Abstract

A homology model of human 3β-HSD type 2 has been developed from homology modeling techniques using Phyre2 server and refi ned by ModRefi ner. The PROCHECK, QMEAN and ProSA-web online tools were carried out to evaluate the stereochemical quality of the model. The Ramachandran plot resulted from PROCHECK showed that 84.5% residues are in the most favored region, 13.7% are in the additional allowed region, 1.5% are in the generously allowed region and 0.3% are in the disallowed region. The QMEAN (Z-score) are 0.509 (-3.006) and Z-score of ProSA-web is -7.10. The negative values of protein fold energies also found in almost all sequences. Furthermore, molecular docking was also applied to validate the model using MOE. The hydrogen bonding interactions with Tyr154, Ser124, and Ser218 are found in all docked substrates as well as known inhibitors (trilostane and epostane). A dataset of azasteroid inhibitors were also docked into the substrate active site of human 3β-HSD2. These docked structures were utilized to construct corresponding docking-based QSAR equation by employing genetic algorithm (GA) statistical analysis. The contructed best QSAR equation has a robust predictive power according to its statistical parameters, hence may be applied to supersede the default scoring function provided by docking software. These results indicate that the human 3β-HSD2 model was successfully evaluated as a good model.

Abstract

Classification technique has already been applied widely in the medical data. One of its applications is for classification of cancer. The accuracy of this technique highly depends on the type of data to be processed (whether the data are separable or non-separable) and the dissimilarity function used. To surmount those hindrances and to improve the accuracy of classification therefore a method named Fuzzy Kernel K-Medoids (FKKM). The method can be used for separable or non separable of data. Based on the research on the concentration data of Zn, Ba, Mg, Ca, Cu, and Se in blood in order to diagnose cancer, FKKM gives better result than the Support Vector Machines Method. This paper will discuss an application of the FKKM method on the concentration data of Zn, Ba, Mg, Ca, Cu, and Se in blood samples and compared with the Support Vector Machines Method for the diagnosis of cancer. Results showed that the FKKM method produced a better result than the Support Vector Machines Method.

Abstract

The leaves of yacon (Smallanthussonchi folius [Poepp. &Endl.] H. Robinson) or in Indonesia known as “Daun insulin” (Insulin leaf), scientifically has various medicinal properties and one of them as antidiabetic. The aim of this research is to know α-glucosidase enzyme inhibitor compound in the ethanol 96% extract of yacon leaves. Fractionation of ethanol extract subjected to column chromatography (SiO2; dichloromethane-methanol-water = 5 : 5 : 1) gave 5 fractions (Fr -1 ~Fr – 5). The results of the -glucosidase enzyme inhibition activity test in vitro against the five fractions obtained the 3rd fraction as the most active of 87.84 %. Purification of the 3rd fraction with preparative thin layer chromatography (SiO2; n-BuOH-ethanol-water = 4 : 1 : 2.2) gave pure isolate as Fr 3-1. Identification of pure isolates Fr 3-1 was carried out by interpreted spectra of Ultraviolet, Fourier Transform-Infra Red, Mass Spectra (from LC-MS) and compared data on the chemical shift of the Nuclear Magnetic Resonance (proton and carbon) and determined as Nystose (GF3) compound. The results of the -glucosidase enzyme inhibition activity showed that Fr 3-1 has IC50 of 21.36 ppm

Abstract

World Health Organization reported that hypertension was in the top rank as global causes of death in 2009. Adherence to the antihypertensive medication reduced risks of various cardiovascular diseases. A cross-sectional study has been carried out to observe profile of the use of antihypertensive and to evaluate patient’s compliance based on Medication Possession Ratio (MPR) at X-Pharmacy Yogyakarta in 2009. Study subjects were the prescription of antihypertensive for patient regularly visits at least 3 times. The MPR was calculated as the ratio of the sum of the days supply of the prescription filled, excluding the latest prescription, divided by number of days filled. Results showed that the class and type of the most commonly prescribed antihypertensives based on the number of units and the highest mean units per prescription was the ACEI (captopril 25mg) and based on the frequency of prescribing was CCB (amlodipin 5mg). The use of generic drugs versus branded medicines 5.6 times in the units, 3.2 times more in the frequency of prescribing, and 1.7 times more generic drugs in mean units per prescription. There was no significant difference between the adherence of patients based on sex, with or without costsharing; as well as single or combination antihypertensive based on MPR (p > 0.05); mean MPR was 0.78 ± 0.15 (range 0.41-1.00); and as many as 50.00 0 had MPR > 0.8 and considered as adherent patients.

Abstract

Several antimalarial drugs may act by producing free radicals, anion superoxide through a redox cycling process with oxygen. The mode of action of chloroquine, primaquine, artemisinin, anthraquinone and xanthones as antimalarial drug is partly increasing the oxidative stress in the parasitized red blood cell.

