Abstract

Andrographis paniculata, commonly known as sambiloto, is empirically used for various treatments, including its ability to inhibit the growth of Mycobacterium tuberculosis (M. tuberculosis). This study aims to evaluate the growth inhibition activity of M. tuberculosis strain H37Rv using sambiloto herb extract (Andrographis paniculata). The sambiloto extract was prepared using the maceration method. M. tuberculosis was grown on MODS medium, and the toxicity of sambiloto (A. paniculata) was analyzed using pharmacokinetic prediction studies (pkCMS). The results showed that at concentrations of 0.255 mg/ml, 1.275 mg/ml, and 2.55 mg/ml of sambiloto extract, M. tuberculosis growth occurred on days 7 to 14. Further observations were made until day 28, and it was found that starting at a concentration of 6.375 mg/ml, sambiloto extract did not show M. tuberculosis growth in MODS medium. The positive control, isoniazid, did not show bacterial growth, while the negative control showed extensive bacterial growth on day 12. The determination of M. tuberculosis growth was based on microscopic observations of the bacterial colonies, focusing on cord formation. In conclusion, this study, the use of an extract with a concentration of 6.375 mg/ml in the MODS method showed no growth of Mycobacterium tuberculosis, indicating that the extract is effective in inhibiting bacterial growth at this concentration. The online pkCSM test conducted in this study showed that the extract used is not cytotoxic, meaning that the extract is safe for body cells and does not cause cellular damage. Therefore, it has the potential to be an adjunct therapy in the treatment of tuberculosis.

Abstract

Ekstrak teripang mengandung glukosamin yang berpotensi dapat menekan peradangan (inflamasi) dan menyembuhkan osteoarthritis. Penelitian ini bertujuan untuk menguji  aktivitas antiinflamasi secara in vitro dan in vivo dari nanopartikel kitosan-ekstrak kering teripang (Stichopus variegatus). Aktivitas antiinflamasi secara in vitro dilakukan dengan metode denaturasi protein Bovine Serum Albumin (BSA). Aktivitas antiinflamasi secara in vivo dilakukan dengan metode induksi karagenan pada telapak kaki tikus dengan parameter pengukuran edema kaki. Penelitian ini menggunakan tikus putih jantan galur Sprague-Dawley dan dibagi menjadi 4 kelompok yaitu kelompok yang diberi nanopartikel kitosan-ekstrak teripang kering dosis 50 mg/KgBB dan 100 mg/Kg BB, kelompok kontrol negatif (aquadest) serta kelompok kontrol positif (natrium diklofenak). Hasilnya menunjukkan bahwa sampel nanopartikel memiliki aktivitas antiinflamasi secara in vitro dengan nilai IC50 sebesar 267,76 bpj. Nanopartikel kitosan-ekstrak teripang kering dosis 50 mg/Kg BB dan 100 mg/KgBB memiliki aktivitas antiinflamasi dengan nilai persentase penghambatan inflamasi secara berturut-turut sebesar 53,37%, dan 49,95 %. Adapun nilai ED50 yaitu sebesar 99,27 mg/KgBB.

Abstract

Sea cucumbers are deep-sea-dwelling invertebrate animals that are rich in nutrients and have been empirically used to prevent various degenerative diseases, especially due to their ability to dampen free radicals. The purpose of this study was to identify, and determine antioxidant activity, total phenolic content, and toxic properties in extracts of several sea cucumber species originating from Tegal Mas Island, Lampung. The identification results of the three sea cucumber species were Stichopus vastus Sluiter (SV), Stichopus monotuberculatus Quay & Gaimard (SM), and Stichopus quadrifasciatus Massin. (SQ). The extraction was performed by kinetic maceration against all parts of the sea cucumber using a 70% ethanol solvent. Antioxidant activity test using the DPPH (2,2-diphenylpicrilhydrazyl) free radical suppression method and determination of the total phenolic content using the Folin-Ciocalteu method. The results showed that SV had the highest total phenolics and antioxidants with the lowest toxicity. However, optimisation of extraction between wet and dry sea cucumbers is still required. Extraction optimisation with various solvents can also be done to gain secondary metabolites more effectively.

Abstract

Scouting activities are activities that internalize good values for students. Although scouts are extracurricular, most of the content of the activities is in the form of instilling pro-social attitudes and actions needed by students as a young generation. Pro-social attitudes are concerned with sharing, cooperating, helping, being honest, generosity, and considering the rights and welfare of others. This study aims to describe the perception of SMPK Bhakti Luhur Malang students towards the contribution of strengthening pro-social attitudes in scout activities, as an effort to prevent violence. The research method applied is a mix method, which combines quantitative approaches to describe student perceptions and qualitative approaches as supporting data. All 28 grade VIII and IX students were respondents, and 4 people consisting of scout leaders, homeroom teachers, and principals became informants in this study. Data were collected using questionnaires referring to the Likert Scale model and interview guide. Quantitative data were analyzed using percentages and graphs, while qualitative data were analyzed using the Miles and Huberman approach. The results stated that students' perceptions of sharing attitudes obtained a score of 79.9%, cooperation and help 82.6%, honesty 70.3%, generosity 76.1%, attention to the rights of others 76.6%, with an average of 78.0% (very high contribution). These results are confirmed by the results of qualitative research which states that between students various toys and stationery, cooperate in helping friends, especially those with special needs, be generous by sharing provisions with other students, empathize with others. On the element of honesty, some students still cheat on exams and give incorrect excuses when arriving late
