Abstract

1,5-bis (3-ethoxy-4-hydroxyphenyl) -1,4-pentadien-3-one (EHP) compound is an analogous compound of curcumin which has antimicrobial activity. In this study, the formulation and evaluation of mouthwash was carried out with EHP as an antimicrobial ingredient. The formulation of mouthwash was made with various concentrations of EHP 3, 6, 12, 16 and 18 ppm. Evaluation of mouthwash included organoleptic test, pH, clarity, density and antimicrobial activity against Staphylococcus aureus, Steptococcus mutans, and Candida albicans. Antimicrobial test was done by agar diffusion method. The results of organoleptic test obtained mouthwash was clear blue liquid, the aroma of mint, and fresh cold taste; pH range of 6.15 - 6.74; density of 1.0419 - 1.0561 g/cm3. The evaluation of mouthwash showed that the mouthwash was stable in storage for 1 month at 40 °C. Antimicrobial tests showed the diameter of inhibitory zone against Staphylococcus aureus (ATCC 6538); Streptococcus mutans (ATCC 31987) and Candida albicans (ATCC 10231) ranged from 6.2 to 8.4 mm at the concentration of EHP 18 bpj. The results showed that EHP compound was potential as antimicrobial ingredient in moutwash formula.

Abstract

Skin premature aging which is characterized by fine wrinkles can be overcome by stimulating collagen production and inhibiting its degradation, and also reducing corneocyte buildup. Madeira vein has been shown to induce collagen production and inhibit its degradation, while glycolic acid increase desquamation. The aim of this research was to optimize W/O/W multiple nanoemulsion formula of Madeira vein-leaves extract in W1 phase and glycolic acid in W2 phase. Glycolic acid was added after conjugated with chitosan to reduce its stinging effect on skin. The W/O/W multiple nanoemulsion was prepared through two emulsification steps: W1/O primary emulsion using high speed homogenizer, and the secondary emulsion using magnetic stirrer. W1/O was optimized including the selection of co surfactant and extract concentration, followed by secondary emulsion optimization using response surface methodology consisted of 2-level factorial and Box-Behnken design. The W1/O consisted of extract, polysorbate 80 as surfactant, PEG 400 as co surfactant and isopropyl myristate as oil phase. The 2-level factorial gave the three significant factors: W1/O sonication time, W1/O concentration, and secondary emulsification stirring time. Box-Behnken optimization for globule size below 400 nm was obtained by 6-10 minutes W1/O sonication time, 12-16% W1/O concentration, and 32-42 minutes stirring time of secondary emulsification.

Abstract

Background: Indonesia is a major producer of several essential oils such as citronella oil, clove oil, cananga oil, vetiver oil, sandalwood oil, and patchouli oil. The Cymbopogon (Poaceae) genus has the most important components of essential oils, namely citral, geraniol, citronellol, citronellal. Geraniol is one of the most important chemical compounds in the aroma and fragrance industry and shows the nature of insecticides and natural insect repellents which show low toxicity and is suggested to represent a new class of chemoprevention agents for cancer, antimicrobial, anti-oxidant, anti-inflammatory and some vascular effects Transdermal drugs also attract the attention of researchers and scientists in the field of formulation. Objective: This study aimed to obtain geraniol compounds obtained from Cymbopogon nardus (L.) Rendle and identification. Methodology: The citronella, essential oil was obtained from the Cymbopogon nardus (L) Rendle plant using a water-steam distillation method, then fractionation was carried out and continued by purification isolation by column chromatography. Results: Based on the interpretation of IR data, Core magnetic resonance (proton and carbon RMI) and mass spectra for isolate 5 were determined as geraniol compounds. Conclusion: Geraniol can be obtained from Cymbopogon nardus (L) Rendle plants by water-steam distillation, fractionation and isolation methods.

Abstract

Radioimmunotherapy is one of modalities means of treatment for cancer that exploits the antibody monoclonal specificity to bind to its receptor and the ability of alfa or beta radiation in destroying cancer cells. Trastuzumab is humanized IgG1 monoclonal antibodies which selectively bind to extracellular domain of Human Epidermal Growth Factor Receptor-2 (HER-2). HER-2 has now became a target receptor for treatment of human breast cancer due to their overexpression in the human breast cancer cell surface. This study was aimed in developing a method for preparation of (DOTA) n-[Dendrimer PAMAM]-(trastuzumabm conjugate, a precursor of (177Lu-DOTA)n-PAMAM-(trastuzumab) m radioimmunoconjugate, which is expected to be potential for breast cancer radioimmunotherapy. The conjugate was prepared through several conjugation steps. Characterization of (DOTA)n-[Dendrimer PAMAM]-(trastuzumab)m which was carried out by using a HPLC equipped size exclusion column (SEC) gave a clean peak with retention time (tR) of 10.53 mins which was slower compared to tR of its precursor unconjugated trastuzumab (10.70 mins). This result indicated that molecular weight of (DOTA)n-[Dendrimer PAMAM]-(trastuzumab)m was bigger than its precursor (unconjugated trastuzumab) and pure. The purity of (DOTA)n-[Dendrimer PAMAM]- (trastuzumab)m was verified by measuring the Rf of 177Lu-radiolabeled-(DOTA)n-[Dendrimer PAMAM]-(trastuzumab)m using instance thin layer chromatography-silica gel (ITLC-SG). The ITLC-SG of (DOTA)n-[Dendrimer PAMAM]-(trastuzumab)m which was radiolabeled with 177Lu- gave a clean peak with Rf of <0.3, while the precursors which were regards as impurities, (free 177Lu in form 177Lu-EDTA, 177Lu-DOTA, and 177Lu- dendrimer PAMAM which was incubated as non specific binding), all of them gave Rf of >0.6. These ITLC results indicated that (DOTA)n-[Dendrimer PAMAM]-(trastuzumab)m radiolabeled with 177Lu was pure.

