Abstract

Catharanthus roseus is one plant recognized as medical potential which can decrease cholesterol. Thepresent study was carried out to evaluate the effect of aqueous leaf extracts of C. roseus on plasma lipidprofil levels in rats cholesterol rich diet and atherogenic index. This study was carried out on 15 SpraqueDawley male rats randomly distributed into five groups (n=3). Rats hypercholesterolemic wereadministrated cholesterol rich diet containing 1% (w/w). Normal control group with normal diet whitoutextracts (K1), positive control hypercholesterolemic group (K2) with cholesterol rich diet whitout extracts,and others three groups (K3-K5) were feed high cholesterol and 1 mL of aqueous leaf extracts of C. roseuswith a dose of 20% (w/v), 40% (w/v) and 80% (w/v) respectively, administered twice daily 1 mL orally.Treatment was given for 28 days. After treatment, the plasma lipids profile such cholesterol total, highdensity lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TGA) level were measured. Theresults showed that during the period of treatment with cholesterol rich diet have produced under condition hypercholesterolemic of rats with average cholesterol levels of 124.33 ± 4.04 mg/dL (K2 group). Treatmentwith extract dose of 80% showed the best results among the other doses in reducing levels of total cholesterol,triglycerides, LDL and raise HDL levels. Extract treatment dose of 80% up to four weeks resulted incholesterol total (79.33±3.51 mg/dL), TGA (72.33±6.65 mg/dL), HDL (55.00±3.60 mg/dL), and LDL(9.87±5.34mg/dL). In addition, value ratio of cholesterol:HDL was 1.4: 1 and atherogenic index value was0.44. These results suggest that extracts of leaf C. roseus optimum doses 80% (w/v) has hipolipidemicactivity in hypercholesterolemic rats and it has low atherogenic index value.

Abstract

Kasus urolithiasis oksalat monohidrat dijumpai pada kucing lokal yang datang ke Rumah sakit Hewan Fakultas Kedokteran Hewan Universitas Udayana dengan anamesis kucing mengalami penurunan nafsu makan. Gejala klinis yang terlihat adanya kifosis ringan. Palpasi di abdomen bagian atas hingga belakang dilakukan dan hewan meronta kesakitan saat palpasi di bagian hypogastrium medial vesica urinaria. Kucing juga menunjukkan ekspresi nyeri saat dilakukan palpasi pada bagian penis saat pemeriksaan uretra dan urinasi. Kucing sedikit-sedikit mengeluarkan urin berdarah dengan konsistensi cair. Pemeriksaan lanjut dilakukan menggunakan ultrasonografi dan urinalisis menggunakan uji cepat dipstik. Hasil pemeriksaan ultrasonografi memperlihatkan adanya bintik bintik putih di vesika urinaria, sedangkan pemeriksaan fisik urin memperlihatkan hematuria dan mikroskopik memperlihatkan adanya kristal oksalat monohidrat. Penanganan yang dilakukan pada kasus ini adalah memberikan antibiotik amoxicillin injeksi dosis 11-22 mg/kg bb untuk mengurangi infeksi pada saluran kemih. Selain itu Nephrolit® yang mengandung Orthosiphon stamineus (kumis kucing) dan beberapa herbal lainnya juga diberikan untuk membantu alkalinisasi dan pelarutan kristal-kristal dalam urin agar mudah dikeluarkan saat urinasi. Hasil penanganan memperlihatkan kucing membaik dalam waktu 10 hari terapi.

Abstract

Organic acids are a mix of various compounds of organic carboxylic acids and are used as an acidifier in chicken feed. Organic acids could increase the digestibility and absorption of protein in broiler chicken, because organic acids can inhibit pathogenic bacterial growth in the gastrointestinal tract. The increase of protein absorption can affect the level of blood urea nitrogen and creatinine in broiler chicken’s blood, which is considered parameters for kidney function. The objective of this study is to determine the Blood urea nitrogen (BUN) and creatinine levels in 24 female broiler chickens in the course of 35 days of organic acids treatment. This study used an experimental method with completely randomized design (RAL) consisting of 4 treatments, the control group were only given finished feed (P0), then administered with Orgacids® as much as 0.1% (P1), 0.2% (P2), and 0.3% (P3). The results showed that the administration of organic acid had no significant effect (P>0.05) on the BUN levels in the P1, P2, and P3 groups against the P0 group. And the administration of organic acids also had no significant effect (P>0.05) on creatinine levels in the P1, P2, and P3 groups against the P0 group. It was concluded that the administration of organic acids in the feed does not affect the levels of BUN and blood creatinine in broilers significantly.

