Abstract

Secondary metabolites of filamentous mold are mycotoxins, which in some situations can develop on foodsderived from plants or from animals. Fusarium, Aspergillus, and Penicillium are the most common types ofmold that produce mycotoxins and also often contaminate human food and animal feed. Aflatoxins includingaflatoxin B1, B2, G1 and G2 are produced by A. flavus and A. parasiticus. M1 and M2 aflatoxins arefound in dairy products. In this study, we used PCR to detect and identify mycotoxigenic fungi material infoods from traditional markets and supermarkets in Surabaya, Indonesia. Samples of chicken meat from atraditional market and from a supermarket (10 pieces each) were placed in a conical tube and crushed in PBS.The crushed samples were centrifuged and the supernatants were cultured on potato dextrose agar (PDA)media and observed using a reverse microscope. DNA was isolated from cultured samples and subjectedto PCR with primers specific for genes encoding aflatoxins. Interestingly, we revealed that the PolymeraseChain Reaction (PCR) analysis showed A. flavus and A. ochraceus were present on chicken meat sold attraditional markets and supermarkets. In sum, enhanced precautions may be needed to ensure that foods soldin traditional markets and supermarkets are free from molds that have the potential to produce mycotoxins.Further studies are needed to detect and identify the prevalence of mycotoxins in the food supply.

Abstract

Iptek bagi Masyarakat (IbM) which performed on a group of local cattle fattening is not qualified yet to solve the problems 1) rice straw as animal feed 2) agricultural waste and industrial waste as feed without forage 3) processing of rice straw into straw tape 4) processing agricultural waste and industrial waste into animal feed without forage 5) processing growth promotor derived from empon-empon and retrieval medicinal plants, and 6) making of cow manure waste into environmentally friendly fertilizer. The purposes of IbM are (1) to process of rice straw into straw tape as a nutritious animal feed (2) without forage feed processing (complete feed) (3) processing of growth promotors (4) local cattle fattening using tape straw and feed without forage as well as growth promotor. Benefits of IbM is to develop human resources and increase income of fattening local cattle farmers with lower production costs. The method used in IbM activities are education and training of local cattle fattening using tape straw feed and feed without forage plus growth promotor, processing waste into environmentally friendly fertilizer on UKMK of local cattle fattening members in Babat and Sugio district in Lamongan. IbM program activities involving 8 students to increase entrepreneurial spirit. The result is enhancement of cattle weight per cow per day on local cattle amounted to 0.578+0.054 kg, ongole crossbreed cattle 0.887+0.100 kg, hybrid limousine cattle for 1.588+0.084 kg and Simmental crossbreed cattle of 1,967+0.255 kg. AbstrakIptek Bagi Masyarakat (IbM) yang dilakukan pada kelompok penggemukan sapi lokal belum memenuhi syarat untuk menyelesaikan masalah 1) jerami padi sebagai pakan ternak 2) limbah pertanian dan limbah industri sebagai pakan tanpa hijauan 3) pengolahan jerami padi menjadi pita jerami 4) pengolahan limbah pertanian dan limbah industri menjadi pakan ternak tanpa pakan ternak 5) mengolah promotor pertumbuhan yang berasal dari empon-empon dan pengambilan kembali tanaman obat, dan 6) pembuatan limbah kotoran sapi menjadi pupuk ramah lingkungan. Tujuan IbM adalah (1) mengolah jerami padi menjadi pita jerami sebagai pakan ternak yang bergizi (2) tanpa pengolahan pakan ternak (pakan lengkap) (3) pengolahan promoter pertumbuhan (4) penggemukan sapi lokal dengan menggunakan jerami dan pakan pita tanpa pakan ternak serta promotor pertumbuhan. Manfaat IbM adalah mengembangkan sumber daya manusia dan meningkatkan pendapatan penggemukan peternak sapi lokal dengan biaya produksi lebih rendah. Metode yang digunakan dalam kegiatan IbM adalah pendidikan dan pelatihan penggemukan sapi dengan pakan jerami dan pakan tanpa pakan ternak ditambah promotor pertumbuhan, pengolahan limbah menjadi pupuk ramah lingkungan pada UKMK penggemukan sapi potong di Kabupaten Babat dan Sugio di Lamongan. Kegiatan program IbM melibatkan 8 siswa untuk meningkatkan semangat kewirausahaan. Hasilnya adalah peningkatan bobot sapi per ekor per hari pada ternak lokal sebesar 0,578 + 0,054 kg, sapi potong silang ongole 0,887 + 0,100 kg, ternak limusin hibrida 1,588 + 0,084 kg dan sapi potong silang Simmental 1.967 + 0.255 kg.

