Abstract

Embung Biting is a native habitat for freshwater fish for Cyprinidae, tilapia, catfish and other endemic fish species. Abundant of natural biological resources including supply of embung water from water sources are very important for meeting the community's water needs. In order to preserve the biological resources of ponds and as a source of fish nutrition for the people of Sukorame Village, it is necessary to be given an understanding to preserve the embung through the introduction of good and environmentally friendly methods of fish farming. This activity has been able to provide an understanding to the community of the importance of sustainability of the embung which is seen in the enthusiasm of the participants and the motivation of the participants to improve entrepreneurial skills in the field of fish farming as alternative livelihoods.

Abstract

Environmental pollution could directly reduce water quality for shrimp cultivation. This study aims to determine how the environmental quality, shrimp population and genetic characteristics of shrimp that live in polluted waters in shrimp ponds in East Java associated with WSSV disease that often attacks on cultivated shrimp. The method used was a descriptive exploratory. Data collection was collected by observation and interview with farmers on disease history. Water was sampled for water quality study and characterize its suitability for shrimp cultivations. Shrimps were also sampled for morphological and genetical study for its susceptibility of WSSV. All samples were taken every week during shrimp cultivation in Malang ans Gresik. Morphological study using scoring method to determine the disease stages on shrimp, while for genetical study using specific primer for ICP11 for WSSV detection, since ICP 11 is expressed when WSSV infection is occur. Samples were taken from 2 shrimp ponds in South Malang and Gresik.The overall water quality is good, except for NO2 and TOM in both seawater shrimp ponds which higher than the freshwater/estuary shrimp ponds. Light infections of WSSV were detected in all seawater ponds both in morphology and genetics. However, in the freshwater/estuary pond only shrimps from freshwater/estuary Gresik which showing light WSSV infection genetically, but not in the morphological signs. Early disease detection is important to control the disease spread.

Abstract

Water pollution can make many problems such as, the incidence of disease and poisoning. Pollution in water can produce free radicals and it is can trigger of disease for aquatic organisms. There is some efforts that can be done to provide this problem, such as chemical compound that can reduce the reaction of free radicals. Antioxidants are one of the chemical compounds that can reduce the activity of free radicals. Eucheuma cottonii is the one of a seaweed that has many in antioxidant compounds, such as phenol compounds, but it is also rich in iodine fiber and other important minerals. The method used in this research is descriptive explorative and experimental method. This research was conducted with several stages of seaweed extraction. Identification of Eucheuma cottonii extract is using FTIR test. The last stage is an antioxidant activity test that includes DPPH test (1,1-diphenyl-2-picrilhidrazil) and Inhibition Concentration 50 (IC 50). The results obtained in this study were based on FTIR test of antioxidant compound in Eucheuma cottonii seaweed extract. The one of compounds that have antioxidant activity include galaktosa-4-sulfat. Based on the results of antioxidant activity test using DPPH obtained that seaweed extract Eucheuma cottonii active as an antioxidant to ward off free radicals in the waters. The concentration of Eucheuma cottonii seaweed extract for preventive 50% concentration of DPPH is 39,926 ppm.

Abstract

White spot syndrome (WSS) adalah penyakit yang secara signifikan menyebabkan tingginya mortalitas dan kerusakan parah pada budidaya udang. Penelitian ini bertujuan mengetahui dampak stres salinitas terhadap prevalensi WSSV dan survival rate udang vannamei (Litopenaeus vannamei). Penelitian ini menggunakan udang vannamei ukuran PL 40 yang diinfeksi WSSV dengan konsentrasi virus 20 μg/ml pada tiga rentang salinitas yang berbeda 0-10 ppt, 11-20 ppt, 21–30 ppt dan di rendam selama 4 jam kemudian dilakukan pengamatan selama 7 hari pasca infeksi dan diukur survival rate dan kuaitas airnya. Hasil pengamatan menunjukkan bahwa seluruh sampel terinfeksi oleh WSSV, ditunjukkan hasil analisa PCR dan gejala klinis yang timbul. Salinitas 0-10 ppt memberikan hasil persentase survival rate terendah jika dibandingkan dengan rentang salinitas yang lainnya yaitu sebesar 7 ekor atau 33% dari jumlah total sampel yang digunakan. Persentase survival rate udang tertinggi pasca infeksi virus WSSV terdapat pada perlakuan salinitas 21-30 ppt yaitu sebesar 13 ekor atau 63% dari jumlah total individu, dan salinitas 11-20 ppt memiliki persentase survival rate medium yaitu sebesar 10 ekor atau 49% dari jumlah total sampel. Stres salinitas mempengaruhi prevalensi WSSV dengan semakin tingginya tingkat infeksi seiring menurunnya rentang salinitas, akan tetapi survival rate semakin tinggi seiring dengan bertambahnya rentang salinitas.

