Abstract

As a marine biological resource macroalgae grow and develop naturally in the waters of Manado Bay, especially in the coastal waters of Molas. This study aims to analyze the community structure of macroalgae in the coastal waters of Molas, Bunaken District, Manado City. Data retrieval in the field using the Line Transect method with quadratic sampling technique performed at the lowest ebb analysis of the data required to obtain an overview of the macroalgae community structure and performed several formulas. Found 6 species consisting of 3 red algae (Rhodophyceae) G. edulis, G. salicornia, Gracilaria sp., 1 brown alga (Phaeophyceae) Padina australis, and 2 green algae (Ulvophyceae) Halimeda macroloba and Chaetomorpha crassa. Keywords: macroalgae, community structure, coastal waters, Molas, Manado Abstrak Sebagai sumberdaya hayati laut makroalga tumbuh dan berkembang secara alamiah di Perairan Teluk Manado, khususnya di pantai Molas. Penelitian ini bertujuan untuk menganalisis struktur komunitas makroalga di perairan pantai Molas Kecamatan Bunaken Kota Manado. Pengambilan data di lapangan menggunakan metode Line Transect dengan teknik sampling kuadrat dilakukan pada surut terendah Analisis data yang diperlukan untuk mendapatkan gambaran struktur komunitas makroalga dan dilakukan beberapa formula. Ditemukan 6 spesies yang terdiri dari 3 alga merah (Rhodophyceae) yaitu G. edulis, G. salicornia, Gracilaria sp., 1 alga coklat (Phaeophyceae) yaitu Padina australis, dan 2 alga hijau (Ulvophyceae) yaitu Halimeda macroloba dan Chaetomorpha crassa. Abstract

Macroalgae are thallus plants that do not have organs in the form of roots, stems, and leaves that are not true. The existence of macroalgae as producer organisms contributes to the life of aquatic biota, especially herbivorous organisms in the waters of Temajo Island, Mempawah Regency, West Kalimantan. This study aims to determine the abundance, relative abundance, relative frequency, diversity index, uniformity index, and dominance index in the waters of Temajo Island. This research was conducted in August 2022, the sampling locations were three stations and were determined using the exploratory method based on the presence of macroalgae found. Macroalgae sampling was carried out using a quadratic transect of 10x10 m and based on the research results obtained 5 families, 3 classes, 4 orders, and 6 genera consisting of Sargassum, Turbinaria, Padina, Halimeda, Codium, and Amphiroa. The results of the analysis found that the genus Sargassum had abundance, relative abundance, and relative frequency (K = 0.72-2.35 ind/m2, KR = 49.99%, F = 0.83) of macroalgae communities in the waters of Temajo Island, Mempawah Regency, Kalimantan West, has a low-medium diversity value, low-high uniformity index and low-high dominance index. Keywords: Community Structure, Macroalga, Temajo Island, Mempawah Abstrak Makroalga merupakan tumbuhan thallus yang tidak memiliki organ-organ berupa akar, batang dan daun tidak sejati. Keberadaan makroalga sebagai organisme produsen memberikan sumbangan bagi kehidupan biota akuatik, terutama organisme-organisme herbivor di perairan Pulau Temajo Kabupaten Mempawah Kalimantan Barat. Tujuan dari penelitian ini yaitu mengetahui kelimpahan, kelimpahan relatif, frekuensi relative, indeks keanekaragaman, indeks keseragaman, dan indeks dominansi di perairan Pulau Temajo. Penelitian ini dilakukan pada bulan Agustus 2022, lokasi pengambilan sampel sebanyak tiga stasiun dan ditentukan menggunakan metode Eksploratif berdasarkan keberadaan makroalga yang ditemukan. Sampling makroalga dilakukan menggunakan transek kuadrat dengan ukuran 10x10 m. Berdasarkan hasil penelitian diperoleh 5 famili, 3 kelas, 4 ordo, dan 6 genera yang terdiri dari Sargassum, Turbinaria, Padina, Halimeda, Codium dan Amphiroa. Hasil Analisa mendapatkan Genus Sargassum memiliki kelimpahan, kelimpahan relatif, dan frekuensi relatif (K = 0,72-2,35 ind/m2, KR = 49,99%, F = 0,83) komunitas makroalga di perairan Pulau Temajo Kabupaten Mempawah Kalimantan Barat, memiliki nilai keanekaragaman rendah-sedang, indeks keseragaman rendah-tinggi dan indeks dominansi rendah-tinggi. Abstract

