Abstract

Coliform bacteria are often referred as indicator organisms to indicate the potential presence of disease-causing bacteria. Escherichia coli is a type of Coliform bacteria. Most Escherichia coli are harmless and commonly found in the intestines of humans and animals. However, some strains can cause illness. Coliform bacteria and Escherichia coli contamination in chicken could be detected using Most Probable Number (MPN) multi-step assay which consists of presumptive, confirmed, and completed test. The aim of this study was to determine the number of Coliform bacteria contamination and to detect Escherichia coli in 15 chicken meat samples collected from 3 traditional markets and 3 modern markets in Pekanbaru. Estimated numbers ofColiform bacteria in samples were varied from 2.1x10 4 MPN/g to >11x10 MPN/g. We found that chicken samples were contaminated by Escherichia coli, which showed colour changes and bubles production in EC broth and the appearence of dark centered colony with metallic shine on EMB Agar.

Abstract

Cellulolytic bacteria are  bacteria that can degrade cellulose into smaller polysaccharides called cellodextrins or completely into glucose units. Glucose is used as a carbon source for bacterial growth. The aims  of this study were to isolate cellulolytic bacteria from estuary water of Siak watershed in  Bengkalis  area and to determine its  cellulolytic activity and specific activity. A total of 13 isolates of cellulolytic bacteria have  been isolated using commercial cellulose substrate containing 1% w/v carboxymethyl cellulose (CMC). We found that 11 of the 13 isolates were Gram negative bacteria, while others were Gram positive. The isolate of C3 from Pelabuhan Roro showed the highest cellulolytic activity [(1.701 ±  0.166)  x 10-3 U/mL]  which was  not significantly different (p≤0.05) with cellulolytic activity of the  A3  isolate  [(1.605 ± 0.041) x 10-3 U/mL]. The C3 isolate also had the highest Cellulolytic specific activity [ (31.037  ± 4026) x 10-3 U/mg] and it was significant (p≥0.05) different with other isolates

Abstract

Cellulolytic bacteria has been isolated from water and sediment sample of Indragiri river waters. Cellulase was produced by cellulolytic bacterial isolates in 1% Carboxy Methyl Cellulose (CMC) liquid medium at 37°C, 150 rpm for 24 h. The specific activity of cellulase was determined by Nelson-Somogy’s method (cellulase enzyme activity) and Lowry’s method (protein concentration). 914-5 sedimen isolates showed the highest activity of the enzyme 12,246.10 -4 ± 0,5885.10 -4 U/mL from 17 other isolates that was significantly different at (P≤0,05) compared with the other isolate.

Abstract

Isolation of cellulolytic bacteria has been investigated because this bacteria producescellulase enzyme which has important role in various industry applications. In this research, isolation of cellulolytic bacteria was originated from sediment samples that was taken from 4 sampling points in estuary watershed of Siak River Bengkalis area and Dumai waters, which were 925 point (Siak River), 929 point (Pelabuhan Roro Pakning), 930 point (Bengkalis Strait), and 931 point (Dumai). Isolates of cellulolytic bacteria were identified with Gram staining method, the enzyme activities were measured with Nelson-Somogyi method. The results of identification showed 1 isolate was Gram positive and 15 isolates were Gram negative. The highest cellulase activity was obtained from 931-3S isolate (0.735±0.03) x 10 -3 U/mL, and it was not significant different with 929-1S, 929-2S, 929-3S, 930-3S isolates.

Abstract

Kelompok Musyawarah Guru Mata Pelajaran (MGMP) IPA Kecamatan Kuantan Tengah, yang menaungi guru-guru IPA Sekolah Menengah Pertama di Kecamatan Kuantan Tengah, Teluk Kuantan memerlukan adanya tim pendamping ataupun penyuluh untuk memberikan materi dan praktek tentang sesuatu yang dapat diaplikasikan di Laboratorium IPA Sekolah. Selain untuk kebutuhan praktikum siswa, materi tersebut juga hendaknya dapat diaplikasikan dan dikembangkan Guru-guru untuk riset dan penulisan karya ilmiah yang merupakan syarat perolehan sertifikasi guru. Pada kegiatan ini, tim penyuluh memberikan teori dan aplikasi teknologi fermentasi sederhana yang dapat diaplikasikan di laboratorium IPA Sekolah Menengah Pertama. Praktek dilakukan dengan membuat bersama fermentasi ubi kayu dari berbagai daerah menggunakan beberapa jenis khamir untuk beberapa variasi waktu, sistem aerasi, dan suhu untuk membuktikan adanya factor yang mempengaruhi produk fermentasi. Prinsip-prinsip sederhana tersebut akan dapat dikembangkan dan diaplikasikan Guru IPA dalam skreening mikroba penghasil enzim tertentu, produksi enzim di skala laboratorium, serta analisis aktivitas enzimnya. Guru dan siswa yang mengikuti kegiatan diberikan buku panduan mengenai prinsip dasar fermentasi dan fermentasi enzim. Kegiatan berlangsung lancar dengan antusias yang tinggi dari peserta yang terlihat dari beragamnya pertanyaan yang diberikan selama kegiatan berlangsung.

