Abstract

L-Arabinose isomerase (L-AI) is a type of intracellular enzyme catalyzes the reversible isomerization reaction of L-arabinose to L-ribulose, besides that it can convert D-galactose to D-tagatose. One of the bacteria that can produce this enzyme is Geobacillus stearothermophilus. The high price of instant media, which reaches IDR 1,500,000 to IDR 3.000.000,- for every 500 g and the abundance of natural resources that can be used as a medium for the growth of microorganisms encourages researchers to find alternative media from materials that are easy to get and do not require expensive costs. The researchers compared the effectiveness of liquid Luria Broth (LB) media and molasses media for enzyme production. The research involved inoculating bacteria in both media types, followed by subculture and incubation. IPTG was added to induce enzyme production, followed by cell harvesting through centrifugation and sonication. Enzyme activity was evaluated using SDS PAGE and the cysteine carbazole method. Molasses showed promising results due to water solubility, enzyme activity, abundance, and cost-effectiveness. Molasses appears as an alternative with active enzymes, water solubility, abundant availability, and significant cost savings, making it a viable choice for enzyme production. Keywords— L- Arabinose Isomerase, Molasses, Enzyme

Abstract

Abstract

Abstract

Spirulina sp. is a highly promising microalgae with substantial protein and pigment content, holds great economic potential across various industries. However, the cultivation of Spirulina sp. often faces the challenge of high production costs, particularly in media formulation. To address this issue, the development of more affordable alternative media formulations is crucial. One potential approach involves utilizing a combination of liquid organic fertilizer derived from Canna indica waste and urea for cultivation. This study aims to determine the optimal ratio of the fertilizers mixture that influences protein and pigment synthesis. The study design employed a completely randomized design with seven treatment groups along with the control. The result shows that the treatment 2A (20 mL/L organic fertilizer; 125 ppm urea) demonstrated the highest dry biomass weight. Treatment 2C (22 mL/L organic fertilizer; 150 ppm urea) exhibited the best specific growth rate and carotenoid production, with the carotenoid production significantly differing from the control. Treatment 1A (20 mL/L organic fertilizer; 100 ppm urea) yielded the highest levels of chlorophyll and phycobiliproteins. Treatment 1B (20 mL/L organic fertilizer; 125 ppm urea) resulted in the highest protein production.

Abstract

Spirulina sp., a type of blue-green microalgae, possesses various beneficial properties such as antioxidants, probiotics, proteins, pigments, and carbohydrates, enabling it to generate a wide range of useful products. Previous studies using commercial fertilizers as the growth medium, have not achieved optimal pigment and protein production. Therefore, this study aims to investigate the impact of combining liquid organic fertilizer derived from Canna indica waste with TSP (Triple Super Phosphate) fertilizer on changes in pigment and protein production in Spirulina sp. The study was conducted for 16 days using a completely randomized design, with four replicates for each of the six treatments, along with a control. The results demonstrated a prolonged logarithmic growth phase, with the highest dry biomass weight observed on the 16th day in the treatment 1A (1,585 gr/L). Treatment 1A exhibited the highest levels of allophycocyanin and phycoerythrin pigments (9,861 mg/L and 4,319 mg/L, respectively), while treatment 1C yielded the highest protein content (0.33%). Statistical analysis on day 12, with a 95% confidence level, revealed significant differences in pigment production between various treatment groups. The study suggests that a phosphor concentration of 20 ppm is optimal for cultivating Spirulina sp.

Abstract

Population increase has an impact on the procurement of food sources. Pakcoy is a vegetable that is in increasing demand. Farmers' response to this condition is to use excessive fertilizer and pesticides, thereby reducing soil fertility. The research aims to obtain a combination of fertilizer and humic acid, which is effective in cultivating B. rappa chinesis L., as well as soil conservation efforts. The methodology was experimental with Randomized Block Design (RBD) on four treatments, K, P1, P2, and P3. The dose of K was 150 g/5 m2, NPK 16:16:16, 1 kg/5 m2 dolomite, and 5 kg/5 m2 manure. The P1 treatment is the same as K but without NPK fertilizer. P2 and P3 treatments were reduced by 50% and 70% of NPK fertilizer, respectively, with the addition of humic acid. The research results show that P2 with local environmental conditions provides optimal results in terms of chlorophyll, stomata, and plant height. Soil fertility was shown by increasing pH to 6.6, K 281 ppm and P 509 ppm, Cation Exchange Capacity (CEC) 31.60 cmolc/kg, soil enzymes of 0.12 mg FDA g-1 soil d.w. h-1, an average number of earthworms of 17. The conclusion is that P2 fertilizer formulation can be applied to cultures as an anticipation of food security.

Abstract

Waste management in traditional markets is a major challenge in the context of urbanization and environmental sustainability. This study aims to analyze the sustainability index of waste management in Pasar Kebayoran Lama, Jakarta, using a three-dimensional approach: ecological, social, and economic. The methods used are literature study and assessment simulation based on indicators relevant to national policies, such as Law No. 18 Year 2008. The analysis showed that the overall sustainability index score was 1.5, indicating unsustainable conditions. The ecological aspect scored 1.3, social 1.6, and economic 1.6, reflecting the low effectiveness of waste management, lack of community participation, and not optimal waste-based economic empowerment. The proposed action plan includes increased education, waste segregation and recycling, and the development of a circular economy model. This study recommends cross-sector collaboration and continuous monitoring to realize a more ethical, efficient, and sustainable market waste management.