Diabetes mellitus (DM) remains a serious global health threat, claiming a million lives every year and affecting nearly 9% of the adult population who suffer from this impaired insulin sensitivity disease. The Dipeptidyl peptidase-4 (DPP-4) enzyme has recently attracted attention because of its crucial role in insulin signaling, making this enzyme an interesting and emerging target for antidiabetic drug discovery. This study aimed to explore reported metabolites from marine algae of the genus Caulerpa as DPP-4 inhibitors through computational studies using CB-dock 2, Protein-Ligand Interaction Profiler, SwissAdme, and pkCMS. Molecular docking allowed the identification of 7 hit compounds with strong binding affinities (8.4 kcal to 9.3 kcal/mol) against DPP-4 target enzyme PDB ID: 3G4I. Four hits showed stronger binding affinity than two DPP-4 specific inhibitors and FDA-approved antidiabetic drugs, sitagliptin (8.4 kcal/mol) and linagliptin (9.0 kcal/mol). Following a molecular modification of the hit compounds using a bioisosterism-like approach and ADMET evaluation with pkCMS, three putative DPP-4 inhibitors were identified. They showed either stronger binding affinities or better ADMET profiles than sitagliptin and linagliptin, suggesting their promising potential as DPP-4 inhibitors. However, further optimized bioisoterism, in silico and in vivo studies, and clinical-based trials are required to confirm their antidiabetic activity. Abstract

Fish nuggets are processed frozen foods, both made from seawater fish and freshwater fish, contain high nutritional value, and are favored by children and adolescents as snacks and as side dishes. This study aimed to determine the physicochemical characteristics of trash fish nuggets added with Eucheuma cottonii seaweed. The physicochemical characteristics include pH, proximate levels such as protein, fat, and carbohydrates, water content and ash content, and the essential mineral content of zinc (Zn). Trash fish nuggets with the addition of wheat flour alone have characteristics, including pH of 6.8-7.1, water content of 40.5%, ash content of 1.04%, protein content of 40.3%, fat content of 7 0.07% and carbohydrate content of 10.12%, zinc content of 2.55 g/g nugget weight, while trash fish nuggets with the addition of a mixture of wheat flour and E. cottonii had a pH of 6.8-7.1, water content of 42.5%, ash content of 1.22%, protein content of 42.9%, fat content of 5.97% and carbohydrate content of 4.11% and zinc content of 4.56 g/g nugget weight. Based on the organoleptic examination, trash fish and E. cottonii nuggets were chewy with a delicious and savory taste. Keywords: characteristics, phytochemistry, E. cottonii, nuggets, trash fish Abstrak Nugget ikan merupakan salah satu olahan pangan frozen food, baik berbahan ikan air laut maupun ikan air tawar, mengandung nilai gizi tinggi dan digemari oleh anak-anak dan remaja sebagai cemilan maupun sebagai lauk. Penelitian ini bertujuan untuk menentukan karakteristik fisikokimia dari nugget ikan rucah yang ditambahkan dengan rumput laut Eucheuma cottonii. Karakteristik fisikakimia meliputi pH, kadar proksimat seperti protein, lemak dan karbohidrat, kadar air dan kadar abu serta kandungan mineral essensial seng (Zn). Nugget ikan rucah dengan penambahan tepung gandum saja memiliki karakteristik antara lain pH sebesar 6,8-7,1, kandungan air sebesar 40,5 %, kadar abu sebesar 1,04 %,  kadar protein sebesar 40,3 %, kadar lemak sebesar 7,07 % dan kadar karbohidrat sebesar 10,12 %, kandungan seng  sebesar 2,55 μg/g bobot nugget, sedangkan nugget ikan rucah dengan penambahan campuran tepung gandum dan E. cottonii memiliki  pH sebesar 6,8-7,1, kandungan air sebesar 42,5 %, kadar abu sebesar 1,22 %,  kadar protein sebesar 42,9 %, kadar lemak sebesar 5,97 % dan kadar karbohidrat sebesar 4,11 % serta kandungan seng sebesar 4,56 μg/g bobot nugget. Berdasarkan pemeriksaan organoleptis menunjukkan produk nugget ikan rucah dan E. cottoni lebih kenyal dengan rasa yang lezat dan gurih. Abstract

