Abstract

This study aimed to investigate the genetic impact of single nucleotide polymorphisms (SNPs) of the sterol regulating element binding factor 1 (SREBF1) and scavenger receptor class B member 1 (SCARB1) genes on carcass and meat characteristics, as well as fatty acid composition, in the Bali cattle. The blood and beef samples used for DNA sequencing, physical assessment, and fatty acid analysis were collected from 95 male Bali cattle. The ultrasound images were analyzed using the Image-J NIH software. A total of 4 SNPs were identified in the SREBF1 gene and 5 SNPs in the SCARB1 gene. The results showed that the 4 SNPs in the SREBF1 gene, namely g.12629T>C, g.12731T>C, g.12881A>G, and g.12986C>T, were associated with heptadecanoic acid (C17:0) and cis-11-eicosanoic acid (C20:1). The SNPs g.12731T>C of the SREBF1 gene was associated with fat content, palmitoleic acid (C16:1), stearic acid (C18:0), cis-11-eicosanoic acid (C20:1), and total fatty acids. Furthermore, 4 SNPs in the SCARB1 gene, including g.72219C>T, g.72380C>A, g.72517G>A, and g.72607C>T correlated with longissimus dorsi thickness (LDT). All SNPs in the SCARB1 gene showed significant associations with cis-10 heptadecanoic acid (C17:1) and cis 8,11,14-eicosatrienoic acid (C20:3n6). The SNP g.72400A>G of the SCARB1 gene was related to caprylic acid (C8:0), lauric acid (C12:0), arachidonic acid (C20:4n6), monounsaturated fatty acids (MUFA), and unsaturated fatty acids (UFA). These results suggested that the identified polymorphisms in the SREBF1 and SCARB1 genes could serve as valuable references for investigating similar genes in other cattle breeds, particularly concerning fatty acids.

Abstract

ABSTRACT ESTIMATE OF BODY WEIGHT FIGHTING AND MEAT GARUT SHEEP AND CROSSBREED WITH MERFOMETRIC ANALYSIS APPROACH The aims of this study were to estimate body weight and to determine the best equation which can be used to estimate body weight of fighting and meat Garut sheep and its crossbreed type at district and outdistrict Garut. This study used primary data of Garut sheep from Garut district consisting of Margawati, Wanaraja and Sukawening. For outdistrict Garut, it was used Garut sheep from Bogor consisting of Ciomas and Cinagara. The total number of sheep collected were 531 heads. The differences of body measurements locations were analyzed by t-test, Correlation, and Regression Multiple Linear Regression and Polynomial Regression (Linear, Quadratic, and Cubic) Analyses. The results showed that body weight of fighting and meat rams and ewes from Wanaraja were the highset than another Garut sheep and crossbreed. The highest body measurements were found from Wanaraja (fighting and meat type) and Sukawening (fighting type) sheep and the lowest body measurements were found from Margawati and Cinagara. Whilst that some body measurements of rams were not different (P>0.05). Correlation coefficient between body weight and various body measurements body lenght, weither height, chest deep and chest circumference of Garut Sheep were highest at district and outdistrict of Garut. The best regression equations to describe the relation ship between body weight and body measurements for rams and ewes Garut sheep at district and outdistrict Garut were multiple regression equations. The highest determination coefficient (100%) was shown for meat type from Wanaraja and the lowest one (93.93%) was shown of fighting type from Sukawening. ABSTRAK Penelitian ini bertujuan untuk menduga bobot badan dan mencari model terbaik dalam menduga bobot badan domba Garut tangkas, pedaging dan persilangannya di daerah Garut dan luar Garut. Total domba yang digunakan pada penelitian ini sebanyak 531 ekor domba Garut dari beberapa lokasi di kabupaten Garut (Margawati, Wanaraja, Sukawening) dan di Luar Garut (Ciomas dan Cinagara). Data dari hasil penelitian dianalisis dengan menggunakan Uji-t, Korelasi, dan Regresi meliputi Analisis Regresi Linier Ganda dan Analisis Regresi Polinomial (Linier, Kuadratik, dan Kubik). Hasil Uji-t menunjukkan, bahwa rataan bobot badan domba tangkas Garut baik jantan maupun betina dari Wanaraja memiliki bobot badan paling besar dibandingkan dengan domba Garut lain dan persilangannya. Ukuran tubuh tertinggi pada domba Garut ditunjukkan pada domba tangkas dan pedaging Wanaraja serta tangkas Sukawening dan ukuran tubuh terkecil ditunjukkan pada domba Garut di Margawati dan Cinagara. Koefisen Korelasi tertinggi antara bobot badan dan ukuran-ukuran tubuh pada domba Garut tangkas, pedaging da persilanganya ditunjukkan antara bobot badan dengan panjang badan, tinggi pundak, dalam dada dan lingkar dada. Persamaan regresi yang paling baik digunakan menduga bobot badan berdasarkan ukuran-ukuran tubuh pada domba Garut tangkas, pedaging dan persilangannya adalah Persamaan Regresi Linier Ganda dengan tingkat akurasi berkisar antara 93,3%-100%. Tingkat akuras tertinggi ditunjukkan pada domba tangkas Wanaraja dengan tingkat akurasi 100% dan terendah ditunjukkan pada domba tangkas di Sukawening dengan tingkat akurasi 93,93%.

