Abstract

Cabai (Capsicum annuum L.) merupakan komoditas hortikultura utama di Indonesia. Organisme pengganggu tanaman (OPT) yang menyerang cabai adalah Colletotrichum sp. penyebab penyakit antraknosa. Bacillus mycoides dapat mengendalikan penyakit antraknosa. Limbah cair tahu dan air kelapa dapat dimanfaatkan sebagai media alternatif perbanyakan B. mycoides. Penelitian disusun dengan rancangan acak lengkap faktorial (RALF) dengan dua faktor dan tiga ulangan. Faktor pertama adalah media alternatif: NB/PDA, limbah cair tahu, air kelapa, dan limbah cair tahu + air kelapa. Faktor kedua adalah umur biakan: 24 jam dan 48 jam. Data dianalisis menggunakan Analisa ragam (Anova) dan apabila terdapat pengaruh yang nyata, maka dilanjutkan dengan uji BNT pada taraf 5%. Hasil penelitian menunjukkan bahwa terdapat interaksi perlakuan yang nyata antara perlakuan jenis media dengan umur biakan terhadap pertumbuhan Bacillus mycoides serta antagonismenya terhadap jamur Colletotrichum sp. Perlakuan B3A1 (media air kelapa dengan umur biakan 24 jam) menghasilkan jumlah koloni sebesar 1.53 ×  CFU/ml dan mengakibatkan daya hambat tertinggi terhadap Colletotrichum sp. sebesar 40.48%.

Abstract

Penyakit layu Fusarium yang disebabkan oleh Fusarium oxysporum merupakan kendala utama dalam budidaya tomat karena dapat menurunkan pertumbuhan dan hasil secara signifikan. Penelitian ini bertujuan untuk mengevaluasi efikasi berbagai formulasi Trichoderma sp. dengan volume inokulasi berbeda dalam mengendalikan layu Fusarium pada tanaman tomat. Metode yang digunakan adalah rancangan acak lengkap (RAL) faktorial.  Faktor pertama adalah jenis formulasi (air kelapa, ekstrak kentang gula, dan rendaman kedelai) dan faktor kedua Adalah volume inokulasi (15 ml dan 25 ml), serta kontrol. Parameter yang diamati meliputi kerapatan spora Trichoderma sp., masa inkubasi penyakit, intensitas serangan, dan tinggi tanaman. Hasil analisis ragam menunjukkan bahwa faktor formulasi dan volume inokulasi memberikan pengaruh nyata terhadap seluruh parameter, tetapi interaksi antara keduanya hanya ditemukan nyata pada parameter intensitas serangan di akhir pengamatan. Formulasi rendaman kedelai menghasilkan kerapatan spora tertinggi dan masa inkubasi penyakit terlama (15 hari). Kombinasi formulasi rendaman kedelai dengan volume inokulasi 25 ml merupakan perlakuan paling efektif dalam menekan intensitas serangan penyakit. Sementara itu, pertumbuhan tinggi tanaman tomat terbaik dicapai pada penggunaan formulasi air kelapa dan volume inokulasi 25 ml. Formulasi Trichoderma sp. berbasis bahan organik lokal berpotensi dikembangkan sebagai biofungisida ramah lingkungan untuk pengendalian layu Fusarium.

Abstract

Background: Ralstonia solanacearum is a bacterium highly adaptive to tropical and subtropical environments and can survive for long periods in the soil. This pathogen has a wide host range and is capable of causing severe damage to tomato plants, even leading to total crop failure until 100%. Therefore, appropriate control of these pathogens using biological agents is necessary. Soybean media is considered suitable for propagating Bacillus sp., Pseudomonas fluorescens, and Trichoderma sp. due to its adequate nutritional content and economic value. Previous studies have addressed microbes or media separately, such as PDA or NA, whereas this study combines microbial consortia and alternative media in a single integrated test. This study aims to examine the compatibility and antagonistic ability of a consortium of Bacillus sp., P. fluorescens, and Trichoderma sp. cultured on a soybean-based alternative medium against the bacterium Ralstonia solanacearum under in vitro conditions. Methodology: Observations were made on microbial compatibility and inhibition ability against R. solanacearum using thee double layer pepper disk diffusion methode. Findings: The compatibility test results showed that the microbial consortium was compatible and grew optimally on the soybean-based alternative medium. The highest compatibility index (CI) was observed in the Bacillus sp. + P. fluorescens treatment (0.402), while the lowest CI was found in the Trichoderma sp. × Bacillus sp. + P. fluorescens combination (0.873). In the antagonism test, the combination of Trichoderma sp. + Bacillus sp. × P. fluorescens produced an average inhibition zone of 17.2 mm, categorized as strong. Meanwhile, the Bacillus sp. + P. fluorescens treatment had the lowest average inhibition zone of 9.3 mm, categorized as moderate. Contribution: This study indicate that the consortium of Bacillus sp., P. fluorescens, and Trichoderma sp. maintains compatibility and antagonistic activity against R. solanacearum when grown on alternative soybean-based media, which has generally been tested on conventional media. This study recommends the utilization of soybean-based media as an alternative for developing microbial consortia of antagonists in the control of bacterial wilt disease

