Abstract

NRAMP-1 protein and INOS enzyme have been shown as deffense mechanism against bacteria and virus infection. These genes have not been investigated in Sentul Selection chickens. This study was aimed to determine the polymorphism of NRAMP-1 and INOS in Sentul Selected chickens. A number of 172 sentul selected chickens was observed for NRAMP-1 and INOS genes using PCR-RFLP. The frequency of genotypes and alleles, polymorphic information content and Hardy-Weinberg equilibrium status (HWE) were analyzed. Polymorphism in NRAMP1|SacI and INOS|AluI were observed. The population was in HWE for both genes. NRAMP1 gene frequencies had higher TC genotypes and INOS gene frequencies had higher CC genotypes (P<0.05) in Selected Sentul chickens. Furthermore, the NRAMP1 and INOS genes, the frequency of allele C was higher than the T allele. It could be concluded that NRAMP-1 and INOS gene has potential to propose has associated with immune traits in Sentul chicken.

Abstract

Kampung chicken is a local Indonesian chicken that comes from red jungle chicken that has been successfully domesticated but has a low productivity. One of method to increase the productivity can be done by crossing a local chicken with a broiler. This research was conducted to examine the growth performance of crossing result G3 interse (pelung, sentul, kampung, broiler) age 1-12 weeks. IPB D-1 G3 chicken measured in this research were 76 DOC, 18 cocks, and 30 hens. T test was used to knowing the differences of body weight, body weight gain, feed consumption, and feed conversion. Based on the research, body weight, growth, and feed conversion of male IPB D-1 chickens was better than female IPB D-1 chickens when 12 weeks age. Body weight of a cock IPB D-1 G4 in 12 week was 1939.9 g and for a hen was 994.9 g. Feed consumption of rooster IPB D-1 G4 3 468.97 g and 3 339.80 in females with fluctuating body weight growth. Chicken feed conversion IPB D-1 lowest G4 1.579 and 6.430 with the highest. Coefficient of variation on all variables IPB D-1 G4 was high. Performance growth of IPB D-1 G4 still diverse.

Abstract

Kampung chicken is Indonesian native chickens that has slow growth. One way to increase growth is crossing. Pelung-sentul-kampung-meat type chicken (IPB D-1) is a chicken from crossing between F1 pelung-sentul (PS) male and kampung-meat type chicken strain cobb (KM) female to produce chicken that has fastly growth and big body size. Factors that need to be examined to find out the bone growth and body structure of chicken is the body size. The aim of the research was to asses body size chicken IPB D-1 generation G4 crosing results with interse at age 2 until 12 weeks. The number of chickens used 33 males and 38 females. The Variables measured every 2 weeks were body weight, shank length, shank circumference, tibia length, femur length, back length, chest circumference, chest width, chest depth, and chest length. Research data were analyzed descriptive. IPB D-1 chicken G5 have a coefficient of diversity 5% to 15% on each variable observed. The fifth generation of IPB D-1 chicken (G5) have  a variation body size. Chest length is the best variable to predict the body weight.

Abstract

Etawah-grade goat is a graded up line between kacang and etawah goats, which well adapted to Indonesia’s humid tropical climate. The good reproductive trait of Etawah-grade does are inherited from kacang goat (prolific goat). This study used six heads of Etawah-grade does at 1–2 years old (I1 dental condition). IPB University. The body variables were withers height, hip height, body length, chest width, chest depth, thurl width, rump length, chest girth, and cannon circumference, whereas for head variables were acrocranion–prosthion, basion–prosthion, lower jaw length, head height, tuber facial left-right, nasion–rhinion, entorbitale left-right, euryon left-right, supraorbitale left-right. Body score and head score were obtained from Principal Component Analysis (PCA). PCR-RFLP technique was used to determine genotypes of fecundity genes BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1) and KISS1 (intron 1) with restriction enzymes AluI (AG|CT), SduI (GDGCH|C), BsrI (ACTGG|) and MwoI (GCCTAAG|TAGC), respectively. Progesterone leves were measured by EIA (enzyme immuno assay). The results showed that the association between crude fat consumption and progesterone profile was found in this study, but it was not related to twin births with certain genotypes of gene fecundity BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1) and KISS1 (intron 1). The progesterone profile during the pregnancy period did not correlate to their body size and head size.