Abstract

Dengan adanya kejadian luar biasa yang pertama virus avian influenza H5N1 tahun 1997, semakin jelas bahwa potensi virulensi virus H5N1 telah meluas ke manusia. Review ini disusun untuk memahami karakteristik virus, siklus replikasi virus, mekanisme virus masuk ke dalam hospes, peran hemaglutinin sebagai determinan patogenisitas, urutan basa hemaglutinin yang berperan dalam memicu peningkatan virulensi dan fungsi dari 6 segmen gen lainnya pada virus avian influenza. Review juga dibuat untuk memahami gambaran gen Hemaglutinin avian influenza virus H5N1 yang bertujuan untuk mengetahui penularan secara efisien dan replikasi virus avian influenza pada manusia, sehingga penularan selanjutnya dapat diantisipasi dengan baik. Kerja sama lintas sektor antara kementerian kesehatan, kementerian koordinator kesejateraan rakyat, kementerian lain, universitas dan organisasi yang berkompeten sangat dibutuhkan untuk mendukung pencegahan penyebaran virus avian influenza H5N1 di Indonesia.

Abstract

The aims of this community dedication: 1) Improve the economy of the villagers of Watubonang and Biting, Badegan Ponorogo district through changing mindsets to improve people’s welfare 2) Increasing the knowledge and skills of the villagers of Watubonang and Biting, Badegan Ponorogo district through technology transfer for making pecticide formulations, antiscabies drugs, antiseptik, compost manure disinfectant, dipping solution based on tobacco waste. The result of the communiy dedication implementation showed an increase in participants on how to manufacture various kinds of tobacco waste products from 6.7% to 93.3% and an increase in the skills of participants on how to manufacture various kinds of tobacco waste products from 6.7% to 93.3%. in addition to the increased knwoledge and skills of the community dedication participants in Watubonang and Biting villages, Badegan Ponorogo district, it showed a decrease in the average cost of using medicines, antiseptiks, and disinfectants per house from 210 thousand rupiah to 42 thousand rupiah. Form the results of community dedication in the villages of Biting and Watubonang, Badegan Ponorogo district, it is hoped that they will become fostered villages of FKH Unair.

Abstract

AbstractIn general, Gracilaria culture still using organic and inorganic fertilizer single. Organic or inorganic fertilizer applied is felt have not yet enough to support the growth of Gracilaria. The aim of this experiment was to know the best compost and NPK combinations which could influence on growth, chlorophyll a and water content of G.verrucosa. Treatment applied was compost and NPK combinations with different doses, that is A treatment (0 gr/l :  2 gr/l), B treatment (0,5 gr/l : 1,5 gr/l), C treatment (1 gr/l : 1gr/l), D treatment (1,5 gr/l : 0,5 gr/l), E treatment (2 gr/l : 0 gr/l) and F treatment (without fertilizer), each treatment was replicated 4 times. Main parameter observed was daily growth rate (%/day), relative length growth (%), chlorophyll a (í¬g/ml) and water content (%) of G.verrucosa. oSupported parameter observed was pH, temperature ( C) and salinity ("°). Data analysis using Analysis of Variance (ANOVA) and Duncan Multiple Range Test. Result of  the experiment indicates that compost and NPK combinations with different doses was significantly influence to growth, chlorophyll a and water content of G. verrucosa. The highest mean of daily weight growth rates was C treatment (1 gr/l : 1gr/l) which was equal to 2,56%/day and the lowest mean was  F treatment (without fertilizer) which was equal to 1,12%/day. The highest mean of relative length growth was C treatment (1 gr/l : 1gr/l) which was equal to 2,21% and the lowest mean was F treatment (without fertilizer) which was equal to 0,60%. The highest mean of chlorophyll a by the end of the experiment was C treatment (1 gr/l : 1gr/l) which was equal to 0,001948 í¬g/ml and the lowest mean was F treatment (without fertilizer) which was equal to 0,001440 í¬g/ml. The lowest mean of water content by the end of the experiment was C treatment (1 gr/l : 1gr/l) which was equal to 28,48 % and the highest mean was F treatment (without fertilizer) which was equal to 30,39%.

Abstract

The purpose of this study was to determine the serological study of Newcastle Disease that infects slaughtered ducks (Anas javanicus) at the East Surabaya Traditional Market. This study was conducted from December 2018 to January 2019. A total of 122 samples were collected from four traditional markets in East Surabaya. The Haemagglutination Inhibition (HI) test was used to determine a positive result with an antibody titer of 4 or more. Samples for the HI test were collected from duck serum that had been treated with chicken red blood cells. The results showed 13 (10.65%) of the 122 samples were positive for ND.

