Abstract

The study aimed at finding out the development of Ascaridia galli (A. gall) eggs that were given aerator treatment and those without aerator treatment into infective eggs and L2 through in vitro culture. Each treatments has 108,000 eggs assigned to 8 groups of 1,000; 2,000; 3,000; 4,000; 5,000; 6,000; 7,000; and 8,000 eggs, respectively with 3 repetitions. Female A. galli were collected from the small intestinal lumen of naturally infected domestic chickens. The eggs collected from the uterus of adult female A. galli were incubated in sterile aquadest at ambient temperature for 45 days (without aerator) and 25 days (with aerator) to obtain the infective eggs and the L2. The number of the infective eggs and hatched L2 were counted under stereo microscope. Data were analysed descriptively. There were 97.740 eggs (90.5%) in the groups without aerator developed into infective eggs and 77,040 eggs (71.3%) developed into the L2. Meanwhile, there were 101,847 eggs (94.3%) in the groups with the aerator developed into the infective eggs and88.722 eggs (82.15%) hatched L2. It is concluded that the eggs collected from worms uterus had high viability and the aerator application shortened the developing period of the A. galli worms.

Abstract

The purpose of this study was to analyze the potential of tomato extract (Lycopersicum esculentum L.) in increasing the Sertoli and spermatogenic cells count of the contralateral testis in white rats (Rattus novergicus) with unilateral torsion. A total of 24 male Wistar white rats, aged 3-4 months old with a weight of 180-200 g were used in this study. The rat samples were randomly divided into 4 treatment groups: Group 1 as a negative control (NC) which consisted of rats without testicular torsion and given tomato extract; Group 2 as a positive control (PC) which consisted of rats with unilateral testicular torsion 720 for 6 hours without given tomato extract; Group 3 as treatment 1 (T1) consisted of rats that were subjected to unilateral testicular torsion 720o for 6 hours and were given tomato extract at a dose of 100 mg/kg BW/day; and Group 4 as treatment 2 (T2) consisted of rats that were subjected to unilateral testicular torsion 720 for 6 hours and were given tomato extract at a dose of 200 mg/kg BW/day. Each group consisted of six rats. Tomato extract was administered orally for 30 days. The observations of the number of Sertoli cells and spermatogenic cells (spermatogonia cells, spermatocytes, and spermatids) were carried out histologically with Hematoxylin-Eosin (HE) staining and the slides were examined with a light microscope. The data obtained was analyzed by ANOVA and continued with Duncan's test. The results showed that the administration of tomato extract significantly increased (P0.05) the number of Sertoli cells and spermatogenic cells of the contralateral testicular seminiferous tubule. Giving tomato extract 100 mg/kg BW/day resulted in an increase in the number of Sertoli cells and better spermatogenic cells than tomato extract 200 mg/kg BW/day. In conclusion the administration of tomato extract after detorsion can increase the number of Sertoli cells and spermatogenic cells of the contralateral testicular seminiferous tubules in white rats subjected to unilateral torsion of 720 for 6 hours

Abstract

This study aimed to examine the mucolytic activity of roselle calyces extract on cow intestinal mucus in vitro. The extraction of rosellecalyces was performed by the subsequence maceration method using different polarity solvent (n-hexane, ethyl acetate, and methanol). Mucolyticactivity of each extract was evaluated by measuring the viscosity of mixture of cow intestinal mucus and phosphate buffer pH 7 compared toacetylcysteine as the standard mucolytic drug. The result showed that the decreasing of cow intestinal mucus and phosphate buffer pH 7 mixturesviscosity was indicated the increasing of mucolytic activity. The n-hexane, ethyl acetate, and methanol extract of roselle calyces exhibit dosedependentmucolytic activity. Mucolytic activity of roselle calyces ethyl acetate extract with dose of 0.8% and roselle calyces methanol extractwith dose of 0.6% showed no significant difference to mucolytic activity from acetylcysteine with dose of 0.1% (positive control). An increaseddose of roselle calyces extract will decrease the viscosity of cow intestinal mucus and phosphate buffer pH 7 mixtures, indicating an increase inmucolytic activity.