Abstract

The aim of this study was to examine semen characteristics of IPB-D1 roosters. A total of 15 IPB-D1 roosters were used in this study, healthy, aged between 6 to 9 months, body weight between 1.2-2.0 kg and placed in individual cages 50 cm × 50 cm × 90 cm. Semen were collected using massage techniques on the back and cloaca rooster. Before collection, cloaca is cleaned using tissue that has been moistened with physiological NaCl solution. The evaluation parameters of semen are macroscopically including volume, color, consistency and pH and microscopic include mass movements, spermatozoa motility (%), spermatozoa morphology (%), concentration and number of spermatozoa per ejaculate. The results showed the semen volume of IPB-D1 was 0.10 ± 0.07 mL, milky white in colour, moderate and dense in concistency and pH 6.97 ± 0.27, Microscopic characteristics of IPB-D1 roosters showed mass spermatozoa movement of 2.73 ± 0.46, spermatozoa motility and concentration were 75.00 ± 6.27% and 3257.50 ± 1303.141×106/mL respectively. It was concluded that IPB-D1 roosters produced good semen quality.

Abstract

IPB-D1 is a chicken cross between male F1 PS (PelungxSentul) with F1 female (Kampung x parent stock cobb). The establishment of chicken IPB-D1 aims to produce commercial meat type local chicken with semi-intensive maintenance management. The aim of this research to observe study assesses the performance of egg production and the quality of IPB D-1 chicken eggs and estimates the value of repeatability of production and haugh units of IPB D-1 chicken eggs. The chicken was observed as many as 30 hens IPB D-1 G7 aged 24-36 weeks and eggs as many as 288 eggs. The production and egg quality data were analyzed descriptively and the repeatability values were analyzed with Anova. Egg weight 40 g, Haugh unit value 87 (Very good), egg production 49.22%. The repeatability value of egg production is 0.2 (Moderate) and the repeatability value of the haugh unit is 0.31 (Medium). IPB D-1 Chicken has the potential to be a good dual-purpose chicken.

Abstract

Fatty acid content and carcass characteristics are two meat quality traits consumers choose to ob tain healthy and high-quality lamb. Genetic improvement of fatty acid content and carcass characteris tics in sheep is important to increase the public appeal of lamb meat. The Biglycan gene (BGN) is a gene that functions to regulate the growth and maintenance of connective tissues, such as bone and cartilage. This study examined the polymorphism of the BGN gene to get a deeper understanding of the correlation between fatty acid content and carcass traits in Indonesian sheep. The PCR-RFLP ap proach was used to detect polymorphism in the BGN gene in meat samples obtained from the longissi mus dorsi region of 115 rams aged between 10 and 12 months. The General Linear Model (GLM) was used test to analyze genotyping through association studies between genotypes and phenotypic attrib utes. The results indicated that the carcass' fatty acid composition, including tridecanoic acid (C13:0), arachidonic acid (C20:4n-6), and polyunsaturated fatty acids (PUFA), as well as the BGN gene with SNP point g. 77807325 G>C (GG and CC genotypes), were significantly (P<0.05) related to these traits. Genotype GG was more associated with carcass percentage traits than other genotypes, whereas genotype CC was best associated with carcass length traits. The BGN gene with SNP point g. 77807325 G>C can be a potential genetic marker for selecting fatty acid (tridecanoic acid C13:0), pol yunsaturated fatty acid (PUFA), arachidonic acid), carcass percentage, and carcass length in sheep meat in Indonesia.

Abstract

The insulin-like growth factor binding-protein 7 (IGFBP7) gene is one of the potential genes related to meat quality. The objective of the current study was to evaluate polymorphism of the IGFBP7 gene (g.72351183A>C) and its association with meat quality traits in the Javanese thin-tailed (JTT) sheep. A comprehensive analysis was conducted on 88 JTT male sheep to examine their fatty acid composition, carcass characteristics, carcass retail cuts, and the physical properties of lamb meat. The polymorphism was detected using the PCR-RFLP technique. The association between the IGFBP7 gene polymorphism and the observed variables of meat quality was evaluated using one-way analysis of variance (ANOVA). The study results indicated that the IGFBP7 gene was polymorphic in JTT sheep population, with the allele distribution conforming to Hardy-Weinberg equilibrium. The AA genotype was found to be predominant. The IGFBP7 gene variants were associated (p<0.05) with erucic acid, linoleic acid, eicosapentaenoic acid, and total polyunsaturated fatty acid (PUFA). Sheep possesing the CC genotype exhibited the highest levels of linoleic acid, eicosapentaenoic acid, and total PUFA in comparison to those with AA and AC genotypes. However, the IGFBP7 gene polymorphism was not associated with carcass characteristics, carcass retail cuts, and physical properties of meat. These findings suggest that the IGFBP7 gene is a promising candidate marker for improving fatty acid composition in JTT sheep.