Abstract

Background: Stem rot disease caused by Fusarium oxysporum is one of the major diseases affecting vanilla plants (Vanilla planifolia A.), which can cause losses of up to 100%. Current management practices rely heavily on synthetic pesticides and often lack precision, raising environmental concerns and need for sustainable disease management strategies. This study aimed to determine the effectiveness of Fobio biopesticide concentration and soil sterilization intervals as control methods in suppressing the development of vanilla stem rot disease. Methodology: The study was conducted in vivo in Wonosalam Village, Jombang Regency using a factorial completely randomized design (CRD) with factors of Fobio biopesticide concentration (5 mL/L, 10 mL/L, and 15 mL/L) and application interval (1-week and 2-week intervals). The data were analyzed statistically using the Analysis of Covariance (ANCOVA). The parameters observed included disease intensity, infection rate, and biopesticide effectiveness. Findings: The results showed that the initial disease intensity had a very significant effect on the final intensity. The treatment with an application interval of once a week reduced disease intensity to 23.89%, lower than the interval of 2-weeks (26.67%) and the control (46.66%). A concentration of 15 ml/L reduced disease intensity to 24.94%, with the highest effectiveness also obtained in the 15 ml/L treatment at 1-week intervals (50.29%). The interaction between interval and concentration was also not significant, indicating that the effect of concentration did not change depending on treatment interval. The biopesticide Fobio was found to be sufficiently effective in suppressing the development of vanilla stem rot, and applications with more frequent intervals showed the highest efficacy. Contribution: This study highlights the importance of integrating biopesticide concentration  and application interval while adjusting for initial disease intensity in vanilla stem rot management.

Abstract

Control of bacterial leaf blight in rice plants is often done by applying chemical pesticides whose future effects will be very bad for the soil and plants that will be planted next. An alternative that can be applied is using the biopesticide Fobio. The aim of this research was to measure the effectiveness of the application of fobio in reducing the intensity of bacterial leaf blight attacks. This research was carried out in June-September 2023 in Diwek District, Jombang Regency. The study used a Randomized Block Design with two factors, namely the Fobio dose consisting of 3 levels, namely P0 (without giving Fobio / 0ml/liter), P5 (dose 5 ml/liter), and P10 (dose 10 ml/liter), the second factor was long The soaking time for seeds using Fobio consists of 3 levels, namely W10 (10 minutes soaking time), W20 (20 minutes soaking time), and W30 (30 minutes soaking time) so there are 9 treatment combinations. Observation parameters include plant height, number of leaves, number of tillers, and intensity of disease attacks. The results of the combination treatment were not significantly different for the number of leaves and number of tillers, for single and combination treatments the plant height and intensity of disease attacks were significantly different. Application of Fobio can increase plant height, number of leaves, and number of tillers on rice plants, and Fobio can suppress bacterial leaf blight attacks that attack rice plants.

Abstract

This study aimed to evaluate the potential of Streptomyces sp. isolated from the rhizosphere of the mangrove Avicennia marina as a biological control agent against Sclerotium rolfsii, the causal agent of basal stem rot in bird’s eye chili (Capsicum frutescens L.). The study was conducted using a non-factorial completely randomized design consisting of a control treatment and root-soaking treatments in a Streptomyces sp. suspension for different durations: 15, 30, and 45 minutes. Observational data were analyzed using ANOVA at the 5% significance level with IBM SPSS Statistics version 27. The results showed that Streptomyces sp. inhibited the growth of S. rolfsii on artificial medium by up to 52.33%, indicating strong antagonistic activity. The in vivo assay further demonstrated that the 45-minute root-soaking treatment (SW3) delayed disease incubation until the final day of observation, prevented the development of disease symptoms throughout the observation period, and enhanced vegetative growth in bird’s eye chili plants. These findings suggest that Streptomyces sp. from the rhizosphere of A. marina has promising potential as a biological control agent for managing basal stem rot caused by S. rolfsii in bird’s eye chili.

Abstract

HIGHLIGHTSSeed encapsulation using alginate and B. cereus BTH21 for biocontrol of wilt disease in eggplant.ABSTRACTBacterial wilt is a major plant disease caused by Ralstonia solanacearum that infects important crops, such as eggplant, causing wilt, stunted growth, and reduced yield. The biocontrol of the pathogen can be done by employing antagonistic bacteria, yet direct inoculation into the soils are often ineffective due to low population of the pathogen in the soils, which also easily washed away by watering activity. Seed encapsulation technique, using alginate, zeolite and peat, containing antagonistic bacteria Bacillus cereus BTH21 to control R. solanacearum is the novelty proposed in this research. Hence, this research aimed to develop alginate seed-encapsulation technique containing Bacillus cells as a mean to biologically control R. solanacearum in eggplant. Endophytic Bacillus cereus BTH21 strains was isolated from healthy eggplant tissue obtained from Kediri, East Java Province, Indonesia. Subsequent isolation, purification and molecular identification confirmed the identity of the strain. Three seed-encapsulation formulations were made: a) control (no encapsulation); b) alginate + zeolite + Bacillus (Al+Z+B); and c) alginate + peat + Bacillus (Al+P+B). A field experiment was designed in a Completely Randomized Design (CRD) where the alginate-encapsulated eggplant seeds were sown along with deliberate R. solanacearum inoculation (107 cfu/mL) into the soils. Observations were carried out every 7 days from 0 – 49 days after sowing (DAS) on several parameters, i.e., incubation period, infection rate, germination percentage, plant height, and number of leaves. The data were analyzed by using analysis of variance (ANOVA) followed by Duncan’s multiple range test at a significance level of P < 0.05. The results showed that both seed encapsulation formulations prolonged incubation period (15 and 13 DAS) compared to the control (10 DAS), reduced infection rate, increased germination rate, as well as higher agronomic performances (plant height, number of leaves) compared to the control at 7 – 49 DAS. Overall, this results indicated the potential of alginate seed-encapsulation techniques containing biocontrol agents to control plant diseases.