The village of Bahoi has a vast expanse of seagrass meadows of approximately 16.50 Ha, located between mangrove and coral reef ecosystems. This study aims to inventory the gastropod species in the seagrass meadow of Bahoi village waters and to determine the gastropod community structure through analysis of species density, relative density, species diversity index, and dominance index. The method used was a transect line method along 50 meters and a 1x1m quadrat pulled towards the sea, repeated three times during low tide. A total of 117 individuals were found, belonging to 26 species (11 families and 17 genera) of gastropods in the seagrass meadow of Bahoi village coastal waters. Based on the results, the highest density value was 1.10 ind/m2 with a relative density of 28.20%. Based on the analysis, the dominance index (D) of gastropods in the seagrass meadow of Bahoi village coastal waters was low, with values of D = 0.019 to 0.041, indicating that there was no specific species dominance in the seagrass meadow of Bahoi village coastal waters. This has an effect on the diversity index with a value of H' = 2.51, which is categorized as moderate. These results show that the diversity of gastropod species in the seagrass meadow of Bahoi village waters is quite diverse due to the absence of specific gastropod species dominance. Abstrak Desa Bahoi memiliki luas hamparan padang lamun ± 16.50 Ha yang berada di antara ekosistem mangrove dan ekosistem terumbu karang. Penelitian ini bertujuan untuk melakukan inventarisasi jenis-jenis gastropoda di hamparan lamun perairan Desa Bahoi, dan mengetahui struktur komunitas gastropoda melalui analisis kepadatan spesies, kepadatan relatif, indeks keanekaragaman spesies, dan indeks dominasi. Metode yang digunakan adalah metode transek garis sepanjang 50 meter dan kuadrat berukuran 1×1m yang ditarik ke arah laut, dan dilakukan ulangan sebanyak 3 kali pada saat air surut.Ditemukan 117 individu yang termasuk ke dalam 26 spesies (11 famili dan 17 genera) gastropoda di hamparan padang lamun perairan pantai desa Bahoi. Berdasarkan hasil penelitian, nilai kepadatan tertinggi sebesar 1,10 ind/m2 dengan kepadatan relatif 28,20%. Berdasarkan hasil analisis, Indeks Dominansi (D) gastropoda di hamparan lamun perairan pantai desa Bahoi tergolong rendah, dengan nilai D=0,019 sampai dengan 0,041 menunjukkan bahwa tidak terdapat dominasi spesies tertentu di hamparan padang lamun perairan pantai desa Bahoi. Hal ini berpengaruh terhadap Indeks Keanekaragaman dengan nilai H’=2,51 sehingga berada pada kategori sedang. Hasil tersebut menunjukkan bahwa keanekaragaman jenis gastropoda di hamparan lamun perairan pantai desa Bahoi cukup beragam dikarenakan tidak adanya dominasi spesies gastropoda tertentu. Abstract

Together with bacterial and viral infection, fungal infection represents the world's top ten killer diseases, desperately requiring new antifungal drugs. This research aimed to evaluate the antifungal activity of the sponge Styllisa flabelliformis against the pathogenic fungi Aspergillus flavus and Aspergillus parasiticus by the standard agar diffusion technique. Three concentrations (1, 10, and 10 mg/mL) for the extract and 0.5 mg/mL for ketoconazole and fluconazole were prepared and evaluated in triplicate against the tested fungi. Whereas ketoconazole poorly inhibited A. parasiticus and fluconazole weakly inhibited A. flavus, the extract of S. flabelliformis exerted antifungal activity against A. parasiticus (6.8 ± 1.8; 8.3 ± 3.2; and 9.5 ± 2.1) mm and A. flavus (6.8 ± 1.1; 11.5 ± 1.4; and 14.3 ± 1.1) mm 1, 10 and 100 mg/mL respectively. PASS analysis showed jasplakinolide as a promising antifungal agent with potential activity (Pa) of 0.736. STITCH analysis further confirmed that jasplakinolide worked by inhibiting the expression of cytoskeleton genes that prevented fungi from synthesizing chitin and inhibiting the formation of the fungi’s cell walls and hyphae, different from the ergosterol synthesis inhibition in ketoconazole and fluconazole, implying the potential of jasplakinolide as an antifungal agent.

