Abstract

ABSTRAK Herba pecut kuda (Stachytarpheta jamaicensis (L.) Vahl) secara tradisional dapat digunakan untuk mengobati berbagai penyakit, seperti infeksi dan batu saluran kencing, diuretik, rheumatik, sakit tenggorokan (faringitis), pembersih darah, datang haid tidak teratur, keputihan, hepatitis A. Penelitian ini bertujuan untuk mengetahui keamanan penggunaan herba pecut kuda pada hewan uji. Ekstrak air herba pecut kuda dibuat dengan metode perebusan dalam air suling selama 30 menit. Pengujian toksisitas akut dilakukan mengacu pada pedoman BPOM. Pengujian toksisitas akut dilakukan pada mencit Swiss Webster jantan dan betina dengan pemberian tunggal ekstrak air herba pecut kuda pada dosis 100, 500, 2.000, 5.000 dan 10.000 mg/kg bb. Pengamatan dilakukan selama 14 hari. Hasil menunjukkan bahwa selama 14 hari setelah pemberian tunggal ekstrak air herba pecut kuda, tidak ada kematian pada seluruh hewan uji. Dapat disimpulkan bahwa LD50 ekstrak air herba pecut kuda lebih besar dari 5.000 mg/kg bb. Kata kunci: pecut kuda, Stachytarpheta jamaicensis, toksisitas akut, ekstrak air, LD50 ABSTRACT Traditionally, Pecut kuda (Stachytarpheta jamaicensis (L.)Vahl) is used to treat various diseases such diuretic and rheumatic. The research objective was to determine safety of water extract S. jamaicensis herbas in mice. Water extract of S. jamaicensis were made using boiled water for 30 minutes. Acute toxicity test was done according to BPOM. Acute toxicity test was performed on Swiss Webster mice with a single administration of the water extract at doses of 100, 500, 2.000, 5.000 and 10.000 mg/kg bw. Observations of animal were done for a total of 14 days. The results showed that during 14 days of observation after single oral administration of water extract of S. jamaicensis up to 10000 mg/kg bw showed no toxicity of all experimental animals. It can be conclude that LD50 of water extract of S. jamaicensis is greater than 5.000 mg/kg bw. Keywords: pecut kuda, Stachytarpheta jamaicensis, acute toxicity, water extract, LD50

Abstract

The effect of a combination of ethanol extract of Physalis angulata L. with antibiotic against Staphylococcus aureus and Klebsiella pneumonie                                                                                           Abstract Infectious disease is one of the biggest health problems not only in Indonesia, but worldwide. Bacteria are important agents in causing infectious diseases. Indonesian people often use antibiotics along with herbal medicines. Cecendet (Physalis angulata L.) is one of the traditional plants that have antibacterial activity and widely used by the Indonesian community. This study aims to test the antibacterial activity of cecendet herbs and their effects when used in combination with various antibiotics. The preparation of P.angulata extract was carried out using reflux method with ethanol 50%. The characteristic examination and phytochemical screening are examined on simplicia and the extract. Determination of antibacterial activity from P.angulata extract was done by microdilution test method by assessing minimum inhibitory concentration (MIC). Determination of the effectiveness of combination of antibiotics with herbal cecendet against test microbes Staphylococcus aureus and Klebsiella pneumoniae was done by using checkerboard method. The results of phytochemical screening showed that simplicia and P.angulata extract contained alkaloids, flavonoids, saponins, polyphenols, monoterpenoids, sesquiterpenes, steroids and triterpenoids. The result of antibacterial activity test showed that P.angulata extract had antibacterial activity against S.aureus and K.pneumoniae with the MIC was 128 μg/mL, 256 μg/mL. The synergistic interaction of S.aureus is demonstrated by the combination of P.angulata herb extract with tetracycline. The additive/indifferent interaction of S.aureus is demonstrated by the combination of P.angulata extracts with ampicillin, and against K.pneumoniae shown by the combination of P.angulata extract of cecendet with ampicillin or tetracycline. Ethanol extract of P.angulata has antibacterial activity and and combination with antibiotics (ampicillin/tetracyclin) can give synergistic effect or additive of S.aureus and K.pneumoniae bacteria. Keywords:  Cecendet, Physalis angulata L., MIC, antibiotic combination