Abstract

Stearoyl-CoA desaturase (SCD) is an integral membrane protein of endoplasmic reticulum (ER) that catalyzes the rate limiting step in the monounsaturated fatty acids from saturated fatty acids. Selection for fatty acids traits based on molecular marker assisted selection is needed to increase a value of chicken meat. This study was designed to analyze expression and associations of SCD gene polymorphisms with fatty acid traits in F2 kampung-broiler chicken cross. A total of 62 F2 kampung-broiler chicken cross (29 males and 33 females) were used in this study. Fatty acid traits were measured at 26 weeks of age. Samples were divided into two groups based on fatty acid traits (the highest and the lowest). Primers in exon 2 region were designed from the genomic chicken sequence. The SNP g.37284A>G was detected and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was then used to genotype. The expression of SCD gene was analyzed using quantitative real time PCR (qRT-PCR). The result showed that there were three genotypes (AA, AG, and GG) found in this study. The SCD|AciI polymorphism was significantly associated with palmitoleic acid (C16:1), fatty acids total and saturated fatty acid in 26 weeks old of F2 kampung-broiler chicken cross (P<0.05). The SCD gene was expressed for polyunsaturated fatty acids in liver tissue in two groups of chickens. In conclusion, the SCD gene could be a candidate gene that affects fatty acids traits in F2 kampung-broiler chicken cross.

Abstract

The Tumor Protein P53 Inducible Nuclear Protein 1 (TP53INP1) gene is expected to be a crucial candidate for mineral content.  This research aimed to analyse the genotype polymorphism and investigate its association with the TP53INP1 gene and sheep mineral content. To analyse gene polymorphisms and conduct an association study, 30 rams of Javanese thin-tailed sheep were utilised. The NlaIII restriction enzyme was applied to investigate the genotype polymorphism of the TP53INP1 gene through Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Regarding the association with analysing TP53INP1, a General Linear Model (GLM) was used. The findings indicate that the TP53INP1 gene shows polymorphism, revealing three distinct genotypes identified by the results: AA, AG, and GG. The genotypes were detected using the Hardy-Weinberg Equilibrium (HWE) principle. According to the association analysis, a significant association (P < 0.05) was found between the TP53INP1 gene and mineral content, particularly iron (Fe). A higher mineral content was associated with the GG genotype, whereas a lower mineral content was related to the AA genotype. The TP53INP1 could be the candidate gene for sheep's mineral content.

Abstract

Pesisir cattle are native to Indonesia and originate from West Sumatra. It is known for its high environmental adaptability and can be further developed using a proteomic approach. The proteomic approach for testing the fertility of Pesisir bulls is an important factor in determining a potentially superior breed. Proteomic studies of reproduction in Pesisir bulls have not been widely conducted. This study aimed to identify and characterize functional sperm proteins in Pesisir bulls that are associated with reproductive processes. Semen samples were collected from 12 pesisir bulls aged 2–4 years and grouped into three age categories. Protein profile analysis was performed using polyacrylamide gel electrophoresis with 1D-SDS-PAGE, followed by proteolytic digestion of the proteins in the gel and protein identification using LC-MS/MS analysis. Protein functions were predicted based on analysis of biological annotations generated using UniProt, Venny, PANTHER, and STRING. Based on the evaluation of fresh semen, sperm motility in Pesisir bulls was found to increase with age. Proteomic analysis successfully identified 334 proteins in the sperm of Pesisir bulls. Among these, two proteins, ZPBP and SPACA3, were identified as involved in reproductive and fertilisation processes. Functionally, both proteins play crucial roles in acrosomal events during fertilisation. Gene Ontology analysis showed that most proteins in sperm are involved in various biological processes, including cellular activities, metabolic processes, and molecular functions related to catalytic activities. In conclusion, SPACA3 and ZPBP proteins were identified as potentially involved in reproductive processes and may serve as fertility markers in Pesisir bulls.