Abstract

Diabetes mellitus is disease with a high prevalence rate and continuously increase in Indonesia. So some researchers began to glance at natural rescource for prevention and treatment of diabetes mellitus. Clove Flower (Syzygium aromaticum) contains eugenol that can chemically lower blood sugar levels. The aimed of this research is to know the effect of eugenol on pancreas histology of diabetic rats. Clove flowers was extracted used maseration method use n-hexan and eugenol was isolated tested with TLC. Alloxan-induced rats were dosed 125 mg / kg BW IP and then the eugenol was administered in dosage 10 mg / kg BW compared with glibenclamid 1,35 mg /Kg BW orally for 14 days. Observation of pancreatic histology results were made pancreatic preparations using Hematoxilin Eosin (HE) staining. The results showed that alloxan-induced rats had pancreatic cell damage. How ever, after being treated with Glibenclamide and Eugenol showed the improvement of damage organs. Giving the eugenol at a dose of 15 mg / kg BW showed the most significant improvement.

Abstract

Formulation of lozenges containing: A) green tea extract; B) green tea extract with sodium ascorbate; and C) green tea extract with grape seed extract have been carried out. Antioxidant activities were compared to the activity of raw active materials. Lozenges were tested for physical characteristics, hedonic values, dissolution rate, and stability during storage, Physical tests indicated that the lozenges comply with the Farmakope Indonesia IV requirements. Determination of IC50 antioxidant activity using DPPI-I free radical scavenger showed that IC50 of formula A, B, and C were 844.04 µg/mL, 221.80 µg/ mL, and C 57.4 µg/mL, respectively. Meanwhile, IC50 value of green tea extract, sodium ascorbate, and grape seed extract were 4.29 µg/mL, 12.95 µg/mL, and 1.96 µg/ml., respectively. Accelerated stability test were carried out based on the EGCG concentration observed by using HPLC and then calculated by Arrhenius equation showed that the shelf life of lozenges of formula A, B, and C Were 2,11 years; 1.87 years; and 2,73 years, respectively. The result showed that the lozenge containing green tea extract and grape seed extract (formula C) is the best formula based on FI IV requirements, antioxidant activity and lozenges stability.

Abstract

Limited accumulation of anti-tuberculosis drugs in macrophages become a barrier to the success of latent tuberculosis therapy. The purpose of this study is to develop a D-mannose modified nanoparticle formula as a targeting agent to the mannose receptors to increase the internalization of rifampicin into macrophages. D-mannose was conjugated with chitosan using an amine reducing agent. Chitosan-D-mannose conjugate was characterized using FTIR. Subsequently, the conjugate was adsorbed onto the nanostructured lipid carrier (NLC) electrostatically. The NLC formula consisted of an ethyl acetate solution of solid-liquid lipid blend and rifampicin and an aqueous solution of chitosan-D-mannose conjugate, which were emulsified using polysorbate 80. Solidification of the NLC-chitosan-mannose nanoparticles was carried out by ionotropic gelation and solvent evaporation. The nanoparticle formula was optimized using Box-Behnken design. The formation of chitosan-D-mannose conjugate was shown by the change of the amide band wave number and the Schiff base formation of the FTIR spectra. The optimum formula of nanoparticles had a diameter of 766.1 ± 57.56 nm with a polydispersity index of 0.32 ± 0.02, encapsulation efficiency of 91.54 ± 0.18% and drug loading of 36.62 ± 0.07%. Rifampicin was released from the nanoparticles at pH 5.2 or 7.4 with a similar rate. This D-mannose modified NLC formula has the potential to be further developed as an intracellular antibiotic targeting to macrophages.

Abstract

Candlenut plant in Indonesia is still limited to traditional uses such as cooking spices and traditional medicines. Only a few research has been done on Candlenut oil. The purpose of this study is to make ointment base variations toward the quality of Candlenutoil ointment with 12.5% concentration andits healing potential teston open wounds in rabbits infected with the Staphylococcus aureus bacteria. This research method is a laboratory experiment using 3 variations of base ointment namely hydrocarbon base, absorption base and water soluble base with Candlenut oil test solution of 12.5%. The results showed the quality test of the ointment with three variations of ointment base fulfilled the requirements of organoleptic test, homogeneity test, and pH test, but Candlenutoil ointment of 12.5% with hydrocarbon base did not meet the scattering power requirement. The results of the ANOVA test showed no significant difference between the base variations of the ointment and the size of wound healing. This shows that the three variations of the 12.5% Candlenut oil ointment base can be used in healing open wounds on rabbits (Oryctolagus cuniculus) infected with the Staphylococcus aureus bacteria.