Abstract

The implementation of the ship maintenance management system on Rear Admiral Jhon Lie's training ship did not run optimally, which impacted the less-than-optimal condition of various training ship equipment because the crew did not carry out the ship maintenance system according to the procedure, resulting in the incident that the ship's anchor winch operation did not function optimally when the ship was anchored in Belang Port, Southeast Minahasa. This study aims to identify and analyze the positive and significant effect of the planned maintenance system on shipping safety. The primary data collection methods used in this study were questionnaires and interviews. In this study, data processing and presentation were in the form of diagrams and path analysis with structural equation modeling (SEM-PLS) using SmartPLS. From the results of the data analysis, the R-square value of shipping safety (Y) is 0.307, which means that the planned maintenance system (X1) and ship maintenance (X2) can affect shipping safety (Y) by 30.7%. Ship maintenance (X2) has a significant effect on sailing safety (Y), with P-values = 0.033 0.05 (hypothesis accepted). The planned maintenance system (X1) has a significant effect on sailing safety (Y), with a P-value of 0.037 0.05 (hypothesis accepted). As a result of the analysis, it was found that the planned maintenance system has a significant effect on shipping safety.

Abstract

This research is to know community structure and gastropod distribution include species density (individual/m2), relative density (%), diversity indices (H’), Evenness indices (E), Richness indices (R), Dominance indices (D) and Dispersion indices (Id).This inestigation was carried out February 2020 in intertidal area, Waleo beach in Minahasa Northern. Quadrat transect placed via area sampling based on substrate difference. Placing quadrat transect was centr on three points that different with distance with point 100 m and distance between quadran 10 m with size quadrat 50 cm2. Based on this research was find 22 species include Cypraea tigris, Terebra areolata, Cymbiola nivosa oblita, Euplica borealis, Turbo argyrostomus, Cypraea moneta, Conus cumingii, Cypraea errones, Tectus fenestrats, Nerita ascensionis, Nerita exuvi, Nerita albicilla, Nerita polita, Nerita Picea, Vasum turbinellus, Lunella cinerea, Nassarius globosus, Nassarius pullus, Vexillum acupictum, Clypeomorus pellucida, Vexillum ebenus, Terebralia sulcata. Five order include Neogastropoda, Littorinimorpha, Cycloneritida, Caenogastropoda, Trochida. Analysis result to species density is the highest 0,40 (individual/m2),relative density, Nerita polita 12,63 %, index of diversity,H’= 3,…, index of evenness, E = 0,97, index of richness, R = 0,22 and index of dominance, range D =0,13-0,18 concluded that no species dominantly. Gastropoda distribution pattern in intertidal area based 3 stations that showed all pattern of clumed distribution. Keywords: Gastropoda, Intertidal, density, spread pattern Abstrak Penelitian ini bertujuan untuk mengetahui struktur komunitas dan pola penyebaran gastropoda meliputi Kepadatan spesies (Ind/m2), Kepadatan Relatif (%),Indeks Keanekaragaman (H’), Indeks Keseragaman (E), Indeks Kekayaan spesies (R), Indeks Dominansi (D), dan indeks Sebaran. Penelitian ini di lakukan pada bulan februari 2020 yang berlokasi di daerah intertidal pantai Waleo Kabupaten Minahasa Utara. Penempatan transek kuadran di tentukan melalui teknik area sampling berdasarkan perbedaan substrat, penempatan transek dan kuadran di pusatkan di tiga titik yang berbeda-beda dengan jarak antara titik 100m dan jarak antara kuadran 10m dengan ukuran kuadran 50x50cm. Dari hasil penelitiaan di temukan 22 spesis meliputi Cypraea tigris, Terebra areolata, Cymbiola nivosa oblita, Euplica borealis, Turbo argyrostomus, Cypraea moneta, Conus cumingii, Cypraea errones, Tectus fenestrats, Nerita ascensionis, Nerita exuvi, Nerita albicilla, Nerita polita, Nerita Picea, Vasum turbinellus, Lunella cinerea, Nassarius globosus, Nassarius pullus, Vexillum acupictum, Clypeomorus pellucida, Vexillum ebenus, Terebralia sulcata. Dan 5 ordo Neogastropoda, Littorinimorpha, Cycloneritida, Caenogastropoda, Trochida. Hasil analisis terhadap kepadatan spesis nilai tertinggi terdapat pada spesis N.polita sebesar 0,40 Ind/m² dan kepadatan realtif tertinggi juga terdapat pada spesis N.polita sebesar 12,63 %, nilai indeka keanekaragaman H’= 9,49 yang merupakan kategori tinggi, nilai dari indeks keseragaman secara keseluruhan (E)=0,97 dalam kondisi , dan nilai indeks kekayaan spesis yaitu 0,22, indeks Dominansi (D) = 0,13 – 0,18 disimpulkan tidak ada yang mendominasi, Pola sebaran gastropoda di daerah intertidal dari ketiga stasiun menunjukan pola sebaran secara mengelompok. Wilayah pantai Waleo Kabupaten Minahasa Utara memiliki suhu 31,3ºC. Derajat keasaman pH yang di peroleh yaitu 7 Salinitas yang di peroleh sebesar 34‰ yang masih dalam kisaran baik untuk gastropoda. Abstract