Abstract

Mystacoleucus marginatus is a type of freshwater fish that is spread in various countries with tropical climates, one of which is Indonesia. This fish can be found in the Brantas River Basin (DAS), East Java. In its development, identification of fish species needs to be done in detail and more deeply, so that the correct identification results can be obtained. This study aims to identify genetic and phylogenetic characteristics of Mystacoleucus marginatus in the Brantas River Basin based on the Cytochrome Oxidase C Subunit I (COI) gene. Test samples were taken from 11 individuals of Mystacoleucus marginatus in the Brantas watershed, from June to August 2018. The results showed that Mystacoleucus marginatus in the Brantas watershed had 2 Haplotypes with Haplotype (Hd)=0.96 diversity values and nucleotides diversity (Pi)=0.00357. These results indicate that the population in the Brantas watershed is interconnected between one location and another sampling location, this is supported by a phylogenetic topology of DNA barcodes showing a monophyletic clade, as well as a haplotype distribution indicating that the sample area is genetics are not isolated from one another. So that the type of Mystacoleucus marginatus scattered along the Brantas watershed is still in one species, which is indicated by the low level of genetic variation. Genetic identification efforts must be carried out continuously so that appropriate information can be obtained for the diversity of fish species, especially endemic fish.

Abstract

The aim of this study was to support policy of government to lessen import and improve export of non migas presented characteristic of genetic of white prawn mains (Penaeus vannamei) so that this product can be yielded in Indonesia with high quality which do not fail with import product. Special goals wishing to be reached at this research is to analyze the genetic characteristic DNA with Polymorphism Chain Reaction (PCR) and primer ICP11. The background of this research was difference among vannamei prawn DNA characteristic that have infected by viruses. With experimental methods ten healthy post larva of white prawn cultured in aquarium and invected by WSSV virus in 1ml/10 l marine water dossage with three time replication during 7 days in culture. To know these infection, the sample were analyze with specific primer ICP11.

Abstract

The pathogenic bacterial infection is one of the problems in the cultivation of vannamei shrimp (Litopenaeus vannamei), causing a high mortality rate of cultured shrimp. The use of antibiotics or chemicals with inappropriate concentrations can harm the aquatic environment, cause resistance, and endanger consumer health because the residues from the chemicals used will periodically accumulate in the body of shrimp. One way to control and prevent shrimp disease is to increase the shrimp immune system by using immunostimulants from seaweed. This study aims to analyze the immunostimulant activity of seaweed extract (Gracilaria sp. and Padina sp.) against vannamei shrimp (L. vannamei) infected with Vibrio harveyi by observing the nonspecific immune system based on its hematological features, namely by counting the number of hemocytes and phagocytic activity. The research was conducted at the Hatchery Unit, Brackish Water Cultivation Development Center (BPAP) Situbondo, East Java. Seaweed sample Gracilaria sp. and Padina sp. purchased from seaweed farmers in Jepara, Central Java. The result of this study shows that supplementation of Gracilaria sp. and Padina sp. at a dose of 10 g/kg of feed can increase the total number of hemocytes and phagocytosis activity of L. vannamei shrimp. The best treatment is Gracilaria sp.