Abstract

Watercress (Nasturtium officinale) and purslane (Portulaca oleracea) are used to increase urination.This preclinical study aims to prove the diuretic effect of a combination of watercress and purslaneextracts. Simplisia of watercress and purslane herbs mixed with various combinations A (1:9), B(1:3) and C (1:1), then extracted in 50% ethanol. The animals were divided into five groups: control,furosemide 3.6 mg/kgBW, A 38 mg/kgBW, B 28 mg/kgBW, and C 10 mg/kgBW. Diuretic assay wascarried out using the modified Lipschitz method with the parameters observed were cumulative urinevolume for 6 and 24 hours, and potassium levels in 24-hour urine. The results showed the onset ofdiuretic effect of furosemide at 60 minutes, A at 150 minutes, B at 240 minutes and C at 90 minutes.Percent effectiveness of the diuretic in the furosemide was 718%, A 212%, B 231%, and C 368% whencompared to the controls. All combinations of extracts increased sodium and potassium secretion whencompared to controls, but this effect was not as strong as furosemide. All combinations of watercressand purslane herbs have a diuretic effect but weakly saluretic when compared to furosemide and thecombination that gives the best effect is combination C.

Abstract

The double burden of malnutrition (obese mothers with stunted children) has become an important focus in the field of public health both in Indonesia and the world. Serang Baru District, Bekasi Regency, is one of the areas in Indonesia that has a significant prevalence of stunting and obesity. This study aims to determine the effectiveness of nutritional education interventions for obese mothers of stunted children on changes in the status of the double burden of malnutrition in the household. The research began with collecting sociodemographic, anthropometric and food intake data for the past 2x24 hours. Respondents who met the inclusion criteria were then given a face-to-face nutrition education intervention every week for 3 months. In the third month, all respondents had their data collected again in the form of anthropometric data and food intake data. The results of the study showed that after the nutritional education intervention was carried out there was a change in the child's HAZ score from -2.87 ± 0.56 to -2.61 ± 0.62, while the mother's BMI was originally 32.52 ± 2.48 to 31.72 ± 2.16 which was significantly different when compared to before the intervention (P<0.05). It can be concluded that nutrition education interventions can reduce the double burden of malnutrition in Serang Baru District, Bekasi Regency.

Abstract

One of the ways to increase profit in small and medium business (UKM) in Cimahi is by diversifying its products, through manufacturing of bar soap derived from cow's milk from UKM. The requirement of bath soap is proper acidity (pH) level, otherwise it will make the skin dry, itchy or even increase irritation. Therefore, this in vivo non-clinical irritation evaluation is conducted to evaluate the effects of bar soap produced by UKM in Cimahi according to the guidelines in rat test animals. The animals is divided into five groups and exposed to soap for four hours. Five soap samples is obtained from UKM in Cipageran area, Cimahi. Observations is made at the hours of 1, 4, 24, 48, and 72 hours after exposure and is continued for 14 days. The measured parameters is erythema and edema, and the primary irritation index. The results showed that bar soap containing cow milk produced by UKM in Cimahi did not cause erythema and edema. Primary irritation index of all bath soap samples was 0.0 . It can be concluded that soap bars containing cow's milk produced by Cimahi City UKM including into the category of very mild irritation (negligigle).

Abstract

Plasmodium is a parasite that causes malaria, which is transmitted to humans through the bite of female Anopheles mosquitoes. Plasmodium that infects red blood cells will cause the formation of new permeability pathway (NPP) in the red blood cell membrane. This change makes the red blood cell membrane more permeable, allowing various nutrients the parasite needs to enter the red blood cells. Resveratrol is a polyphenol compound that is reported to have antimalarial activity by inhibiting the growth of Plasmodium berghei. This study aims to activate the activity of resveratrol in inhibiting NPP. The study began with the preparation of blood suspensions that showed infection and non-infection, then test preparations were added and induced using sorbitol, and then hemolysis was measured using a UV-visible spectrophotometer. The test group consisted of normal groups, control and various concentrations of resveratrol, namely 1.25, 2.5, 5, 10 and 20 μM. The observation parameters in this study were the percentage of hemolysis inhibition. Based on the study’s results, the percentage of hemolysis inhibition increased with increasing resveratrol concentration. The IC50 value of the NPP inhibition activity test of resveratrol was 1.025 μM. This study concludes that resveratrol can inhibit NPP in erythrocytes infected with Plasmodium berghei.