Abstract

Kampung chicken is one of Indonesia native chickens which has slow growth and it needs to be improved. Growth is a quantitative trait which is controled by many genes. One of the genes which plays a crucial role in controling growth is IGF2 gene. The objective of this study was to analyze the association of IGF2 gene polymorphisms with growth traits in kampung chicken. A total of 56 male and female chicks were maintained for 16 weeks and genotyped using PCR-RFLP method.Result showed there was TC muation on IGF2 gene and it was polymorphic in kampung chicken. Frequency of heterozigous TCwas the highest than the others. There was no association between IGF2 gene polymorphismswith growth traits in kampung chicken. It can be caused by the synonimus mutation on IGF2 sequence that does not alter the amino acid. Bot of the nucleotides coded for histidine. Abstract

The Mx gene is wellknown as marker genetic for chicken resistance of virus infection. The aim of this study was to analyze the mx gene polymorphisms in selected kampung chicken. A total of 25 heads of kampung chicken, which had been selected for ND virus resistant, were identified by PCR-RFLP method.The Mx gene used Hpy8I restriction enzyme where digested at GTN|NAC nucleotides. There were two  alleles, A and G with three genotypes AA, AG dan GG. The result showed that 69.2% of kampung chicken were resistant toward the virus infection. This population was in Hardy-Weinberg equilibrium (x2< x20.05).Keywords: polymorphisms, Mx gene, selected kampung chicken

Abstract

The aim ofthis study was to examine the preservability of local chicken semen in extender added with various level extra virgin olive oil (EVOO).Semen was collected from three roosters using massage method. Immediately after collection the semen evaluated macro and microscopically. Semen demonstrated   motility, individually divided into five tubes. Each of them diluted with ringer fructose that supplemented with 0%, 4%, 6%, 8% or 10% EVOO. Semen was stored in  refrigerator (5 oC) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results revealed that supplementing the diluent of roosters semen with EVOO and then storing it for different storage periods (, , ,  or  hours) resulted in significant (p .) improvement in spermatooa motility and viabilityin comparison with the control group.Moreover, used of 8% EVOO surpassed other treatments afterin vitro storage for up to 60 hoursand resulted fertilizing ability up to 73.81%.This research conclude that 8% EVOO can be used as antioxidant source for roosters semen during preservation.Keywords : extender, local chicken, extract virgin olive oil, sperma

Abstract

Kampung chicken is Indonesian native chicken that potential to be developed as commercialchicken.  The large-scale breeders less interest to breed kampung chicken because it has slow growth toreach slaughter weight than commercial meat type. Therefore, kampung chicken’s performance needto be improved by crossing with commercial meat type that has fast growth. This research purposedto observed the effect performance of first generation (F1) from commercial meat type crossing withkampung and their reciprocal.  Research conducted performance of crossing chicken in age 0-12 weeksage. T test used for knowing difference body weight mean, body weight gain, feed consumption, and feedconvertion rate.  The result of this research were not significant between KB cock with BK cock andKB hen with BK hen on body weight, body weight gain, feed consumption, and feed convertion rate.Whereas mortality percentage BK chicken was higher than KB chicken althought less of mortality.  KBand BK cocks had better performance than hens. Reciprocal crossing between kampung chicken andcommercial meat type had performance better than kampung chicken, so crossing had been increasedgenetic quality of kampung chicken.

Abstract

The experiment aimed to observe growth performance of 3 native chicken crossed. The materials used in this experiment were 22 day old chick (DOC) resulted of natural mating between male pelung and female sentul chicken (PS) and 113 DOC resulted of natural mating between male sentul and female kampung chicken (SK). Feeds were given during experiment divided into 3 phases that were BR-21E 100% for chicken between 0-3 weeks of age, rice bran 20% and BR-21E 80% for 4 week age, and grower feeds rice bran 40% and BR-21E 60% for chicken between 5-12 weeks. Variables which recorded weekly were body weight until 12 weeks of age, weight gain, feed consumption, feed conversion, and mortality. The t-test was used to compare data of performance between two sexes and between two cross system (PS and SK). The results showed that body weight at 12 weeks of crossbred (PS) male was significantly higher than SK (P<0.01) also female (P>0.05). Feed consumption and feed conversion were not significantly different (P>0.05). The mortality rate during the experiment were 4.54% for crossbred PS and 7.07% for SK. SK and PS can be applied to increase meat production.
