Abstract

The study was aimed to determine the anti-inflammatory activity of water extract of the Luvunga sarmentosa stem in an animal model. Twenty-five Wistar rats were divided into five groups (n=5). Group 1 was administered 0.9% normal saline (negative control), group 2 was administered 150 mg/kg diclofenac sodium (positive control), and groups 3 to 5 were administered 50, 300, and 550 mg/kg BW of L. sarmentosa extract, respectively. Carrageenan was injected subcutaneously into each rat's subplantar region of the left hind paw. The paw volume was measured using a plethysmometer. The results showed that the water extract of L. sarmentosa stem (doses of 50, 300, and 550 mg/kg BW) significantly reduced the paw edema volume from the 4th to 5th hour compared to the negative control. The percent inhibition of edema at the 5th hour is 47.45; 46.95; 50.39%. The first phase of the edema (1st and 2nd hour) was not affected by the extract. Meanwhile, diclofenac sodium decreased paw edema volume from the 1st to 5th hour with a percent inhibition of 95.90% at the 5th hour. The histopathology result is relevant to the percentage inhibition of edema. Treatment with L. sarmentosa extract showed slight improvement, destruction of epidermal tissue, hyperkeratotic skin, and subepidermal edema. Meanwhile, positive control showed no inflammatory signs with normal keratin, subepidermal, and subcutaneous layers. The water extract of L. sarmentosa stem has anti-inflammatory activity. This extract effectively reduces the paw edema volume in the late phase with decreased neutrophil infiltration.

Abstract

Luvunga sarmentosa, commonly known as saluang belum, is widely used in Kalimantan to relieve pains, rheumatism, boost the immune system, and fever. The research on the free radical scavenging and analgesic effect of the L. sarmentosa stem extract has not been reported. This study aimed to evaluate the free radical scavenging and analgesic activity of the ethanol extract of L. sarmentosa. The L. sarmentosa stem was extracted using 70% ethanol and tested for free radical scavenging using the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and analgesic activity, acetic acid-induced writhing test, and hot plate test in an animal model. The results showed that the 70% ethanol extract of the L. sarmentosa had an anti-free radical scavenging and analgesic activity. The extract has weak free radical scavenging with an IC50 value of 293.45 µg/mL. Analgesic activity using the writhing test indicated that the extract significantly reduced the writhes count after oral administration in a dose-dependent manner compared to the negative control. Extract at a dose of 550 mg/kg BW can reduce the writhing test by 67.60% compared to others. In contrast, the diclofenac sodium reduced the number of writhes by 74.74%. While in a hot plate, the extract at a dose of 550 mg/kg BW produced a maximum possible analgesia (MPA) of 17.64%, lower than the MPA of diclofenac sodium (51.01%). Analgesic activity of the extract has higher inhibition on the writhing test than on the hot plate. The extract could be responsible for the peripheral mechanism by inhibiting the prostaglandin biosynthesis.

Abstract

Hepatitis C Virus (HCV) infection is a serious public health problem since HCV is the ribonucleic acid (RNA) virus that  easy to mutate. The HCV standard treatment  has rapidly developed but the possibility of resistance and effectiveness of treatment needs to be considered. The medicinal plants are a source of various compounds that may potentially cure diseases including infectious diseases. Since a long years ago, medicinal plants were famous as an inherited treatment that believed to cure the disease. One of the medicinal plants is Scoparia dulcis (S. dulcis) that belongs to Scrophulariaceae family and traditionally used as remedies for digestive problems, hypertension, diabetes mellitus, bronchitis, and as an analgesic & antipyretic agent. The previous report showed that S. dulcis was known active as an antiviral against Herpes Simplex Virus (HSV) type 1 in vitro and in vivo. The aim of the study is to determine the biactivity potential of S. dulcis against HCV. Scoparia dulcis was extracted using 80% ethanol (EE) then further separated by liquid-liquid fractionation using dichloromethane (DCMF), ethyl acetate (EAF), butanol solvent (BF) and water (WF). The in vitro anti-HCV analysis was performed with Huh7it cells and HCV JFH1 (genotype 2a) by determining inhibition concentration 50 (IC50). The toxicity (Cytotoxicity Concentration 50, CC50) test was performed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and mechanism of action were analyzed using time addition experiment.   Phytochemical groups as the suspected active compounds of S. dulcis were identified by Thin Layer Chromatography (TLC) and observed under UV 254 nm, UV 365 nm, before and after sprayed using H2SO4 10% and heated at 105oC for 5 minutes. The IC50 test result of 80% EE and DCMF showed anti-HCV activity with a value of 12.7±4.8 µg/ml and 5.8±0.69 µg/ml, while EAF, BF, and AF respectively resulted in IC50 value of  >100 µg/ml that suggested there was no inhibition effect on HCV JFH1.  The DCMF was the most active fraction but toxic to the cell with CC50 value >23 µg/ml and selectivity index (SI) >3.9. According to the time addition experiment data, DCMF of S. dulcis inhibited post entry step HCV JFH1 infection that it means the possibility was to inhibit virus replication and or virion release. Scoparia dulcis contain chlorophyll, flavonoids and terpenoids as the suspected active compounds for inhibition of HCV JFH1 infecton. Futher study of post-entry inhibitions of HCV infection was needed.

