Abstract

Mangrove, Rhizophora mucronata, grows in the intertidal area, which contains much organic matter and varying salinity. The organic matter content is influenced by the bacterial community that inhabits the ecosystem, but information regarding the bacterial community, especially in the mangrove root system, is not widely available. There are several challenges in completing this information, one of which is that the method used is still in a conventional form. Developments in environmental DNA analysis can support and complement this information. However, this method must be optimized because the organic matter content and salt variations affect the extraction results. Thus, this study aimed to determine the optimal approach for extracting bacterial DNA from mangrove sediments. The analysis used two methodologies: manual DNA extraction techniques based on buffer modification and DNA extraction kits. There were four different treatments, namely the soil DNA isolation plus kit (M1), the fecal / soil microbial quick-DNA miniprep kit (M2), glass powder with charcoal (M3), and glass powder with skimmed milk (M4). DNA samples were obtained from each method and assessed for concentration and purity using a nanodrop. In addition, the resulting DNA's quality was analyzed using 1.5% agarose. The results obtained were in the M2 treatment, which showed optimal results compared to the others. M2 uses a bead-based beating and spin column method to achieve optimal DNA concentration through high molecular weight. The DNA obtained was also protein-free, and several samples were contaminated with humic acid, namely KL.S1, KL.S4, and T7.S4.Keywords:Bacteria 16SBead beatingDNA ExtractionSedimentSpin column

Abstract

Perancangan  ini bertujuan untuk merancang sebuah wisata mangrove ecopark yang  ada  di  Torosiaje Kabupaten Pohuwato  dengan  menggunakan sebuah  pendekatan Arsitektur Ekologi.   Wisata merupakan sebuah kegiatan yang dilakukan seseorang atau sekelompok orang untuk melepas penat atau berekreasi dan bersantai. Wisata juga menjadi salah satu kegiatan yang dapat membantu peningkatan pendapatan daerah. Adapun penelitian pada perancangan ini dilakukan dengan cara observasi, dokumentasi dan studi kepustakaan. Dari hasil data-data yang ada  kemudian menghasilkan sebuah rancangan kawasan wisata mangrove. Adapun tahap yang dilakukan sebelum perancangan yaitu pembuatan konsep pendekatan yang di gunakan agar kemudian akan menghasilkan sebuah gambar perancangan Mangrove Ecopark. Adapun pendekatan yang di gunakan yaitu pendekatan ekologi yang bertujuan agar pembangunan Ecopark tidak merusak lingkungan dan menggunakan material yang dapat di perbaharui sehingga tidak menyebabkan pencemaran pada lingkungan. Penerapan Arsitektur Ekologi pada perancangan terletak pada penataan bangunan yang berada di antara pepohonan mangrovesehingga nantinya tidak merusak kawasan mangrove yang ada.

Abstract

Employing Mind Mapping Technique to Facilitate Secondary School Student to Write Narrative Text At The First Grade In the 2020/2021 Academic Year. This research was conducted on class XI students of SMA Negeri 1 Wongsorejo Banyuwangi. This research used Class Action Research with research design using Pre-Test and Post-Test in cycle 1 and cycle 2. The data of this study were in the form of student scores both pre-test and post-test. The data collection technique was carried out by researchers by collecting student scores, observation sheets on student activities both before research, and after research, interviews with students and teachers on learning before and after using the mind mapping method. The results showed that: learning using the mind mapping method was classified as very supportive of students' abilities in learning writing narrative text. This is evidenced by the increase in student test results from cycle 1 with an average result of 72.79 students with a completeness level of 59%, increasing in cycle 2 with an average value of 79.26 with a completeness level reaching 85%. Student responses to learning using the mind mapping method are very happy, and students are quite active in group learning activities. In cycle 1 students do individual learning activities but the results are still not optimal because students still do not understand the mind mapping method, but in cycle 2 with group learning students experience an increase in ability because students can discuss with their group friends.