Abstract

This study aims to determine the effect of transportation on changes in total protein levels in Bali cattle after a trip to the Pesanggaran Slaughterhouse (RPH) Denpasar. Transportation can lead to stress and dehydration. To diagnose the occurrence of stress can be done by biochemical tests. The biochemical test carried out in this study was to measure the levels of total protein in the blood. The object used is Bali cattle that have not been rested after transportation as many as 20 heads at the Denpasar Pesanggaran Slaughterhouse. Blood was taken through the auricular vein and put into a vacutainer tube containing a gel separator to separate serum from blood. Blood serum samples were taken to the Bali Provincial Health Laboratory for immediate examination of total protein levels using the Photometer 5010V5+ machine. The total protein content of the examined samples had the lowest level of 5.8 g/dL and the highest 8.5 g/dL with an average of 7.5 g/dL. From the results of the study, the total protein content of Bali cattle after transportation was considered normal, this indicated that there was no effect of transportation on the total protein content.

Abstract

A medical examination is a mandatory treatment received by police dogs to monitor health, that performance in the field becomes optimal. Regular blood tests are importance to get health information. Platelets are one of the importance indicators in the blood test results in the form of Platelet values (PLT), Mean Platelet Volume (MPV), Platelet Distribution Width (PDW), and Platelet Crit (PCT). This value can establish a diagnosis and become a health surveillance parameter in police dogs. This study is conduct observationally to determine the picture of platelets in police dogs in the Malang City Resort police. The sample came from all police dogs in the Malang City Resort Police, which amounted to 10 tails. Blood samples were read using the Haematology Analyzer RT-7600 machine in Clinical Pathology Laboratory at Brawijaya University. The result of the study showed that in the value of PLT, five police dog blood samples had thrombocytopenia and five others within the range of typical values. Only one specimen is below the range of expected values at the MPV value. At the PDW value, all samples are in the expected range value. On PCT value, all specimens are also in the normal range of values. It can conclude that the study of PLT values in 5 police dogs had thrombocytopenia in the MPV value of 1 sample below the normal range in addition to the PDW and PCT values of all specimens within the normal range. The need for follow-up platelet examinations such as liver and bone marrow biopsies for further check-ups. The addition of a routine blood check agenda on police dogs needs to be made.

Abstract

Saliva adalah cairan tubuh unik yang mudah diakses, dan memiliki berbagai peran dalam menjaga kesehatan tubuh. Karena kelebihan ini, saliva memiliki potensi yang besar untuk dikembangkan menjadi sampel biologis yang non-invasif dalam dunia kedokteran hewan, dimana kesejahteraan hewan selalu diupayakan. Meskipun saliva anjing sudah cukup banyak diteliti, namun belum ada penelitian mengenai komponen seluler pada saliva anjing. Penelitian ini bertujuan untuk mengetahui jenis sel yang dapat ditemukan pada saliva anjing normal. Penelitian ini menggunakan objek penelitian berupa spesimen saliva dari anjing sehat. Sampel saliva dari 15 anjing kemudian dibuat menjadi 15 sediaan segar sebelum diperiksa menggunakan mikroskop. Pemeriksaan mikroskopis dilakukan menggunakan perbesaran 400x untuk mengidentifikasi jenis sel yang dapat ditemukan pada saliva anjing. Data yang didapatkan akan dilaporkan secara deskriptif kualitatif. Hasil penelitian menunjukkan bahwa pada saliva anjing dapat ditemukan beberapa sel yaitu sel epitel, sel darah putih, mikroorganisme, serta komponen bukan sel yaitu kristal. Sel yang ditemukan merupakan sel yang berasal dari rongga mulut, sedangkan pembentukan kristal pada saliva anjing disebabkan oleh kandungan mineral serta pH saliva anjing yang bernilai tinggi. Perlu dilakukan penelitian lanjutan agar sel dapat teridentifikasi dengan lebih baik.