Abstract

Background: The urgency generated by drug-resistant strains of malaria parasites has accelerated the need to find a new anti-malarial drug. Recent studies have shown that 80% ethanol extract Acmena acuminatissima has suitable antimalarial activities. Aim: To analyze the inhibitory effect of A. acuminatissima extract at the developmental stage of P. falciparum in infected erythrocytes in vitro. Methods: the synchronized culture of P. falciparum was incubated along with A. acuminatissima extract, and compared with negative controls. The effect on parasite growth was determined by observing the level of parasitemia and accumulation of various infective stages seen on thin blood smears after the incubation period of 12, 24, 36, and 48 hours using a light microscope with 1000 times magnification. Results: Within 48 hours of observation the difference was mainly in the proportion of ring-stage which was 2.75% in culture with the test compound and 41.41% in the negative control which showed that ethanol extract of A. acuminatissima inhibited the growth of parasites at the schizont stage into the ring stage. In trophozoite and schizonts stages of culture with test compounds, enlargement of digestive vacuole and vesiculation is obtained. Conclusion: A. acuminatissima inhibits the development of intraerythrocyt parasites at the schizonts stage and induced morphological changes.

Abstract

Abstrak Desa Bengkaung adalah desa yang terletak di kecamatan Batu Layar, Lombok Barat, Nusa Tenggara Barat.  Banyak potensi pada desa Bengkaung yang masih bisa dikembangkan diantaranya adalah sumber daya alam yang berlimpah. Namun, pemanfaatan sumber daya alam ini belum maksimal, yang disebabkan oleh kurangnya pengetahuan masyarakat mengenai manfaat dan  pengolahannya. Maka dilakukan pengabdian masyarakat yang berlangsung 2 tahap yaitu seminar terkait pojok herbal dan pelatihan pembuatan teh dan sirup dengan bahan kayu manis, kunyit, temu lawak, asam jawa, kapulaga untuk sediaan sirup serta bangle pada sediaan teh herbal yang berfungsi untuk meningkatkan kesehatan masyarakat. Kegiatan tersebut dilaksanakan  pada 25 Oktober 2024 dengan mitra terdiri dari  anggota IAI  kota Mataram dan kader PKK desa Bengkaung. Adapun materi pelatihan diberikan dengan metode ceramah dan metode praktik. Evaluasi dilakukan dengan cara pemberian pre-test dan post-test, kemudian dianalisis secara kuantitatif. Berdasarkan hasil pre-test dan post-test, diperoleh nilai rata-rata sebesar 60,94 dan 79.37, yang menunjukkan adanya peningkatan pengetahuan mitra terhadap materi penyuluhan yang diberikan. Teh daun salam dan JKP-Bangle dievaluasi dengan pengukuran kadar antioksidan dengan DPPH dan FRAP assay. Hasil analisis menunjukkan bahwa kadar antioksidan the daun salam dan JKP-Bangle dengan FRAP assay memperoleh kadar tertinggi tertinggi nilai masing-masing sebesar 1359.84±13.47 mg GAE/g berat kering dan 396.73±16.79 mg GAE/g berat kering. Aktivitas antioksidan yang tinggi ini mengindikasikan potensi manfaat kesehatan dalam menangkal stres oksidatif yang berhubungan dengan penyakit neurodegeneratif, kardiovaskular, dan diabetes. Kata kunci: Desa Bengkaung; pojok herbal; tanaman obat; antioksidan. Abstract Bengkaung Village is a village located in Batu Layar sub-district, West Lombok, West Nusa Tenggara.  There is a lot of potential in Bengkaung Village that can still be developed, including abundant natural resources. However, the utilization of these natural resources has not been maximized, this is caused by a lack of public knowledge regarding their benefits and processing. So community service was carried out which took place in 2 stages, namely seminars related to the herbal corner and training in making tea and syrup using cinnamon, turmeric, ginger, tamarind, cardamom for syrup and bangle preparations, as well as bangles for herbal tea preparations which function to improve health. public. This activity was carried out on 24-26 October 2024 with partners consisting of IAI members from Mataram city and PKK cadres from Bengkaung village. The training material is provided using the lecture method and the Experimental Learning method (direct implementation learning). Evaluation is carried out by giving a pre-test and post-test, then analyzed quantitatively. Based on the results of the pre-test and post-test, an average score of 60.94 and 79.37 was obtained, which indicates an increase in partners' knowledge of the extension material providedrn how to make innovative products from mangosteen fruit so that they can be produced commercially. Thus, in the long term it is expected to increase the income of the Songgon Village residents. Antioxidant levels of bay leaf tea and JKP-Bangle were evaluated using DPPH and FRAP assays. Analysis results showed that the highest antioxidant levels measured by the FRAP assay were 1359.84±13.47 mg GAE/g dry weight for bay leaf tea and 396.73±16.79 mg GAE/g dry weight for JKP-Bangle. This high antioxidant activity indicates potential health benefits in combating oxidative stress associated with neurodegenerative, cardiovascular, and diabetes-related diseases. Keywords: Songgon Village; herbal corner; medicinal plants; antioxidants.

Abstract

Excessive fatigue and burnout are serious issues frequently experienced by healthcare workers and patients with chronic illnesses, particularly within high-pressure healthcare environments. These conditions not only impair the performance of medical personnel but also negatively affect the therapeutic outcomes of patients. To address this problem, a community service activity was conducted through educational outreach on fatigue and burnout management, utilizing acupressure, traditional remedies, and aromatherapy. The program involved 49 participants, including healthcare workers and Surabaya's chronic disease patients. Held on September 11, 2024, the activity employed lectures and two-way discussions as its primary methods. The material covered included traditional medicine, basic acupressure techniques, and training in roll-on and massage oil aromatherapy preparation. A structured module was developed to enhance learning effectiveness, and participants' comprehension was evaluated through pre- and post-tests. Statistical analysis using a paired t-test revealed a significant improvement in participants' understanding (p <0.01), with an average score increase of 19.8%. This initiative demonstrates that practical and interactive education can effectively improve participants' knowledge and skills in managing fatigue and burnout. Therefore, the outcomes of this activity are essential as a promotive and preventive effort to support the physical and mental well-being of vulnerable groups in healthcare settings.