Abstract

This study aims to determine the effect of hepatitis B vaccine of spleen weight and white pulp diameter in rat (Rattus norvegicus). This study used 20 male wistar rats aged 8 weeks with an average weight of 200-250 grams, which was divided into four groups. Control group (P0) was given PBS solution, treatment group 1 (P1) was given type 1 hepatitis B vaccine, treatment group 2 was given type 2 hepatitis B vaccine, and treatment group 3 (P3) was given type 3 hepatitis B vaccine. Dose administered 0.4 ml and given repeatedly at days 0,30, and 60. Animals model to euthanize on day 75 for sampling. The spleen organ is weighed after the necropsing process and then the spleen is stored for the preparation of histopathologic with HE staining (Hematoxylin Eosin). The data obtained are weight data of spleen organ and spleen white pulp diameter. The results showed the highest weight of spleen organ 0.92 gram in group P0 and the largest white pulp diameter 34,34 μm in group P1. The results of data analyzed using one-way ANOVA (Analysis of Variance) and continued with Duncan alpha test. One Way Anova and Duncan alpha test showed that there were non significantly different between groups (p>0.05). The conclusion is we know that hepatitis B vaccine type 1,2, and 3 can't give the effect for spleen weight and white pulp diameter in rat (Rattus norvegicus).

Abstract

The aim of this research was to know histopathological changes of kidney of broiler chicken exposed to chronic heat stress. Twenty broilers were divided randomly into 2 groups, 10 broilers each are being exposed to chronic heat stress for 21 days. The first 21 days were the adaptation period in a chamber with temperature 24-28o C and humidity 40-55%. After 21 days, continue with exposure to heat stress in a chamber with temperature 36-40o C and humidity 50-65% in 8 hours per day. After exposed to chronic heat stress, kidney tissues were processed, and kidney tissue histopathological changes were evaluated by using the Klopfleisch modified scoring method. The data was analyzed by Mann – Whitney Test. The result of this research showed that chronic heat stress exposure causing the presence of degeneration of tubular epithelial cell, necrosis of tubular epithelial cell, necrosis of glomerular and interstitial infiltration. From the analysis data the overviewof multiparametric showed that when control group compared with the treatment group showed significantly difference (p<0.05).

Abstract

This study aims to determine the anthelmintic activity ethanol extract of porang tubers (A. oncophillus) against F. gigantica worms in vitro. There were five treatments and each treatment was done in five replications and used 10 F. gigantica. The treatments of this research were K- with CMC Na 1%, K+ with Albendazole 2.4 mg/ml, P1 with extract concentration 5%, P2 with extract concentration 10%, P3 with extract concentration 20%. The results showed that the extract of Porang tuber (A. oncophillus) had an anthelmintic effect against F. gigantica worms in vitro. In the extract with a concentration of 20%, there were anthelmintic properties that almost the same as Albendazole. The higher the extract concentration, the higher the anthelmintic properties. The longer the immersion time, the higher the number of dead worms. The morphological changes was evaluated by light microscopic examination and the results showed many histopathological changes on the morphology of F. gigantica. The results indicate that the possible use of the tubers as a potential anthelmintic against F. gigantica.

Abstract

The aim of this study was to know the effect of Mimosa pudica root extract on histopathological appearance of Rattus norvegicus brain induced by Naja sputatrix venom. Thirty rats were divided into 5 groups. There were 2 control groups and 3 treatment groups, which was given 250, 500, and 1000 mg/kg BW of Mimosa pudica root extract orally. The first 7 days each group was adapted to the environment. On the 8th day, the treatment was started by injecting Naja sputatrix LD50 (0,13 L/gram BW) IM in gluteus muscle, continued with giving Mimosa pudica root extract orally for the treatment groups 5 minutes after venom injection. 6 hours after the last treatment, rats were killed by cervical dislocation, injected with formalin 10% in the heart, then necropsied. Histopathological evaluation was done to score brain damage based on meningitis, perivascular cuffing, and necrotic cells using HE stain with 1000x magnification. The result showed 1000 mg/kg BW dosage of Mimosa pudica root extract can reduce brain damage based on meningitis, perivascular cuffing, and necrotic cells in Rat (Rattus norvegicus) caused by Naja sputatrix venom and gave significant difference (p < 0.05) among the treatment groups.