Abstract

Rubus fraxinifolius can be founded in the mountainous regions of West Java. Previous studies have shown that this plant has potent antioxidant activity. The content of polyphenol compounds in this species contributes substantially to antioxidant activity, and the extraction method will affect the content of the compound. In this study an evaluation of the extraction method effect on leaves, young fruits and stems of R. fraxinifolius on the content of polyphenols and their antioxidant activity, have been done. Method: The leaves, fruits, and stems of R. fraxinifolius were extracted by maceration, reflux, and soxhlet using methanol as a solvent. Each extract was determined the total polyphenol content using Folin-Ciocalteu reagent and antioxidant activity test using DPPH reduction method. Result: The highest extract yield was R. fraxinifolius leaf reflux extract (30.20%). The highest total polyphenol content was given by the Soxhlet extract of leaf R. fraxinifolius (48.79 mg GAE / g extract). The highest antioxidant activity was leaf Soxhlet extract of R. fraxinifolius (98.29% at 100 ppm). Conclusion: From this study it was shown that the methanol extract of R. fraxinifolius leaves extracted by Soxhlet gave the highest polyphenol content and antioxidant activity.

Abstract

Cutleaf groundcheny (Physalis angulata Linn.) is a popular traditional medicine. This experiment was conducted to investigate the possible role of the herbal extract as an anticancer agent using T47D breast cancer cell line. Cytotoxicity assay was caried out by direct counting method. Breastcancer T47D cell lines Were treated with 70% ethanol extract from cutleaf groundcherry fruit with serial dilution of 2000, 1000, 500, 250, 125, 62.5, and 31.25 µg/ml for 24 hours of incubation at 37°C As a positive control doxorubicin was also tested on the breast cancer T47D cell lines with serial dilution of 20, 10, 5, 2.5, 1.25, 0.625, 0.312, and 0.156 µg/ml for 24 hours of incubation at 37°C The amount of living cells were observed and counted, death percentage was then determined and probit analysis was used to determine the LC50 value. The LC50 value for ethanol extract from the fruits was 28,02 µg/ml While doxorubicin as the positive control was 0,113 µg/ml. It is concluded that the cutleaf groundcherry (Physalis angulata) fruits have a cytotoxic activity toward T47D (breast cancer) cell lines which confirmed previous toxicity experiment using brine shrimp lethality test (BSLT) method.

Abstract

Plants of Piperaceae family are widespread in the tropics region and have been widely used as medicinal plants, these includes kemukus (Piper cubeba), kiseureuh (Piper aduncum), and Java chili (Piper retrofractum). The purpose of this study was to evaluate the immunomodulatory activity of ethanol extracts of several plants from the Piperaceae family in Balb/C strain mice with carbon clearance method. The study was conducted by dividing the experimental animals into 12 groups : normal control group, immunosuppressant comparison, immunostimulant comparison, and low, moderate, and high dose of kemukus, kiseureuh, and Java chili, resepctively. Immunomodulatory activity was measured by phagocytosis index, peripheral blood leukocyte levels, and splenic leukocyte levels. The results showed that ethanol kemukus extract (Piper cubeba) acts as an immunosuppressant, increasing the dose increased the immunosuppressant effect. Low doses of ethanol extract of kiseureh (Piper aduncum) also acts as immunosuppressant, whereas at medium and high doses has a tendency to be immunostimulant. Ethanol extract of Java chili (Piper retrofractum) acts as an immunostimulant, with the highest activity achieved at moderate doses. Ethanol extract of kemukus (Piper cubeba), kiseureh (Piper aduncum), and Java chili (Piper retrofractum) affect the number of peripheral blood granulocyte components, neutrophils levels affects the increase in phagocytic effects. Ethanol extract of kemukus (Piper cubeba), kiseureh (Piper aduncum), and Java chili (Piper retrofractum) did not affect the number of splenic blood granulocyte components.