Abstract

Penelitian ini bertujuan menginterpretasikan sonogram dengan kelainan organ hepatobiliari pada anjing. Pemeriksaan dilakukan pada 17ekor anjing dengan menggunakan brightness mode dan color flow Doppler ultrasonografi. Berdasarkan hasil interpretasi pada sonogram,ditemukan 7 kasus abnormalitas hati dan 10 kasus abnormalitas kantung empedu. Abnormalitas yang ditemukan pada hati, terdiri atas tumor, hepatitis, dan kongesti hati. Kasus tumor hati ditandai dengan hepatomegali, tekstur hati tidak homogen, dan adanya m assa pada parenkim hati. Sonogram kasus hepatitis ditunjukkan oleh adanya hepatomegali dan peningkatan akti vitas vaskularisasi hati. Sonogram kasus kongesti hati ditunjukkan oleh pembesaran diameter dan peningkatan echogenicity dinding pembuluh darah hati. Abnormalitas kantung empedu yang ditemukan antara lain cholecystitis, cholelithiasis dan mucocele. Penebalan dinding dengan atau tanpa edema merupakan citra yang ditemukan pada kasus cholecystitis. Sonogram kasus cholelithiasis menunjukkan adanya massa hyperechoic di dalam lumen kantung empedu disertai acoustic shadowing. Sonogram kasus mucocele menunjukkan adanya massa hypoechoic yang terdapat di dalam kantung empedu.

Abstract

ABSTRACT The study aimed to find out the effect of Eurycoma longifolia Jack on testosterone level and spermatozoa quality of Etawa crossbreed goat. Goats were divided into two treatment groups, three goats each. Group 1 (K1) was control group which given orally 20 ml of distilled water every morning at 9:00 am for six days, and group 2 (K2) was administered with Eurycoma longifolia with the dose of 90 mg/kg bw in 20 ml of distilled water orally every morning at 9:00 am for six days. The concentration of testosterone was measured on day 1, 3, and 6 using the method of enzyme-linked immunosorbent assay (ELISA). The quality of sperm consisted of concentration and percentage of life sperm. The results showed that level of testosterone on K2 increased started from day 1 (9.36 ng/ml) to day 6 (12.43 ng/ml) (P0.05). The percentage of life spermatozoa increased higher on K2 compare to K1 on day 3 to day 6, that was 88.6 and 89.8% on K1 to 91.0 and 92.7% on K2. In conclusion, the administration of pasak bumi with the dose of 90 mg/kg bw in 20 ml of distilled water for 6 days able to increase testosterone levels and percentage of life sperm of Etawa crossbreed goat.

Abstract

Penelitian ini bertujuan melakukan karakterisasi molekuler antigenic site terhadap isolat virus avian influenza (AI) Balai Penyidikan dan Pengujian Veteriner (BPPV) Regional III Lampung dari tahun 2008-2013. Amplifikasi RNA dilakukan dengan teknik reverse transcription polymerase chain reaction (RT-PCR) menggunakan 4 pasang primer referens dari Australian Animal Health Laboratory (AAHL) Geelong Australia (HA10, HA20, dan HA30) dan dilanjutkan dengan proses pengurutan. Analisis hasil pengurutan dengan menggunakan perangkat lunak MEGA versi 5.05 yang meliputi multiple alignment, deductive amino acids prediction, dan phylogenic tree analysis diperoleh hasil perbedaan genetik antar isolat Lampung dari tahun 2003-2013 ditemukan berkisar 1,1-9,1% dengan tingkat homologi mencapai 90,9-98,9%. Variasi genetik ditemukan adanya substitusi pada posisi 53 (R53K), 126 (D126E), 136 (P136), 138 (H138Q, dan H138L), 140 (R140K, R140S, dan R140N), 141 (S141P), dan 189 (K189R). Berdasarkan analisis filogenic tree isolat Lampung tahun 2008-2011 termasuk ke dalam clade 2.1.3. Analisis filogenik isolat AI tahun 2012-2013 yang menginfeksi unggas air mempunyai homologi sekitar 98,5-99,1% dibandingkan dengan isolat AI yang menginfeksi unggas air asal Jawa dan termasuk ke dalam clade 2.3.2.1.

