that growth above it. Naturally, pioneer plants could grow to replace lost plants. Pioneer plants can be used as a reference for land recovery because these plants could grow on damaged land because they are associated with Plant Growth Promoting Rhizobacteria (PGPR). In addition, the presence of pioneer plants could inhibit soil erosion due to its root ability to hold the soil from scouring water. This study aimed to determine the characteristics of the initial natural succession process, namely the pioneer post-limestone mining community. This research was conducted by purposive random sampling method at three mining locations, namely Banjaragung Village, 5 years after mine, Maibit Village, 8 years after mine, and Rengel Village, 7 years after mine. Samples were taken for each of the 10 plots of 1x1 m size. The results of this study obtained 5 types of plants with the highest INP, namely: Lamium barbatum, Chromolena odoratum, Tridax procumbens, Waltheria indica, and Digitaria sanguinalis. While the highest diversity index is in the Village of Maibit, Rengel Sub-District, Tuban Regency after 8 years post-mining, which is 2.54.

Abstract

Padina australis is a brown algae that has secondary metabolites such as phenols, tannins, flavonoids, saponins, and alkaloids that have antibacterial potential. This study aims to determine the antibacterial activity of Padina australis extract against spoilage bacteria of tuna (Euthynnus affinis) from genus Staphylococcus sp. and Bacillus sp. through disc diffusion method. Data on the diameter of the inhibition zone were analyzed statistically. The results showed that Padina australis extract was able to produce an inhibition zone in both isolates, but the antibacterial activity was relatively weak (≤ 5mm). The 60% extract concentration produced the highest inhibition zone of 2.07 mm in Staphylococcus sp. and 2.23 mm Bacillus sp. isolates, while the positive control produced strong to very strong antibacterial activity, with diameters of 19.28 mm and 33.76 mm. The results of statistical analysis showed that variations in extract concentration did not significantly affect the inhibition zone in both isolates (p = 0.119 and p = 0.160). The low antibacterial activity is likely due to limited content of bioactive compounds, the extraction method, and suboptimal diffusion of the extract on the agar medium. Padina australis extract has the potential as a natural antibacterial, but its effectiveness still needs to be improved with further optimization.

Abstract

Daging ayam merupakan produk hewani yang memiliki kandungan protein dan nutrisi yang tinggi sehingga berperan dalam pemenuhan gizi masyarakat. Namun, daging ayam termasuk bahan pangan yang mudah rusak karena kandungan air dan protein yang tinggi, sehingga menjadi media yang baik bagi pertumbuhan mikroorganisme. Kontaminasi bakteri pada daging ayam dapat terjadi akibat penanganan yang kurang higienis, mulai dari peternakan, penyembelihan, pengolahan, hingga penyimpanan. Salah satu bakteri patogen yang dapat mengontaminasi daging ayam adalah bakteri Salmonella sp. yang merupakan indikator penting dalam keamanan pangan. Bakteri Salmonella sp. dapat menyebabkan infeksi saluran pencernaan atau penyakit salmonellosis. Oleh karena itu, diperlukan pengawasan dan pengujian mikrobiologi untuk memastikan mutu dan keamanan daging ayam sebelum dikonsumsi. Penelitian ini bertujuan untuk mengevaluasi keamanan pangan daging ayam pada Rumah Potong Ayam di daerah Jawa Timur berdasarkan indikator keberadaan bakteri Salmonella sp.. Pengujian dilakukan terhadap delapan sampel daging ayam dengan metode yang mengacu pada SNI 2897:2008. Penelitian ini dilakukan dengan beberapa tahapan meliputi tahap pra-pengayaan, pengayaan, isolasi dan identifikasi, serta uji konfirmasi/biokimia. Berdasarkan hasil pengujian, seluruh sampel daging ayam dinyatakan negatif terhadap cemaran bakteri Salmonella sp. per 25gram sampel. Meskipun pada tahap isolasi awal terdapat satu sampel yang diduga menunjukkan karakteristik Salmonella sp. pada media XLD, hasil uji biokimia lanjutan menunjukkan bahwa sampel tersebut bukan merupakan bakteri Salmonella sp., dan dengan demikian daging ayam dari Rumah Potong Ayam tersebut telah memenuhi persyaratan mutu mikrobiologis dan aman untuk dikonsumsi.
