Abstract

Australian guava fruit (Psidium guajava L.) is thought to have antioxidant activity. Antioxidants are compounds that are useful for treating oxidative damage caused by free radicals in the body. The aim of this study was to determine the secondary metabolite content and IC50 value on the antioxidant activity of extracts and fractions from Australian guava fruit. This research used four solvents, including 70% ethanol extract, n-hexane fraction, ethyl acetate fraction and water fraction with varying concentrations of 50 ppm, 100 ppm, 150 ppm, 200 ppm and 250 ppm. The antioxidant test was carried out using a UV-Vis spectrophotometry tool with the DPPH method at a wavelength of 515 nm with an incubation time of 15 minutes. Antioxidant activity tests show that Australian guava fruit is effective as an antioxidant in capturing free radicals. The IC50 value for the ethanol extract is 0.784 µg/mL, the n-hexane fraction is 2.448 µg/mL, the ethyl acetate fraction is 1.015 µg/mL and the water fraction is 0.676 µg/mL. All samples were included in the very strong category, but the water fraction had the best antioxidant activity among the ethanol extract, n-hexane and ethyl acetate fractions.

Abstract

Antioxidants are compounds prevent oxidative damage by stabilizing free radicals by donating electrons. Phenolics are compounds that can be used as antioxidants because they can counteract free radicals. This study aims to determine the total phenolic content and antioxidant activity of the extract and fraction of the kalalawit (Uncaria gambir Roxb) stem. Extract preparation by maceration using 96% ethanol solvent. Testing for phenolic content with the Folin Ciocalteu method using UV-Vis spectrophotometry on extracts. Antioxidant activity test using the FRAP method on extracts with concentrations of 80, 90, 100, 110, and 120 ppm. The test results showed that the extract of the bajakah kalalawit stem (Uncaria gambir Roxb) had a phenolic content of 31.734 mg GAE/gr sample, there was antioxidant activity in the extract and fraction of the stem of the kalalawit (Uncaria gambir Roxb) indicated by the formation of a purplish blue color when reacted with FRAP solution , and the IC50 value of the ethanol extract was 29.842 ppm which was in the very strong category, water fraction was 39.865 ppm which was in the very strong category, n-hexane fraction was 75.319 ppm which was in the strong category and ethyl fraction was 12.572 ppm which was very strong.

Abstract

Indonesia kaya akan sumber daya alam, termasuk diantaranya tanaman herbal yang telah dipercaya oleh masyarakat untuk digunakan sebagai obat. Salah satu obat herbal yang banyak dikenal masyarakat adalah Jamu Kunyit Asam. Jamu ini dijual dengan cara digendong oleh pedagang di pasar tradisional. Jamu Kunyit Asam terdiri dari kunyit ( Curcuma domestica ) dan asam ( Tamarindus indica ). Kunyit memiliki kandungan zat aktif yang paling umum diketahui adalah kurkumin, yang memiliki aktivitas biologis sebagai analgetik, antipiretik dan antiinflamasi. Penelitian ini bertujuan untuk mengetahui kadar kurkumin pada jamu kunyit asam yang ada di Desa Palur.Metode penelitian ini menggunakan KLT-Densitometri. Sampel penelitian adalah produsen jamu gendong kunyit asam yang berada di Desa Palur. Jenis penelitian ini menggunakan analisa kualitatif yaitu dengan metode KLT dan Kuantitatif menggunakan metode KLT-Densitometri.Hasil Penelitian menunjukkan bahwa sampel jamu kunyit asam memiliki nilai Rf = 0,5 dan kadar kurkumin dari metode KLT-Densitometri pada sampel 1 sebesar 47,68ppm, sampel 2 sebesar 41,11ppm. Berdasarkan penelitian ini dapat disimpulkan bahwa kadar kurkumin dari produsen jamu gendong kunyit asam adalah mendekati sama.Kata Kunci : Kurkumin, Jamu, KLT-Densitometri, Kunyit Asam

Abstract

Saat ini mencuci tangan dengan sabun dan air mengalir bukanlah satu-satunya carauntuk membersihkan tangan, terdapat cara yang lebih praktis yaitu menggunakan cairan gelantiseptik yang dapat digunakan kapan dan dimana saja tanpa harus dibilas dengan air, yangdikenal dengan hand sanitizer. Penambahan zat aktif pada sediaan yang berasal dari ekstraktumbuhan yang memiliki aktivitas antibakteri dapat memberikan efek penghambatan padapertumbuhan bakteri seperti Staphylococcus aureus. Daun sirsak (Annona muricata L.)mengandung senyawa flavonoid, saponin, tanin, dan alkaloid, serta tanaman sereh (Cymbopogoncitratus) memiliki kandungan kimia antara lain minyak atsiri, saponin, polifenol dan flavonoid,kedua tanaman ini memiliki aktivitas antibakteri yang cukup besar.Pada penelitian ini, dilakukan kombinasi Ekstrak Etanol Daun Sirsak (Annona muricataL.) dan Ekstrak Etanol Batang Sereh (Cymbopogon citratus) untuk memformulasikan gelhandsanitizer dengan bahan alami. Uji karakteristik gel yang dilakukan adalah uji viskositas, ujidaya sebar, uji pH, uji organoleptis dan uji homogenitas. Uji aktivitas antibakteri dilakukandengan pengukuran zona hambat menggunakan metode disc diffusion test terhadap variasiformula yaitu F1, F2, dan F3.Hasil penelitian didapatkan kombinasi ekstrak etanol daun sirsak (Annona mricata L.)dan ekstrak etanol batang sereh (Cymbopogon citratus) pada konsentrasi 2,5%, 5%, 10% denganperbandingan ekstrak 2:1 dapat diformulasikan menjadi sediaan gel hand sanitizer yang baik danmemenuhi persyaratan sediaan gel yang baik. Kombinasi kedua ekstrak ini memberikan efekantibakteri yang lebih baik, dibuktikan dengan konsentrasi 10% kombinasi ekstrak memiliki efekantibakteri terhadap bakteri Staphylococcus aureus dengan nilai zona hambat 16 mm yangmanatermasuk kategori kuat.

Abstract

Free radicals work by binding to molecules or cells in the body and are very dangerous because they can damage cells in the body and trigger disease. Antioxidants are compounds that can counteract the effects of free radicals. This study aims to determine the antioxidant activity of ethanol extract and the n-hexane, ethyl-acetate and water fractions of Australian guava peel (Psidium guajava L.). Extraction was carried out by maceration method using 70% ethanol. The skin of the Australian guava (Psidium guajava L.) contains alkaloids, flavonoids, tannins, triterpenoids and saponins. Fractionation was carried out with solvents based on polarity differences obtained by the n-hexane fraction, ethyl acetate fraction, and air fraction. Antioxidant test by inhibiting DPPH free radicals (1,1-Diphenyl-2-Picrylhydrazyl) on ethanol extract and n-hexane fraction, ethyl acetate fraction and air at several concentrations, namely 50 ppm, 100 ppm, 150 ppm, 200 ppm, and 250 ppm. The IC50 calculation results obtained for the ethanol extract were 4.406 µg/mL, the n-hexane fraction was 4.131 µg/mL, the ethyl acetate fraction was 3.812 µg/mL and the air fraction was 3.312 µg/mL. The antioxidant activity with the best IC50 value was the fraction air.
