Abstract

This study aims to develop an Electronic Student Worksheet (E-LKPD) based on Higher Order Thinking Skills (HOTS) using a Problem Based Learning (PBL) approach on coastal environment material at SMA Negeri 2 Tilamuta. The background of this study is the low level of student involvement and the limited availability of teaching materials that foster critical thinking and problem-solving abilities, especially regarding coastal ecosystem topics. The research applies the Research and Development (R&D) method, specifically using the ADDIE model at the development stage, which includes needs analysis, product design, and limited trials involving 26 grade X students and a Biology teacher. Data collection techniques included questionnaires, observations, and interviews, analyzed using both quantitative and qualitative approaches. The results revealed that the developed E-LKPD was highly practical, with teacher response reaching 96.9% and student response 94.5%. Observations also showed the implementation of teacher and student activities reached 100%, 100%, and 95% across three meetings. These findings indicate that the HOTS-based E-LKPD effectively encourages student engagement, enhances conceptual understanding, and creates a meaningful, enjoyable learning experience. Thus, the E-LKPD is suitable to be used as an adaptive, interactive, and contextual learning tool that meets the demands of 21st-century education, especially in enhancing students' critical thinking and problem-solving skills.

Abstract

EDNRB gene is a gene located on chromosome 13q22, plays a role in the signaling pathway during the development of the ENS intestinal nerves. Mutations in the EDNRB gene in humans are associated with Hirschsprung Disease (HD). Mutations in the EDNRB gene are the second most common cause after the RET gene in cases of HD with a percentage of 5–10%. To detect mutations, PCR products are needed. Primers are one of the basic components of PCR, while optimization of annealing temperature and optimization of primer concentration are factors in the success of primer attachment to the target gene. This study aims to design EDNRB exon 4 gene primers and determine the optimum annealing temperature and primer concentration to amplify the EDNRB gene. Primer design with Geneious Prime, and good primer criteria are analyzed in silico. Optimization of annealing temperature and primer concentration is carried out with gradient PCR and variations in primer concentration. The results of this study obtained the best primers, namely the forward primer 5'- CAGTAAGTGTGGCCTGAAAG-3' and the reverse primer 5'-GTGGAACCGAAGTGACTAGA-3', which amplified the EDNRB gene exon 4. The optimum PCR conditions for amplification of exon 4 of the EDNRB gene were at an annealing temperature of 57.2 ℃ and at a primer concentration of 0.5 μM.

Abstract

Tissue staining is necessary to show the structure and morphology of the tissue, namely Hematoxylin Eosin (H&E) staining. However, eosin is carcinogenic when used in the long term. The aim of this research is expected to develop the use of natural materials as an alternative for Hematoxylin-Eosin histological staining. One of the color pigments that can be obtained from natural materials is the anthocyanin pigment found in strawberries. The anthocyanin extract from strawberries, which is acidic like eosin, will bind to positively charged molecules in the basic cytoplasm. The design of this study uses a simple experimental design (Posttest Only Design). Based on the Mann Whitney U test, a significant result was obtained with a value of sig.<.001 <0.05, indicating a significant difference in the results between the preparations stained with hematoxylin and eosin and those stained with hematoxylin and anthocyanin extract. Thus, it can be concluded that statistically, staining with hematoxylin and anthocyanin extract cannot be used as a substitute for hematoxylin eosin. Nevertheless, the results of staining the preparations using hematoxylin and anthocyanin extract still produce color in the cytoplasm, and are sufficiently contrasting to distinguish between the color of the cytoplasm and the color of the nucleus.

Abstract

The global urgency for sustainable energy solutions amid climate change and fossil fuel depletion has driven innovation in Microbial Fuel Cells (MFCs). This study investigates the synergistic interactions within a thermophilic microbial consortium (MS9 & MS12) to optimize energy efficiency in MFCs. By varying the inoculum ratios (1:1, 1:2, and 1:3), the research evaluates how microbial population balance impacts bioelectricity production. The MFCs were operated for 24 hours, with voltage measurements recorded every 2 hours. Results revealed that a 1:3 (MS9:MS12) ratio achieved the highest voltage output, peaking at 20 hours before declining, indicating metabolic synergy and efficient substrate utilization. Descriptive analysis highlighted the critical role of cross-feeding mechanisms and functional complementarity between species in enhancing electron transfer. The findings align with modular co-cultivation principles, emphasizing the importance of engineered microbial ratios for stable system performance. This study provides a protocol for designing balanced thermophilic consortia, advancing sustainable bioenergy strategies through optimized microbial interactions.