Abstract

This research aims to evaluate the protective potency of freeze-dried egg yolk powder on Garut ram sperm during the liquid semen preservation process. Semen with good sperm quality was divided into three groups and diluted using the following Tris diluents: fresh egg yolk-Tris (FEY-Tris), commercial egg yolk powder-Tris (CEY-Tris), and freeze-dried egg yolk powder-Tris (DEY-Tris). Semen that had been diluted was observed every 12 hours until sperm progressive motility was 50%. Sperm quality was tested for progressive motility percentages, viability, intact plasma membrane (IPM), and intact acrosome (IA). Fresh egg yolk had a particle size of 14460.00330.76 nm, with polydispersity index value (PI) and zeta potential (ZP) being 1.000.11 and -9.220.30 mV, respectively. The particle size of commercial egg yolk powder was 877.90168.86 nm with PI value 0.340.04 and ZP-28.71.24 mV. The particle size of freeze-dried egg yolk powder was 1296.0086.73 nm, with PI value 0.700.04 and ZP-34.50.64 mV. Progressive motility percentage showed that DEY-Tris diluent managed to survive to 51.116.06% for 168 hours, compared to FEY-Tris (51.256.74%) which survived for 156 hours, although no longer than CEY-Tris diluent (53.476.33%) which survive for 180 hours (P0.05). Higher sperm motility was also supported by viability percentage, IPM, and IA, which were all higher (P0.05). In conclusion, freeze dried egg yolk powder mixed with Tris buffer was able to maintain sperm quality during the liquid semen preservation process.

Abstract

This study aimed to analyze MSP-1 gene in mice erythrocyte infected with Plasmodium berghei using polymerase chain reaction (PCR). The result showed the existence of 462 bp DNA band which was assumed to encode MSP-1 protein with molecular weight of 19 kDa that can only be found in erythrocyte infected with Plasmodium berghei. BLASTn analysis showed that PbMSP-1 obtained in this study have 100% similar identity with mRNA of PbMSP-1 partial sequence (462 bp), 93% similarity with PbMSP-1 complete sequence (5750 bp and 5376 bp), and 87% similarity with PbMSP-1 incomplete sequence (333 bp).

Abstract

Penelitian bertujuan mengetahui kualitas spermatozoa ayam kampung dalam pengencer NaCl fisiologis dengan lama simpan yang berbedapada suhu kamar. Materi yang digunakan dalam penelitian ini adalah semen ayam kampung berumur 1-1,5 tahun. Lama simpan pada suhu kamar adalah 0, 30, 60, 90, dan 120 menit dengan 10 kali ulangan. Rasio pengenceran semen dengan pengencer 1:10. Variabel yang diamati yaitu motilitas dan viabilitas spermatozoa. Metode yang digunakan adalah metode percobaan menggunakan rancangan acak lengkap (RAL). Hasil analisis statistik menunjukkan bahwa lama simpan semen ayam kampung pada suhu kamar dengan pengencer NaCl fisiologis selama 0, 30, 60, 90, dan 120 menit berpengaruh sangat nyata (P0,01) terhadap motilitas individu dan viabilitas. Rata-rata motilitas individu pada penyimpanan selama 0, 30, 60, 90, dan 120 menit masing-masing adalah 68,5+5,5; 58,5+5,79; 49,5+7,25; 39,5+8,9; dan 26+11,7% sedangkan rata-rata viabilitas masing-masing adalah 83,1+3,63; 77,6+3,47; 67,8+4,4; 57,7+7,7; dan 45,6+7,19%. Dapat disimpulkan bahwa motilitas dan viabilitas spermatozoa ayam kampung mengalami penurunan secara bertahap seiring dengan lama simpan pada suhu kamar.

Abstract

This study aims to determine the effect of the substitution of bovine serum albumin (BSA) with cattle blood serum and egg white in the diluent of Cauda epididymal Plasma 2 (CEP-2) on sperm quality of Limousin cattle during cooling at 3-5 C. The research material used was rejected Limousin bull sperm (motility of 50-60%) from Artificial Insemination Centre Singosari, Malang. This research was a laboratory experiment using a randomized block design which was composed of six treatments with 10 replications, those were T0 as controls ((90% CEP-2 with BSA + 10% egg yolk); T1 (83.84% CEP-2 + 6.16% cattle blood serum + 10% egg yolk); T2 (81.84% CEP-2 + 8.16% cattle blood serum + 10% egg yolk); dan T3 (90% CEP-2 + 0,4% egg white + 10% egg yolk); T4 (90% CEP-2 + 0.8% egg white + 10% egg yolk); and T5 (90% CEP-2 without BSA + 10% egg yolk). Parameters measured were the percentage of motility, viability, and abnormality of sperms. Results of research after 48 hours of storage showed that the percentage of sperm motility in T0, T1, T2, T3, T4, and T5 were 40.505.90, 3636.16, 34.006.58, 40.503.69, 38.503.37, and 38.504.12, respectively, while the percentage of sperms viability were 75.164.30, 70.502.88, 73.802.80, 74.803.30, 75.133.13, and 74.034.13, respectively, and the percentage of sperms abnormality were 10.142.34, 10.621.34, 11.332.00, 10.942.82, 10.021.95, and 10.781.96, respectively.