Abstract

Penelitian ini bertujuan melakukan isolasi dan identifikasi serotipe lokal Escherichia coli (E. coli) O157:H7 dan uji profil hemolisisnya pada media agar darah. Isolasi bakteri dilakukan dengan media eosin methylene blue agar (EMBA), dilanjutkan dengan identifikasi pada media selektif sorbitol MacConkey agar (SMAC) dan uji konfirmasi menggunakan uji aglutinasi lateks O157 serta uji antiserum H7 sebagai konfirmasi akhir dari E. coli O157:H7. Gambaran hemolisis diuji dengan menumbuhkan isolat pada media agar darah domba. Hasil penelitian menunjukkan bahwa 7 isolat (8,54%) dari 82 sampel feses ayam teridentifikasi E. coli O157:H7 dan memperlihatkan profil enterohemolisis seperti halnya isolat kontrol ATCC 43894. Dari hasil penelitian disimpulkan bahwa isolat lokal E. coli O157:H7 hasil isolasi dari feses ayam diketahui memiliki patogenitas yang tinggi terkait dengan dihasilkannya enterohemolisin yang merupakan marka/penanda kemampuan dari isolat untuk menghasilkan faktor virulensi Shiga like toxin.

Abstract

Escherichia coli O157:H7 has been identified as one of the most devastating microorganism causing diseases in human. The infection by this bacteria causes a wide spectrum of clinical manifestations ranging from asymptomatic, diarrhea or bloody diarrhea, up to serious clinical conditions such as hemorrhagic colitis, and hemolytic uremic syndrome. Poultry is known as one of its reservoir besides cattle and pig. Moreover, the use of antibiotic in animal feed as growth promoters is common worldwide. Due to its potential threat to public health, sensitivity test against various antibiotics and studying the plasmid DNA profiles are necessary. The study was initiated by cultivation of isolates, followed by test of antimicrobial susceptibility against various antibiotics and then by analyzing the plasmid DNA profiles. Results of study showed that among of 7 E. coli O157:H7 local isolates originated from chicken feces, as many as 42.9%; 14.3%; 14.3%; and 14.3% were resistant to 2; 3; 4; and 5 various of antibiotics, respectively. Base on the type of antibiotic, 85.7%; 71.4%; and 42.9% showed resistance to Methicillin, Penicillin G, Doxycycline and Streptomycin. Plasmid DNA of each isolates showed characters variation too, ranging from 2 to 4 bands with molecular weight ranging from 19.457 bp; 15.137 bp, 5.749 bp, 4.118 bp, 3.500 bp, 2.957 bp and 2.024 bp. Results of study indicated that local isolates of E. coli O157:H7 isolated from chicken showed multiple antibiotic resistance against various antibiotics, as well as its plasmid DNA profiles.

Abstract

Lactic acid bacteria SR4 strain is known to have potential as a probiotic source according to the previous study. The isolate derived from the gastric’s juice of bali cattle has characteristics such as its resistance to growth at low pH, namely pH 2, 3, and 4, its tolerance to bile concentration of 0.6 mM, and its inhibition against Gram-positive and negative bacteria. On the other hand, a complete molecular analysis of the isolate has never been carried out. Based on these considerations, the molecular analysis of the LAB SR4 strain becomes interesting to present. The study was initiated by cultivating LAB SR4 isolate on an MRS broth medium. The isolate was further confirmed molecularly through analysis of the 16S rRNA gene which was initiated by isolating genomic DNA followed by PCR reaction using universal primers 27F and 1492R. The PCR product of the 16S rRNA gene was sequenced and further analyzed using the MEGA 5.2 software. The data were then analyzed descriptively and presented in the form of tables or figures. The results showed that the LAB strain SR4 was identified as a Pediococcus pentosaceus. This strain potentially to develops as a source of bacteriocin and it can be applied as a bio preservative, improvement of the meat taste, inhibition of inflammation, and also as antagonism of cancer.