This study aims to determine the sex ratio, gonad maturity level, gonad maturity index, and fecundity of dolphinfish Coryphaena hippurus.  Samples were obtained from the catches of fishermen who landed in Kalinaun Village. Fish and gonadal weight were measured with a digital scale with an accuracy of 0,1 gram and fork length was measured to an accuracy of 0.1 cm. Samples obtained were 60 individuals, 16 males with a size range of 40.5-57.4 cm and 44 females with a size range of 42.9-72.6 cm.  The sex ratio of males and females was 1:2.75.  The level of gonadal maturity of all samples was level II (developing) III (maturation) and IV (mature) for both males and females, level I (undeveloped) and V (saline) were not found. The gonadal maturity level was dominated by maturity level III (maturation) every month, except June when level IV for males was dominated.  The gonadal maturity index value of developing (level II) was 0.79 ± 0.37, level III was 3.62 ± 1.33 and level IV was 4.66 ± 0.65 for females. Level  II 0.28 ±0.18, level III 0.47 ±0.16 and level IV 0.63 ±0.06 for male. Fecundity ranged from 28,313-554,023 with an average of 229,839 ±139,967. Abstrak             Penelitian ini bertujuan untuk mengetahui nisbah kelamin, tingkat tematangan gonad, indeks kematangan gonad dan fekunditas ikan lemadang Coryphaena hippurus. Sampel diambil dari tangkapan nelayan yang didaratkan di Desa Kalinaun.  Berat ikan diukur dengan timbangan digital berketelitian 0,1 gram dan Panjang cagak diukur sampai ketelitian  0,1 cm. Sampel yang diperoleh sebanyak 60 individu, jantan 16 individu dengan kisaran ukuran 40,5-57,4 cm dan betina 44 individu dengan kisaran ukuran 42,9-72,6 cm.  Perbandingan kelamin jantan dan betina adalah 1:2,75.  Tingkat kematangan gonad dari seluruh sampel adalah tingkat kematangan II (berkembang) III (pematangan) dan IV (matang) baik jantan maupun betina, tingkat kematangan I (belum berkembang) dan V(salin)  tidak ditemukan. Tingkat kematangan gonad didominasi tingkat kematangan III (pematangan) setiap bulan, kecuali bulan Juni didiminasi TKG IV untuk jantan.  Nilai IKG  betina TKG II 0,79 ±0,37, TKG III 3,62 ±1,33 dan TKG IV 4,66 ±0,65. Ikan jantan TKG II 0,28 ±0,18, TKG III 0,47 ±0,16 dan TKG IV 0,63 ±0,06. Fekunditas berkisar 28.313-554.023 dengan rata-rata  229.839 ±139.967.

Abstract

Reservoirs are water resources that function for the needs of the community. Reservoir maintenance is carried out to maintain the conservation of water resources and inhibit sedimentation. This study aims to determine the participation of the Ngudi Tirto Lestari community group in Sobokerto Village in maintaining the Cengklik reservoir. This research uses the theory of community participation according to Keith Davis with 3 indicators: individual mental and emotional involvement, motivation to contribute, and individual responsibility. The method used is qualitative with a descriptive approach. Data collection is done by observation, interview, and literature study. The results of the study are Community Group Participation in maintaining the Cengklik reservoir as a water resource conservation area in Sobokerto Village is quite good. In individual mental and emotional involvement, community groups are aware of social activities and the important benefits of maintaining the Cengklik reservoir. The encouragement of contribution motivation, community groups received encouragement from the Sobokerto Village government and PT Pertamina Patra Niaga Adi Soemarmo through socialization and training activities regarding reservoir maintenance. However, there is still a lack of responsibility for reservoir maintenance caused by the collision of other individual activities, so it is necessary to hold bounding between members.
