Abstract

One of the problems in using ganyong (Canna edulis Kerr)as food is the presence of tannin, an antinutritional substance, that can reduce the nutritional quality of ganyong. The purpose of this study was aimed to analyze the effect of fermenting ganyong using Lactobacillus Plantarumbacteria on the chemical compounds and the reduction of tannin level in its flour. Theresults showed that the optimum conditions of fermenting ganyong were at pH 6, the temperature of 40 oC, and at the fermentation time of 36 hours. At the optimum conditions, Lactobacillus Plantarumwas able to decrease tannin content in ganyong from 2.53 mg/mL to 0.84 mg/mL. The reduction of tannin content is due to the activity of tannase produced by the microorganisms. Fermentation caused the enrichment protein content, from 1.87% to 2.01% and the reduction of starch, amylose, and amylopectin contents to 51%, 16.82%, and 38.08% from 60.19%, 18.27%, and 43.49% respectively. SDS-PAGE results showed that the protein profiles changed in ganyong sample after the fermentation process. Protein with a molecular weight of 72.49 kDa which showed in the non-fermented ganyong, did not appear in the fermented ganyong, and protein band intensities also showed changes.

Abstract

Abstract

Objective: This study aimed to assess the quality of complete feed wafers with different plant protein sources for a feed of small ruminants. Methods:The research was conducted in May-September 2022. The research method used a completely randomized design (CRD) with 4 treatments and 4 replications. The method used in this study was Completely Randomized Design (CRD) with 4 treatments, namely R0 = Wafer with an animal protein source (Fish meal) (Control), R1 = Wafers with protein source Sesbania grandiflora, R2 = Wafers with protein source Gliricidia sepium. R3=Wafer with protein source Leucaena leucocephala. The research variables consisted of physical quality (moisture content, wafer density, specific gravity, water absorption) and nutritional quality (dry matter, organic matter, crude protein, crude fat, crude fiber, and nitrogen free extracts). Data analysis according to the Analysis of Variance procedure using SPSS version 21.Results:The results showed that the use of plant protein sources had no significant effect (P>0.05) on water content and wafer density but had a significant effect (P<0.05) on specific gravity and water absorption capacity. The use of plant protein sources can increase the value of water content, wafer density, and specific gravity and reduce water absorption capacity. Meanwhile, the crude protein (CP) content of the wafer had no significant effect (P>0.05) and produced the same value as using fish meal. Other nutritional variables had a significant effect (P<0.05) such as DM, OM, EE and NFE which increased and CF decreased on the use of plant protein sources in making feed wafers compared to the use of fish meal. Conclusions: It was concluded that the use of legumes Sesbania grandiflora, Gliricidia sepium, and Leucaena leucocephala as a source of vegetable protein in wafers could replace fish meal. The three types of legumes had the same effect on the physical and nutritional quality of the resulting product.

Abstract

The processing of velvet beans (Mucuna pruriens) from Timor is traditionally done by boiling them repeatedly twelve times before consumption due to concerns about their antinutrient content. On the other hand, earlier research indicated that velvet beans possess bioactive compounds that may serve as antioxidant and antidiabetic agents. The purpose of this study was to identify the bioactive components in extracts of velvet beans grown by the Timorese community, assess the overall levels of secondary metabolite compounds, and evaluate their bioactivities through in vitro antioxidant and hyperglycemia tests. Velvet beans samples were extracted using maceration, soaking them in ethanol and n-hexane solvents, followed by qualitative phytochemical analysis. Additionally, quantitative determination of secondary metabolites was performed, along with pharmacological activity tests, which included assessing antioxidant activity and antidiabetic effects through the spectrophotometric method. The findings revealed that the local Timor velvet beans extract contained flavonoid compounds at a concentration of 12.77 ± 3.10 mg QE/g, while the levels of phenolic and tannin compounds in the extract could not be quantified due to an absorbance lower than the y-intercept. The DPPH test demonstrated that the antioxidant activity of the local Timor velvet beans extract was measured at 1,964.51 ppm, which is categorized as very weak in comparison to the standard ascorbic acid solution that registered a value of 5.39, classified as very strong. The antidiabetic assessment indicated that the extract could inhibit the activity of the α-glucosidase enzyme at a concentration of 1,000 ppm by 28.60 ± 1.10%, which is categorized as very weak relative to the results from acarbose. Further investigation is necessary to optimize the bioactive compounds and identify the content of anti-nutritional compounds, as well as to explore effective methods of processing local Timor velvet beans that can reduce the antinutritional compounds.