Abstract

Center for Fisheries Production, Inspection and Certification (PPISHP) is a unit technical implementer within the Department of Food Security, Maritime Affairs and Agriculture of DKI Jakarta Province. PPISHP DKI Jakarta Province does not yet have a comprehensive performance measurement. Performance measurement only uses an assessment on the financial aspect, financial measures alone are not enough to evaluate the overall performance of the agency. Non-financial performance such as productivity and the ability of employees to produce products or services that can meet consumer desires is neglected. To measure the performance of an agency, a comprehensive performance measurement method is needed to achieve the desired goal. Therefore, the Balanced Scorecard method is deemed necessary to be applied to PPISHP DKI Jakarta Province.

Abstract

The piloted-village grant of University of Riau is a community development program which is granted for University of Riau academics to build a village in the sectors of economics and health. In this program, Desa Pulau Banjar Kari was chosen due to their active in developing their agricultural and health. In this program, the academic team from University of Riau supervised the community how to prepare and produce organic fertilizer based on Effective Microorganism technology (active EM, EM5, bokashi, and fermented-plant extracts). The products were applied into their garden as well as for commercial. Furthermore, the product was applied in increasing medicinal plants values.  In order to achieve the aim of this program, the team built a home for the preparation of the product, providing all the equipment and materials and as well as knowledge transfer by doing workshop. The monitoring and evaluation of the programs were done by visiting the area and do evaluation such as observation of the product, the sustainability of product and medicinal plants garden (TOGA). In conclusion, the program was successfully where the people in Desa Pulau Banjar Kari is able to produce the product by themselves and they have been transferring the knowledge to students and people from different villages.

Abstract

Over the past decade, the use of biological agents such as plants, cyanobacteria, bacteria, and fungi for synthesis of metal nanoparticles has been developed. The aim of this study was to investigate antibacterial activity of Cucumis melo L. peel extract and its nanoparticles formulation against Eschericia coli. The nanoparticles were made using silver nitrate with the ratio between C. melo L. extract and silver nitrate aqueous solution (1 mM) were 1:10 and 1:15. The formation of silver nanoparticles was observed after microwaved for 30, 60, 90, 120, 150, and 180 seconds by visible spectrophotometry analysis. Phytochemical screening revealed the presence of flavonoid and terpenoid within the extract. However, the characteristic of surface plasmon resonance band, which occurs in the range of 410-500 nm were not found in the nanoparticle extract, even though the reaction time was extended to 330s. Antibacterial activity against E. coli of the extract and its nanoparticle formulations was determined using Resazurin microtiter assay and compared to Amoxsan® as positive control. The highest E. coli inhibition was exhibited by the nanoparticles (79.8739±0.3859), followed by the extract (65.2821±0.9949). The nanoparticles and the extract have potent antibacterial activity compared to positive control (84.5519 ± 0.2544). In conclusion, the antibacterial activity of the C. melo L. silver nanoparticles formulation was better than its extract.Keywords: Cucumis melo, silver nanoparticles, UV-Vis spectra

Abstract

Phaleria macrocarpa (Scheff.) Boerl is originated from Papua Island, Indonesia. There have been only limited attempts to explore the biological properties of this plant in relation to their medicinal use. This study aimed to examine antioxidant activity of P. macrocarpa fruit. Extraction of pericarp and mesocarp of P. macrocarpa were performed using soxhlet method with ethyl acetate as the solvent. Antioxidant activity was characterized in various in vitro model systems, including DPPH and ferric reducing antioxidant assay. We found that the highest amount of phenolic compounds and flavonoids were found in the pericarp (58.3±0.07 mg/g DW and 127.8±1.08 mg/g DW, respectively). The results showed that pericarp had higher antioxidant activity (IC50= 122.4±1.14 µg/ml) compared to mesocarp (IC50=175.48 ±1.75 µg/ml). In conclusion, the result of this study indicated the possible application of P. macrocarpa as a source of natural antioxidant compound.Keywords: antioxidant, Phaleria macrocarpa, phenolic compounds, flavonoids

Abstract

Waterborne-disease masih memberikan angka prevalensi yang tinggi terutama di negara sedang berkembang. Keberadaan E. coli di perairan memberikan indikasi adanya cemaran pathogen dari feses hewan ataupun manusia. Penelitian ini bertujuan untuk menganalisa kekerabatan dan indeks resistensi antimikroba dari E.coli yang telah diisolasi dari DAS Siak, sungai terbesar yang melewati Provinsi Riau, yang masih digunakan oleh sebagian warga untuk aktivitas sehari-hari. Dendrogram yang dihasilkan dari gabungan 3 primer RAPD-PCR mampu membedakan tiap isolate uji. Intensitas penggunaan air sungai oleh warga dan kuatnya arus mempengaruhi distribusi mikroba indicator sanitasi tersebut dari Tapung hingga Perawang. Sifat resistensi antibiotika masih tergolong rendah (MAR index 0,142), namun eritromisin bukan merupakan antibiotika pilihan untuk diare akibat E.coli disebabkan seluruh isolate resisten terhadap antibiotika ini.