Abstract

Tanaman cabai rawit (Capsicum frutencens L) merupakan salah satu komoditas utama tanaman hortikultura yang dibudidayakan secara komersial di daerah tropis yang bernilai ekonomi tinggi di Indonesia. Satu diantara penyakit penting pada cabai rawit adalah penyakit antraknosa. Antraknosa merupakan penyakit utama tanaman cabai yang disebabkan oleh jamur Colletotrichum sp. Penggunaan pestisida sintetik yang kurang bijaksana ternyata banyak merugikan manusia dan agroekosistem. Oleh karena itu, perlu dilakukan upaya pengendalian penyakit yang ramah lingkungan dengan penggunaan agensia hayati. Beberapa penelitian menunjukkan jamur endofit dapat digunakan untuk meningkatkan ketahanan inang dari serangan patogen serta dapat memacu pertumbuhan tanaman dengan menghasilkan metabolit sekunder dan enzim tertentu. Penelitian ini dilaksanakan dari bulan April hingga Juni 2021. Tempat pelaksanaan penelitian di Laboratorium Kesehatan Tanaman Fakultas Pertanian Universitas Pembangunan Nasional “Veteran” Jawa Timur untuk In Vitro. Sedangkan, untuk In Vivo dilaksanakan di Screenhouse Kebun Bibit Wonorejo Surabaya. Penelitian ini bertujuan untuk mengetahui potensi jamur endofit dari tanaman cabai rawit sehat yang mampu menghambat pertumbuhan jamur Colletotrichum sp. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) non faktorial. Berdasarkan pengamatan, jamur endofit asal tanaman cabai rawit berpotensi menekan pertumbuhan jamur Colletotrichum sp. penyebab penyakit antraknosa.

Abstract

Background: Bacillus sp. as a biological agent can be an environmentally friendly disease control solution by producing antimicrobial compounds. However, expensive culture media is an obstacle in its use. Alternative media made from natural materials such as legumes are considered potential because they are rich in nutrients and affordable. This study aims to find the best alternative media that support the growth and viability of Bacillus sp. isolate Bcz 30. Method: This research was conducted at the Plant Health Laboratory of the National Development University “Veteran” East Java in July-November 2024. This research used a completely randomized design (RAL) with one stage of in vitro testing. The implementation of the test stages includes testing the growth of Bacillus sp. isolate Bcz 30 on each alternative media, testing the viability of Bacillus sp. isolate Bcz 30 on liquid media then the two highest viability results on liquid media will be continued for viability tests on different formulations, namely solid and liquid formulas. Analysis of the data obtained using the variance analysis test (ANOVA), if the data presented is significantly different, it will be continued with the BNJ test at the 5% level. Result: Based on the growth test, tolo bean and lamtoro gung media can replace NA media. Based on the liquid formula viability test, tolo bean and lamtoro gung media with 30% concentration had the highest viability results. Based on the formulation test, the solid formula can maintain the viability of Bacillus sp. than the liquid formula. Conclusion: Bean and lamtoro gung media with 30% concentration formulated with solid formula is the best alternative media for Bacillus sp. isolate Bcz 30.

Abstract

 Low rice productivity due to the use of low-grade seeds. The quality of the seeds can suffer a decrease caused by the attack of pathogenic fungi carried by the seeds both at the time of planting and storage, which will have an impact on the disruption of plant growth. Biopriming using Trichoderma sp. be an effort to control because it is antagonistic to pathogens. The success of biopriming is also influenced by the duration of seed priming. This study aims to examine the duration of soaking rice seeds with Trichoderma sp. the most effective for lowering seed-borne pathogenic infections. The study was conducted by providing seed soaking treatment using Trichoderma sp. suspension for 6 hours, 12 hours, and 24 hours with PDA medium incubation test method and Growing on Test. The parameters tested are the percentage of infection rate and the percentage of germination. Identification results showed that pathogenic fungi carried by seeds included Rhizopus sp., Fusarium sp., Aspergillus sp., and Mucor sp. The results showed the duration of soaking rice seeds with Trichoderma sp. more effective in decreasing the infection rate is at a 24-hour duration. However, the duration of soaking rice seeds with Trichoderma sp. there was no noticeable difference for the percentage parameter of seed germination.