Abstract

The use of Leucaena leucochepala as an antidiabetic has already been known in society, but the support experiment was very rare. The purpose of this experiment is to know whether the extract of L. leucochepala semen have an effect to the blood lipid profile of a NIDDM rat which is streptozotocin (stz) induced. It is made in 6 groups, each group consist of 5 rats. Group 1: Normal control, group 2: Negative control (stz), group 3: Positive control (clorpropamid), group 4: Extract of Leucaena semen low dose (0.25 g/kg BW), group 5: Extract of Leucaena semen medium dose (0.5 g/kg BW) and group 6: Extract Leucaena semen high dose (1 g/kg BW). Rats are made hiperglycemia with iv induction of stz 50 mg/kg BW. Chlorpropamid and Leucaena semen extract are given orally. The experiment results of blood sugar level, total cholesterol, triglyceride, HDL and LDL are analyzed by ANOVA and continued with BNT test (p < 0.05). The result from the experiment is the high reduction of blood sugar level from the Leucaena semen extract high dose in amount of 65.30%. Results from the total cholesterol level of group 1:61.60 mg/dl, group 2: 91.60 mg/dl, group 3: 62.20 mg/dl, group 4: 76.00 mg/dl, group 5: 65.40 mg/dl and group 6: 55.80 mg/dl. Results from the triglyceride level of group 1: 83.40 mg/dl, group 2: 201.40 mg/dl, group 3: 133.40 mg/dl, group 4: 114.80 mg/dl, group 5: 93.20 mg/dl and group 6: 80.40 mg/dl. Results from the HDL level of group 1: 47.60 mg/dl, group 2: 26.40 mg/dl, group 3: 35.20 mg/dl, group 4: 43.40 mg/dl, group 5:58.40 mg/dl and group 6: 90.40 mg/dl. Results from the LDL level of group 1: -2.68 mg/dl, group 2:26.72 mg/dl, group 3: 0.32 mg/dl, group 4: 9.64 mg/dl, group 5:-11.64 mg/dl and group 6: -50.16 mg/dl.

Abstract

Diabetes melitus (DM) merupakan kelompok penyakit metabolik yang bila tidak terkendali dengan baik dapat menimbulkan komplikasi yang berbahaya. Pengobatan yang rasional pada penderita DM diduga dapat mengendalikan glukosa darah karena pasien menerima obat sesuai dengan kebutuhan klinisnya. Selain pengobatan yang rasional, penderita DM harus mampu melakukan self-care yang merupakan perawatan diri secara mandiri untuk meningkatkan kualitas hidup dan mencegah komplikasi. Penelitian ini bertujuan untuk mengetahui hubungan antara rasionalitas pengobatan dan self-care dengan pengendalian glukosa darah pasien DM tipe II di Poli Rawat Jalan Rumah Sakit (RS) Bina Husada Cibinong. Penelitian ini menggunakan metode deskriptif analitis dengan desain cross sectional. Pengambilan data dilakukan secara retrospektif maupun prospektif terhadap 109 pasien DM tipe 2 selama periode Maret-Mei 2015. Digunakan kuesioner Diabetes Self-Management Questionnaire untuk mengakses self-care. Hasil penelitian menunjukkan data demografi pasien DM tipe 2 adalah 57,80% wanita dan 42,20% pria dengan kisaran usia terbanyak yaitu 46-55 tahun (49,54%). Persentase pengobatan yang rasional cukup tinggi yaitu 61,47%, sedangkan self care pasien sebagian besar (69,72%) berada pada kriteria “baik” dan sisanya (30,28%) pada kriteria “cukup”. Pada uji korelasi Chi-square independen ditemukan tidak adanya hubungan antara rasionalitas pengobatan dan self care dengan pengendalian glukosa darah (p > 0,05). Hal ini menunjukkan kemungkinan adanya faktor lain yang dapat mempengaruhi pengendalian glukosa darah.

Abstract

Chamomile (Chamomilla recucita L.) that contain apigenin as an active substance are known as hair-lightening agent. The application of these flowers as such is not convenient, so the incorporation of its extract in shampoos - for cleaning of both scalp and hair - gives a better outcome. In this study, six formulas Were applied, each containing 5% of chamomile extract with different concentrations of HPMC (Methocel® F4M) and other additives. The shampoos were evaluated during a six Week-storage on their physico-chemical properties (pH, viscosity, rheological characteristic at 28-30°C [room temperaturel, 40°C, and 6-7°C [chilled temperature], surface tension, foam-forming in hard and distilled water as Well as sensory properties [discoloration, odor, homo genenity]. The results showed that HPMC of 2% gave an optimum viscosity, no discoloration, no detected change on homogenenity as Well as odor until the end of six-Week storage. The viscosity the shampoo with 2% HPMC at 28-30°C was 2060.00-2080.00 cPs,at 40°C 1040.00-1080.00 cPs, and at 6-7°C 2420.00-2480.00 cPs. Having rheological characteristic of pseudoplastic, the shampoo’s surface tension Was 28.99-29.88 dyne/cin, with density ranged 1,0428- 1.0459, pH 6.26-6.34, height of foam in hard Water ranged 3.30-3.45 cm and in distilled 3.40-3.60 cm. The density, surface tension, and pH of the shampoo were in agreement with the requirements. Key words: Hydroxypropyl methyl cellulose, chamomile, shampoo, viscosity.