Abstract

Tujuan dari penelitian ini adalah untuk memperoleh profil hematosit udang Vannamei yang diberikan imunostimulan berupa alga laut melalui pakan terhadap infeksi penyakit WSSV.Penelitian ini dilakukan diLaboratorium Fakultas Perikanan dan Ilmu Kelautan dan Laboratorium Sentral Ilmu Hayati Universitas Brawijaya Malang. Pemberian pakan dengan alga laut sebagai imunostimulan dengan jenis alga yang berbeda sebagai berikut :Kontrol positif (udang diinfeksi WSSV tanpa suplemen pakan alga laut); Kontrol negative (udang tanpa infeksi WSSV dan tanpa suplemen alga laut); A (suplementasi pakan dg Sargassum sp 10 g/kg pakan); B (suplementasi pakan dg Padina sp 10 g/kg pakan); C (suplementasi pakan dg Eucheuma cottonii 10 g/kg pakan); D (suplementasi pakan Garcillaria sp 10 g/kg pakan) dengan 3 ulangan. Parameter yang diamati adalah sel Hyalin, granular, semi granular, THC, kelulushidupan dan SOD.Suplementasi pakan dengan ekstrak alga meningkatkan jumlah sel hyaline, granular dan semi granular, serta THC.Kelusluhidupan udang dan SOD mampu ditingkatkan antara 36-108 unit/mL pada suplementasi alga

Abstract

Aquaculture activities cannot be separated from obstacles that can lead to failure of aquaculture, including the presence of microplastics. Microplastic polystyrene (styrofoam) is a type of plastic that is commonly found in aquatic organisms (35%). Microplastics in fish can cause hormonal disturbances and high vacuolation in gonads which can cause apoptosis in gonadal cells. This study aimed to determine the effect of microplastics exposure on catfish (Clarias gariepinus) feed on the gonadal histology of male catfish. The research design used an experimental method with a Completely Randomized Design (CRD) which was divided into four treatments and three replications, namely 3% of the feed rate with treatments A (5% microplastics), B (10% microplastics), C (15% microplastics), and K (0% microplastics). Microplastic exposure treatment was given to fish by mixing it with commercial feed for 30 days. The results showed that microplastic exposure had a significant effect on the 15th and 30th days of the study on the Gonad Maturity Level (GML) and Gonado Somatic Index (GSI) of male catfish (C. gariepinus) in treatments A, B and C compared to controls (K). Treatment C (Microplastics 15%) had the lowest GML and GSI values, while treatment K (Microplastics 0%) had the highest GML and GSI values. Treatments A, B, and C experienced changes and delays in the development of gonadal cell structure, while control (K) developed well. This is presumably because the nutritional needs of control fish (K) were more fulfilled than treatments A, B and C exposed to microplastics.

Abstract

Produksi limbah Industri dan juga buangan limbah rumah tangga yang semakin meningkat dan terjadi terus-menerus, menjadikan sungai tempat pembuangan terakhir sehingga dapat mencemari biota yang berada disekitaran sungai tersebut. Masyarakat di Desa Kupang memanfaatkan aliran sungai tersebut sebagai sumber air untuk budidaya rumput laut Gracilaria sp. Salah satu untuk mengetahui cemaran logam berat pada perairan tersebut dengan mengecek keberadaan kadar Metallothionein pada rumput laut, karena sifat Metallothionein adalah protein yang dapat mengikat logam esensial maupun non esensial. Penelitian ini bertujuan untuk mengetahui kadar Metallothionein pada Gracilaria sp. yang terekspresi pada bagian pangkal dan ujung rumput laut Gracilaria sp. Metode yang digunakan adalah metode deskriptif. Analisis pengujian kadar Metallothionein menggunakan Eliza rider. Hasil penelitian ini bahwa kondisi perairan budidaya tambak Gracilaria sp. untuk parameter suhu, pH, DO sesuai dengan kondisi perairan tambak, tetapi pada parameter salinitas tidak sesuai yaitu melebihi SNI 40 ppt, karena penguapan yang tinggi pada saat musim kemarau, sehingga kadar garamnya tinggi. Kualitas perairan sangat berpengaruh dengan keberadaan Enzim Metallothionein pada pangkal dan ujung rumput laut, Kadar enzim Metallothionein tertinggi terdapat pada bagian ujung Gracilaria sp. yaitu berkisar antara 2.026-2.734 mg/mL. Terjadinya penyerapan yang tinggi karena pada bagian ujung terdapat banyak percabangan kecil dan masih muda sehingga proses penyerapannya masih kuat, sedangkan pada bagian pangkal terdiri dari sel-sel yang sudah tua dengan percabangan yang sedikit dan jumlahnya juga sedikit.