Abstract

Diabetes is characterized as a hyperglycemic condition impacted by β-cell dysfunction and insulin deficiency. Black soybean (Glycine soja (L.) Merr.) is widely known as an origin of nutritious food that has shown activities in preventing cardiovascular disease and reducing hyperglycemia. This research aimed to evaluate the potential of black soybeans ethanol extract (BSEE) as an α-amylase and α-glucosidase activity inhibitor. Black soybean seeds were extracted using the Soxhlet method with 50% ethanol as a solvent. The extract Soybean seeds were screened for the presence of phytochemicals. Inhibitory activity of α-amylase and α-glucosidase enzymes was tested in vitro with acarbose as a control. The absorbance measurement was conducted at 565 nm and 400 nm, respectively. BSEE contained alkaloids, flavonoids, polyphenols, saponins, quinones, tannins, and terpenoids. The results indicated that BSEE exhibited a weak inhibitory effect of α-amylase enzyme activity, with an IC50 value of 360.37 ± 20.80 µg/ml, in contrast to acarbose, which showed a significantly lower IC50 of 4.02 ± 0.56 µg/ml. Meanwhile, BSEE was classified as an active inhibitor of α-glucosidase enzyme activity, presenting 25.67 ± 0.27 µg/mL IC50 value, while acarbose demonstrated 10.85 ± 0.5 µg/mL IC50 value. In conclusion, BSEE inhibits α-amylase and α-glucosidase.

Abstract

Oral health faces significant challenges due to increasing dental infections by pathogenic microbes such as Porphyromonas gingivalis, Streptococcus mutans, and Candida albicans. Topical antioxidants in toothpaste, gel, and mouthwash are often used to treat dental diseases. There is a growing interest in finding natural alternatives for oral hygiene without side effects compared to conventional toothpaste formulations. Nigella sativa (black cumin) has various pharmaceutical efficacy, making it a valuable plant-based source medicinal compound. This study evaluated black cumin ethanol extract's (BCSE) antimicrobial and antioxidant activities. Antibacterial effectiveness was evaluated through Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC), and disc diffusion assays against P. gingivalis, S. mutans, and C. albicans. Antioxidant activity was reported based on DPPH scavenging, ABTS reduction, H2O2 scavenging, and NO scavenging assays. Black cumin seed ethanolic extract (BCSE) showed antibacterial activity of 4.49 mm, 4.33 mm, and 7.12 mm for P. gingivalis, S. mutans, and C. albicans, respectively, with zones of inhibition increasing in a concentration-dependent manner (p<0.05). MIC and MBC evaluations also revealed that MIC for S. mutans was achieved at 12.5% BCSE concentration, while P. gingivalis and C. albicans required 25%, and MBC reached 100%. The IC50 for DPPH, ABTS, NO, and H2O2 were 25.41, 69.93, 87.18, and 95.16 μg/ml, respectively. The antioxidant activity increased in a concentration-dependent manner, especially at 100 µg/ml of BCSE. Based on these results, BCSE could be an optional ingredient in toothpaste.

Abstract

Introduction: Tarum areuy (Marsdenia tinctoria) is a plant whose efficacy has not been widely explored. Traditionally used for infections and inflammation related disease.. Aims: To determine the anti-inflammatory effect of M. tinctoria leaf water extract on animals induced by carrageenan.Methods: Extract preparation by boiling in water. Animals were divided into groups, namely control, comparison (diclofenac sodium 4.5 mg/kgBW) and M. tinctoria leaf water extract (20, 40, 80 mg/kgBW). Induction of 0.1 mL of 1% carrageenan was given as much as intraplantar and the test preparation was given orally. Inflammatory and inflammatory inhibition percentages were measured every 30 minutes for 240 minutes. The t-test was utilized in data analysis.  Result: All test doses of M. tinctoria leaf water extract was effective to reduce inflammation in comparison to the control (p<0.05). A dose of 20 mg/kgBW was the optimal anti-inflammatory.. Conclusion: M. tinctoria leaf water extract has the potential as an anti-inflammatory.
